1Isolation,characterization,and phylogenic analysis of three new severe fever with thrombocytopenia syndrome bunyavirus strains derived from Hubei Province,China显示文摘Hubei Province is a major epidemic area of severe fever with thrombocytopenia syndrome bunyavirus(SFTSV) in China. However, to date, a few SFTSV strains have been isolated from Hubei Province, preventing effective studies of epidemic outbreaks. Here, we report three confirmed patients(2015–2016) with typical symptoms of severe fever with thrombocytopenia syndrome disease(SFTS) who were farmers resident in different regions in Hubei Province. Three new SFTSV strains were isolated from the serum samples of each patient. Characterization of viral growth properties showed that there were no significant differences in virus production. All strains were completely sequenced, and phylogenetic analysis showed that unlike the other strains from Hubei province, which belonged to the SFTSV C3 genotype, one of the three strains belonged to the SFTSV C2 genotype. 2Changes in nitrogen budget and potential risk to the environment over 20 years(1990–2010) in the agroecosystems of the Haihe Basin, China显示文摘The nitrogen balance can serve as an indicator of the risk to the environment of nitrogen loss from agricultural land. To investigate the temporal and spatial changes in agricultural nitrogen application and its potential threat to the environment of the Haihe Basin in China, we used a database of county-level agricultural statistics to calculate agricultural nitrogen input, output,surplus intensity, and use efficiency. Chemical fertilizer nitrogen input increased by 51.7% from1990 to 2000 and by 37.2% from 2000 to 2010, concomitant with increasing crop yields.Simultaneously, the nitrogen surplus intensity increased by 53.5% from 1990 to 2000 and by16.5% from 2000 to 2010, presenting a continuously increased environmental risk. Nitrogen use efficiency decreased from 0.46 in 1990 to 0.42 in 2000 and remained constant at 0.42 in 2010,partly due to fertilizer composition and type improvement. This level indicates that more than half of nitrogen inputs are lost in agroecosystems. Our results suggest that although the improvement in fertilizer composition and types has partially offset the decrease in nitrogen use efficiency, the environmental risk has still increased gradually over the past 20 years, along with the increase in crop yields and nitrogen application. 3Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 4Catalysis under shell: Improved CO oxidation reaction confined in Pt@h-BN core-shell nanoreactors显示文摘Core-shell nanostructures consisting of active metal cores and protective shells often exhibit enhanced catalytic performance, in which reactants can access a small part of the core surfaces through the pores in the shells. In this study, we show that Pt nanoparticles (NPs) can be embedded into few-layer hexagonal boron nitride (h-BN) overlayers, forming Pt@h-BN core-shell nanocatalysts. The h-BN shells not only protect the Pt NPs under harsh conditions but also allow gaseous molecules such as CO and O2 to access a large part of the Pt surfaces through a facile intercalation process. As a result, the Pt@h-BN nanostructures act as nanoreactors, and CO oxidation reactions with improved activity, selectivity, and stability occur at the core-shell interfaces. The confinement effect exerted by the h-BN shells promotes the Pt-catalyzed reactions. 5Abnormal Behavior Detection and Recognition Method Based on Improved ResNet Model显示文摘The core technology in an intelligent video surveillance system is that detecting and recognizing abnormal behaviors timely and accurately.The key breakthrough point in recognizing abnormal behaviors is how to obtain the effective features of the picture,so as to solve the problem of recognizing them.In response to this difficulty,this paper introduces an adjustable jump link coefficients model based on the residual network.The effective coefficients for each layer of the network can be set after using this model to further improving the recognition accuracy of abnormal behavior.A convolution kernel of 1×1 size is added to reduce the number of parameters for the purpose of improving the speed of the model in this paper.In order to reduce the noise of the data edge,and at the same time,improve the accuracy of the data and speed up the training,a BN(Batch Normalization)layer is added before the activation function in this network.This paper trains this network model on the public ImageNet dataset,and then uses the transfer learning method to recognize these abnormal behaviors of human in the UTI behavior dataset processed by the YOLO_v3 target detection network.Under the same experimental conditions,compared with the original ResNet-50 model,the improved model in this paper has a 2.8%higher accuracy in recognition of abnormal behaviors on the public UTI dataset.Huifang Qian Mengmeng Zheng Xuan Zhou 2020Computers, Materials & Continua2020,,12:3

