| 2 | A Highly Pathogenic Strain of Porcine Deltacoronavirus Caused Watery Diarrhea in Newborn Piglets显示文摘Porcine deltacoronavirus(PDCoV) is a newly identified virus that causes watery diarrhea in newborn piglets and results in significant economic losses to the pig industry. Since first reported in Hong Kong in 2012, PDCoV has been subsequently detected in USA, South Korea, Thailand, and China's Mainland. Here we isolated a strain of PDCoV, named CHN-GD-2016,from the intestinal content of a diseased newborn piglet with severe diarrhea in a pig farm in Guangdong, China. PDCoV CHN-GD-2016 could be identified by immunofluorescence with PDCoV specific rabbit antisera, and typical crown-shaped particles with spiky surface projections of this PDCoV were observed with electron microscopy. Genomic analysis showed that the PDCoV CHN-GD-2016 was closely related to other Chinese PDCoV strains, with the highest sequence similarity with the strain CHN/Tianjin/2016. Importantly, inoculation of newborn piglets with 1×10~5 TCID_(50) of CHN-GD-2016 by oral feeding successfully reproduced clear clinical symptoms, including vomiting, dehydration, and severe diarrhea in piglets. In addition, the virus RNA in rectal swabs from 1 to 7 days post inoculation was detected, macroscopic and microscopic lesions in small intestine were observed, and viral antigen was also detected in the small intestines with immunohistochemical staining. | 3 | Characterization and source analysis of indoor/outdoor culturable airborne bacteria in a municipal wastewater treatment plant显示文摘The potential health risks of airborne bacteria emission from a wastewater treatment process have been concerned. However, few studies have investigated the differences in community structure between indoor and outdoor bacteria. In this work, the characterization of airborne bacteria was studied in a municipal wastewater treatment plant in Beijing, China. Two indoor(i.e., fine screen room and sludge dewatering house) and two outdoor(i.e., aeration tank and control site) sampling sites were selected. An Andersen six-stage impactor was used for collecting culturable airborne bacteria in the air, and Illumina MiSeq sequencing was conducted to track the emission source of the culturable airborne bacteria. The results indicate that, compared with the outdoor aeration tank site, the concentrations of culturable airborne bacteria in the indoor fine screen room with poor ventilation were more than ten times higher and the particle size was about twice as large. The community structures of indoor and outdoor culturable airborne bacteria were obviously different. Enterobacteriaceae and opportunistic pathogens were detected in indoor culturable airborne bacteria, with wastewater and sludge dewatering machine identified as the primary sources. Conversely,Enterobacteriaceae and opportunistic pathogens were not detected in outdoor culturable airborne bacteria. Outdoor high wind speed might have resulted in rapid dilution and mixing of culturable airborne bacteria generated from the aeration tank with the ambient air. | 4 | Characterization of the airborne bacteria community at different distances from the rotating brushes in a wastewater treatment plant by 16S rRNA gene clone libraries显示文摘Biological risks of bioaerosols emitted from wastewater treatment processes have attracted wide attention in the recent years. However, the culture-based analysis method has been mostly adopted for detecting the bacterial community in bioaerosols, which may result in the underestimation of total microorganism concentration as not all microorganisms are cultivable. In this study, oligonucleotide fingerprinting of 16S rRNA genes was applied to reveal the composition and structure of the bacterial community in bioaerosols from an Orbal oxidation ditch in a Beijing wastewater treatment plant (WWTP). Bioaerosols were collected at different distances from the aerosol source, rotating brushes, and the sampling height was 1.5 m which is the common respiratory height of a human being. The bacterial communities of bioaerosols were diverse, and the lowest bacterial diversity was found at the sampling site just after the rotating brush rotating brush. A large proportion of bacteria in bioaerosols were affiliated with Proteobacteria and Bacteroidetes. Numerous bacteria present in the bioaerosols also emerged in water, indicating that the bacterial community in the bioaerosols was related to that of the aerosols' sources. The forced aeration of rotating brushes brought about observably distinct bacterial communities between sampling sites situated before and after the rotating brush. Isolation sources of closest relatives in bioaerosols clone libraries were associated with the aqueous environment in the WWTP. Common potential pathogens in bioaerosols as well as those not reported in previous research were also analyzed in this study. | 6 | Cloned pigs derived from somatic cell nuclear transferembryos cultured in vitro atlow oxygen tension显示文摘Pig cloning has great potential to human xenotransplantation. The present study was designed to establish a more efficient system for producing cloned pigs by somatic cell nuclear transfer (SCNT). Our approach was as follows: SCNT embryos were reconstructed by using fetal fibroblasts of Chinese miniature pig as