| 1 | Discovering small molecules as Wnt inhibitors that promote heart regeneration and injury repair显示文摘There are intense interests in discovering pro regenerative medicine leads that can promote cardiac differentiation and regeneration,as well as repair damaged heart tissues.We have combined zebrafish embryo-based screens with cardiomyogenesis assays to discover selective small molecules that modulate heart development and regeneration with minimal adverse effects.Two related compounds with novel structures,named as Cardiomogen 1 and 2(CDMG1 and CDMG2),were identified for their capacity to promote myocardial hyperplasia through expansion of the cardiac progenitor cell population.We find that Cardiomogen acts as a Wnt inhibitor by targeting p-catenin and reducing Tcf/Lef-mediated transcription in cultured cells.CDMG treatment of amputated zebrafish hearts reduces nuclear p-catenin in injured heart tissue,increases cardiomyocyte(CM)proliferation,and expedites wound healing,thus accelerating cardiac muscle regeneration.Importantly,Cardiomogen can alleviate the functional deterioration of mammalian hearts after myocardial infarction.Injured hearts exposed to CDMG1 display increased newly formed CMs and reduced fibrotic scar tissue,which are in part attributable to the^-catenin reduction.Our findings indicate Cardiomogen as a Wnt inhibitor in enhancing injury-induced CM proliferation and heart regeneration,highlighting the values of embryo-based small molecule screens in discovery of effective and safe medicine leads. | Shuying Xie Wenbin Fu Guangju Yu Xueli Hu Kaa Seng Lai Xiangwen Peng Yating Zhou Xuejiao Zhu Ptamen Christov Leah Sawyer Terri T.Ni Gary A.Sulikowski Zhongzhou Yang Ethan Lee Chunyu Zeng Wei E.Wang Tao P.Zhong | 2020 | Journal of Molecular Cell Biology2020,12,1: | 4 |

| 9 | Determination of Fosthiazate Residue in Ginger by Gas Chromatography显示文摘[Objectives] An analytical method was established for determining fosthiazate residue in ginger by gas chromatography with flame photometric detector(GC-NPD) to evaluate the safety of fosthiazate in ginger. [Methods] The fosthiazate residue in ginger was extracted with dichloromethane, and subjected to sodium chloride salt-out and liquid-liquid extraction with hexane and dichloromethane(the plant and soil samples had no need for the later two steps). The analysis was performed on gas chromatograph equipped with a flame photometric detector(GC-FPD), and the fosthiazate residue was quantitatively determined by calculating the peak area. [Results] The results of degradation dynamics showed that the half-life of fosthiazate was 7.2-8.7 d in ginger plants, and was 9.9-19.8 d in the soil. The results of the final residue test showed that after applying the fosthiazate granules according to the recommended high dose(active component 58.2 g a.i/m^2,WK) and low dose(38.8 g a.i/m^2,WK) once, the final residual amounts of fosthiazate in the ginger and soil were estimated to be <0.02 mg/kg and <0.02-0.103 mg/kg, respectively, while no fosthiazate residue was detected in all control samples(<0.02 mg/kg). The maximum residue limit(MRL) of fosthiazate in ginger was not established in China, and was 0.2 mg/kg in Japan. [Conclusions] The method was proved to meet the basic requirements of pesticide residue analysis on sensitivity, accuracy and precision and have good linearity. | 11 | Real-time detection of LMP1/LMP1 interaction in MβCD-induced apoptosis of nasopharyngeal carcinoma cells using FRET method显示文摘Latent mermbrane protein 1(LMP1)is known as an oncoprotein in nasopharyngeal carcinoma(NPC)cells,which is considered to have a strong association with growth,invasion and metastasis of NPC cells through lipid rafts.Met hy-A-cyclodextrin(M3CD)can disrupt lipid rafts through cholesterol depletion.In this study,we revealed that MICD induced apoptosis in two kinds of NPC cells including CNE1 cells,a LMP1 negative nasopharyngeal squamous carcinoma cell line,and CNE1-LMP1 cells,a LMP1-positive nasopharyngeal squamous carcinoma cell line.Furthermore,the impact of MBCD on LMP1 was investigated by fuorescence resonance energy transfer(FRET)method in NPC cells.Synchronized tempo spatial and spectral detection of LMP1/LMP1 interaction were performed using fluorescence microscope and spectrograph.FRET efficiency indicated that LMP1/LMP1 interaction gradully enhanced after M9CD treatment.MTT assays showed that M8CD caused strong cytotoxicity in CNE1 cells,but cauused relatively weaker cytotoxicity in CNE1-LMP1 cells,which indicated that LMP1 may regulate sensitivity of NPC cells to MBCD.Then,detection of cleaved caspase-3 in two kinds of NPC cells indicated that LMP1 may inhibit activation of caspase 3.These results strongly suggested that MBCD can induce apoptosis of NPC cells,but enhancing of LMP1/LMP1 interaction may likely resist apoptosis through inhibiting cleavage of caspase-3. | Zhiwei Wu Qing Ye Yimei Huang Yating Lin Mengmeng Zheng Xiaoyan Wang Shusen Xie Juqiang Lin | 2019 | Journal of Innovative Optical Health Sciences2019,,6: | 0 |

| 16 | A Predictive Nomogram for Predicting Improved Clinical Outcome Probability in Patients with COVID-19 in Zhejiang Province,China显示文摘The aim of this research was to develop a quantitative method for clinicians to predict the probability of improved prognosis in patients with coronavirus disease 2019(COVID-19).Data on 104 patients admitted to hospital with laboratory-confirmed COVID-19 infection from 10 January 2020 to 26 February 2020 were collected.Clinical information and laboratory findings were collected and compared between the outcomes of improved patients and non-improved patients.The least absolute shrinkage and selection operator(LASSO)logistics regression model and two-way stepwise strategy in the multivariate logistics regression model were used to select prognostic factors for predicting clinical outcomes in COVID-19 patients.The concordance index(C-index)was used to assess the discrimination of the model,and internal validation was performed through bootstrap resampling.A novel predictive nomogram was constructed by incorporating these features.Of the 104 patients included in the study(median age 55 years),75(72.1%)had improved short-term outcomes,while 29(27.9%)showed no signs of improvement.There were numerous differences in clinical characteristics and laboratory findings between patients with improved outcomes and patients without improved outcomes.After a multi-step screening process,prognostic factors were selected and incorporated into the nomogram construction,including immunoglobulin A(IgA),C-reactive protein(CRP),creatine kinase(CK),acute physiology and chronic health evaluation II(APACHE II),and interaction between CK and APACHE II.The C-index of our model was 0.962(95%confidence interval(CI),0.931-0.993)and still reached a high value of 0.948 through bootstrapping validation.A predictive nomogram we further established showed close performance compared with the ideal model on the calibration plot and was clinically practical according to the decision curve and clinical impact curve.The nomogram we constructed is useful for clinicians to predict improved clinical outcome probability for each COVID-19 patient,which may facilitate personalized counselling and treatment. | Jiaojiao Xie Ding Shi Mingyang Bao Xiaoyi Hu Wenrui Wu Jifang Sheng Kaijin Xu Qing Wang Jingjing Wu Kaicen Wang Daiqiong Fang Yating Li Lanjuan Li | 2022 | Engineering2022,8,1: | 0 |
