| 1 | Isolation and phylogenetic assignation of actinomycetes in the marine sediments from the Arctic Ocean显示文摘Actinomycetes in five marine sediments collected from the Arctic Ocean at depths of 43 to 3 050 m were cultivated using a variety of media. A total of 61 actinomycete colonies with substrate mycelia only were observed, and no colonies with aerial mycelia were observed under aerobic conditions at 15 ℃. From these colonies, 28 were selected to represent different morphological types.Denaturing gradient gel electrophoresis(DGGE) was used to check the purity of isolates and select representatives for subsequent sequencing. Phylogentic analyses based on nearly full-length 16S ribosomal RNA gene (rDNA) sequences indicated that the actinomycetes isolated were accommodated within genus Rhodococcus of family Nocardiaceae, genus Dietzia of family Dietziaceae, genera Janibacter and Terrabacter of family Instrasporangiaceae and genera Kocuria and Arthrobacter of family Micrococcaceae. One of the strains(P27-24) from the deep-sea sediment at depth of 3 050 m was found to be identical in 16S rDNA sequence(1474/1474)with the radiation-resistant Kocuria rosea ATCC 187T isolated from air. More than half of the isolates showed the similarities ranging from 99.5% to 99.9% in 16S rDNA sequence to dibenzofran-degrading, butyl 2-ethylhexanoate-hydrolysising and nitrile-metabolizing actinomycetes. All the strains isolated were psychrotolerant bacteria and grew better on the media prepared with natural seawater than on the media prepared with deionized water. Three of them (Dietzia sp. P27-10, Rhodococcus sp. | 2 | Inhibitory effect of Salicornia europaea on the marine alga Skeletonema costatum显示文摘Exploiting the negative biochemical interference between plants and algal species has been suggested as a method to control harmful algal blooms.In this work,we investigated the inhibitory effect of the salt marsh halophyte Salicornia europaea against the marine alga Skeletonema costatum.S.europaea suppressed the growth of S.costatum in a nutrient-sufficient co-culture system,indicating that the inhibition of algal growth was because of the phytotoxic effect of S.europaea,rather than nutrient competition.We tested aqueous and organic extracts from S.europaea roots against S.costatum.The organic extracts inhibited growth and affected the cell size and chlorophyll a content of S.costatum in a dose-dependent manner.Among the three tested organic extracts,the methanol extract had the greatest effects on S.costatum,followed by butanol extract,and then the chloroform extract.Two flavonoids,rutin and quercetin-3-D-glucoside,were identified in the methanol extract by high performance liquid chromatography.The concentration of rutin was much higher than that of quercetin-3-D-glucoside.In an algal bioassay,rutin inhibited the growth of S.costatum and the inhibitory effect increased with increasing rutin concentration and with decreasing initial algal density.Therefore,we concluded that S.europaea negatively affects the growth of S.costatum,and that rutin,a metabolite of S.europaea,may play a role in this inhibitory effect. | JIANG Dan HUANG LingFeng LIN YongQing NIE LingLing LV SuLian KUANG TingYun LI YinXin | 2012 | Science China(Life Sciences)2012,55,6: | 8 |
| 3 | Molecular genetic diversity of bacteria in the bottom section of arctic sea ice from the Canada Basin显示文摘PCR-DGGE approach was used to analyze bacterial diversity in the bottom section of seven arctic sea ice samples colleted from the Canada Basin. Thirty-two 16S rDNA sequences were obtained from prominent DGGE bands. The closest relatives of these sequences are found to be those of cultivated or uncultured bacteria from antarctic or arctic sea ice. Phylogenetic analysis clustered these sequences or phylotypes within α- proteobacteria, γ-proteobacteria and CFB (cytophaga-flexibacter-bacteroides) group. Sequences belonging to γ-proteobacteria were dominant and members of the CFB group were highly abundant. | 4 | Interaction of Restin with transcription factors显示文摘Restin, a member of melanoma-associated antigen superfamily gene, was first cloned from differentiated leukemia cell induced by all trans-retinoic acid, and was able to inhibit cell proliferation, but the molecular mechanism was not clear. Since Restin was localized in cell nucleus, and its homolog member, Necdin (neuronal growth suppressor factor), could interact with transcription factors p53 and E2F1, we proposed that Restin might also function as Necdin through interacting with some transcription factors. In this study, transcription factors p53, AP1, ATFs and E2Fs were cloned and used in the mammalian two-hybrid system to identify their in- teraction with Restin. The results showed that only ATF3 had a strong interaction with Restin. It is interesting to know that ATF3 was an important transcription factor for G1 cell cycle initiation in physiological stress response. | 5 | Isolation and characteristics of one marine psychrotrophic cellulase-generating bacterium Ar/w/b/75°/10/5 from Chuckchi Sea,Arctic显示文摘Microorganisms living in polar zones play an important part as the potential source of organic activity materials with low temperature characteristics in the biotechnological applications. A psychrotrophic bacterium (strain Ar/w/b/75°/10/5), producing cellulase at low temperatures during late-exponential and early-stationary phases of cell growth, was isolated from sea ice-covered surface water in Chuckchi Sea, Arctic. This bacterium, with rod cells, was Gram-negative, slightly halophilic. Colony growing on agar plate was in black. Optimum growth temperature was 15℃. No cell growth was observed at 35℃ or above. Optimum salt concentration for cell growth was between 2 and 3 % of sodium chloride in media. Maximal cellulase activity was detected at a temperature of 35℃ and pH8. Cellulase was irreversibly inactivated when incubated at 55℃ within 