| 1 | Roux-en-Y gastric bypass promotes expression of PDX-1 and regeneration of β-cells in Goto-Kakizaki rats显示文摘AIM:To study the effects of Roux-en-Y gastric bypass(RYGB) on the expression of pancreatic duodenal homeobox-1(PDX-1) and pancreatic β-cell regeneration/neogenesis,and their possible mechanisms in diabetics.METHODS:Three groups of randomly selected nonobese diabetic Goto-Kakizaki(GK) rats were subjected to RYGB,sham-RYGB and sham-operation(sham-op) surgery,respectively.The rats were euthanized at postoperative 1,2,4 and 12 wk.Their pancreases were resected and analyzed using reverse transcription polymerase chain reaction to detect the mRNA of PDX-1.Anti-PDX-1 immunohistochemical(IHC) staining and Western blotting were used to detect the protein of PDX-1.Double IHC staining of anti-Brdu and-insulin was performed to detect regenerated β-cells.The index of double Brdu and insulin positive cells was calculated.RESULTS:In comparison with sham-RYGB and sham-op groups,a significant increase in the expressions of PDX-1 mRNA in RYGB group was observed at all experimental time points(1 wk:0.378 ± 0.013 vs 0.120 ± 0.010,0.100 ± 0.010,F = 727.717,P < 0.001;2 wk:0.318 ± 0.013 vs 0.110 ± 0.010,0.143 ± 0.015,F = 301.509,P < 0.001;4 wk:0.172 ± 0.011 vs 0.107 ± 0.012,0.090 ± 0.010,F = 64.297,P < 0.001;12 wk:0.140 ± 0.007 vs 0.120 ± 0.010,0.097 ± 0.015,F = 16.392,P < 0.001);PDX-1 protein in RYGB group was also increased significantly(1 wk:0.61 ± 0.01 vs 0.21 ± 0.01,0.15 ± 0.01,F = 3031.127,P < 0.001;2 wk:0.55 ± 0.00 vs 0.15 ± 0.01,0.17 ± 0.01,F = 3426.455,P < 0.001;4 wk:0.39 ± 0.01 vs 0.18 ± 0.01,0.22 ± 0.01,F = 882.909,P < 0.001;12 wk:0.41 ± 0.01 vs 0.20 ± 0.01,0.18 ± 0.01,F = 515.833,P < 0.001).PDX-1 mRNA and PDX-1 protein production showed no statistical significance between the two sham groups.Many PDX-1 positive cells could be found in the pancreatic islets of the rats in RYGB group at all time points.In addition,the percentage of Brdu-insulin double staining positive cells was higher in RYGB group than in the other two groups(1 wk:0.22 ± 0.13 vs 0.03 ± 0.06,0.03 ± 0.06,P < 0.05;2 wk:0.28 ± 0.08 vs 0.00 ± 0.00,0.03 ± 0.06,P < 0.05;4 wk:0.24 ± 0.11 vs 0.07 ± 0.06,0.00 ± 0.00,P < 0.001;12 wk:0.20 ± 0.07 vs 0.03 ± 0.06,0.00 ± 0.00,P < 0.05).CONCLUSION:RYGB can increase the expression of pancreatic PDX-1 and induce the regeneration of β-cells in GK rats.The associated regeneration of islet cells may be a possible mechanism that how RYGB could improve type 2 diabetes mellitus. | Zhen Li,Dong-Fei Li,Jing-Xing Dai,Yu Bai,Lin Yuan,Southern Medical University,Institute of Basic Medical Anatomy National Key Disciplines,Guangzhou 510515,Guangdong Province,China Hong-Ya Zhang,Lu-Xian Lv,Wen-Qiang Li,Second Affiliated Hospital,Xinxiang Medical College,Henan Province Key Laboratory of Biological Psychiatry,Xinxiang 453002,Henan Province,China Ou Sha,Department of Anatomy,Faculty of Medicine,Shenzhen University,Shenzhen 518060,Guangdong Province,China | 2010 | World Journal of Gastroenterology2010，16，18： | 14 |
| 2 | Relationship between osmotic stress and polyamines conjugated to the deoxyribonucleic acid-protein in wheat seedling roots显示文摘The contents of covalently conjugated polyamines (CC-PAs) and noncovalently conju- gated polyamines (NCC-PAs) to deoxyribonucleic acid-protein (DNP) isolated from wheat (Triticum aestivum L.) seedling roots under osmotic stress were detected. Results showed that after osmotic stress treatment for 7 d, the levels in NCC-spermine (NCC-Spm) and NCC-spermidine (NCC-Spd) of drought-tolerant Yumai No. 18 cv. increased more markedly than that of drought-sensitive Yangmai No. 9 cv., while the NCC-putrescine (NCC-Put) could not be statistically detected in two cultivars. Exogenous Spm treatment alleviated osmotic stress injury to Yangmai No. 9 cv. seedlings, coupled with marked increases of NCC-Spm and NCC-Spd levels of this cultivar. Under PEG osmotic stress, the concomitant treatment of drought-tolerant Yumai No. 18 cv.seedlings with methylglyoxyl-bis (guanylhydrazone) (MGBG), an inhibitor of S-adenosylmethionine decarboxylase (SAMDC), aggravated osmotic stress injury to this cultivar, coupled with market decreases of the NCC-Spm and NCC- Spd levels. The levels in CC-Put and CC-Spd of drought-tolerant Yumai No. 18 cv. increased more markedly than that of drought-sensitive Yangmai No. 9 cv. Under osmotic stress. The treatment of drought-tolerant Yumai No. 18 cv. seedlings with phenanthrolin (o-Phen), an inhibitor of transglutaminase (TGase), aggravated osmotic stress injury to this cultivar, coupled with a reduction of sum contents of CC-Put+CC-Spd. | 4 | Object oriented land cover classification using