1B-box proteins:Pivotal players in light-mediated development in plants^FA显示文摘Light signals mediate a number of physio-logical and developmental processes in plants, such as flowering, photomorphogenesis, and pigment accu-mulation. Emerging evidence has revealed that a group of B-box proteins (BBXs) function as central players in these light-mediated developmental processes. B-box proteins are a class of zinc-coordinated transcription factors or regulators that not only directly mediate the transcription of target genes but also interact with various other factors to create a complex regulatory network involved in the precise control of plant growth and development. 3Integration method of TINs and Grids for multi-resolution surface modeling显示文摘Highly detailed surface models and their real-time applications are increasingly popular in architecture,construction and other design and engineering fields.However,new and related problems have emerged concerning the efficient management of the resulting large datasets and the seamless integration of heterogeneous data.Moreover,the increasingly common requirements of local high-fidelity modeling combined with large-scale landscapes lead to difficulty in the seamless multi-resolution representation of hybrid triangulated irregular networks(TINs)and Grids.This paper presents a hybrid data structure with high-efficiency and a related organizational method for the seamless integration of multi-resolution models.This approach is characterized by(1)a self-adaptive algorithm for feature-preserving surface partitioning,(2)an efficient hybrid index structure for combined Grid and TIN surfaces,and(3)a view-dependent scheduling strategy with access to Grids of necessary resolution,giving priority to the dynamic loading of TINs.Experiments using typical real design datasets of highway constructions are able to achieve accuracy-preserved and real-time availability of results that prove the validity and efficiency of this approach.Xie XIAO Xu WEIPING Zhu QING Zhang YETING Du ZHIQIANG 2013Geo-Spatial Information Science2013,16,1:2

7Proximity-enabled covalent binding of IL-2 to IL-2Rα selectively activates regulatory T cells and suppresses autoimmunity显示文摘Interleukin-2(IL-2)is a pleiotropic cytokine that orchestrates bidirectional immune responses via regulatory T cells(Tregs)and effector cells,leading to paradoxical consequences.Here,we report a strategy that exploited genetic code expansion-guided incorporation of the latent bioreactive artificial amino acid fluorosulfate-L-tyrosine(FSY)into IL-2 for proximity-enabled covalent binding to IL-2Rαto selectively promote Treg activation.We found that FSY-bearing IL-2 variants,such as L72-FSY,covalently bound to IL-2Rαvia sulfur-fluoride exchange when in proximity,resulting in persistent recycling of IL-2 and selectively promoting the expansion of Tregs but not effector cells.Further assessment of L72-FSY-expanded Tregs demonstrated that L72-FSY maintained Tregs in a central memory phenotype without driving terminal differentiation,as demonstrated by simultaneously attenuated expression of lymphocyte activation gene-3(LAG-3)and enhanced expression of programmed cell death protein-1(PD-1).Subcutaneous administration of L72-FSY in murine models of pristane-induced lupus and graft-versus-host disease(GvHD)resulted in enhanced and sustained therapeutic efficacy compared with wild-type IL-2 treatment.The efficacy of L72-FSY was further improved by N-terminal PEGylation,which increased its circulatory retention for preferential and sustained effects.This proximity-enabled covalent binding strategy may accelerate the development of pleiotropic cytokines as a new class of immunomodulatory therapies.Bo Zhang Jiaqi Sun Yeshuang Yuan Dezhong Ji Yeting Sun Yudong Liu Shengjie Li Xingxing Zhu Xunyao Wu Jin Hu Qiu Xie Ling Wu Lulu Liu Boyang Cheng Yuanjie Zhang Lingjuan Jiang Lidan Zhao Fei Yu Wei Song Min Wang Yue Xu Shiliang Ma Yunyun Fei Lihe Zhang Demin Zhou Xuan Zhang 2023Signal Transduction and Targeted Therapy2023,8,2:0
8Friction measurement of aircraft wing based on optimized FlowNet2.0显示文摘Few studies have applied the deep optical flows model to global friction measurements of aircraft wing.This study used an optimized FlowNet2.0 model to measure friction of wing based on fluorescent oil film,which achieved the first integration of deep learning and friction measurement.Two input images of the traditional FlowNet2.0 model were extended to multiple images so that more flow features and details could be with them.It is the specific part of optimization that will also further improve the measurement accuracy of FlowNet2.0.Simulation experimental results show that the optimized FlowNet2.0 model reduces the Mean Absolute Percentage Error(MAPE)error by 8.51%and increases Root Mean Square Error(RMSE)by only 0.0138 when compared to the hybrid optical flow method,which indicate that the optimized FlowNet2.0 model has great potential for friction measurement.Measurements in continuous transonic wind tunnel tests demonstrate that FlowNet2.0 can calculate clearer and more accurate flow velocity than hybrid optical flow,and the solved friction magnitude distribution is consistent with the actual flow,which will be of great practical application in wind tunnel engineering.Hongjiang QIAN Zhiyong HUANG Jian WANG Yeting XU Xiucheng DONG Jiebin SHEN 2023Chinese Journal of Aeronautics2023,36,11:0