donors and in vitro matured oocytes of prepubertal gilts as recipients. Reconstructed em- bryos were induced by electrical fusion/activation and cultured in BSA-containing North Carolina State University 23 medium (NCSU-23) or Porcine Zygote Medium (PZM-3) at the gas condition of 5% CO2, 7% O2, 88% N2. A total of 230 cloned embryos were transferred to three surrogate sows, producing three piglets. One of them is apparently healthy. The clonal provenance of the piglet was indicated by its coat color and confirmed by DNA microsatellite analysis. | 8 | Sulfur dioxide and o-xylene co-treatment in biofilter:Performance, bacterial populations and bioaerosols emissions显示文摘Sulfur dioxide(SO_2) and benzene homologs are frequently present in the off-gas during the process of sewage sludge drying. A laboratory scale biofilter was set up to co-treat SO_2 and o-xylene in the present study. SO_2 and o-xylene could be removed simultaneously in a single biofilter. Their concentration ratio in the inlet stream influenced the removal efficiencies. It is worth noting that the removal of SO_2 could be enhanced when low concentrations of o-xylene were introduced into the biofilter. Pseudomonas sp., Paenibacillus sp., and Bacillus sp. were the main functional bacteria groups in the biofilter. Sulfur-oxidizing bacteria(SOB) and o-xylene-degrading bacteria(XB) thrived in the biofilter and their counts as well as their growth rate increased with the increase in amount of SO_2 and o-xylene supplied. The microbial populations differed in counts and species due to the properties and components of the compounds being treated in the biofilter. The presence of mixed substrates enhanced the diversity of the microbial population. During the treatment process, bioaerosols including potentially pathogenic bacteria, e.g., Acinetobacter lwoffii and Aeromonas sp., were emitted from the biofilter. | 11 | Animal diversities and characteristics of environmental change revealed by skeletons unearthed at Zhongba Site of Chongqing City,China显示文摘Nearly 200000 animal skeletons are unearthed in the Unit T0202 from Zhongba Site of Zhongxian County.According to the analysis of 129165 specimens,these skeletons may be classified into 5 kinds, namely,Mammalia,Osteichthys,Aves,Amphibian and Reptilia,which belong to 13 orders,28 families and 42 genera.In this paper,based on archeological dating and AMS 14 C data,through statistically analyzing the unearthed skeletons and studying the change of the smallest individual numbers,these research results detected the following:1)In almost all the time of 2370―200 BC,in Zhongba region, some animals distributed widely,such as Muntiacus sp,Elaphodus cephalophus and Muntiacus sp. inhabiting in glade and grassland,Scrofa sp.and Canis familiaris raised by the ancient people,rodentia rabbit and Rattus rattus,which suggested that a fairly good ecological environment of forest and grassland was preserved at that period and the predecessors began to raise domestic animals from 1750 to 1000 BC,which has lasted until today.2)Rhinoceros only lived during 2000―1750 BC,1000― 700 BC and 700―500 BC,which indicates that the ecological environment of grassland and wetland might be better in these phases.3)Macaca sp.and Ursus arctos appeared only after 1750―1000 BC, which may show that the forest condition is better for animals to live during 1750―200 BC.4)The smallest individual numbers of Bublus sp.,bos sp.and otters emerged during about 2370―1750 BC, which perhaps infers that water area during the early period was wider than that of the late period.5) Since skeletons of rhinoceros are discovered in strata of Zhongba Site during 2000―1750 BC and 1000―500 BC,according to the climate and ecology environment which rhinoceros live in now,the average annual temperature and precipitation during 2000―1750 BC and 1000―500 BC are supposed to be probably higher than that of today.Although Zhu Kezhen considered that the first low temperature period in the past 5000 years of China was between 1100 BC and 850 BC,massive pollen of Morus, Ulmus,Fagus,Quercus,Castanea,etc.were found in Dajiuhu peat at that time,which should indicate that the climate was still moderate for living things to live at least in Dajiuhu basin and Zhongba Site.6) Because skeletons of Gervus albirostris were unearthed during 310―200 BC at Zhongba Site,based onthe climate and the ecological environment those animals live in now,the average annual temperature and the average annual precipitation in Zhongba area between 310 BC and 200 BC should be lower than those of today,which is confirmed by the TOC research of Zhongba Site strata. | ZHU Cheng MA ChunMei OUYANG Jie LI ZhongXuan YIN Qian SUN ZhiBin HUANG YunPing R.K.FLAD LI Lan LI YuMei | 2008 | Chinese Science Bulletin2008,53,S1: | 4 |

