12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 2A Deep Adaptive Learning Method for Rolling Bearing Fault Diagnosis Using Immunity显示文摘The extraction of rolling bearing fault features using traditional diagnostic methods is not sufficiently comprehensive and the features are often chosen subjectively and depend on human experience. In this paper, an improved deep convolutional process is used to extract a set of features adaptively. The hidden multi-layer feature of deep convolutional neural networks is also exploited to improve the extraction features. A deterministic detection of low-confidence samples is performed to ensure the reliability of the recognition results and to decrease the rate of false positives by evaluating the diagnosis of the deep convolutional neural network. To improve the efficiency of the continuous learning elements of the rolling bearing fault diagnosis, a clone learning strategy based on cloning and mutation operations is proposed. 4Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE.Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu 2019Cellular & Molecular Immunology2019,16,3:15
5The microbiome in inflammatory bowel diseases:from pathogenesis to therapy显示文摘Inflammatory bowel disease(IBD)has become a global disease with accelerating incidence worldwide in the 21st century while its accurate etiology remains unclear.In the past decade,gut microbiota dysbiosis has consistently been associated with IBD.Although many IBDassociated dysbiosis have not been proven to be a cause or an effect of IBD,it is often hypothesized that at least some of alteration in microbiome is protective or causative.In this article,we selectively reviewed the hypothesis supported by both association studies in human and pathogenesis studies in biological models.Specifically,we reviewed the potential protective bacterial pathways and species against IBD,as well as the potential causative bacterial pathways and species of IBD.We also reviewed the potential roles of some members of mycobiome and virome in IBD.Lastly,we covered the current status of therapeutic approaches targeting microbiome,which is a promising strategy to alleviate and cure this inflammatory disease.Sheng Liu Wenjing Zhao Ping Lan Xiangyu Mou 2021Protein & Cell2021,12,5:15
6CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12
7Numerical simulation of low pressure die-casting aluminum wheel显示文摘The FDM numerical simulation software,ViewCast system,was employed to simulate the low pressure die casting(LPDC)of an aluminum wheel.By analyzing the mold-filling and solidification stage of the LPDC process,the distribution of liquid fraction,temperature field and solidification pattern of castings were studied.The potential shrinkage defects were predicted to be formed at the rim/spoke junctions,which is in consistence with the X-ray detection result.The distribution pattern of the defects has also been studied.A solution towards reducing such defects has been presented.The cooling capacity of the mold was improved by installing water pipes both in the side mold and the top mold.Analysis on the shrinkage defects under forced cooling mode proved that adding the cooling system in the mold is an effective method for reduction of shrinkage defects.Mi Guofa Liu Xiangyu Wang Kuangfei Fu Hengzhi 2009China Foundry2009,6,1:11
8Progress of regional oceanography study associated with western boundary current in the South China Sea显示文摘Recent progress of physical oceanography in the South China Sea (SCS) associated with the western boundary current (WBC) and eddies is reviewed in this paper. It includes Argo observations of the WBC, eddy detection in the WBC based on satellite images, cross-continental shelf exchange in the WBC, eddy-current interaction, interannual variability of the WBC, air-sea interaction, the SCS throughflow (SCSTF), among others. The WBC in the SCS is strong, and its structure, variability and dynamic processes on seasonal and interannual time scales are yet to be fully understood. In this paper, we summarize progresses on the variability of the WBC, eddy-current interaction, air-sea interaction, and the SCSTF achieved in the past few years. Firstly, using the drifting buoy observations, we point out that the WBC becomes stronger and narrower after it reaches the central Vietnam coast. The possible mechanisms influencing the ocean circulation in the northern SCS are discussed, and the dynamic mechanisms