| 1 | 苎麻遗传转化再生体系的建立显示文摘苎麻(BoehmerianiveaL.Guad)是荨麻科苎麻属宿根型草本植物,主要分布在热带和亚热带地区,为我国特产,是重要的纺织纤维和饲料作物。由于苎麻为杂合体,其遗传背景十分复杂,有性杂交等常规方法难以取得育种上的突破,因此,探讨利用基因工程技术改良苎麻品种具有重要意义。本研究以苎麻品种圆叶青叶盘为外植体,选用MS培养基为基本培养基,附加激素BA、IAA、GA3和NAA,通过研究不同激素浓度组合对外植体愈伤诱导及不定芽分化的影响以及不同激素配比对生根的作用,建立了较好的遗传转化再生体系。试验结果表明圆叶青叶盘最适诱导愈伤培养基为MS+6-BA2.0mg/L+GA30.4mg/L+IAA0.2mg/L+蔗糖30g/L+琼脂7.5g/L;分化培养基为MS+6-BA2.0mg/L+GA30.4mg/L+蔗糖30g/L+琼脂7.5g/L;小苗的最佳生根培养基为MS+NAA0.2mg/L+IAA0.1mg/L+蔗糖30g/L+琼脂7.5g/L。在抗生素浓度梯度筛选实验中获得50mg/L卡那霉素可以抑制叶盘的分化;40mg/L卡那霉素可以抑制再生植株的生根。筛选出的遗传转化再生体系,为进一步利用基因工程技术对苎麻进行遗传改良打下基础。 | 孔华 郭安平 章霄云 姚庆荣 郭运玲 贺立卡 Kong Hua Guo Anping Zhang Xiaoyun Yao Qingrong Guo Yunling He Lika | 2006 | 分子植物育种2006,4,2: | 16 |
| 2 | WIP1 Phosphatase Plays a Critical Neuroprotective Role in Brain Injury Induced by High-Altitude Hypoxic Inflammation显示文摘The hypobaric hypoxic environment in highaltitude areas often aggravates the severity of inflammation and induces brain injury as a consequence. However, the critical genes regulating this process remain largely unknown. The phosphatase wild-type p53-induced phosphatase 1(WIP1) plays important roles in various physiological and pathological processes, including the regulation of inflammation in normoxia, but its functions in hypoxic inflammation-induced brain injury remain unclear.Here, we established a mouse model of this type of injury and found that WIP1 deficiency augmented the release of inflammatory cytokines in the peripheral circulation and brain tissue, increased the numbers of activated microglia/macrophages in the brain, aggravated cerebral histological lesions, and exacerbated the impairment of motor and cognitive abilities. | 3 | Integrin-interacting protein Kindlin-2 induces mammary tumors in transgenic mice显示文摘Kindlin-2, an integrin-interacting protein, regulates breast cancer progression. However, currently, no animal model to study the role of Kindlin-2 in the carcinogenesis of mammary gland is available. We established a Kindlin-2 transgenic mouse model using a mammary gland-specific promoter, mammary tumor virus(MMTV) long terminal repeat(LTR). Kindlin-2 was overexpressed in the epithelial cells of the transgenic mice. The mammary gland ductal trees were found to grow faster in MMTV-Kindlin-2 transgenic mice than in control mice during puberty. Kindlin-2 promoted mammary gland growth as indicated by more numerous duct branches and larger lumens, and more alveoli were formed in the mammary glands during pregnancy under Kindlin-2 overexpression. Importantly, mammary gland-specific expression of Kindlin-2 induced tumor formation at the age of 55 weeks on average. Additionally, the levels of estrogen receptor and progesterone receptor were decreased, whereas human epidermal growth factor receptor 2 and β-catenin were upregulated in the Kindlin-2-induced mammary tumors. | 4 | Geometric active contour based approach for segmentation of high-resolution spaceborne SAR images显示文摘Segmentation is the key step in auto-interpretation of high-resolution spaceborne synthetic aperture radar(SAR) images. A novel method is proposed based on integrating the geometric active contour(GAC) and the support vector machine(SVM)models. First, the images are segmented by using SVM and textural statistics. A likelihood measurement for every pixel is derived by using the initial segmentation. The Chan-Vese model then is modified by adding two items: the likelihood and the distance between the initial segmentation and the evolving contour. | 6 | Imaging and Spectral Characteristics of Amyloid Plaque Autofluorescence in Brain Slices from the APP/PS1 Mouse Model of Alzheimer’s Disease显示文摘Amyloid deposits are one of the hallmark pathological lesions of Alzheimer's disease(AD). They can be visualized by thioflavin-S, silver impregnation,Congo red staining, and immunohistochemical reactions.However, that amyloid deposits generate blue autofluorescence(auto-F) has been ignored. Here, we report that visible