| 2 | The KDM4B–CCAR1–MED1 axis is a critical regulator of osteoclast differentiation and bone homeostasis显示文摘Bone undergoes a constant and continuous remodeling process that is tightly regulated by the coordinated and sequential actions of bone-resorbing osteoclasts and bone-forming osteoblasts.Recent studies have shown that histone demethylases are implicated in osteoblastogenesis;however,little is known about the role of histone demethylases in osteoclast formation.Here,we identified KDM4B as an epigenetic regulator of osteoclast differentiation.Knockdown of KDM4B significantly blocked the formation of tartrate-resistant acid phosphatase-positive multinucleated cells.Mice with myeloid-specific conditional knockout of KDM4B showed an osteopetrotic phenotype due to osteoclast deficiency.Biochemical analysis revealed that KDM4B physically and functionally associates with CCAR1 and MED1 in a complex.Using genome-wide chromatin immunoprecipitation(ChIP)-sequencing,we revealed that the KDM4B–CCAR1–MED1 complex is localized to the promoters of several osteoclast-related genes upon receptor activator of NF-κB ligand stimulation.We demonstrated that the KDM4B–CCAR1–MED1 signaling axis induces changes in chromatin structure(euchromatinization)near the promoters of osteoclast-related genes through H3K9 demethylation,leading to NF-κB p65 recruitment via a direct interaction between KDM4B and p65.Finally,small molecule inhibition of KDM4B activity impeded bone loss in an ovariectomized mouse model.Taken together,our findings establish KDM4B as a critical regulator of osteoclastogenesis,providing a potential therapeutic target for osteoporosis. | Sun-Ju Yi You-Jee Jang Hye-Jung Kim Kyubin Lee Hyerim Lee Yeojin Kim Junil Kim Seon Young Hwang Jin Sook Song Hitoshi Okada Jae-Il Park Kyuho Kang Kyunghwan Kim | 2021 | Bone Research2021,9,3: | 1 |

| 6 | A Novel Transparent and Auditable Fog-Assisted Cloud Storage with Compensation Mechanism显示文摘This paper introduces a new fog-assisted cloud storage which can achieve much higher throughput compared to the traditional cloud-only storage architecture by reducing the traffics toward the cloud storage. The fog-storage service providers are transparency to end-users and therefore, no modification on the end-user devices is necessary. This new system is featured with(1) a stronger audit scheme which is naturally coupled with the proposed architecture and does not suffer from the replay attack and(2) a transparent and efficient compensation mechanism for the fog-storage service providers. We provide rigorous theoretical analysis on the correctness and soundness of the proposed system. | 7 | Ursolic acid enhances autophagic clearance and ameliorates motor and non-motor symptoms in Parkinson’s disease mice model显示文摘Protein aggregation and the abnormal accumulation of aggregates are considered as common mechanisms of neurodegeneration such as Parkinson’s disease(PD).Ursolic acid(UA),a natural pentacyclic triterpenoid compound,has shown a protective activity in several experimental models of brain dysfunction through inhibiting oxidative stress and inflammatory responses and suppressing apoptotic signaling in the brain.In this study,we investigated whether UA promoted autophagic clearance of protein aggregates and attenuated the pathology and characteristic symptoms in PD mouse model.Mice were injected with rotenone(1 mg·kg^(−1)·d^(−1),i.p.)five times per week for 1 or 2 weeks.We showed that rotenone injection induced significant motor deficit and prodromal non-motor symptoms accompanied by a significant dopaminergic neuronal loss and the deposition of aggregated proteins such as p62 and ubiquitin in the substantia nigra and striatum.Co-injection of UA(10 mg·kg^(−1)·d^(−1),i.p.)ameliorated all the rotenone-induced pathological alterations.In differentiated human neuroblastoma SH-SY5Y cells,two-step treatment with a proteasome inhibitor MG132(0.25,2.5μM)induced marked accumulation of ubiquitin and p62 with clear and larger aggresome formation,while UA(5μM)significantly attenuated the MG132-induced protein accumulation.Furthermore,we demonstrated that UA(5μM)significantly increased autophagic clearance by promoting autophagic flux in primary neuronal cells and SH-SY5Y cells;UA affected autophagy regulation by increasing the phosphorylation of JNK,which triggered the dissociation of Bcl-2 from Beclin 1.These results suggest that UA could be a promising therapeutic candidate for reducing PD progression from the prodromal stage by regulating abnormal protein accumulation in the brain. | Yeojin Bang Yoonjung Kwon Mihyang Kim Soung Hee Moon Kiwon Jung Hyun Jin Choi | 2023 | Acta Pharmacologica Sinica2023,44,4: | 0 |
