1Cross-talk between NMDA and GABA_A receptors in cultured neurons of the rat inferior colliculus显示文摘Neuronal ion channels of different types often do not function independently but will inhibit or potentiate the activity of other types of channels,a process called cross-talk.The N-methyl-D-aspartate receptor (NMDA receptor) and the γ-aminobutyric acid type A receptor (GABAA receptor) are important excitatory and inhibitory receptors in the central nervous system,respectively.Currently,cross-talk between the NMDA receptor and the GABAA receptor,particularly in the central auditory system,is not well understood.In the present study,we investigated functional interactions between the NMDA receptor and the GABAA receptor using whole-cell patch-clamp techniques in cultured neurons from the inferior colliculus,which is an important nucleus in the central auditory system.We found that the currents induced by aspartate at 100 μmol L-1 were suppressed by the pre-perfusion of GABA at 100 μmol L-1,indicating cross-inhibition of NMDA receptors by activation of GABAA receptors.Moreover,we found that the currents induced by GABA at 100 μmol L-1 (IGABA) were not suppressed by the pre-perfusion of 100 μmol L-1 aspartate,but those induced by GABA at 3 μmol L-1 were suppressed,indicating concentration-dependent cross-inhibition of GABAA receptors by activation of NMDA receptors.In addition,inhibition of IGABA by aspartate was not affected by blockade of voltage-dependent Ca2+ channels with CdCl2 in a solution that contained Ca2+,however,CdCl2 effectively attenuated the inhibition of IGABA by aspartate when it was perfused in a solution that contained Ba2+ instead of Ca2+ or a solution that contained Ca2+ and 10 mmol L-1 BAPTA,a membrane-permeable Ca2+ chelator,suggesting that this inhibition is mediated by Ca2+ influx through NMDA receptors,rather than voltage-dependent Ca2+ channels.Finally,KN-62,a potent inhibitor of Ca2+/calmodulin-dependent protein kinase II (CaMKII),reduced the inhibition of IGABA by aspartate,indicating the involvement of CaMKII in this cross-inhibition.Our study demonstrates a functional interaction between NMDA and GABAA receptors in the inferior colliculus of rats.The presence of cross-talk between these receptors suggests that the mechanisms underlying information processing in the central auditory system may be more complex than previously believed.CONG DanNi TANG ZhengQuan LI LongZhu HUANG YiNa WANG Jun CHEN Lin 2011Science China(Life Sciences)2011,54,6:9

4Synthetic biology of plant natural products: From pathway elucidation to engineered biosynthesis in plant cells显示文摘Plant natural products(PNPs)are the main sources of drugs,food additives,and new biofuels and have become a hotspot in synthetic biology.In the past two decades,the engineered biosynthesis of many PNPs has been achieved through the construction of microbial cell factories.Alongside the rapid development of plant physiology,genetics,and plant genetic modification techniques,hosts have now expanded from single-celled microbes to complex plant systems.Plant synthetic biology is an emerging field that combines engineering principles with plant biology.In this review,we introduce recent advances in the biosynthetic pathway elucidation of PNPs and summarize the progress of engineered PNP biosynthesis in plant cells.Furthermore,a future vision of plant synthetic biology is proposed.Although we are still a long way from overcoming all the bottlenecks in plant synthetic biology,the ascent of this field is expected to provide a huge opportunity for future agriculture and industry.Xiaoxi Zhu Xiaonan Liu Tian Liu Yina Wang Nida Ahmed Zhichao Li Huifeng Jiang 2021Plant Communications2021,2,5:4
