1The Reference Genome Sequence of Scutellaria baicalensis Provides Insights into the Evolution of Wogonin Biosynthesis显示文摘Scutellaria baicalensis Georgi is important in Chinese traditional medicine where preparations of dried roots,'Huang Qin,' are used for liver and lung complaints and as complementary cancer treatments. We report a high-quality reference genome sequence for S. baicalensis where 93% of the 408.14-Mb genome has been assembled into nine pseudochromosomes with a super-N50 of 33.2 Mb. Comparison of this sequence with those of closely related species in the order Lamiales, sesamum indicum and Salvia splendens,revealed that a specialized metabolic pathway for the synthesis of 4'-deoxyflavone bioactives evolved in the genus Scu-tellaria. We found that the gene encoding a specific cinnamate coenzyme A ligase likely obtained its newfunc- tion following recent mutations, and that four genes encoding enzymes in the 4'-deoxyflavone pathway are present as tandem repeats in the genome of S. baicalensis. Further analyses revealed that gene duplications, segmental duplication, gene amplification, and point mutations coupled to gene neo- and subfunctionaliza-tions were involved in the evolution of 4'-deoxyflavone synthesis in the genus Scutellaria. Our study not only provides significant insight into the evolution of specific flavone biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the development of tools for enhancing bioactive productivity by metabolic engineering in microbes or by molecular breeding in plants. 3Surgical Management of Retinal Detachment Resulting from Macular Hole in a Setting of High Myopia显示文摘Purpose:To evaluate the anatomical and visual outcomes of pars plana vitrectomy (PPV) with internal limiting membrane (ILM).peeling and use of retinal tamponade for retinal detachments resulting from macular hole (MHRD) in highly myopic eyes. Methods:.Twenty-nine highly myopic patients.(29 eyes) underwent PPV with ILM peeling and retinal tamponade for MHRD were enrolled. Demographics and best-corrected visual acuity.(BCVA).were measured preoperatively and at final follow-up. Anatomical success and macular hole closure were analyzed. Results:.Patients' mean age of patients was 58.7±10.6 years, mean follow-up was 11.7±7.4 months. Twenty three eyes (23/26,88.5%)undergoing primary PPV combined with ILM peeling had successful initial retinal reattachment, including 19 eyes (19/19, 100%) with silicone oil tamponade and in 4 eyes (4/7,57.1%).with sulfur hexafluoride.(C3F8).tamponade. Overall anatomical success was achieved in 27 eyes (27/29, 93.1%)..The macular hole closure was observed in 17 eyes (17/26, 65.4%) with final anatomical success. Compared to preoperative BCVA,.the mean postoperative BCVA in the eyes with anatomical success was significantly improved (P = 0.007, Wilcoxon signed rank test). 5Loss of Wip1 aggravates brain injury after ischaemia/reperfusion by overactivating microglia显示文摘Background and purpose The inflammatory response mediated by microglia/macrophages is closely related to cerebral ischaemia/reperfusion injury.Wild-type p53-induced protein phosphatase 1(Wip1),a serine/threonine phosphatase,is expressed in various tissues.A growing number of reports have suggested that Wip1 is a negative regulator of inflammation in peripheral tissue;however,its role in the central nervous system(CNS)remains unclear.This study aimed to clarify whether Wip1 can inhibit CNS inflammation by regulating microglia/macrophage functions after ischaemic injury.Methods A model of middle cerebral artery occlusion and reperfusion was established in mice.CNS inflammation was simulated by lipopolysaccharide treatment of primary microglia.Laser speckle imaging was used to monitor regional cerebral blood flow.Behavioural outcomes were assessed with a TreadScan gait analysis system.TTC staining was used to evaluate the infarct volume,and western blotting and immunofluorescence staining were applied to detect the phenotypical transformation of microglia.ELISA was performed to detect the levels of inflammatory factors.Results Wip1 expression was increased after ischaemia/reperfusion.Wip1-knockout(KO)mice displayed more severe brain injury than wild-type mice,as indicated by aggravated motor dysfunction,greater brain infarct volumes and higher expression of inflammatory cytokines(interleukin-6 and tumour necrosis factor alpha)in the brain.We also found that Wip1 depletion increased microglial/macrophage activation in both in vitro and in vivo models,which all showed activation of microglia/macrophages.Lentivirus-Ppm1d reversed the injury induced by Wip1-KO.Conclusions Our results suggest that Wip1 may inhibit neuroinflammation by inhibiting microglial/macrophage activation after brain ischaemia/reperfusion injury.Feng Yan Xiang Cheng Ming Zhao Shenghui Gong Ying Han Liping Ding Di Wu Yumin Luo Wei Zuo Lingling Zhu Ming Fan Xunming Ji 2021Stroke & Vascular Neurology2021,6,3:4
