| 1 | LncRNA NEAT1 promotes colorectal cancer cell proliferation and migration via regulating glial cell-derived neurotrophic factor by sponging miR-196a-5p显示文摘Colorectal cancer (CRC)is the one of the most common malignant tumors worldwide.Recent years,widespread long non-coding RNAs (IncRNAs)have been discovered and are known to regulate gene expression in cancers.However,the roles and underlying mechanisms of IncRNA in CRC remain largely unclear.Here,we firstly revealed that repression of IncRNA nuclear paraspeckle assembly transcript 1 (NEAT1)inhibited cell proliferation and migration in HCT116.cells and overexpression of NEAT1 promoted cell proliferation and migration in SW480 cells using CCK8 assay and transwell assay.Then,we found that suppression of NEAT1 increased the miR- 196a-5p expression in HCT116 cells,while elevation of NEAT1 decreased the miR-196a-5p expression in SW480 cells using qPCR assay.Furthermore,miR-196a-5p could bind to the predicted binding site of NEAT1.We then found that miR-196a-5p was involved in the role of NEAT1 in CRCs.In addition,we demonstrated that miR-196a-Sp mimics inhibited the glial cell-derived neurotrophic factor (GDNF)expression in HCT116 cells and meanwhile,miR-196a-5p inhibitor promoted GDNF expression in SW480 cells using qPCR and western blot analysis.Then,we proved that miR-196a- 5p exerted its function via regulating GDNF expression in CRCs.Ultimately,our study demon- strated that NEAT1 exerted its role via miR-196a-5p/GDNF axis in CRCs.In summary,this work provided the first evidence of a NEAT1/miR-196a-5p/GDNF regulatory pathway in CRC. | Feng Zhong Wei Zhang Yang Cao Qian Wen Yue Cao Bidan Lou Jinxiang Li Wenying Shi Yinghan Liu Rong Luo and Cheng Chen | 2018 | Acta Biochimica et Biophysica Sinica2018,50,12: | 5 |
| 2 | Enhanced removal of organic matter and typical disinfection byproduct precursors in combined iron–carbon micro electrolysis-UBAF process for drinking water pre-treatment显示文摘The organic matter and two types of disinfection byproduct(DBP) precursors in micropolluted source water were removed using an iron–carbon micro-electrolysis(ICME)combined with up-flow biological aerated filter(UBAF) process. Two pilot-scale experiments(ICME-UBAF and UBAF alone) were used to investigate the effect of the ICME system on the removal of organic matter and DBP precursors. The results showed that ICME pretreatment removed 15.6% of dissolved organic matter(DOM)and significantly improved the removal rate in the subsequent UBAF process. The ICME system removed 31% of trichloromethane(TCM) precursors and 20% of dichloroacetonitrile(DCAN) precursors. The results of measurements of the molecular weight distribution and hydrophilic fractions of DOM and DBP precursors showed that ICME pretreatment played a key role in breaking large-molecular-weight organic matter into low-molecular-weight components, and the hydrophobic fraction into hydrophilic compounds, which was favorable for subsequent biodegradation by UBAF.Three-dimensional fluorescence spectroscopy(3D-EEM) further indicated that the ICME system improved the removal of TCM and DCAN precursors. The biomass analysis indicated the presence of a larger and more diverse microbial community in the ICME-UBAF system than for the UBAF alone. The high-throughput sequencing results revealed that domination of the genera Sphingomonas, Brevundimonas and Sphingorhabdus contributed to the better removal of organic matter and two types of DBP precursors. The HPV E6 gene is essential for the oncogenic potential of HPV.The recgulation of apoptosis by oncogene has been relatel to carcinogenesis closely; therefore, the modulation of E6 on cellular apoptosis has become a hot research topic recently. Inactivation of the pro-apoptotic tumor suppressor p53 by E6 is an important mechanism by which E6promotes cell growth; it is expected that inactivation of p53 by E6 should lead to a reduction in cellular apoptosis,numerous studies showed that E6 could in fact sensitize cells to apoptosis. The molecular basis for apoptosis modulation by E6 is poorly understood. | 4 | Screening and identification of mimotope of gastric cancer associated antigen MGb1-Ag显示文摘AIM: Using a monoclonal antibody against gastric cancer antigen named MGbl to screen a phage-displayed random peptide library fused with coat protein plII in order to get some information on mimotopes.lV^37BODS: Through affinity enrichment and EUSA screening,positive clones of phages were amplified. 