5Study of brain morphology change in Alzheimer’s disease and amnestic mild cognitive impairment compared with normal controls显示文摘Background With an aggravated social ageing level, the number of patients with Alzheimer's disease (AD) is gradually increasing, and mild cognitive impairment (MCI) is considered to be an early form of Alzheimer's disease. How to distinguish diseases in the early stage for the purposes of early diagnosis and treatment is an important topic. Aims The purpose of our study was to investigate the differences in brain cortical thickness and surface area among elderly patients with AD, elderly patients with amnestic MCI (aMCI) and normal controls (NC). Methods 20 AD patients, 21 aMCIs and 25 NC were recruited in the study. FreeSurfer software was used to calculate cortical thickness and surface area among groups. Results The patients with AD had less cortical thickness both in the left and right hemisphere in 17 of the 36 brain regions examined than the patients with aMCI or NC. The patients with AD also had smaller cerebral surface area both in the left and right hemisphere in 3 of the 36 brain regions examined than the patients with aMCI or NC. Compared with the NC, the patients with aMCI only had slight atrophy in the inferior parietal lobe of the left hemisphere, and no significant difference was found. Conclusion AD, as well as aMCI (to a lesser extent), is associated with reduced cortical thickness and surface area in a few brain regions associated with cognitive impairment. 7Analysis of Expression Profiles of Long Noncoding RNAs and mRNAs in A549 Cells Infected with H3N2 Swine Influenza Virus by RNA Sequencing显示文摘Long noncoding RNAs(lncRNAs)participate in regulating many biological processes.However,their roles in influenza A virus(IAV)pathogenicity are largely unknown.Here,we analyzed the expression profiles of lncRNAs and mRNAs in H3N2-infected cells and mock-infected cells by high-throughput sequencing.The results showed that 6129 lncRNAs and 50,031 mRNA transcripts in A549 cells displayed differential expression after H3N2 infection compared with mock infection.Among the differentially expressed lncRNAs,4963 were upregulated,and 1166 were downregulated.Functional annotation and enrichment analysis using gene ontology and Kyoto Encyclopedia of Genes and Genomes databases(KEGG)suggested that target genes of the differentially expressed lncRNAs were enriched in some biological processes,such as cellular metabolism and autophagy.The up-or downregulated lncRNAs were selected and further verified by quantitative real-time polymerase chain reaction(RT-qPCR)and reverse transcription PCR(RT-PCR).To the best of our knowledge,this is the first report of a comparative expression analysis of lncRNAs in A549 cells infected with H3N2.Our results support the need for further analyses of the functions of differentially expressed lncRNAs during H3N2 infection.Yina Zhang Tianqi Yu Yingnan Ding Yahui Li Jing Lei Boli Hu Jiyong Zhou 2020Virologica Sinica2020,35,2:2

9Multi-environment QTL mapping of crown root traits in a maize RIL population显示文摘Crown root traits,including crown root angle(CRA),diameter(CRD),and number(CRN),are major determining factors of root system architecture,which influences crop production.In maize,the genetic mechanisms determining crown root traits in the field are largely unknown.CRA,CRD,and CRN were evaluated in a recombinant inbred line population in three field trials.High phenotypic variation was observed for crown root traits,and all measured traits showed significant genotype–environment interactions.Singleenvironment(SEA)and multi-environment(MEA)quantitative trait locus(QTL)analyses were conducted for CRA,CRD,and CRN.Of 46 QTL detected by SEA,most explained less than 10%of the phenotypic variation,indicating that a large number of minor-effect QTL contributed to the genetic component of these traits.MEA detected 25 QTL associated with CRA,CRD,and CRN,and 2 and 1 QTL were identified with significant QTL-by-environment interaction effects for CRA and CRD,respectively.A total of 26.1%(12/46)of the QTL identified by SEA were also detected by MEA,with many being detected in more than one environment.These findings contribute to our understanding of the phenotypic and genotypic patterns of crown root traits in different environments.The identified environment-specific QTL and stable QTL may be used to improve root traits in maize breeding.Pengcheng Li Yingying Fana Shuangyi Yina Yunyun Wanga Houmiao Wang Yang Xu Zefeng Yang Chenwu Xu 2020The Crop Journal2020,8,4:1