The incomplete reprogramming of donor cell nuclei has been implicated as a primary reason for the low efficiency of SCNT. DNA methylation is a major epige-netic modification of the genome that regulates crucial aspects of genome function,including estab-lishment of genomic imprinting. In order to make sure whether the DNA methylation reprogramming is efficient in SCNT animals,we analyzed the DNA methylation status of two imprinting genes,H19 and Xist,in lungs of deceased SCNT bovines that died within 48 h of birth using bisulfite sequencing analysis. | 18 | Water level variation characteristics under the impacts of extreme drought and the operation of the Three Gorges Dam显示文摘Under the influence of a climate of extreme drought and the Three Gorges Dam (TGD) operation, the water levels in the middle and lower reaches of the Yangtze River in 2006 and 2011 changed significantly compared with those in the extreme drought years of 1978 and 1986. To quantitatively analyze the characteristics of water level variations in 2006 and 2011, a new calculation method was proposed, and the daily water level and discharge from 1955-2016 were collected in this study. The findings are as follows: in 2006 and 2011, the water level in the dry season significantly increased, but that in the flood season obviously decreased compared with the levels in 1978 and 1986. Here, we described this phenomenon as “no low-water-level in dry season, no high-water-level in flood season”. | 19 | DVL mutations identified from human neural tube defects and Dandy-Walker malformation obstruct the Wnt signaling pathway显示文摘Wnt signaling pathways,including the canonical Wnt/β-catenin pathway,planar cell polarity pathway,and Wnt/Ca2+signaling pathway,play important roles in neural development during embryonic stages.The DVL genes encode the hub proteins for Wnt signaling pathways.The mutations in DVL2 and DVL3 were identified from patients with neural tube defects(NTDs),but their functions in the pathogenesis of human neural diseases remain elusive.Here,we sequenced the coding regions of three DVL genes in 176 stillborn or miscarried fetuses with NTDs or Dandy-Walker malformation(DWM)and 480 adult controls from a Han Chinese population.Four rare mutations were identified:DVL1 p.R558 H,DVL1 p.R606 C,DVL2 p.R633 W,and DVL3 p.R222 Q.To assess the effect of these mutations on NTDs and DWM,various functional analyses such as luciferase reporter assay,stress fiber formation,and in vivo teratogenic assay were performed.The results showed that the DVL2 p.R633 W mutation destabilized DVL2 protein and upregulated activities for all three Wnt signalings(Wnt/β-catenin signaling,Wnt/planar cell polarity signaling,and Wnt/Ca2+signaling)in mammalian cells.In contrast,DVL1 mutants(DVL1 p.R558 H and DVL1 p.R606 C)decreased canonical Wnt/β-catenin signaling but increased the activity of Wnt/Ca2+signaling,and DVL3 p.R222 Q only decreased the activity of Wnt/Ca2+signaling.We also found that only the DVL2 p.R633 W mutant displayed more severe teratogenicity in zebrafish embryos than wild-type DVL2.Our study demonstrates that these four rare DVL mutations,especially DVL2 p.R633 W,may contribute to human neural diseases such as NTDs and DWM by obstructing Wnt signaling pathways. | Lingling Liu Weiqi Liu Yan Shi Ling Li Yunqian Gao Yunping Lei Richard Finnell Ting Zhang Feng Zhang Li Jin Huili Li Wufan Tao Hongyan Wang | 2020 | Journal of Genetics and Genomics2020,47,6: | 2 |
| 20 | Carbon and nitrogen stable isotope analyses of mammal bone fossils from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River,China显示文摘Based on AMS 14 C dating data,carbon and nitrogen isotope analyses were conducted on mammal bone collagen of deer,cattle and pigs from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River.These analyses were conducted to reconstruct palaeodiets of mammals,palaeoecology,palaeoenviroment and previous human activities in the study area.Results show that the collagen loss of bone did not change the in vivo isotopic composition of carbon and nitrogen stable isotopes,and most of the bone fossils were well preserved.The bone collagen of samples from deer had a mean δ 13 C of-23.1‰ and a mean δ 15 N of 4.7‰,suggesting that deer subsisted in a closed habitat and fed on branches and leaves.The bone collagen of cattle had a mean δ 13 C of-19.6‰ and a mean δ 15 N of 5.2‰,which indicates that cattle subsisted in an open habitat and fed on grasses and stems.The δ 13 C values show that both deer and cattle fed on C 3 plants and lived in the same ecosystem,but the t-test results show that deer δ 13 C and δ 15 N values were both more negative than those of cattle,indicating that they inhabited different niches.The δ 13 C and δ 15 N values of cattle partially overlapped those of deer,suggesting some competition in diets between them.The t-tests show that the δ 13 C and δ 15 N values of pigs were more positive than those of cattle and deer,which signifies that pigs occupied a higher trophic level compared to cattle and deer.The wide range of pig δ 13 C values demonstrates that pig trading had been taking place from early Neolithic Age to late Bronze Age.There were no significant differences in deer δ 13 C and δ 15 N values among different archaeological periods,making it clear that climatic,ecological and environmental conditions were kept relatively stable from 2200 to 4200 a BP.This stability may have been responsible for the extensive and complete cultural layers at the Zhongba site.The minimum number of samples required to estimate the mean δ 13 C values of deer,pigs and cattle are 8,73 and 16,respectively,and for mean δ 15 N values of deer,pigs and cattle,the minimum numbers are 4,5 and 6,respectively. | TIAN XiaoSi ZHU Cheng SUN ZhiBin SHUI Tao HUANG YunPing FLAD Rowan Kimon LI YuMei | 2011 | Chinese Science Bulletin2011,56,2: | 2 |
