| 1 | Calibration procedures for the CHASE/HIS science data显示文摘The Hα line is an important optical line in solar observations containing the information from the photosphere to the chromosphere. To study the mechanisms of solar eruptions and the plasma dynamics in the lower atmosphere, the Chinese Hα Solar Explorer(CHASE) was launched into a Sun-synchronous orbit on October 14, 2021. The scientific payload of the CHASE satellite is the Hα Imaging Spectrograph(HIS). The CHASE/HIS acquires, for the first time, seeing-free Hα spectroscopic observations with high spectral and temporal resolutions. It consists of two observational modes. The raster scanning mode provides full-Sun or region-of-interest spectra at Hα(6559.7-6565.9 ?) and Fe I(6567.8-6570.6 ?) wavebands. The continuum imaging mode obtains full-Sun photospheric images at around 6689 ?. In this paper, we present detailed calibration procedures for the CHASE/HIS science data, including the dark-field and flat-field correction, slit image curvature correction, wavelength and intensity calibration, and coordinate transformation. | 2 | Enhancing single-cell hyaluronic acid biosynthesis by microbial morphology engineering显示文摘Microbial morphology engineering is a novel approach for cell factory to improve the titer of target product in bio-manufacture.Hyaluronic acid(HA),a valuable glycosaminoglycan polymerized by HA synthase(HAS),a membrane protein,is particularly selected as the model product to improve its single-cell HA-producing capacity via morphology engineering.DivIVA and FtsZ,the cell-elongation and cell division related protein,respectively,were both down/up dual regulated in C.glutamicum via weak promoter substitution or plasmid overexpression.Different from the natural short-rod shape,varied morphologies of engineered cells,i.e.small-ellipsoid-like(DivIVA-reduced),bulb-like(DivIVA-enhanced),long-rod(FtsZ-reduced)and dumbbell-like(FtsZ-enhanced),were observed.Applying these morphology-changed cells as hosts for HA production,the reduced expression of both DivIVA and FtsZ seriously inhibited normal cell growth;meanwhile,overexpression of DivIVA didn't show morphology changes,but overexpression of FtsZ surprisingly change the cell-shape into long and thick rod with remarkably enlarged single-cell surface area(more than 5.2-fold-increase).And finally,the single-cell HA-producing capacity of the FtsZ-overexpressed C.glutamicum was immensely improved by 13.5-folds.Flow cytometry analyses verified that the single-cell HAS amount on membrane was enhanced by 2.1 folds.This work is pretty valuable for high titer synthesis of diverse metabolic products with microbial cell factory. | Yukun Zheng Fangyu Cheng Bo Zheng Huimin Yu | 2020 | Synthetic and Systems Biotechnology2020,5,4: | 3 |
| 3 | Effects of 2, 4-Dichlorophenoxy Acetic Acid andLight on Growth of Gerbera (Gerbera jamesoniicv. Daxueju) Callus显示文摘This study was conducted to determine effects of 2,4-dichlorophenoxy acetic acid(2,4-D) and light on growth of gerbera(Gerbera jamesonii cv. Daxueju) callus. Callus was induced from both petiole and leaf explants of gerbera on Murashige and Skoog medium supplemented with 3% sucrose and various concentrations of 2,4-D and placed under light and dark. Callus induction percentage, callus size and callus fresh and dry weights were efficiently higher when using petiole as explant. MS medium supplemented with 1.5 mg/L 2,4-D showed the highest callus induction percentage of 96.70%. Callus induced under light had larger weight mass. | 4 | On the technologies of Hα imaging spectrograph for the CHASE mission显示文摘The Hα imaging spectrograph(HIS) is the scientific payload of the first solar space mission, the Chinese Hα solar explorer(CHASE), supported by the China National Space Administration(CNSA). The CHASE/HIS achieves, for the first time in space, Hα spectroscopic observations with high spectral and temporal resolutions. Separate channels for the raster scanning mode(RSM) and continuum imaging mode(CIM) are integrated into one, and a highly integrated design is achieved through multiple folding of the optical path and ultra-light miniaturized components. The design of HIS implements a number of key technologies such as high-precision scanning of the optical field of view(FOV), high-precision integrated manufacturing inspection, a large-tolerance pre-filter window, and full-link solar radiation calibration. | 6 | Ultrabroad-band,white light emission from carbon dot-based materials with hybrid fluorescence/phosphorescence for single component white light-emitting diodes显示文摘Benefiting from the large Stokes shift between fluorescence and phosphorescence,fluorescence/phosphorescence dual-emitting carbon dots(CDs)have gradually entered at the stage of single-phase white light-emitting diodes(WLEDs)as‘green material'.However,most of the developed dual-emitting CDs have weak phosphorescence,short emission wavelength and narrow emission band,resulting in relatively bluish white light emission and low color rendering index(CRI).Herein,an ultrabroad-band fluorescence/phosphorescence dual-emitting CD-based material(UB-CD@BA)is prepared by thermal treatment of boric acid(BA)and CDs with large conjugated structure.The stable covalent bonding between CDs and BA,as well as three-dimensional spatial restriction effect of selfpolymerization BA molecules around CDs during long-term heating efficiently rigidified the single/triplet excited states of CDs from non-radiative deactivation,thus producing strong dual emissive materials with the high phosphorescence quantum yield of 21%.Remarkable,the prepared UB-CD@BA powders exhibit bright pure white light emission with Commission Internationale de l'Eclairage(CIE)coordinates of(0.32,0.33)and the highest reported full width at half maximum of 250 nm.Based on the unique characteristics of UB-CD@BA,it was used as a color conversion layer to prepare a WLED with CIE coordinates of(0.35,0.33)and the CRI value of 87. | Yuchen Li Qijun Li Shuai Meng Yukun Qin Dengke Cheng Hailing Gu Zifei Wang Yunxia Ye Jing Tan | 2023 | Chinese Chemical Letters2023,34,5: | 0 |