12Laboratory investigation of coal sample permeability under the coupled effect of temperature and stress显示文摘The stress and temperature sensitivities of coal reservoirs are critical geological factors affectingcoalbed methane(CBM)well exploitation;in particular it is important to reduce or eliminate their influence on coal reservoir permeability.To investigate coal permeability behavior at various effective stresses and temperatures,CH4 permeability tests were conducted on raw coal samples under a varying effective stress of 2.0-8.0 MPa under five different temperatures(25℃-65℃)in the laboratory.The results show that the permeability of the coal samples exponentially decreases with increasing effective stress or temperature,which indicates obvious stress and temperature sensitivity.Through a dimension-less treatment of coal permeability,effective stress,and temperature,a new stress sensitivity index S and tempera-ture index ST are proposed to evaluate coal stress and temperature sensitivity evaluation parameters.These new parameters exhibit integrality and uniqueness,and,in combination with stress sensitivity coefficient αk,tempe-rature sensitivity coefficient αT,and the permeability damage rate PDR,the sensitivities of coal permeability to stress and temperature are evaluated.The results indicate that coal sample stress sensitivity decreases with increasing effective stress,while it first decreases and then increases with increasing temperature.Additionally,coal sample temperature sensitivity shows a downward trend when temperature increases and fluctuates when effective stress increases.Finally,a coupled coal permeability model considering the impacts of effective stress and temperature is established,and the main factors affecting coal reservoir permeability and their control mechanism are explored.These results can provide some theoretical guidance for the further development of deep CBM.Yina YU Zhaoping MENG Jiangjiang LI Yixin LU Caixia GAO 2022Frontiers of Earth Science2022,16,4:1

14Adoptive transfer of Pfkfb3-disrupted hematopoietic cells to wild-type mice exacerbates diet-induced hepatic steatosis and inflammation显示文摘Background and objectives:Hepatic steatosis and inflammation are key characteristics of non-alcoholic fatty liver disease(NAFLD).However,whether and how hepatic steatosis and liver inflammation are differentially regulated remains to be elucidated.Considering that disruption of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(Pfkfb3/iPfk2)dissociates fat deposition and inflammation,the present study examined a role for Pfkfb3/iPfk2 in hematopoietic cells in regulating hepatic steatosis and inflammation in mice.Methods:Pfkfb3-disrupted(Pfkfb3^(+/-))mice and wild-type(WT)littermates were fed a high-fat diet(HFD)and examined for NAFLD phenotype.Also,bone marrow cells isolated from Pfkfb3^(+/-)mice andWT mice were differentiated into macrophages for analysis of macrophage activation status and for bone marrow transplantation(BMT)to generate chimeric(WT/BMT-Pfkfb3^(+/-))mice in which Pfkfb3 was disrupted only in hematopoietic cells and control chimeric(WT/BMT-WT)mice.The latter were also fed an HFD and examined for NAFLD phenotype.In vitro,hepatocytes were co-cultured with bone marrowderived macrophages and examined for hepatocyte fat deposition and proinflammatory responses.Results:After the feeding period,HFD-fed Pfkfb3^(+/-)mice displayed increased severity of liver inflammation in the absence of hepatic steatosis compared with HFD-fed WT mice.When inflammatory activation was analyzed,Pfkfb3^(+/-)macrophages revealed increased proinflammatory activation and decreased anti-proinflammatory activation.When NAFLD phenotype was analyzed in the chimeric mice,WT/BMT-Pfkfb3^(+/-) mice displayed increases in the severity of HFD-induced hepatic steatosis and inflammation compared with WT/BMT-WT mice.At the cellular level,hepatocytes co-cultured with Pfkfb3^(+/-) macrophages revealed increased fat deposition and proinflammatory responses compared with hepatocytes co-cultured with WT macrophages.Conclusions:Pfkfb3 disruption only in hematopoietic cells exacerbates HFD-induced hepatic steatosis and inflammation whereas the Pfkfb3/iPfk2 in nonhematopoietic cells appeared to be needed for HFD feeding to induce hepatic steatosis.As such,the Pfkfb3/iPfk2 plays a unique role in regulating NAFLD pathophysiology.Xin Guo Bilian Zhu Hang Xu Honggui Li Boxiong Jiang Yina Wang Benrong Zheng Shannon Glaser Gianfranco Alpini Chaodong Wu 2020Liver Research2020,4,3:1