9Effect of serum on PEGylated quantum dots:Cellular uptake and intracellular distribution显示文摘Protein adsorption is closely related with the interactions between nanoparticles and physiological systems,and further influences the cellular uptake and distribution of nanoparticles in cells.Although polyethylene glycol(PEG)ylation can largely reduce specific protein adsorption,some protein components in whole serum still interact with nanoparticles.In this work,PEGylated quantum dots(QDs) were used for investigating the quantitative and qualitative relationships of fetal bovine serum(FBS) and the cellular uptake/intracellular distribution in human hepatoma(HepG2) cell line.Nondenaturing polyacrylamide gel electrophoresis and two dimensional electrophoresis were used to analyze the adsorption of protein by PEGylated QDs.Quantitative cellular uptake of PEGylated QDs was determined by fluorescence activated cell sorting(FACS) with different FBS concentrations and incubating durations.The intracellular location of PEGylated QDs in HepG2 cells was observed using a confocal laser scanning microscope(CLSM) and a transmission electron microscope(TEM).This work will be helpful to understand the interaction between nanoparticles and cells with serum.Yeting Jian Xianghui Xu Yunkun Li Zhongwei Gu 2013Progress in Natural Science:Materials International2013,23,6:0
10PIFs‑and COP1‑HY5‑mediated temperature signaling in higher plants显示文摘Plants have to cope with the surrounding changing environmental stimuli to optimize their physiological and devel-opmental response throughout their entire life cycle.Light and temperature are two critical environmental cues that fluctuate greatly during day-night cycles and seasonal changes.These two external signals coordinately control the plant growth and development.Distinct spectrum of light signals are perceived by a group of wavelength-specific photoreceptors in plants.PIFs and COP1-HY5 are two predominant signaling hubs that control the expression of a large number of light-responsive genes and subsequent light-mediated development in plants.In parallel,plants also transmit low or warm temperature signals to these two regulatory modules that precisely modulate the responsive-ness of low or warm temperatures.The core component of circadian clock ELF3 integrates signals from light and warm temperatures to regulate physiological and developmental processes in plants.In this review,we summarize and discuss recent advances and progresses on PIFs-,COP1-HY5-and ELF3-mediated light,low or warm temperature signaling,and highlight emerging insights regarding the interactions between light and low or warm temperature signal transduction pathways in the control of plant growth.Yeting Bian Li Chu Huan Lin Yaoyao Qi Zheng Fang Dongqing Xu 2022Stress Biology2022,2,1:0
11COP1 SUPPRESSOR 6 represses the PIF4 and PIF5 action to promote light-inhibited hypocotyl growth显示文摘Light signaling precisely controls photomorphogenic development in plants.PHYTOCHROME INTERACTING FACTOR 4 and 5(PIF4 and PIF5)play critical roles in the regulation of this developmental process.In this study,we report CONSTITUTIVELY PHOTOMORPHOGENIC 1 SUPPRESSOR 6(CSU6)functions as a key regulator of light signaling.Loss of CSU6 function largely rescues the cop1-6 constitutively photomorphogenic phenotype.CSU6 promotes hypocotyl growth in the dark,but inhibits hypocotyl elongation in the light.CSU6 not only associates with the promoter regions of PIF4 and PIF5 to inhibit their expression in the morning,but also directly interacts with both PIF4 and PIF5 to repress their transcriptional activation activity.CSU6 negatively controls a group of PIF4-and PIF5-regulated gene expressions.Mutations in PIF4 and/or PIF5 are epistatic to the loss of CSU6,suggesting that CSU6 acts upstream of PIF4 and PIF5.Taken together,CSU6 promotes light-inhibited hypocotyl elongation by negatively regulating PIF4 and PIF5 transcription and biochemical activity.Hongxia Lan Yueqin Heng Jian Li Mengdi Zhang Yeting Bian Li Chu Yan Jiang Xuncheng Wang Dongqing Xu Xing Wang Deng 2022Journal of Integrative Plant Biology2022,64,11:0
