| 1 | Efficient production of omega-3 fatty acid desaturase (sFat-1)-transgenic pigs by somatic cell nuclear transfer显示文摘Omega-3(ω-3) fatty acid desaturase transgenic pigs may improve carcass fatty acid composition. The use of transgenic pigs is also an excellent large animal model for studying the role of ω-3 fatty acids in the prevention and treatment of coronary heart disease and cancer. Transgenic pigs carrying synthesized fatty acid desaturase-1 gene (sFat-1) from Caenorhabditis briggsae by somatic cell nuclear transfer (SCNT) were produced for the first time in China. Porcine fetal fibroblast cells were transfected with a sFat-1 expression cassette by the liposome-mediated method. Transgenic embryos were reconstructed by nuclear transfer of positive cells into enucleated in vitro matured oocytes. A total of 1889 reconstructed embryos were transferred into 10 naturally cycling gilts. Nine early pregnancies were established, 7 of which went to term. Twenty-one piglets were born. The cloning efficiency was 1.1% (born piglets/transferred embryos). The integration of the sFat-1 gene was confirmed in 15 live cloned piglets by PCR and Southern blot except for 2 piglets. Expression of the sFat-1 gene in 12 of 13 piglets was detected with RT-PCR. | 3 | Distribution of Pacific-origin water in the region of the Chukchi Plateau in the Arctic Ocean in the summer of 2003显示文摘The upper ocean thermohaline structures in the region of the Chukchi Plateau are analyzed with the hydrographic data collected by the Chinese National Arctic Research Expedition in the summer of 2003. Three types of the Pacific-origin water were found in the upper ocean, Alaska Coastal Water (ACW), summer Bering Sea Water (sBSW) and winter Bering Sea Water (wBSW) are indicated by two maximums temperature and one minimum temperature, piling up from the upper to the lower respectively. The extreme warm ACW with a maximum temperature of 1.62 ℃ was found in the southwestern Canada Basin at a depth of about 50 m. A pathway of the ACW into the basin from its adjacent area did not existed in the expedition period. So it is speculated that the extreme warm feature of the ACW was formed locally in 2003. The relative weak ACW occurred to the east of the Chukchi Cap and in the southern Chukchi Abyssal Plain. The latter one might originate from a warm downwelling that existed in a small canyon at the shelf break of the Chukchi Sea. The sBSW without the ACW was found only at the southwestern flank of the Chukchi Cap. The ACW and the sBSW were not found in the northernmost station at 81°N,which indicates the north boundary of the upper Pacific-origin water in the Canada Basin. The wBSW, which existed in all deep stations,was exactly uniform at temperature. | 5 | Cyclooxygenase-2 promotes angiogenesis by increasing vascular endothelial growth factor and predicts prognosis in gallbladder carcinoma显示文摘AIM: To investigate the relationships between theexpression of cydooxygenase-2 (COX-2), vascular endothelial growth factor (VEGF) and the degree of vascularization, clinicopathologic feature, survival time of patients with gallbladder carcinomas.METHODS: Sixty-four gallbladder carcinoma specimens were evaluated for COX-2, VEGF expression by immunohistochemical methods. Microvessel counts (MVC) were determined using CD34. The relationships between COX-2,VEGF expression, CD34-stained MVC, clinicopathologic features and survival time were analyzed. The correlations between COX-2 and VEGF expression, CD34-stained MVC were also investigated.RESULTS: COX-2, VFGF immunoreactivity were observed in 71.9% (46/64) and 54.7% (35/64) specimens,respectively. The average MVC in 64 cases of gallbladder carcinoma was 57±14 per high power vision field. The status of MVC was closely correlated with Nevin staging, tumor differentiation and lymph node metastasis (P<0.01,0.002, and 0.003, 0.000, respectively). Increased VEGF expression was significantly correlated with tumor differentiation (poorly and moderately>well differentiated, P<0.05, P = 0.016). Clinical stages had no relation with the expression of VEGF (P>0.05, P = 0.612). There was a positive correlation between COX-2 expression and clinical stages. The positive rate of COX-2 was higher in cases of Nevin stages S4-S5 (81.8%) than in those of Nevin stages S1-S3 (50.0%) with a statistical significance (P<0.01, P = 0.009). The expression of