light-induced auto-F of senile plaques(SPs) was detected and validated with conventional methods. Brain slices from APP/PS1(amyloid precursor protein/presenilin1) transgenic mice were mounted on slides, rinsed,coverslipped and observed for details of the imaging and spectral characteristics of the auto-F of SPs. Then the slices were treated with the above classic methods for comparative validation. We found that the SP auto-F was greatest under blue-violet excitation with a specific emission spectrum, and was much easier, more sensitive, and reliable than the classic methods. | 7 | Analysis on the Variation of Urban Heat Island Intensity and Its Relationships with Air Pollutants in Kunming,China显示文摘[Objective]The research aimed to study variation of the urban heat island(UHI) intensity and its relationships with air pollutants in Kunming.[Method]Observation data of the air temperature series from Kunming station and surrounding Taihuashan meteorological station were used to investigate the variation characteristics of UHI intensity in Kunming during 2001-2010.Meanwhile,relationship between UHI intensity and air quality on the basis of concentration data for primary air pollutants(SO2,NO2 and PM10) from environmental monitoring station of Kunming was studied in this paper.[Result]Annual mean UHI intensity increased at the rate of 0.110 ℃/10 a,and UHI intensity was higher in dry season than that in rainy season.The change trend of air pollutants level in Kunming was in accordance with that of the UHI intensity.When UHI intensity reached peak value,air pollutants showed high concentration.UHI intensity had significantly positive correlation with air pollutants level.Annual mean UHI intensity had significantly positive correlation with PM 10 level under the likelihood of 95%,but monthly mean UHI intensity had significantly positive correlation with NO2 level under the likelihood of 99%.In addition,annual statistics days with Grade I air quality obviously decreased with the strengthenment of UHI intensity.Therefore,air quality in Kunming City turned more deteriorated with the enhancement of UHI intensity.[Conclusion] The research findings provided a valid scientific basis for the mitigation of UHI and improvement of the atmospheric environment in Kunming. | Yunling He Hengyu Wang | 2013 | Meteorological and Environmental Research2013,4,1: | 0 |
| 8 | Highly crystalline and water-wettable benzobisthiazole-based covalent organic frameworks for enhanced photocatalytic hydrogen production显示文摘Two-dimensional covalent organic frameworks are promising for photocatalysis by virtue of their structural and functional diversity,but generally suffer from low activities relative to their inorganic competitors.To fulfill their full potential re quires a rational tailoring of their structures at different scales as well as their surface properties.Herein,we demonstrate benzobisthiazole-based covalent organic frameworks as a superior photocatalyst for hydrogen production.The product features high crystallinity with ordered 2.5-nm-wide cylindrical mesopores and great water wettability.These structural advantages afford our polymeric photo catalyst with fast charge carrier dynamics as evidenced by a range of spectroscopic characterizations and excellent catalytic performances when suspended in solution or supported on melamine foams.Under visible-light irradiation,it enables efficient and stable hydrogen evolution with a production rate of 487μmol h^(-1)(or a mass-specific rate of 48.7 mmol g^(-1)h^(-1))—far superior to the previous state of the art.We also demonstrate that hydrogen production can be stoichiometrically coupled with the oxidation conversion of biomass as exemplified by the conversion of furfuryl alcohol to 2-furaldehyde. | Wei Huang Yongpan Hu Zhengyuan Qin Yujin Ji Xuan Zhao Yunling Wu Qing He Youyong Li Chunfeng Zhang Jun Lu Yanguang Li | 2023 | National Science Review2023,10,1: | 0 |