6Evolution and Domestication Footprints Uncovered from the Genomes of Coix显示文摘Coix lacryma-jobi,a plant species closely related to Zea and Sorghum,is an important food and medicinal crop in Asia.However,no reference genome of this species has been reported,and its exact phylogeny within the Andropogoneae remains unresolved.Here,we generated a high-quality genome assembly of coix comprising~1.73 Gb with 44485 predicted protein-coding genes.We found coix to be a typical diploid plant with an overall 1-to-1 syntenic relationship with the Sorghum genome,despite its drastic genome expansion(~2.3-fold)due mainly to the activity of transposable elements.Phylogenetic analysis revealed that coix diverged with sorghum~10.41 million years ago,which was~1.49 million years later than the divergence between sorghum and maize.Resequencing of 27 additional coix accessions revealed that they could be unambiguously separated into wild relatives and cultivars,and suggested that coix experienced a strong genetic bottleneck,resulting in the loss of about half of the genetic diversity during domestication,even though many traits have remained undomesticated.Our data not only provide novel comparative genomic and evolutionary insights into the Andropogoneae lineage,but also an important resource that will greatly benefit molecular breeding of this important crop.Hongbing Liu Junpeng Shi Zexi Cai Yumin Huang Menglu Lv Huilong Du Qiang Gao Yi Zuo Zhaobin Dong Wei Huang Rui Qin Chengzhi Liang Jinsheng Lai Weiwei Jin 2020Molecular Plant2020,13,2:4

8A novel integrated non-targeted metabolomic analysis reveals significant metabolite variations between different lettuce (Lactuca sativa. L) varieties显示文摘Lettuce is an important leafy vegetable that represents a significant dietary source of antioxidants and bioactive compounds.However,the levels of metabolites in different lettuce cultivars are poorly characterized.In this study,we used combined GC×GC-TOF/MS and UPLC-IMS-QTOF/MS to detect and relatively quantify metabolites in 30 lettuce cultivars representing large genetic diversity.Comparison with online databases,the published literature,standards as well using collision cross-section values enabled putative identification of 171 metabolites.Sixteen of these 171 metabolites(including phenolic acid derivatives,glycosylated flavonoids,and one iridoid)were present at significantly different levels in leaf and head type lettuces,which suggested the significant metabolomic variations between the leaf and head types of lettuce are related to secondary metabolism.A combination of the results and metabolic network analysis techniques suggested that leaf and head type lettuces contain not only different levels of metabolites but also have significant variations in the corresponding associated metabolic networks.The novel lettuce metabolite library and novel non-targeted metabolomics strategy devised in this study could be used to further characterize metabolic variations between lettuce cultivars or other plants.Moreover,the findings of this study provide important insight into metabolic adaptations due to natural and human selection,which could stimulate further research to potentially improve lettuce quality,yield,and nutritional value.Xiao Yang Shiwei Wei Bin Liu Doudou Guo Bangxiao Zheng Lei Feng Yumin Liu Francisco A.Tomás-Barberán Lijun Luo Danfeng Huang 2018Horticulture Research2018,5,1:3
9Engineering of dendritic dopant-free hole transport molecules:enabling ultrahigh fill factor in perovskite solar cells with optimized dendron construction显示文摘Developing dopant-free hole-transporting materials(HTMs)for high-performance perovskite solar cells(PVSCs)has been a very active research topic in recent years since HTMs play a critical role in optimizing interfacial charge carrier kinetics and in turn determining device performance.Here,a novel dendritic engineering strategy is first utilized to design HTMs with a D-A type molecular framework,and diphenylamine and/or carbazole is selected as the building block for constructing dendrons.All HTMs show good thermal stability and excellent film morphology,and the key optoelectronic properties could be fine-tuned by varying the dendron structure.Among them,MPA-Cz-BTI and MCz-Cz-BTI exhibit an improved interfacial contact with the perovskite active layer,and non-radiative recombination loss and charge transport loss can be effectively suppressed.Consequently,high power conversion efficiencies(PCEs)of 20.8%and 21.35%are achieved for MPA-Cz-BTI and MCz-Cz-BTI based devices,respectively,accompanied by excellent long-term storage stability.More encouragingly,ultrahigh fill factors of 85.2%and 83.5%are recorded for both devices,which are among the highest values reported to date.This work demonstrates the great potential of dendritic materials as a new type of dopant-free HTMs for high-performance PVSCs with excellent FF.Wei Chen Yang Wang Bin Liu Yajun Gao Ziang Wu Yongqiang Shi Yumin Tang Kun Yang Yujie Zhang Weipeng Sun Xiyuan Feng Frédéric Laquai Han Young Woo Aleksandra B.Djurisic Xugang Guo Zhubing He 2021Science China Chemistry2021,64,1:2