30 min. Enzyme can be kept stable at the temperature no higher than 25℃. | 6 | Phylogenetic diversity of dimethylsulfoniopropionate-dependent demethylase gene dmdA in distantly related bacteria isolated from Arctic and Antarctic marine environments显示文摘Dimethylsulfoniopropionate(DMSP) is mainly produced by marine phytoplankton as an osmolyte, antioxidant,predator deterrent, or cryoprotectant. DMSP is also an important carbon and sulfur source for marine bacteria.Bacteria may metabolize DMSP via the demethylation pathway involving the DMSP demethylase gene(dmdA) or the cleavage pathway involving several different DMSP lyase genes. Most DMSP released into seawater is degraded by bacteria via demethylation. To test a hypothesis that the high gene frequency of dmdA among major marine taxa results in part from horizontal gene transfer(HGT) events, a total of thirty-one bacterial strains were isolated from Arctic Kongsfjorden seawater in this study. Analysis of 16S rRNA gene sequences showed that,except for strains BSw22118, BSw22131 and BSw22132 belonging to the genera Colwellia, Pseudomonas and Glaciecola, respectively, all bacteria fell into the genus Pseudoalteromonas. DmdA genes were detected in five distantly related bacterial strains, including four Arctic strains(Pseudoalteromonas sp. BSw22112, Colwellia sp.BSw22118, Pseudomonas sp. BSw22131 and Glaciecola sp. BSw22132) and one Antarctic strain(Roseicitreum antarcticum ZS2–28). Their dmdA genes showed significant similarities(97.7%–98.3%) to that of Ruegeria pomeroyi DSS–3, which was originally isolated from temperate coastal seawater. In addition, the sequence of the gene transfer agent(GTA) capsid protein gene(g5) detected in Antarctic strain ZS2–28 exhibited a genetically closely related to that of Ruegeria pomeroyi DSS–3. Among the five tested strains, only Pseudomonas sp. BSw22131 could grow using DMSP as the sole carbon source. | 7 | Diversity of the aerobic anoxygenic phototrophy gene pufM in Arctic and Antarctic coastal seawaters显示文摘Aerobic anoxygenic phototrophic(AAP) bacteria serve important functions in marine carbon and energy cycling because of their capability to utilize dissolved organic substrates and harvest light energy.AAP bacteria are widely distributed in marine environments,and their diversity has been examined in marine habitats.However,information about AAP bacteria at high latitudes remains insufficient to date.Therefore,this study determined the summer AAP bacterial diversity in Arctic Kongsfjorden and in the Antarctic coastal seawater of King George Island on the basis of puf M,a gene that encodes a pigment-binding protein subunit of the reaction center complex.Four puf M clone libraries were constructed,and 674 positive clones were obtained from four investigated stations(two in Kongsfjorden and two in the Antarctic Maxwell Bay).Arctic clones were clustered within the Alphaproteobacteria,whereas Antarctic clones were classified into the Alphaproteobacteria and Betaproteobacteria classes.Rhodobacteraceae-like puf M genes dominated in all samples.In addition,sequences closely related to puf M encoded on a plasmid in Sulfitobacter guttiformis were predominant in both Arctic and Antarctic samples.This result indicates the transpolar or even global distribution of puf M genes in marine environments.Meanwhile,differences between the Arctic and Antarctic sequences may prove polar endemism.These results indicate the important role of Rhodobacteraceae as AAP bacteria in bipolar coastal waters. | ZENG Yinxin DONG Peiyan QIAO Zongyun ZHENG Tianling | 2016 | Acta Oceanologica Sinica2016,35,6: | 3 |

| 12 | Human activity during the late Pleistocene in the Lop Nur region, northwest China: Evidence from a buried stone artifact显示文摘The Lop Nur region, in the east part of Tarim Basin, was an important transportation junction between west and east,north and south Eurasia. However, previous studies on prehistoric human activity have concentrated mostly on the Bronze Age,whereas that during the Stone Age remains largely unresearched. Here, we present a new direct evidence of human activity in the late Pleistocene, recorded on a grinding stone buried in a lacustrine sediment section of the Lop Nur region. The grain size distribution of the sediment section indicates that the site was probably in the center of a lake with weak hydrodynamic environment. Therefore, the stone artifact can only be carried to here by people instead of river and it was never move as soon as left here. Results of radiocarbon dating, the evident stratigraphic relations between the stone artifact and the sedimentary formation, indicate that the human activity could extend to approximately 13 ka BP. Furthermore, the results of starch-grain and use-wear analyses suggest that ancient humans gathered seeds of Triticeae, roots, and tubers and used the grinding stone to simply process selected plant as plant foodstuffs during this period. | 19 | Phylogenetic diversity and phenotypic characterization of cultivable bacterioplankton isolated from polar oceans显示文摘A set of 27 marine planktonic bacteria isolated from the polar regions was characterized by 16S rDNA sequencing and physiological and biochemical testing. More than half of these bacteria were positive for caseinase, gelatinase and β-glucosidase, and could utilize glucose, maltose or malic acid as carbon source for cell growth. Twelve isolates expressed nitrate reduction activities. Except for one antarctic isolate BSw10175 belonging to Actinobacteria phylum, these isolates were classified as γ-Proteobacteria, suggesting that γ-Proteobacteria dominated in cultivable marine bacterioplankton at both poles. Genus Pseudoalteromonas was the predominant group in the Chukchi Sea and the Bering Sea, and genus Shewanella dominated in cultivable bacterioplankton in the Prydz Bay. With sequence similarities above 97%, genus Psychrobacter was found at both poles.
