| 4 | Indoor Allergen Levels and Household Distributions in Nine Cities Across China显示文摘Objective Chinese allergic subjects have high levels of sensitization to house dust mite(HDM) and other indoor allergens. This study quantifies common indoor allergen levels in Chinese households. Methods Dust samples were collected from nine cities. Major allergens Der p 1 and Der f 1 from Dermatophagoides pteronyssinus and D. farinae, and specific antigens of Blomia tropicalis, Tyrophagus putrescentiae, Acarus siro, and cockroach species Blattella germanica and Periplaneta americana were measured by ELISA. Results HDM allergens were found in dust samples from bedding in 95% of the Chinese households. The median levels varied from <0.006 to 9.2 μg/g of dust, depending on the city. The percentages of households having HDM allergen levels associated with the risk of developing allergy sensitization and asthma were 65% and 25%, respectively. Specific antigens of the storage mite and cockroach were only found in samples from the southern and tropical regions of China. Levels of mite allergens were generally higher in samples from bedding compared to samples from the living room, even for storage mites, whereas levels of cockroach antigens were higher in the living room samples. Conclusion HDM allergens are present in bedding dust samples from most Chinese households. Cities in southern and central China have relatively high levels of HDM major allergens compared to cities in northern and western China. | 5 | Establishment of the diploid gynogenetic hybrid clonal line of red crucian carp × common carp显示文摘This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2) of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes. Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. | 6 | Discrimination of abiogenic and biogenic alkane gases显示文摘We have combined the analytical data of the carbon isotope distribution pattern, R/Ra and CH4/3He values of abiogenic and biogenic (referring to the thermogenic and bacterial or microbial) alkane gases in China with those of alkane gases from USA, Russia, Germany, Australia and other countries. | 8 | Indirect mineral carbonation of titanium-bearing blast furnace slag coupled with recovery of TiO_2 and Al_2O_3显示文摘Large quantities of CO_2 and blast furnace slag are discharged in the iron and steel industry. Mineral carbonation of blast furnace slag can offer substantial CO_2 emission reduction and comprehensive utilization of the solid waste.This paper describes a novel route for indirect mineral carbonation of titanium-bearing blast furnace(TBBF) slag,in which the TBBF slag is roasted with recyclable(NH_4)_2SO_4(AS) at low temperatures and converted into the sulphates of various valuable metals, including calcium, magnesium, aluminium and titanium. High value added Ti-and Al-rich products can be obtained through stepwise precipitation of the leaching solution from the roasted slag. The NH_3 produced during the roasting is used to capture CO_2 from flue gases. The NH_4HCO_3 and(NH_4)_2CO_3 thus obtained are used to carbonate the CaSO_4-containing leaching residue and MgSO_4-rich leaching solution, respectively. In this study, the process parameters and efficiency for the roasting, carbonation and Ti and Al recovery were investigated in detail. The results showed that the sulfation ratios of calcium,magnesium, titanium and aluminium reached 92.6%, 87% and 84.4%, respectively, after roasting at an AS-to-TBBF slag mass ratio of 2:1 and 350 °C for 2 h. The leaching solution was subjected to hydrolysis at 102 °C for 4 h with a Ti hydrolysis ratio of 95.7% and the purity of TiO_2 in the calcined hydrolysate reached 98 wt%.99.7% of aluminium in the Ti-depleted leaching solution was precipitated by using NH_3. The carbonation products of Ca and Mg were CaCO_3 and(NH_4)_2 Mg(CO_3)_2·4H_2O, respectively. The latter can be decomposed into MgCO_3 at 100–200 °C with simultaneous recovery of the NH_3 for reuse. In this process, approximately 82.1% of Ca and 84.2%of Mg in the TBBF slag were transformed into stable carbonates and the total CO_2 sequestration capacity per ton of TBBF slag reached up to 239.7 kg. | 9 | The Impacts of Mosquito Density and Meteorological Factors on Dengue Fever Epidemics in Guangzhou, China, 2006-2014: a Time-series Analysis显示文摘Objective To explore the associations between the monthly number of dengue fever(DF) cases and possible risk factors in Guangzhou, a subtropical city of China. Methods The monthly number of DF cases, Breteau Index(BI), and meteorological measures during 2006-2014 recorded in Guangzhou, China, were assessed. A negative binomial regression model was used to evaluate the relationships between BI, meteorological factors, and the monthly number of DF cases. Results A total of 39,697 DF cases were detected in Guangzhou during the study period. DF incidence presented an obvious