| 11 | A novel pore-fracture dual network modeling method considering dynamic cracking and its applications显示文摘Unconventional reservoirs are normally characterized by dual porous media, which has both multi-scalepore and fracture structures, such as low permeability or tight oil reservoirs. The seepage characteristicsof such reservoirs is mainly determined by micro-fractures, but conventional laboratory experimentalmethods are difficult to measure it, which is attribute to the dynamic cracking of these micro-fractures.The emerging digital core technology in recent years can solve this problem by developing an accuratepore network model and a rational simulation approach. In this study, a novel pore-fracture dualnetwork model was established based on percolation theory. Fluid flow in the pore of two scales, microfracture and matrix pore, were considered, also with the impact of micro-fracture opening and closingduring flow. Some seepage characteristic parameters, such as fluid saturations, capillary pressure, relative permeabilities, displacement efficiency in different flow stage, can be predicted by proposedcalculating method. | 12 | Determining Critical Support Discharge of a Riverhead and River Network Analysis: Case Studies of Lhasa River and Nyangqu River显示文摘A riverhead is the demarcation point of continuous water channel and seasonal channel, which is characterized by a critical flow that can support a continuous water body. In this study, the critical support discharge(CSD) is defined as the critical steady flows required to form the origin of a stream. The CSD is used as the criterion to determine the beginning of the riverhead, which can be controlled by hydro-climate factors(e.g., annual precipitation, annual evaporation, or minimum stream flow in arid season). The CSD has a close correlation with the critical support/source area(CSA) that largely affects the density of the river network and the division of sub-watersheds. In general, river density may vary with regional meteorological and hydrological conditions that have to be considered in the analysis. In this paper, a new model referring to the relationship of CSA and CSD is proposed, which is based on the physical mechanism for the origin of riverheads. The feasibility of the model was verified using two watersheds(Duilongqu Basin of the Lhasa River and Beishuiqu Basin of the Nyangqu River) in Tibet Autonomous Region to calculate the CSA and extract river networks. A series of CSAs based on different CSDs in derived equation were tested by comparing the extracted river networks with the reference network obtained from a digitized map of river network at large scales. Comparison results of river networks derived from digital elevation model with real ones indicate that the CSD(equal to criterion of flow quantity(Q_c)) are 0.0028 m^3/s in Duilongqu and 0.0085 m^3/s in Beishuiqu. Results show that the Q_c can vary with hydro-climate conditions. The Q_c is high in humid region and low in arid region, and the optimal Q_c of 0.0085 m^3/s in Beishuiqu Basin(humid region) is higher than 0.0028 m^3/s in Duilongqu Basin(semi-arid region). | 14 | Mapping a stable adult-plant stripe rust resistance QTL on chromosome 6AL in Chinese wheat landrace Yibinzhuermai显示文摘Stripe rust,caused by Puccinia striiformis f.sp.tritici(Pst),is one of the most important diseases threatening the yield and stability of wheat production in China and many other countries.Identification and utilization of new genes for durable stripe rust resistance are important for ongoing control of this disease.The objectives of this study were to identify quantitative trait loci(QTL)associated with adult-plant stripe rust resistance in the Chinese wheat landrace Yibinzhuermai(YBZR)and to provide wheat breeders with new sources of potentially durable resistance.A total of 117 recombinant inbred lines(RILs)(F5:8)derived from a cross between YBZR and highly susceptible cultivar Taichung 29(TC29)were assessed for stripe rust severity in field experiments at Wenjiang in 2016 and 2017 and Chongzhou in 2016,2017,2018,and 2019 in Sichuan following inoculation with a mixture of current Pst races.The RILs were genotyped using the Wheat55K single nucleotide polymorphism(SNP)array.Three QTL were identified on chromosome arms 6AL,5BL and 7DS.QYr.YBZR-6AL and QYr.YBZR-7DS conferred major effects in all field environments,explaining 10.6%to 14.7%and 11.5%to 21.2%of phenotypic variation,respectively.The QTL on 5BL and 7DS likely correspond to previously known QTL,whereas QYr.YBZR-6AL is probably novel.Haplotype analysis revealed that the resistance allele at QYr.YBZR-6AL was present in 2.8%of 324 Chinese wheat landraces.SNP markers closely linked with QYr.YBZR-6AL were converted to kompetitive allele-specific PCR markers and validated in the RIL population and a subset of 92 wheat cultivars.QYr.YBZR-6AL and its markers should be useful in breeding programs to improve the level and durability of stripe rust resistance. | Mei Deng Li Long Yukun Cheng Fangjie Yao Fangnian Guan Yuqi Wang Hao Li Zhien Pu Wei Li Qiantao Jiang Yuming Wei Jian Ma Houyang Kang Pengfei Qi Jirui Wang Youliang Zheng Yunfeng Jiang Guoyue Chen | 2022 | The Crop Journal2022,10,4: | 0 |