10 phage clones were selected after three rounds of biopanning and the ability of specific binding of the positive phage clones to MGb1-Ab were detected by ELISA assay (DNA sequencing was performed and the amino acid sequences were deduced)By blocking test, specificity of the mimic phage epitopes was identified.RESULTS: There were approximately 200 times of enrichment about the titer of bound phages after three rounds of biopanning procedures. DNA of 10 phage clones after the third biopanning was assayed and the result showed that the positive clones had a specific binding activity to MGbl-Ab and a weak ability of binding to control mAb or to mouse IgG. DNA sequencing of 10 phage clones was performed and the amino acid sequences were deduced.According to the homology of the amino acid sequences of the displayed peptides, most of the phage clones had motifs of H(x)Q or L(x)S. And these 10 phage clones could also partly inhibit the binding of MGbl-Ab to gastric cancer cell KATO-Ⅲ. The percentage of blocking was from (21.0±1.6) %to (39.0±2.7) %.CONCLUSION: Motifs of H(x)Q and L(x)S selected and identified show a high homology in the mimic epitopes of gastric cancer associated antigen. | 5 | Improvement of Soil Moisture Simulation in Eurasia by the Beijing Climate Center Climate System Model from CMIP5 to CMIP6显示文摘This study provides a comprehensive evaluation of historical surface soil moisture simulation(1979-2012)over Eurasia at annual and seasonal time scales between two medium-resolution versions of the Beijing Climate Center Climate System Model(BCC-CSM)—one that is currently participating in phase 6 of the Coupled Model Intercomparison Project(CMIP6),i.e.,BCC-CSM2-MR,and the other,BCC-CSM1.1m,which participated in CMIP5.We show that BCC-CSM2-MR is more skillful in reproducing the climate mean states and standard deviations of soil moisture,with pattern correlations increased and biases reduced significantly.BCC-CSM2-MR performs better in capturing the first two primary patterns of soil moisture anomalies,where the period of the corresponding time series is closer to that of reference data.Comparisons show that BCC-CSM2-MR performs at a high level among multiple models of CMIP6 in terms of centered pattern correlation and“amplitude of variation”(relative standard deviation).In general,the centered pattern correlation of BCC-CSM2-MR,ranging from 0.61 to 0.87,is higher than the multi-model mean of CMIP6,and the relative standard deviation is 0.75,which surmounts the overestimations in most of the CMIP6 models.Due to the vital role played by precipitation in land-atmosphere interaction,possible causes of the improvement of soil moisture simulation are further related to precipitation in BCC-CSM2-MR.The results indicate that a better description of the relationship between soil moisture and precipitation and a better reproduction of the climate mean precipitation by the model may result in the improved performance of soil moisture simulation. | Yinghan SANG Hong-Li REN Xueli SHI Xiaofeng XU Haishan CHEN | 2021 | Advances in Atmospheric Sciences2021,38,2: | 2 |
| 6 | AT_1 receptor in rostral ventrolateral medulla mediating blunted baroreceptor reflex in spontaneously hypertensive rats显示文摘AIM: To determine the role of AT1 receptor in the rostral ventrolateral medulla (RVLM) in mediating the blunted baroreceptor reflex in spontaneously hypertensive rats (SHR). METHODS: Intravenous injections of graded doses of phenylephrine (1, 5, 10, 20, and 40μg/kg) increased the blood pressure to elicit the baroreceptor reflex in both SHR and normotensive Wistar rats anesthetized with urethane. The baroreceptor reflex sensitivity (BRS) was determined before and after microinjection of Ang II, losartan, or AT1 receptor antisense oligodeoxynucleotides into the RVLM. AT1 receptor protein level in the RVLM was measured by Western blotting. RESULTS: The BRS was significantly decreased in SHR compared with normal rats. Bilateral microinjection of AT~ receptor antagonist losartan (250 nmol/h) into the RVLM partly reversed the blunted BRS in SHR, but had no significant effect on the BRS in normal rats. Ang II (1.5 nmol/h) significantly inhibited the BRS in normal rats, which was completely abolished by pretreatment with losartan. However, no significant change in the BRS was observed after microinjection of Ang Ⅱ into the RVLM in SHR. Bilateral microinjection of AT1 receptor antisense oligodeoxynucleotides (ASODN) into the RVLM partially recovered the blunted BRS in SHR after 3 h, but no significant change in the BRS was observed in normal rats. The AT1 receptor protein level significantly decreased after administration of ASODN. | 7 | Identification of microsatellites in cattle unigenes显示文摘To identify EST-SSR molecular markers, 41,986 cattle UniGene sequences from NCBI were mined for analyzing SSRs. A total of 1,831 SSRs were identified from 1,666 ESTs, which represented an average density of 19.88 kb per SSR. The frequency of EST-SSRs was 4.0%. The dinucleotide repeat motif was the most abundant SSR, accounting for 54%, followed by 22%, 13%, 7% and 4%, respec- tively, for tri-, hexa-, penta- and tetra-nucleotide repeats. Depending upon the length of the repeat unit, the length of microsatellites varied from 14 to 86 bp. Among the di- and tri-nucleotide repeats, AC/TG (57%) and AGC (12%) were the most abundant type. Annotation of EST-SSRs was also carried out.
