2Simultaneous Detection of 13 Key Bacterial Respiratory Pathogens by Combination of Multiplex PCR and Capillary Electrophoresis显示文摘Objective Lower respiratory tract infections continue to pose a significant threat to human health. It is important to accurately and rapidly detect respiratory bacteria. To compensate for the limits of current respiratory bacteria detection methods, we developed a combination of multiplex polymerase chain reaction(PCR) and capillary electrophoresis(MPCE) assay to detect thirteen bacterial pathogens responsible for lower respiratory tract infections, including Streptococcus pneumoniae, Haemophilus influenzae, Moraxella catarrhalis, Pseudomonas aeruginosa, Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, Mycoplasma pneumoniae, Legionella spp., Bordetella pertussis, Mycobacterium tuberculosis complex, Corynebacterium diphtheriae, and Streptococcus pyogenes. Methods Three multiplex PCR reactions were built, and the products were analyzed by capillary electrophoresis using the high-throughput DNA analyzer. The specificity of the MPCE assay was examined and the detection limit was evaluated using DNA samples from each bacterial strain and the simulative samples of each strain. This assay was further evaluated using 152 clinical specimens and compared with real-time PCR reactions. For this assay, three nested-multiplex-PCRs were used to detect these clinical specimens. Results The detection limits of the MPCE assay for the 13 pathogens were very low and ranged from 10^(-7) to 10^(-2) ng/μL. Furthermore, analysis of the 152 clinical specimens yielded a specificity ranging from 96.5%-100.0%, and a sensitivity of 100.0% for the 13 pathogens. Conclusion This study revealed that the MPCE assay is a rapid, reliable, and high-throughput method with high specificity and sensitivity. 3Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 6Hierarchical Ti_(3)C_(2)T_(x)@ ZnO Hollow Spheres with Excellent Microwave Absorption Inspired by the Visual Phenomenon of Eyeless Urchins显示文摘Ingenious microstructure design and rational composition selection are effective approaches to realize high-performance microwave absorbers,and the advancement of biomimetic manufacturing provides a new strategy.In nature,urchins are the animals without eyes but can“see”,because their special structure composed of regular spines and spherical photosensitive bodies“amplifies”the light-receiving ability.Herein,inspired by the above phenomenon,the biomimetic urchin-like Ti_(3)C_(2)T_(x)@ZnO hollow microspheres are rationally designed and fabricated,in which ZnO nanoarrays(length:~2.3μm,diameter:~100 nm)as the urchin spines are evenly grafted onto the surface of the Ti_(3)C_(2)T_(x) hollow spheres(diameter:~4.2μm)as the urchin spherical photosensitive bodies.The construction of gradient impedance and hierarchical heterostructures enhance the attenuation of incident electromagnetic waves.And the EMW loss behavior is further revealed by limited integral simulation calculations,which fully highlights the advantages of the urchin-like architecture.As a result,the Ti_(3)C_(2)T_(x)@ZnO hollow spheres deliver a strong reflection loss of−57.4 dB and broad effective absorption bandwidth of 6.56 GHz,superior to similar absorbents.This work provides a new biomimetic strategy for the design and manufacturing of advanced microwave absorbers.Yan‑Qin Wang Hai‑Bo Zhao Jin‑Bo Cheng Bo‑Wen Liu Qiang Fu Yu‑Zhong Wang 2022Nano-Micro Letters2022,14,5:5
7Characterization of Avian Influenza A(H7N9) Virus Prevalence in Humans and Poultry in Huai′an,China:Molecular Epidemiology,Phylogenetic,and Dynamics Analyses显示文摘Objective To trace the source of human H7N9 cases in Huai'an and elucidate the genetic characterization of Huai'an strains associated with both humans and birds in live poultry market.Methods An enhanced surveillance was implemented when the first human H7N9 case was confirmed in Huai'an.Clinical specimens,cloacal swabs,and fecal samples were collected and screened by real-time reverse transcription-polymerase chain reaction(RT-PCR) for H7N9 virus.The positive samples were subjected to further RT-PCR and genome sequencing.The phylodynamic patterns of H7N9 virus within and separated from Huai'an and evolutionary dynamics of the virus were analyzed.Results Six patients with H7N9 infection were previously exposed to live poultry market and presented symptoms such as fever(>38.0 °C) and headaches.Results of this study support the hypothesis that live poultry markets were the source of human H7N9 exposure.Phylogenetic analysis revealed that all novel H7N9 viruses,including Huai'an strains,could be classified into two distinct clades,A and B.Additionally,the diversified H7N9 virus circulated in live poultry markets in Huai'an.Interestingly,the common ancestors of the Huai'an H7N9 virus existed in January 2012.The mean nucleotide substitution rates for each gene segment of the H7N9 virus were(3.09-7.26)×10-3 substitutions/site per year(95% HPD:1.72×10-3 to 1.16×10-2).Conclusion Overall,the source of exposure of human H7N9 cases in Huai'an was live poultry market,and our study highlights the presence of divergent genetic lineage of H7N9 virus in both humans and poultry specimens in Huai'an.YANG Peng Fei YAN Qing Li LIU Chun Cheng XING Ya Dong ZHANG Min Hui GAO Qiang YU Hao YAO Hai Bo HE Nan Jiang 2016Biomedical and Environmental Sciences2016,29,10:3

13Construction of the Subtracted cDNA Library of Striatal Neurons Treated with Long-term Morphine显示文摘Objective To construct a morphine tolerance model in primarily cultured striatal neurons,and screen the differentially expressed genes in this model using suppression subtractive hybridization (SSH).Methods Sbtracted cDNA libraries were constructed using SSH from normal primarily cultured striatal neurons and long-term morphine treated striatal neurons (10-5 mol/L for 72 hours).To check reliability of the cell culture model,RT-PCR was performed to detect the cAMP-responsive element-binding protein (CREB) mRNA expression.The subtracted clones were prescreened by PCR.The clones containing inserted fragments from forward libraries were sequenced and submitted to GenBank for homology analysis.And the expression levels of genes of interest were confirmed by RT-PCR.Results CREB mRNA expression showed a significant increase in morphine treated striatal neurons (62.85±1.98) compared with normal striatal neurons (28.43±1.46,P<0.01).Thirty-six clones containing inserted fragments were randomly chosen for sequence analysis.And the 36 clones showed homology with 19 known genes and 2 novel genes.The expression of 2 novel genes,mitochondrial carrier homolog 1 (Mtch1;96.81±2.04 vs.44.20±1.31,P<0.01) and thymoma viral proto-oncogene 1 (Akt1;122.10±2.17 vs.50.11±2.01,P<0.01),showed a significant increase in morphine-treated striatal neurons compared with normal striatal neurons.Conclusions A reliable differential cDNA library of striatal neurons treated with long-term morphine is constructed.Mtch1 and Akt1 might be the candidate genes for the development of morphine tolerance.Bo Bai Hai-qing Liu Jing Chen Ya-lin Li Hui Du Hai Lu Peng-li Yu 2011Chinese Medical Sciences Journal2011,26,1:1

18Synthesis of A Natural Cytotoxic Alkaloid Artabotrine and its Analogue显示文摘Artabotrine 1 is a novel N-methoxylated 4,5-dioxoaporphine alkaloid which has been reported to be cytotoxic against P-388 cells. The total synthesis of 1 was carried out, and one of its analogue, N-methoxycepharadione B 2, was also synthesized.