8Novel SFTSV Phylogeny Reveals New Reassortment Events and Migration Routes显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV),the causative agent of a febrile human disease,was first identified from central and eastern provinces in China,and later in Japan and South Korea.Hubei Province is one of the major SFTS epidemic areas in the central part of China.This study reported the isolation of 11 new SFTSV strains from patients in Hubei Province collected in 2017.Extensive phylogenetic analyses were conducted based on the complete coding sequences of SFTSV segments including the new strains.It was suggested that five different SFTSV genotypes were circulating in Hubei,and 15 reassortment patterns and migration pathways correlated with each genotype were identified,which was more than previously recognized.Hubei Province was more involved in the evolutionary events of SFTSV than that previously thought in which the evolutionary events of SFTSV were reported to be independent from those in other epidemic regions.Further divergence of SFTSV strains was suggested by pairwise comparison of SFTSV sequences from each genotype and sequence identity normalized to representative strain in genotype C1.Subsequently,amino acid variations specific for genotype(s),strain(s),or cluster(s)were inspected,which may be related to differential biological activity of SFTSV strains/genotypes.In conclusion,we analyzed the current status of SFTSV phylogeny in Hubei Province and discussed the possible events correlated to SFTSV evolution.It provided an in-depth insight into SFTSV evolution,raising concerns for the use of proper SFTSV strains in future studies.Xiaoli Wu Mingyue Li Yanfang Zhang Boyun Liang Junming Shi Yaohui Fang Zhengyuan Su Mengmeng Li Wenjing Zhang Ling Xu Jun Wang Qiaoli Wu Shuang Tang Hualin Wang Tao Zhang Cheng Peng Xin Zheng Fei Deng Shu Shen 2021Virologica Sinica2021,36,2:2
9Selective coordination and localized polarization in graphene quantum dots: Detection of fluoride anions using ultra-low-field NMR relaxometry显示文摘The development of ultra-sensitive methods for detecting anions is limited by their low charge to radius ratios, microenvironment sensitivity, and p H sensitivity. In this paper, a magnetic sensor is devised that exploits the controllable and selective coordination that occurs between a magnetic graphene quantum dot(GQD) and fluoride anion(F–). The sensor is used to measure the change in relaxation time of aqueous solutions of magnetic GQDs in the presence of F–using ultra-low-field(118 μT) nuclear magnetic resonance relaxometry. The method was optimized to produce a limit of detection of 10 nmol/L and then applied to quantitatively detect F–in domestic water samples. More importantly, the key factors responsible for the change in relaxation time of the magnetic GQDs in the presence of F–are revealed to be the selective coordination that occurs between the GQDs and F–as well as the localized polarization of the water protons. 11Sporophytic control of anther development and male fertility by glucose-6-phosphate/phosphate translocator 1(OsGPT1) in rice显示文摘Coordination between the sporophytic tissue and the gametic pollen within anthers is tightly controlled to achieve the optimal pollen fitness. Glucose-6-phosphate/phosphate translocator(GPT) transports glucose-6-phosphate, a key precursor of starch and/or fatty acid biosynthesis, into plastids. Here, we report the functional characterization of Os GPT1 in the rice anther development and pollen fertility. Pollen grains from homozygous osgpt1 mutant plants fail to accumulate starch granules, resulting in pollen sterility. Genetic analyses reveal a sporophytic effect for this mutation. Os GPT1 is highly expressed in the tapetal layer of rice anther. Degeneration of the tapetum, an important process to provide cellular contents to support pollen development, is impeded in osgpt1 plants. In addition, defective intine and exine are observed in the pollen from osgpt1 plants. Expression levels of multiple genes that are important to tapetum degeneration or pollen wall formation are significantly decreased in osgpt1 anthers. Previously, we reported that At GPT1 plays a gametic function in the accumulation of lipid bodies in Arabidopsis pollen. This report highlights a sporophytic role of Os GPT1 in the tapetum degeneration and pollen development. 