COX-2 did not vary with differentiation (P>0.05, P = 0.067). Statistically significant differences were also observed according to lymph node metastasis, COX-2 expression and VEGF expression(P<0.01, 0.000, and 0.001, respectively). There was no relation between VEGF, COX-2 expression, MVC and the age and sex of patients. MVC and VEGF positive rate in the COX-2 positive gallbladder carcinoma tissue was higher than that in the COX-2 negative tissue (P<0.05, 0.000, and 0.032, respectively). Patients with VEGF, COX-2 positive tumors had a significantly shorter survival time than those with negative tumors (P<0.05, 0.004, 0.01, respectively).CONCLUSION: Augmented tumor neovascularization induced by VEGF may be one of the several effects of COX-2 responsible for poor prognosis of human gallbladder carcinoma. | 6 | Spatiotemporal patterns of the fish assemblages downstream of the Gezhouba Dam on the Yangtze River显示文摘An explicit demonstration of the changes in fish assemblages is required to reveal the influence of damming on fish species.However,information from which to draw general conclusions regarding changes in fish assemblages is insufficient because of the limitations of available approaches.We used a combination of acoustic surveys,gillnet sampling,and geostatistical simulations to document the spatiotemporal variations in the fish assemblages downstream of the Gezhouba Dam,before and after the third impoundment of Three Gorges Reservoir(TGR).To conduct a hydroacoustic identification of individual species,we matched the size distributions of the fishes captured by gillnet with those of the acoustic surveys.An optimum threshold of target strength of 50 dB re 1 m 2 was defined,and acoustic surveys were purposefully extended to the selected fish assemblages(i.e.,endemic Coreius species) that was acquired by the size and species selectivity of the gillnet sampling.The relative proportion of fish species in acoustic surveys was allocated based on the composition(%) of the harvest in the gillnet surveys.Geostatistical simulations were likewise used to generate spatial patterns of fish distribution,and to determine the absolute abundance of the selected fish assemblages.We observed both the species composition and the spatial distribution of the selected fish assemblages changed significantly after implementation of new flow regulation in the TGR,wherein an immediate sharp population decline in the Coreius occurred.Our results strongly suggested that the new flow regulation in the TGR impoundment adversely affected downstream fish species,particularly the endemic Coreius species.To determine the factors responsible for the decline,we associated the variation in the fish assemblage patterns with changes in the environment and determined that substrate erosion resulting from trapping practices in the TGR likely played a key role. | TAO JiangPing GONG YuTian TAN XiChang YANG Zhi CHANG JianBo | 2012 | Science China(Life Sciences)2012,55,7: | 12 |
| 7 | Water mass of the northward throughflow in the Bering Strait in the summer of 2003显示文摘The temperature and salinity data obtained by the Chinese national arctic research expedition (CHINARE2003) are used to study the water structure in the Bering Strait and ambient regions. Four water masses appeared in the research region: the intermediate Bering Sea water mass (IBWM), the Alaska coastal water (ACW), the Anadyr water (AW) and the Bering shelf water (BSW). The AW originates from the IBWM, but the upper layer water has been greatly altered. In the cruise on 28/29 July 2003, there were only the BSW and ACW in a section across the Bering Strait (BS section), but in the September 12/13 cruise, the AW, BSW and ACW flowed parallelly into the Bering Strait. The upper waters of these water masses were all altered due to ice melting, runoff, solar radiation, and wind mixing. The waters in the central and northern parts of Bering Strait stratified by two uniform layers,were expressed as the typical feature of the water masses originating from the pacific. A two-layer structure also dominated the vertical stratification in most part of the Chukchi Sea. | 8 | IL-10-Producing Type 1 Regulatory T Cells and Allergy显示文摘As an important subset of regulatory T(Treg) cells,IL-10-producing type 1 regulatory T cells(Tr1) ,have some different features to thymic-derived naturally occurring CD4+CD25+Foxp3+ Treg cells(nTreg cells) . Similar to nTreg cells,Tr1 also play important roles in the control of allergic inflammation in several ways. There is a fine balance between Tr1 and Th2 responses in healthy subjects. Skewing of allergic-specific effctor T cells to a Tr1 phenotype appears to be a critical event in successful allergen-specific immunotherapy and glucocorticoids and β2-agonists treatment. Tr1 suppress Th2 cells and effector cells of allergic inflammation,such as eosinophils,mast cells,basophils,through producing IL-10,and perhaps TGF-β. | 10 | Green fluorescent protein (GFP) transgenic pig produced by somatic cell nuclear transfer显示文摘Transgenic somatic cell nuclear transfer is a very promising route for producing transgenic farm ani-mals. Research on GFP transgenic pigs can provide useful information for breeding transgenic pigs, human disease models and human organ xenotransplantation. In this study, a liposomal transfection system was screened and transgenic embryos were reconstructed by nuclear transfer of GFP positive cells into enucleated in vitro matured oocytes. The development of reconstructed embryos both in vitro and in vivo was observed, and GFP expression was determined. The results showed that porcine fe-tal-derived fibroblast cells cultured with 4.0 μL/mL liposome and 1.6 μg/mL plasmid DNA for 6 h re-sulted in the highest transfection rate (3.6%). The percentage of GFP reconstructed embryos that de-veloped in vitro to the blastocyst stage was 10%. Of those the GFP positive percentage was 48%. Re-constructed transgenic embryos were transferred to 10 recipients. 5 of them were pregnant, and 3 de-livered 6 cloned piglets in which 4 piglets were transgenic for the GFP as verified by both GFP protein expression and GFP DNA sequence analysis. The percentage of reconstructed embryos that resulted in cloned piglets was 1.0%; while the percentage of piglets that were transgenic was 0.7%. | 11 | Investigation of MHD power generation with supersonic non-equilibrium RF discharge显示文摘Magnetohydrodynamic(MHD) power generation with supersonic non-equilibrium plasma is demonstrated. Capacitively coupled radio frequency(RF) discharge(6 MHz, maximum continual power output of 200 W) was adopted to ionize the Mach number 3.5(650 m/s),0.023 kg/m3 airflow. In a MHD channel of 16 mm*10mm*20 mm, MHD open voltage of 10 V is realized in the magnetic field of 1.25 T, and power of 0.12 m W is extracted steadily and continuously in the magnetic field of 1 T. The reasons for limited power generation are proposed as:low conductivity of RF discharge; large touch resistance between MHD electrode and plasma;strong current eddies due to flow boundary layer. | 13 | Correlation of P-glycoprotein expression with poor vascularization in human gallbladder carcinomas显示文摘AIM: To investigate the relationship between the expression of P-glycoprotein (P-gp) and the degree of vascularization in gallbladder carcinomas.METHODS: P-gp was stained with streptavidin-peroxidase complex immunohistochemical method in routine paraffinembedded sections of gallbladder carcinomas. Microvessel counts (MVC) were determined using factor-Ⅷ-related antigens.RESULTS: The average MVC in 32 cases of gallbladder carcinomas was (34±10)/HP. The value of MVC was closely correlated with Nevin staging and tumor differentiation (P<0.01 and P<0.05). The total expression rate of P-gp was 62.5 %. The P-gp expression rate in cases of Nevin staging S1-S3 (78.6 %) was higher than that of S4-S5 (50.0 %) with no statistical significance. The P-gp expression rate was not correlated with tumor differentiation or pathologic types. The value of MVC in P-gp (+) cases was markedly lower than that in P-gp (-) cases (P<0.01). The positive rate of P-gp was significantly higher in cases of smaller MVC than those of bigger MVC (P<0.05).CONCLUSION: MVC may be used as one of the important parameters to reflect the biological behaviors of gallbladder carcinomas. | 14 | Relationship between P-glycoprotein and CD44 expression in esophageal carcinoma显示文摘Objective: To investigate the relationship between P-glycoprotein (P-gp) and adhesion molecule CD44 expression as well as their clinical significance in esophageal carcinoma. Methods: To examine the expressed level of P-gp and CD44 by flow cytometry (FCM) in the operated samples of 70 cases with esophageal carcinoma and their normal mucosa of esophageal