2Cholesterol-tuned liposomal membrane rigidity directs tumor penetration and anti-tumor effect显示文摘Recently, liposomes have been widely used in cancer therapeutics, but their anti-tumor effects are suboptimal due to limited tumor penetration. To solve this problem, researchers have made significant efforts to optimize liposomal diameters and potentials, but little attention has been paid to liposomal membrane rigidity. Herein, we sought to demonstrate the effects of cholesterol-tuned liposomal membrane rigidity on tumor penetration and anti-tumor effects. In this study, liposomes composed of hydrogenated soybean phospholipids(HSPC), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000](DSPE-PEG2000) and different concentrations of cholesterol were prepared. It was revealed that liposomal membrane rigidity decreased with the addition of cholesterol. Moderate cholesterol content conferred excellent diffusivity to liposomes in simulated diffusion medium, while excessive cholesterol limited the diffusion process. We concluded that the differences of the diffusion rates likely stemmed from the alterations in liposomal membrane rigidity, with moderate rigidity leading to improved diffusion. Next, the in vitro tumor penetration and the in vivo anti-tumor effects were analyzed. The results showed that liposomes with moderate rigidity gained excellent tumor penetration and enhanced anti-tumor effects. 3Deletion of SMARCA4 impairs alveolar epithelial type II cells proliferation and aggravates pulmonary fibrosis in mice显示文摘Alveolar epithelial cells(AECs)injury and failed reconstitution of the AECs barrier are both integral to alveolar flooding and subsequent pulmonary fibrosis(PF).Nevertheless,the exact mechanisms regulating the regeneration of AECs post-injury still remain unclear.SMARCA4 is a part of the large ATP-dependent chromatin remodelling complex SWI/SNF,which is essential for kidney and heart fibrosis.We investigates SMARCA4 function in lung fibrosis by establishing PF mice model with bleomycin firstly and found that the expression of SMARCA4 was mainly enhanced in alveolar type II(ATII)cells.Moreover,we established an alveolar epithelium-specific SMARCA4-deleted SP-C-rtTA/(tetO)7-Cre/SMARCA4f/f mice(SOSM4D/D)model,as well as a new SMARCA4-deleted alveolar type II(ATII)-like mle-12 cell line.We found that the bleomycin-induced PF was more aggressive in SOSM4D/D mice.Also,the proliferation of ATII cells was decreased with the loss of SMARCA4 in vivo and in vitro.In addition,we observed increased proliferation of ATII cells accompanied by abnormally high expression of SMARCA4 in human PF lung sections.These data uncovered the indispensable role of SMARCA4 in the proliferation of ATII cells,which might affect the progression of PF.Danyi Peng Daozhu Si Rong Zhang Jiang Liu Hao Gou Yunqiu Xia Daiyin Tian Jihong Dai Ke Yang Enmei Liu Yujun Shi Q.Richard Lu Lin Zou Zhou Fu 2017Genes & Diseases2017,4,4:3

7Magnetic covalent organic framework nanospheres-based miRNA biosensor for sensitive glioma detection显示文摘Sensitive detection and accurate diagnosis/prognosis of glioma remain urgent challenges.Herein,dispersed magnetic covalent organic framework nanospheres(MCOF)with uniformed Fe3O4 nano-assembly as cores and high-crystalline COF as shells were prepared by monomer-mediated in-situ interface growth strategy.Based on the unique interaction between MCOF and hairpin DNA,a fluorescent signal amplified miRNA biosensor was constructed.It could realize the sensitive detection of miRNA-182 in different matrixes,where the detection limit,linearity range and determination coefficient(R^(2))in real blood samples reached 20 fM,0.1 pM-10 pM and 0.991,respectively.Also,it possessed good stability and precision as observed from the low intra-day/inter-day RSD and high extraction recovery.As a result,it could quantify miRNA-182 in serum of glioma patients,the concentration of which was significantly higher than that of healthy people and obviously decreased after surgery.Finally,a proof-of-concept capillary chip system using this biosensor was proposed to realize the visualized detection of miRNA-182 in microsample.These findings suggest a robust way for sensitive detection and accurate diagnosis/prognosis of glioma.Dong Liang Xiaoyi Zhang Yi Wang Taotao Huo Min Qian Yibo Xie Wenshuai Li Yunqiu Yu Wei Shi Qianqian Liu Junle Zhu Chun Luo Zhijuan Cao Rongqin Huang 2022Bioactive Materials2022,7,8:1