that induce the countercurrent in the region of northern branch of WBC in winter are also studied quantitatively using momentum balance. The geostropic component of the WBC was diagnosed using the ship observation along 18°N, and we found that the WBC changed significantly on interannual time scale. Secondly, using the ship observations, two anti-cyclonic eddies in the winter of 2003/2004 in the northern SCS, and three anti-cyclonic eddies in the summer of 2007 along 18°N were studied. The results show that the two anti-cyclonic eddies can propagate southwestward along the continental shelf at the speed of first Rossby wave (~0.1 ms-1 ) in winter, and the interaction between the three anti-cyclonic eddies in summer and the WBC in the SCS is preliminarily revealed. Eddies on the continental shelf of northern SCS propagated southeastward with a maximum speed of 0.09 ms-1 , and those to the east of Vietnam coast had the largest kinetic energy, both of which imply strong interaction between eddy activity and WBC in the SCS. Thirdly, strong intraseasonal variability (ISV) of sea surface temperature (SST) near the WBC regions was found, and the ISV signal of SST in winter weakens the ISV signal of latent heat flux by 20%. 9Potential targets for protecting against hippocampal cell apoptosis after transient cerebral ischemiareperfusion injury in aged rats显示文摘Mitochondria play an important role in neuronal apoptosis caused by cerebral ischemia, and the role is mediated by the expression of mitochondrial proteins. This study investigated the involvement of mitochondrial proteins in hippocampal cell apoptosis after transient cerebral ischemia-reperfusion injury in aged rats using a comparative proteomics strategy. Our experimental results show that the aged rat brain is sensitive to ischemia-reperfusion injury and that transient ischemia led to cell apoptosis in the hippocampus and changes in memory and cognition of aged rats. Differential proteomics analysis suggested that this phenomenon may be mediated by mitochondrial proteins associated with energy metabolism and apoptosis in aged rats. 10Protective effects of Jiayan Kangtai granules on autoimmune thyroiditis in a rat model by modulating Th17/Treg cell balance显示文摘OBJECTIVE: To investigate the protective effects of Jiayan Kangtai(JYKT) granules, consisting of 9 Chinese herbs, in a rat model of autoimmune thyroiditis(AIT), and the possible underlying mechanism.METHODS: Female Lewis rats(6-8 weeks) were randomly apportioned to 5 groups of 10, including a normal control. AIT was induced in the untreated AIT-model group, and rats treated subsequently with daily low, medium, or high dose JYKT granules. After 12 weeks, plasma levels of thyroid autoantibodies and morphological changes in the thyroid were detected by enzyme-linked immunosorbent assay and histological examination, respectively. The presence of interleukin(IL)-6,IL23 p19, and IL-2 in thyroid tissue was assessed by immunohistochemical staining. The percentages of T helper(Th)17 cells and regulatory T cells(Tregs) in the peripheral blood were analyzed by flow cytometry. Relevant levels of cytokines and proteins were examined via bead-based multiplex flow cytometry and ELISA, respectively. Expressions of genes and proteins regulated by Th17 cells and Tregs were shown by real-time PCR and Western blot.RESULTS: Compared to the control, AIT-model rats had higher plasma concentrations of thyroid autoantibodies. The high-dose JYKT rats showed significantly lower levels of thyroid autoantibodies compared with the AIT model group. Rats in the AIT-JYKT groups also had fewer thyroid lesions and less lymphocytic infiltration, a lower percentage of Th17 cells, and a higher percentage of Tregs, compared with the AIT-model. Rats given high-dose JYKT had a significantly lower Th17/Treg ratio compared with the AIT model. 