11Ubiquitin-specific protease 24 promotes EV71 infection by restricting K63-linked polyubiquitination of TBK1显示文摘TANK-binding kinase 1(TBK1)is an essential protein kinase for activation of interferon regulatory factor 3(IRF3)and induction of the type I interferons(IFN-I).Although the biochemical regulation of TBK1 activation has been studied,little is known about how enterovirus 71(EV71)employs the deubiquitinases(DUBs)to regulate TBK1 activation for viral immune evasion.Here,we found that EV71 infection upregulated the expression of ubiquitinspecific protease 24(USP24).Further studies revealed that USP24 physically interacted with TBK1,and can reduce K63-linked polyubiquitination of TBK1.Knockdown of USP24 upregulated TBK1 K63-linked polyubiquitination,promoted the phosphorylation and nuclear translocation of IRF3,and in turn improved IFN-I production during EV71 infection.As a consequence,USP24 knockdown dramatically inhibited EV71 infection.This study revealed USP24 as a novel regulator of TBK1 activation,which promotes the understanding of immune evasion mechanisms of EV71 and could provide a potential strategy for treatment of EV71 infection.Lichao Zang Jin Gu Xinyu Yang Yukang Yuan Hui Guo Wei Zhou Jinhong Ma Yan Chen Yumin Wu Hui Zheng Weifeng Shi 2023Virologica Sinica2023,38,1:2

14Influence of cementite spheroidization on relieving the micro-galvanic effect of ferrite-pearlite steel in acidic chloride environment显示文摘The corrosion behavior of the as-received steel and the spheroidized steel in acidic chloride environment was investigated. The results indicate the corrosion mode and corrosion rate of two steels are diverse due to their difference in microstructure. For as-received steel with ferrite-pearlite microstructure, severe localized corrosion happens on the pearlite regions, and plenty of cathodic cementite remains in the pits, further strengthening the micro-galvanic effect and accelerating the corrosion rate. 15Analysis of Lymph Node Metastases of 1,526 Cases with Thoracic Esophageal and Cardiac Carcinomas: A Random Sampling Report froni the Fourth Hospital of Hebei Medical University from 1996 to 2004显示文摘OBJECTIVE To summarize the regular pattern and state oflymph node metastasis of patients with esophageal and cardiaccarcinomas,so as to analyze factors influencing lymph nodemetastasis.METHODS Clinical data collected from 1,526 thoracicesophageal and cardiac carcinoma patients who were admitted inthe Fourth Hospital of Hebei Medical University during a periodfrom January 1996 to December 2004,were randomly selectedand an Access Database of the patient's information was set up.Eight clinico-pathologic factors,including the patient's age,tumorlocation and size,pathological classification,the depth of tumorinvasion,vascular tumor embolus (VTE),the state of surroundingorgan encroachment and the status of tumor residues,wereidentified.A correlation between these factors and metastases wasstatistically analyzed using SPSS13.0 software.RESULTS Lymph node metastatic sites from esophagealcarcinomas included the thoracic and abdominal cavity.Lymphnode metastasis from the superior esophageal carcinomasmainly occurred in the neck and thoracic cavity.There was atwo-way lymph node metastasis in the patients with the middleesophageal carcinoma.The inferior esophageal carcinomas mainlymetastasized to the paraesophageal,paragastric cardia,and leftgastric artery lymph nodes.The rate and degree of the metastasisfrom the inferior esophageal carcinomas were significantly highercompared to those of the superior and the middle esophagealcarcinomas (P<0.0125).The degree of abdominal lymph node metastasis fromcarcinomas of the gastric cardia was significantly higher comparedwith that of esophageal carcinomas.In the group with carcinomaof the gastric cardia,the rate and degree of the lymph nodemetastases in the paragastric cardia and left gastric artery weresignificantly higher compared to the group with esophagealcarcinoma (P<0.05).Paraesophageal lymph node metastasis fromcarcinomas of the gastric cardia in the thoracic cavity frequentlyoccurred,too,and the degree of the metastasis was similar to thatof esophageal carcinoma.There was no significant difference inthe rate and degree of the paraesophageal lymph-node metastasisbetween the group with carcinoma of the gastric cardia comparedto those with esophageal carcinoma (P>0.05).Multifactoriallogistic regression analysis showed that the tumor size,depth oftumor encroachment,VTE,and tumor residues could all bringabout obvious impact on lymph-node metastases (P<0.05).CONCLUSION Lymph node metastasis from superioresophageal carcinomas mainly occurs in the neck and thoraciccavity.The middle esophageal carcinomas presented a two-waylymph-node