9A CMOS 4-Element Ku-Band Phased-Array Transceiver显示文摘This paper presents a Ku-Band fully differential 4-element phased-array transceiver using a standard 180-nm CMOS process.Each transceiver is integrated with a 5-bit phase shifter and 4-bit attenuator for high-resolution radiation manipulation.The front-end system adopts time-division mode,and hence two low-loss T/R switches are included in each channel.At room temperature,the measured root-mean-square(RMS)phase error is less than 5.5°.Furthermore,the temperature influence on passive switched phase shifters is analyzed.Meanwhile,an extra phase-shifting cell is developed to calibrate phase error varying with the operating temperatures.With the calibration,the RMS phase error is reduced by 7°at-45℃,and 5.4°at 85℃.The RMS amplitude error is less than 0.92 dB at 15-18 GHz.In the RX mode,the tested gain is 9.6±1.1 dB at 16.5 GHz with a noise figure of 10.9 dB,and the input P1dB is-15 dBm,while the single-channel's gain and output P1dB in the TX mode are 11.3±0.4 dB and 9.4 dBm at 16.1 GHz,respectively.The whole chip occupies an area of 5×4.2 mm^(2) and the measured isolation between each two adjacent channels is lower than-23.1 dB.ZHANG Xiaoning YU Yiming ZHAO Chenxi LIU Huihua WU Yunqiu KANG Kai 2022Chinese Journal of Electronics2022,31,6:0

11Functional flax fiber with UV-induced switchable wettability for multipurpose oil-water separation显示文摘The large number of oily wastewater discharges and oil spills are bringing about severe threats to environment and human health.Corresponding to this challenge,a functional PAA-ZnO-HDTMS flax fiber with UV-induced switchable wettability was developed for efficient oil-water separation in this study.The developed flax fiber was obtained through PAA grafted polymerization and then ZnO-HDTMS nanocomposite immobilization.The as-prepared PAA-ZnO-HDTMS flax fiber was hydrophobic initially and could be switched to hydrophilic through UV irradiation.Its hydrophobicity could be easily recovered through being stored in dark environment for several days.To optimize the performance of the PAA-ZnO-HDTMS flax fiber,the effects of ZnO and HDTMS concentrations on its switchable wettability were investigated.The optimized PAA-ZnO-HDTMS flax fiber had a large water contact angle(∼130°)in air and an extremely small oil contact angle(∼0°)underwater initially.After UV treatment,the water contact angle was decreased to 30°,while the underwater oil contact angle was increased to more than 150°.Based on this UV-induced switchable wettability,the developed PAA-ZnO-HDTMS flax fiber was applied to remove oil from immiscible oil-water mixtures and oil-in-water emulsion with great reusability for multiple cycles.Thus,the developed flax fiber could be further fabricated into oil barrier or oil sorbent for oil-water separation,which could be an environmentally-friendly alternative in oil spill response and oily wastewater treatment.Xiujuan Chen Yunqiu Liu Gordon Huang Chunjiang An Renfei Feng Yao Yao Wendy Huang Shuqing Weng 2022Frontiers of Environmental Science & Engineering2022,16,12:0

15The complete and fully-phased diploid genome of a male Han Chinese显示文摘Since the release of the complete human genome,the priority of human genomic study has now been shifting towards closing gaps in ethnic diversity.Here,we present a fully phased and well-annotated diploid human genome from a Han Chinese male individual(CN1),in which the assemblies of both haploids achieve the telomere-to-telomere(T2T)level.Comparison of this diploid genome with the CHM13 haploid T2T genome revealed significant variations in the centromere.Outside the centromere,we discovered 11,413 structural variations,including numerous novel ones.We also detected thousands of CN1 alleles that have accumulated high substitution rates and a few that have been under positive selection in the East Asian population.Further,we found that CN1 outperforms CHM13 as a reference genome in mapping and variant calling for the East Asian population owing to the distinct structural variants of the two references.Comparison of SNP calling for a large cohort of 8869 Chinese genomes using CN1 and CHM13 as reference respectively showed that the reference bias profoundly impacts rare SNP calling,with nearly 2 million rare SNPs miss-called with different reference genomes.Finally,applying the CN1 as a reference,we discovered 5.80 Mb and 4.21 Mb putative introgression sequences from Neanderthal and Denisovan,respectively,including many East Asian specific ones undetected using CHM13 as the reference.Our analyses reveal the advances of using CN1 as a reference for population genomic studies and paleo-genomic studies.This complete genome will serve as an alternative reference for future genomic studies on the East Asian population.Chentao Yang Yang Zhou Yanni Song Dongya Wu Yan Zeng Lei Nie Panhong Liu Shilong Zhang Guangji Chen Jinjin Xu Hongling Zhou Long Zhou Xiaobo Qian Chenlu Liu Shangjin Tan Chengran Zhou Wei Dai Mengyang Xu Yanwei Qi Xiaobo Wang Lidong Guo Guangyi Fan Aijun Wang Yuan Deng Yong Zhang Jiazheng Jin Yunqiu He Chunxue Guo Guoji Guo Qing Zhou Xun Xu Huanming Yang Jian Wang Shuhua Xu Yafei Mao Xin Jin Jue Ruan Guojie Zhang 2023Cell Research2023,33,10:0