13Improved clinical outcomes of rhG-CSF-mobilized blood and marrow haploidentical transplantation compared to propensity score-matched rhG-CSF-primed peripheral blood stem cell haploidentical transplantation:a multicenter study显示文摘The effects of haploidentical rhG-CSF-mobilized blood and marrow transplantation(HBMT) on hematological malignances are well established. Previous prospective single-center studies have demonstrated better survival after HBMT versus haploidentical rhG-CSF-mobilized peripheral blood stem cell transplantation(HPBSCT) for acute leukemia(AL) not in remission(NR) or in more than the second complete remission(>CR2). To test the hypothesis that HBMT is still superior to HPBSCT for patients with AL, multiple myeloma(MM), or non-Hodgkin lymphoma(NHL) in CR1/CR2 and for patients with chronic myeloid leukemia in the first and second chronic phase lacking a matched donor, we designed a propensity score method-based multicenter study.Hematopoietic recovery, acute graft-versus-host disease(aGVHD), and chronic GVHD were comparable between the HBMT group(n=168) and the HPBSCT group(n=42). No significant differences were found in non-relapse mortality rate(20.17%±3.58%and 27.24%±7.16%, P=0.18) or relapse rate(19.96%±3.72% and 28.49%±8.25%, P=0.32) between the HBMT group and the HPBSCT group. HBMT recipients had better overall survival(65.0%±4.2% and 54.2%±8.3%, P=0.037) and disease-free survival(59.9%±4.6% and 44.3%±8.7%, P=0.051). Multivariate analysis showed that HPBSCT was associated with poorer DFS(HR(95%CI), 1.639(0.995–2.699), P=0.052). 14Donor-Derived CD19-Targeted T Cell Infusion Eliminates B Cell Acute Lymphoblastic Leukemia Minimal Residual Disease with No Response to Donor Lymphocytes after Allogeneic Hematopoietic Stem Cell Transplantation显示文摘Leukemia relapse is still the leading cause of treatment failure after allogeneic hematopoietic stem cell transplantation (allo-HSCT) for B cell acute lymphoblastic leukemia (B-ALL). Relapsed patients with BALL after allo-HSCT have a very short median survival. Minimal residual disease (MRD) is predictive of forthcoming hematological relapse after hematopoietic stem cell transplantation (HSCT);furthermore, eliminating MRD effectively prevents relapse. Donor lymphoblastic infusion (DLI) is the main established approach to treat B-ALL with MRD after allo-HSCT. However, about one-third of patients with MRD are non-responsive to DLI and their prognosis worsens. Although donor-derived cluster of differentiation (CD)19-directed chimeric antigen receptor-modified (CAR) T cells (CART19s) can potentially cure leukemia, the efficiency and safety of infusions with these cells have not yet been investigated in patients with MRD after HSCT. Between September 2014 and February 2018, six patients each received one or more infusions of CART19s from HSCT donors. Five (83.33%) achieved MRD-negative remission, and one case was not responsive to the administration of CAR T cells. Three of the six patients are currently alive without leukemia. No patient developed acute graft-versus-host disease (aGVHD), and no patient died of cytokine release syndrome. 15A transient bulk flow model with circular whirl motion for rotordynamic coefficients of annular seals显示文摘Bulk flow model with perturbation simplification has been used to calculate rotordynamic coefficients in annular seals which have significant influences on the dynamic behavior of rotors in turbomachinery. In this work, a transient bulk flow model with arbitrary rotor motion is developed, and the boundary conditions and friction factor in the model are calibrated with steady Computational Fluid Dynamics(CFD) analysis. The numerical solution scheme is developed based on the finite element method to obtain the transient reaction force in the seal clearance. With a periodic circular rotor orbit, the transient forces at multiple whirling frequencies are used to evaluate the rotordynamic coefficients. The leakage flowrate of CFD analysis has good agreement with experimental results and the calibrated parameters in bulk flow model are dependent on operating conditions. Although CFD calibration improves the accuracy of the perturbed bulk flow model, the direct damping is overestimated and the cross-coupled damping is underestimated. 16Spontaneous apoptosis of cells in therapeutic stem cell preparation exert immunomodulatory effects through release of phosphatidylserine显示文摘Mesenchymal stem cell(MSC)-mediated immunomodulation has been harnessed for the treatment of human diseases,but its underlying mechanism has not been fully understood.Dead cells,including apoptotic cells have immunomodulatory properties.It has been repeatedly reported that the proportion of nonviable MSCs in a MSC therapeutic preparation varied from 5-50%in the ongoing clinical trials.It is conceivable that the nonviable cells in a MSC therapeutic preparation may play a role in the therapeutic effects of MSCs.We found that the MSC therapeutic preparation in the present study had about 5%dead MSCs(DMSCs),characterized