metastasis (both the upwards and the downwards),and the lymph node metastasis from inferior esophagealcarcinomas mainly occurred in the thoracic and abdominal cavities.The metastases of carcinoma of the gastriccardia were most commonly found in the abdominalcavity,with frequent paraesophageal lymph-nodemetastasis.The sufficient attention should be paidto neck lymph node clearance in cases of esophagealcarcinoma.What is of the greatest concern is theclearance of the left gastric artery lymph nodes,andalso in cases of gastric cardia carcinoma,clearance,the paraesophageal lymph nodes.With an increasein the tumor size and depth of tumor encroachment,and occurrence of VTE and tumor residual cells,therisk of lymph node metastasis is significantly raised (P<0.05).Wei Liu Xishan Hao Yong Chen Haixin Li Shijie Wang Peizhong Wang Hng Jin Liyun Guan Qian Fan Linan Song Yumin Ping Xianli Meng Rui Wang Junfeng Liu Xiaoling Wang 2008Chinese Journal of Clinical Oncology2008,5,6:1
16Clinical manifestations and outcomes in severe ulcerative colitis显示文摘In order to evaluate the clinical manifestations and outcomes of severe ulcerative colitis(UC),we retrospectively reviewed 41 patients with severe UC from 144 consecutively hospitalized UC cases from 1988 to 2004.Data recorded included onset,symptoms,signs,laboratory results,endoscopic,radiologic and pathologic findings,the clinical treatment process and follow-up.Of these severe cases,92.7%(38/41)had pancolitis.Clinically,36.9%(15/41)were categorized as first onset type,36.9%(15/41)were chronic persistent and 26.8%(11/41)were chronic recurrent.Steroids played a main role in the remission of severe UC(61.0%).Thirty-one cases(75.6%)were relieved by drug therapy.Seven cases(17.1%)progressed to the need for operation.An early age of onset,pancolitis,low hemoglobin and serum albumin levels,and the need for intravenous steroids tended to be associated with the need for surgery.In conclusion,most of the severe UC patients respond well to drug therapy,but for individuals who are unresponsive to drug therapy,or for those depending on steroids,after a reasonable duration of treatment,the necessity for surgery should be considered.YANG Xuesong YAO Wei LIU Wenbin LI Jun LU Yumin 2007Frontiers of Medicine2007,1,2:1
17The applications of single-atom alloys in electrocatalysis:Progress and challenges显示文摘The development of cost-effective and highly efficient electrocatalysts to accelerate distinct electrochemical reactions is essential to help the industry to achieve a low-carbon footprint.Single-atom alloys(SAAs)with the characteristics of unique electronic structures,well-defined active sites,and maximum atom utilization demonstrate promising potential to replace traditional noble metal catalysts.SAAs are expected to tailor the adsorption properties of reaction species,thus promoting electrocatalytic behaviors.Herein,representative synthetic strategies including wet chemistry,galvanic replacement,dealloying,and atomic layer deposition are introduced,followed by a summary of applications of SAAs in hydrogen evolution reaction,oxygen evolution reaction,oxygen reduction reaction,carbon dioxide reduction reaction,and ethanol electro-oxidation to provide an in-depth understanding of the structure–activity relationship.Moreover,the challenges and perspectives in this emerging field of SAAs are discussed.Yumin Da Rui Jiang Zhangliu Tian Xiaopeng Han Wei Chen Wenbin Hu 2023SmartMat2023,4,1:1

20The synergy between cementite spheroidization and Cu alloying on the corrosion resistance of ferrite-pearlite steel in acidic chloride solution显示文摘In this work,the corrosion behavior of medium-carbon steels(45,45 Cu and 45 Cuq steels)in acidic chloride environment was investigated.The results indicated that the micro-galvanic effect between the anodic ferrite matrix phase and the cathodic cementite secondary phase notably affected the corrosion resistance of the three steels.For 45 steel,serious pitting corrosion happened in and around the pearlite regions,and a large number of lamellar cementite was fixed in the corrosion pits.Meanwhile,the continuously increasing superficial area of cathodic cementite enhanced the micro-galvanic corrosion,resulting in a rapidly increase in corrosion rate with time.While for 45 Cu and 45 Cuq steels,macroscopic uniform corrosion occurred,and the cementite accumulation was markedly reduced as compared with 45 steel,thus the micro-galvanic effect was weakened and the corrosion rate was decreased accordingly.Among these,45 Cuq steel showed the most stable and excellent corrosion resistance during long-term corrosion,indicating the occurrence of a synergistic effect between cementite spheroidization and Cu alloying,thereby significantly improving the corrosion resistance of 45 steel.Hu Liu Jie Wei Junhua Dong Yangtao Zhou Yiqing Chen Yumin Wu Subedi Dhruba Babu Aniefiok Joseph Umoh Wei Ke 2021Journal of Materials Science & Technology2021,,25:1