incision, and to evaluate their relationship with clinicopathological factors. Results: Among the 70 cases with esophageal car- cinoma, the expression of P-gp in the 27 cases (38.6%) was negative (positive cells <25%); 11 cases (15.7%) were 25%–40% expression of P-gp positive cells; 14 cases (20%) were 41%–60% expression of P-gp positive cells; 18 cases (25.7%) were the high expression (positive cells >60%) of P-gp. Of the cases with the tumor sizes being more than 4 cm, the expression of CD44 showed a significant difference (P<0.05) in 25 cases with P-gp positive, compared with 19 cases with P-gp negative. Of the cases with high-mild differentiated esophageal carcinoma, the expression of CD44 showed a significant difference (P<0.05) in 22 cases with P-gp positive, compared with 17 cases with P-gp negative. Of the cases with clinical III--IV stage, the expression of CD44 showed a significant difference (P<0.05) in 26 cases with P-gp positive, compared with 10 cases with P-gp negative. Of the cases with lymph node metastasis, the CD44 expression showed a significant difference (P=0.050) in 27 cases with P-gp positive, compared with 11 cases with P-gp negative. Of the cases of the patients’ age being more than 56 years, the expression of CD44 showed a significant difference (P<0.01) in 27 cases with P-gp positive, compared with 12 cases with P-gp negative. When the P-gp and CD44 expression were positive, the clinical II stage and III–IV stage in esophageal carcinoma was showed a significant difference (P<0.05). | 15 | Effects of Heating and Hot Extrusion Process on Microstructure and Properties of Inconel 625 Alloy显示文摘The effects of the heating process and hot extrusion on the microstructure and properties of inconel 625 alloy were studied. The experimental results showed that the properties of Inconel 625 alloy could be improved through the heating process and hot extrusion concomitant with a reduced corrosion rate. The M23C6 carbide, generated in the heating process, was retained and distributed at the grain boundary during the process of hot extrusion, which had an important influence on both elongation and corrosion resistance. The improvement of the comprehensive properties of the material, as measured by a tensile test at room temperature, was correlated with the dissolution of segregation Nb. A typical ductile fracture changed to a cleavage fracture where secondary cracks could be clearly seen. With the increase of the extrusion ratio, the real extrusion temperature was higher, which led to more dissolution of the M23C6 carbide, decreased the number of secondary cracks, enhanced the effect of solid solution strengthening, and reduced the intergranular corrosion rate. Under the condition of a high extrusion ratio and a high extrusion speed, the less extrusion time made it possible to obtain organization with a smaller average grain size. Moreover, in this case, the M23C6 carbide and segregated Nb did not have enough time to diffuse. | 16 | Surface Heat Budget and Solar Radiation Allocation at a Melt Pond During Summer in the Central Arctic Ocean显示文摘The heat budget of a melt pond surface and the solar radiation allocation at the melt pond are studied using the 2010 Chinese National Arctic Research Expedition data collected in the central Arctic. Temperature at a melt pond surface is proportional to the air temperature above it. However, the linear relationship between the two varies, depending on whether the air temperature is higher or lower than 0℃. The melt pond surface temperature is strongly influenced by the air temperature when the latter is lower than 0℃. Both net longwave radiation and turbulent heat flux can cause energy loss in a melt pond, but the loss by the latter is larger than that by the former. The turbulent heat flux is more than twice the net longwave radiation when the air temperature is lower than 0℃. More than 50% of the radiation energy entering the pond surface is absorbed by pond water. The reinforcement particles were mainly γ Al 2O 3, some θ Al 2O 3 and a little α Al 2O 3. Thermodynamics analyses show that the chemical reactions are 3Cu 2O+2Al=6Cu+Al 2O 3 or 3CuO+2Al=3Cu+Al 2O 3. A related equilibrium diagram was drawn. In situ techniques involve a chemical reaction resulting in the formation of a very fine and thermodynamically stable reinforcing ceramic phase within a metal matrix.