by apoptotic cells.Namely,1×10^(6) MSCs in the preparation contained about 5×10^(4) DMSCs.We found that the treatment with even 5×10^(4) DMSCs alone had the equal therapeutic effects as with 1×10^(6) MSCs.This protective effect of the dead MSCs alone was confirmed in four mouse models,including concanavalin A(ConA)-and carbon tetrachloride(CCI4)-induced acute liver injury,LPS-induced lung injury and spinal cord injury.We also found that the infused MSCs died by apoptosis in vivo.Furthermore,the therapeutic effect was attributed to the elevated level of phosphatidylserine(PS)upon the injection of MSCs or DMSCs.The direct administration of PS liposomes(PSLs)mimic apoptotic cell fragments also exerted the protective effects as MSCs and DMSCs.The Mer tyrosine kinase(MerTK)deficiency or the knockout of chemokine receptor C-C motif chemokine receptor 2(CCR2)reversed these protective effects of MSCs or DMSCs.These results revealed that DMSCs alone in the therapeutic stem cell preparation or the apoptotic cells induced in vivo may exert the same immunomodulatory property as the'living MSCs preparation'through releasing PS,which was further recognized by MerTK and participated in modulating immune cells.Xuemei He Weiqi Hong Jingyun Yang Hong Lei Tianqi Lu Cai He Zhenfei Bi Xiangyu Pan Yu Liu Lunzhi Dai Wei Wang Canhua Huang Hongxin Deng Xiawei Wei 2021Signal Transduction and Targeted Therapy2021,6,8:6

18Efficient and stable tin-based perovskite solar cells by introducing π-conjugated Lewis base显示文摘Tin-based perovskite solar cells(TPSCs)as the most promising candidate for lead-free PSCs have incurred extensive researches all over the world.However,the crystallization process of tin-based perovskite is too fast during the solution-deposited process,resulting in abundant pinholes and poor homogeneity that cause serious charge recombination in perovskite layer.Here,we employed theπ-conjugated Lewis base molecules with high electron density to systematically control the crystallization rate of FASnI3 perovskite by forming stable intermediate phase with the Sn-I frameworks,leading to a compact and uniform perovskite film with large increase in the carrier lifetime.Meanwhile,the introduction of theπ-conjugated systems also retards the permeation of moisture into perovskite crystal,which significantly suppresses the film degradation in air.These benefits contributed to a stabilizing power conversion efficiency(PCE)of 10.1%for the TPSCs and maintained over 90%of its initial PCE after 1000-h light soaking in air.Also,a steady-state efficiency of 9.2%was certified at the accredited test center.Tianhao Wu Xiao Liu Xin He Yanbo Wang Xiangyue Meng Takeshi Noda Xudong Yang Liyuan Han 2020Science China Chemistry2020,63,1:6
19INTERVENTIONAI DIAGNOSIS AND TREATMENT OF VASCULOGENEIC PULSATILE TINNITUS显示文摘Objective To retrospectively study clinical features and diagnostic imaging of vasculogeneic pulsatile tinnitus, and the feasibility and efficacy of transvascular interventional treatment for this condition. Methods Data from 82 cases of arterial or venous pulsatile tinnitus were reviewed. DSA characteristics and possible pathophysiological mechanisms of pulsatile tinnitus in these cases were studied. Diagnoses in this group included intracranial arterovenous fistula(AVF)(n=3), spontaneous skull base dural AVF(n=16), traumatic carotid-cavernous sinus fistula(n=5), subclavian artery stenosis(n=2), internal carotid artery stenosis(n=3), intracranial arterial stenosis(n=1), kinked and/or elongated vertebrobasilar artery(n=2), venous sinus diverticulum(n=2), venous sinus stenosis on the dominant drainage side(n=46) and occipital sinus stenosis(n=2).Treatments included embolization and stenting using coils, NBCA glue, Balt balloons, self-expansion stents and intracranial micro-stents via either the femoral artery or femoral vein. Results Procedures were successful in all cases with no surgery-related complications. Tinnitus disappeared within 2 days after the procedure in all cases. Follow up duration was 5-36 months. Recurrence occurred in 4 cases of arterial tinnitus within 3 months following the initial procedure, which improved after revision embolization or symptom management. There was no recurrence in venous tinnitus cases following stent plastic or stent-coiling embolization treatments. Conclusions Endovascular intervention provides a new approach to the diagnosis and treatment of intractable pulsatile tinnitus.
