| 2 | Effects of Prestressing of the Ring Gear in Interference Fit on Flexural Fatigue Strength of Tooth Root显示文摘When the gearbox body interference is connected to the ring gear, prestressing occurs in the ring gear, which has a significant impact on the strength and life of the gear. Research on the prestressing of the inner ring gear is in the preliminary stage, and the distribution rule of the prestressing and the influence of each parameter on the interference prestressing have not been derived. In this paper, based on the method of calculating the prestressing of the thick cylinder in interference fit, the ring gear is found to be equivalent to a thick cylinder, and the distribution rule of prestressing of the ring gear in the interference fit is inferred. Then, by modeling and analyzing the gearbox body and ring gear in the interference fit using ABAQUS, the distribution rule of prestressing the ring gear in the interference fit is obtained through a numerical simulation. Finally, the prestressing of the ring gear in the interference fit is measured using X-ray di raction, and the distribution rule of prestressing of the ring gear in the interference fit is obtained through analysis. Compared with the distribution rule of prestressing in theory, numerical simulation, and experiment, the theoretical distribution rule of prestressing is amended through a statistical method, and a more accurate formula of prestressing is obtained. Through the calculation of the stress and bending moment in the dangerous section of the ring gear through prestressing, the formula for checking the tooth root flexural fatigue strength in the interference fit prestressing is inferred. | 5 | Single-cell dissection,hdWGCNA and deep learning reveal the role of oxidatively stressed plasma cells in ulcerative colitis显示文摘Ulcerative colitis(UC)develops as a result of complex interactions between various cell types in the mucosal microenvironment.In this study,we aim to elucidate the pathogenesis of ulcerative colitis at the single-cell level and unveil its clinical significance.Using single-cell RNA sequencing and high-dimensional weighted gene co-expres-sion network analysis,we identify a subpopulation of plasma cells(PCs)with significantly increased infiltration in UC colonic mucosa,characterized by pronounced oxidative stress.Combining 10 machine learning approaches,we find that the PC oxidative stress genes accurately distinguish diseased mucosa from normal mucosa(independent external testing AUC=0.991,sensitivity=0.986,specificity=0.909).Using MCPcounter and non-negative matrix factorization,we identify the association between PC oxidative stress genes and immune cell infiltration as well as patient heterogeneity.Spatial transcriptome data is used to verify the infiltration of oxidatively stressed PCs in colitis.Finally,we develop a gene-immune convolutional neural network deep learning model to diagnose UC mucosa in different cohorts(independent external testing AUC=0.984,sensitivity=95.9%,specificity=100%).Our work sheds light on the key pathogenic cell subpopulations in UC and is essential for the development of future clinical disease diagnostic tools. | Shaocong Mo Xin Shen Baoxiang Huang Yulin Wang Lingxi Lin Qiuming Chen Meilin Weng Takehito Sugasawa Wenchao Gu Yoshito Tsushima Takahito Nakajima | 2023 | Acta Biochimica et Biophysica Sinica2023,55,11: | 0 |
| 6 | Determination of Total Flavonoids in Leaves of Tetracera asiatica显示文摘[Objectives] This study was conducted to determine and compare the contents of flavonoids in the leaves of Tetracera asiatica , an ethnic medicine from different habitats, in order to provide a basis for the quality control of its medicinal materials. [Methods] The flavonoid compounds in the leaves of T. asiatica were reflux-extracted in a water bath, and the total flavonoid content in T. asiatica was determined by ultraviolet-visible spectrophotometry. [Results] The total flavonoid content in T. asiatica leaves varied according to different habitats. The highest content was found in Gaofeng (19.41%) of Guangxi, followed by Huizhou (16.88%) of Guangdong and Hong Kong (16.76%). The lowest total flavonoid content was found in Shangcuntang (3.91%) of Bobai County, Guangxi, followed by Beiliu (4.15%), Guangxi. The total flavonoid contents in the samples ranged from 3.91% to 19.41%, with an average content of about 12.36%.