seasonal pattern, with most cases occurring from June to November. The current month's BI, average temperature(Tave), previous month's minimum temperature(Tmin), and Tave were positively associated with DF incidence. A threshold of 18.25 °C was found in the relationship between the current month's Tmin and DF incidence. Conclusion Mosquito density, Tave, and Tmin play a critical role in DF transmission in Guangzhou. | 10 | Effect of Jinhua Qinggan granules on novel coronavirus pneumonia in patients显示文摘OBJECTIVE: To evaluate the effectiveness and safety of Jinhua Qinggan granules in the treatment of patients with novel coronavirus pneumonia(COVID-19).METHODS: Eighty cases of COVID-19 diagnosed from January 24 to February 17, 2020 in Beijing YouAn Hospital Affiliated to Capital Medical University were retrospectively analyzed. All 80 patients received symptomatic and supportive treatment.Among them, 44 patients took Jinhua Qinggan granules(treatment group) within 24 h of admission, and the remaining 36 patients either did not take Jinhua Qinggan granules or took the granules for less than 2 d(control group). In this study, we compared the duration of viral nucleic acid detection and of pneumonia absorption improvement between the two groups.RESULTS: Among the 80 cases, 37 were male(46%)and 43 were female(54%) with age ranging from15 to 86 years, with an average age of 51.19 years.The average duration of viral nucleic acid detection was(7 ± 4) d in the Jinhua Qinggan administration group and(10 ± 4) d for the control group(P =0.010), following which, nucleic acid tests were negative. Of the two groups, 56.82% in the Jinhua Qinggan treatment group and 27.78% in the control group demonstrated negative nucleic acid tests within 7 d or less. The 7-day viral clearance rate was significantly higher in the Jinhua Qinggan group compared with the control group(P = 0.009). Furthermore, the pneumonia recovery time indicated by chest CT was(8 ± 4) d in the Jinhua Qinggan group, which was significantly shorter than the control group, at(10 ± 5) d(P = 0.021). | 12 | A heavy mineral viewpoint on sediment provenance and environment in the Qiongdongnan Basin显示文摘Based on heavy mineral data in core samples from eleven drillings, supplemented by paleontological, element geochemical and seismic data, the evolution of sediment provenance and environment in the Qiongdongnan Basin(QDNB) was analysed. The results show that the basement in the QDNB was predominantly composed of terrigenous sediments. Since the Oligocene the QDNB has gradually undergone transgressions and evolution processes in sedimentary environment from terrestrial-marine transitional to littoral-neritic, neritic, and bathyal roughly. The water depth showed a gradually increasing trend and was generally greater in the southern region than that in the northern region in the same time. With changes in sedimentary environment, provenances of the strata(from the Yacheng Formation to the Yinggehai Formation) showed principal characteristics of multisources, evolving from autochthonous source, short source to distant source step by step. During the Early Oligocene, the sediments were mainly proximal basaltic pyroclastic source and adjacent terrigenous clastic source, afterwards were becoming distant terrigenous clastic sources, including Hainan Island on the north,Yongle Uplift on the south, Shenhu Uplift on the northeast, the Red River System on the northwest and Indochina Peninsula on the southwest, or even a wider region. The Hainan Island provenance began to develop during the Early Oligocene and has become a main provenance in the QDNB since the Middle Miocene. The provenances from Yongle Uplift and Shenhu Uplift most developed from the Late Oligocene to the Early Miocene and gradually subsided during the Middle Miocene. During the Late Miocene, as a main source of sediments filled in the central canyon, the Red River System provenance added to the QDNB massively, whose impact terminated at the end of the Pliocene. The western Yinggehai Basin(YGHB) provenance derived from Indochina Peninsula had developed from the Pliocene on to the Pleistocene. | 14 | Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation. | BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin | 2014 | Science China(Life Sciences)2014,57,2: | 9 |