15Polyphosphate Accelerates Transformation of Nonstructural Carbohydrates to Improve Growth of ppk-Expressing Transgenic Rice in Phosphorus Deficiency Culture显示文摘Crop yield and quality are often limited by the amount of phosphate fertilizer added to infertile soils,a key limiting factor for sustainable development in modern agriculture.The polyphosphate kinase(ppk)gene-expressing transgenic rice with a single-copy line(ETRS)is constructed to improve phosphate fertilizer utilization efficiency for phosphorus resource conservation.To investigate the potential mechanisms of the increased biomass in ETRS in low phosphate culture,ETRS was cultivated in a low inorganic phosphate(Pi)culture medium(15μmol/L Pi,LP)and a normal Pi culture medium(300μmol/L Pi,CP),respectively.After 89 d of cultivation in different concentrations of phosphate culture media,the total phosphorus,polyphosphate(polyP),biomass,photosynthetic rate,nonstructural carbohydrate(NSC)contents,related enzyme activities,and related gene expression levels were analyzed.The results showed that ETRS had a high polyP amount to promote the photosynthetic rate in LP,and its biomass was almost the same as the wild type(WT)in CP.The NSC content of ETRS in LP was higher than that of WT in LP,but slightly lower than that of WT in CP.PolyP notably promoted the sucrose phosphate synthase activities of ETRS and significantly down-regulated the expression levels of sucrose transporter genes(OsSUT3 and OsSUT4),resulting in inhibiting the transport of sucrose from shoot to root in ETRS.It was concluded that polyP can stimulate the synthesis of NSCs in LP,which improved the growth of ETRS and triggered the biological activities of ETRS to save phosphate fertilizer.Our study provides a new way to improve the utilization rate of phosphate fertilizer in rice production.ZHU Jinling WEI Ruping WANG Xin ZHENG Chaoqun WANG Mengmeng YANG Yicheng YANG Liuyan 2023Rice science2023,30,3:1

17Sustainability of SARS-CoV-2 Induced Humoral Immune Responses in COVID-19 Patients from Hospitalization to Convalescence Over Six Months显示文摘Understanding the persistence of antibody in convalescent COVID-19 patients may help to answer the current major concerns such as the risk of reinfection,the protection period of vaccination and the possibility of building an active herd immunity.This retrospective cohort study included 172 COVID-19 patients who were hospitalized in Wuhan.A total of404 serum samples were obtained over six months from hospitalization to convalescence.Antibodies in the specimens were quantitatively analyzed by the capture chemiluminescence immunoassays(CLIA).All patients were positive for the anti-SARS-Co V-2 Ig M/Ig G at the onset of COVID-19 symptoms,and the Ig G antibody persisted in all the patients during the convalescence.However,only approximately 25%of patients can detect the Ig M antibodies,Ig M against N protein(NIg M)and receptor binding domain of S protein(RBD-Ig M)at the 27 th week.The titers of Ig M,N-Ig M and RBD-Ig M reduced to 16.7%,17.6%and 15.2%of their peak values respectively.In contrast,the titers of Ig G,N-Ig G and RBD-Ig G peaked at 4–5 th week and reduced to 85.9%,62.6%and 87.2%of their peak values respectively at the end of observation.Dynamic behavior of antibodies and their correlation in age,gender and severity groups were investigated.In general,the COVID-19 antibody was sustained at high levels for over six months in most of the convalescent patients.Only a few patients with antibody reducing to an undetectable level which needs further attention.The humoral immune response against SARS-Co V-2 infection in COVID-19 patients exhibits a typical dynamic of acquired immunity.Yang Zheng Qing Zhang Ashaq Ali Ke Li Nan Shao Xiaoli Zhou Zhiqin Ye Xiaomin Chen Shanshan Cao Jing Cui Juan Zhou Dianbing Wang Baidong Hou Min Li Mengmeng Cui Lihua Deng Xinyi Sun Qian Zhang Qinfang Yang Yong li Hui Wang Yake Lei Bo Yu Yegang Cheng Xiaolin Tong Dong Men Xian-En Zhang 2021Virologica Sinica2021,36,5:1