ALS and GeoEye imagery over mining area显示文摘An object oriented coal mining land cover classification method based on semantically meaningful image segmentation and image combination of GeoEye imagery and airborne laser scanning (ALS) data was presented. First, DEM, DSM and nDSM (normalized Digital Surface Model, nDSM) were extracted from ALS data. The GeoEye imagery and DSM data were combined to create segmented objects based on neighbor regions merge method. Then 10 kinds of objects were extracted. Different kinds of vegetation objects, including crop, grass, shrub and tree, can be extracted by using NDVI and height value of nDSM. Water and coal pile field was extracted by using NDWI and the standard deviation of DSM method. Height differences also can be used to distinguish buildings from road and vacant land, and accurate building contour information can be extracted by using relationship of neighbor objects and morphological method. | 6 | AI_4C_3 Hydration Thermochemical Analysis for Burned Carbon-containing Refractories with Al显示文摘In this paper, X-ray diffractogram analysis and SEM observation of Al4 C3 formed at high temperature from carbon-containing refractories with Al have been carried out.Aluminum added to carbon-containing refractories reacts with C(s)to form Al4 C3(s) gradually during heating from 600 ℃ to 1200℃.It is considered that the interlocked structure of Al4 C3 plate crystals promotes the outstanding increase of hot modulus of rupture of carbon-containing refractories with Al. The HMOR of carbon-containing refractories added with Al additive from 0 to 5wt% increases by 2.8 times being from 6.5MPa to 18.2MPa.After a thermochemical calculation for hydration reaction processes of Al4 C3 and H2O(g), the equilibrium partial pressure chart of H2O(g)in H2O-Al4C3-Al(OH)3 system vs various temperatures has been attained . | 9 | Geochronology and Geochemical Characteristics of Intrusion in the Jinchangliang Gold Deposit,Inner Mongolia and Their Tectonic Significance显示文摘Discovered and mined in recent years, the Jinchangliang gold deposit has not yet been studied in its genetic type. In this paper, the geological features of ore deposit, S isotopic composition, metallogenic age and elements geochemical of the granite closely related to mineralization were discussed. The results of the geological features of ore deposit and S isotopic composition show that ore-bearing hydrothermal solution was closely related with the intrusion of magmatic. The granite is characteristic of high silica SiO_2=72.38%–72.98%, high aluminum and Al_2O_3=14.22%–14.35%, low calcium CaO=0.16% –0.26%, and low value of FeOT/MgO(6.86–7.73), and rich in alkalis Na_2O+K_2O=9.11%–9.24%, suggesting that it is high-K calc-alkaline, highly fractionated, weak aluminum A-type granite. The REE patterns are inclined to right and show intense fractionation between LREE and HREE, without obvious negative Eu anomaly(δEu=0.80–0.84). The primitive mantle-normalized spidergrams are characterized by depletion of Ba, U, Ta, Nb, Zr, Ti and P, which implies that the granite has the characteristics of the crust-mantle mixing. S isotopes also indicate that the material source of gold deposit is closely related to the granite rocks. The LA-ICP-MS Zircon UPb age of the Damiao rock mass medium-fine grained monzogranite(belonging to the early IndoChina) is(245±1) Ma. It shows that Jinchangliang gold deposit was not formed in Yanshanian, but the early Indo-China. | 12 | Fluorescence and Photosensitizing Behavior of N-(p-Phenoxyphenyl)methacrylamide and Its Polymer显示文摘A novel acrylic monomer bearing aromatic ethereality group N-(p -phenoxy-phenyl) methacrylamide(MAPOA) was synthesized and polymerized. The fluorescence spectra of MAPOA and its polymer were studied, showing that the polymer displays a stronger fluorescence than its monomer at the same chromophore concentration. The fluorescence of P(MAPOA) was quenched by adding electron deficiency monomers such as MMA, AN etc .. It is another example of showing the 'fluorescence structural self-quenching effect' as we reported previously, from which it is demonstrated again that this phenomenon is not an accidental one but a common effect for acrylic monomers bearing electon-donating chromophores. The photopolymerization of AN sensitized by MAPOA and P(MAPOA) was studied. The kinetic equation of the polymerization was obtained and represented as follows: R _p∝ 0.38 × 0.78 , R _p∝ 0.33 × 0.71 The overall activation energies were determined to be E_ a =24\^22 kJ/mol(MAPOA) and E_ a =26\^31 kJ/mol respectively. The fluorescence analysis of the resulted P(AN) showed that the sensitizer also entered into the P(AN) chains.