| 15 | β-arrestin 2 attenuates lipopolysaccharide-induced liver injury via inhibition of TLR4/NF-κB signaling pathwaymediated inflammation in mice显示文摘AIM To study the role and the possible mechanism of β-arrestin 2 in lipopolysaccharide(LPS)-induced liver injury in vivo and in vitro.METHODS Male β-arrestin 2^(+/+) and β-arrestin 2^(-/-)C57 BL/6 J mice were used for in vivo experiments, and the mouse macrophage cell line RAW264.7 was used for in vitro experiments. The animal model was established via intraperitoneal injection of LPS or physiological sodium chloride solution. Blood samples and liver tissues were collected to analyze liver injury and levels of pro-inflammatory cytokines. Cultured cell extracts were collected to analyze the production of pro-inflammatory cytokines and expression of key molecules involved in the TLR4/NF-κB signaling pathway.RESULTS Compared with wild-type mice, the β-arrestin 2 knockout mice displayed more severe LPS-induced liver injury and significantly higher levels of proinflammatory cytokines, including interleukin(IL)-1β, IL-6, tumor necrosis factor(TNF)-α, and IL-10. Compared with the control group, pro-inflammatory cytokines(including IL-1β, IL-6, TNF-α, and IL-10) produced by RAW264.7 cells in the β-arrestin 2 si RNA group were significantly increased at 6 h after treatment with LPS. | 17 | Criteria for hypersonic airbreathing propulsion and its experimental verification显示文摘Hypersonic airbreathing propulsion is one of the top techniques for future aerospace flight, but there are still no practical engines after seventy years’ development. Two critical issues are identified to be the barriers for the ramjet-based engine that has been taken as the most potential concept of the hypersonic propulsion for decades. One issue is the upstream-traveling shock wave that develops from spontaneous waves resulting from continuous heat releases in combustors and can induce unsteady combustion that may lead to engine surging during scramjet engine operation.The other is the scramjet combustion mode that cannot satisfy thrust needs of hypersonic vehicles since its thermos-efficiency decreases as the flight Mach number increases. The two criteria are proposed for the ramjet-based hypersonic propulsion to identify combustion modes and avoid thermal choking. A standing oblique detonation ramjet(Sodramjet) engine concept is proposed based on the criteria by replacing diffusive combustion with an oblique detonation that is a unique pressure-gain phenomenon in nature. The Sodramjet engine model is developed with several flow control techniques, and tested successfully with the hypersonic flight-duplicated shock tunnel.The experimental data show that the Sodramjet engine model works steadily, and an oblique detonation can be made stationary in the engine combustor and is controllable. | 18 | Formation and biological characterization of three new types of improved crucian carp显示文摘The improved tetraploids (G1×AT) were obtained by distant crossing and gynogenesis and the high-body individuals accounted for 2% among G1×AT. After mating with each other, the high-body in- dividuals produced three kinds of bisexual fertile diploid fishes: high-body red crucian carp, high-body fork-like-tails goldfish and gray common carp. The high-body red crucian carp mating with each other formed three types of improved crucian carp (ICC) including improved red crucian carp (IRCC), im- proved color crucian carp (ICCC) and improved gray crucian carp (IGCC). The phenotypes, chromo- some numbers, gonadal structure and fertility of the three kinds of ICC and their offspring were observed. All the three kinds of ICC possessed some improved phenotypes such as higher body, smaller head and shorter tail. The ratios of the body height to body length of IRCC, ICCC and IGCC were 0.54, 0.51 and 0.54, respectively. All of them were obviously higher than that of red crucian carp 0.41 (P<0.01). Three kinds of ICC had the same chromosome number as red crucian carp with 100 chromosomes. All the ICC possessed normal gonads producing mature eggs or sperm, which was important for the production of an improved diploid population. Compared with red crucian carp, all the ICC had stronger fertility such as higher gametes production, higher fertilization rate and higher hatchery rate. Three types of improved diploid fish population were generated from the three kinds of ICC by self-crossing, respectively. The ICC can serve as ornamental fish and edible fish. They are also ideal parents to produce triploids by mating with tetraploids. Only recently, with the rapid development of information technologies and the increasing accumulation of data, human beings can learn more about the unknown world with data-driven methods. Given data timeliness, there is a growing awareness of the importance of real-time data. There are two categories of technologies accounting for data processing: batching big data and streaming processing, which have not been integrated well. Thus,we propose an innovative incremental processing technology named after Stream Cube to process both big data and stream data. Also, we implement a real-time intelligent data processing system, which is based on real-time acquisition, real-time processing, real-time analysis, and real-time decision-making. The real-time intelligent data processing technology system is equipped with a batching big data platform, data analysis tools, and machine learning models.
