| 3 | Phase I trial of human umbilical cord-derived mesenchymal stem cells for treatment of severe bronchopulmonary dysplasia显示文摘Severe bronchopulmonary dysplasia(BPD)is a chronic lung disorder that primarily affects premature babies with extremely low birth weight and involves in multiple organ system;no effective pharmacotherapy for this disease exists,and mortality remains high.Based on the evidence from previous preclinical studies and phase I clinical trials,this study aims to test the safety of intravenous application of a single dose of human umbilical cord-derived mesenchymal stem cells(hUC-MSCs)in patients with severe BPD.The Mesenchymal Stem cells for Bronchopulmonary Dysplasia Treatment(MSBDT)trial is a single center,open-label,dose-escalation phase I clinical trial.Severe BPD patients were enrolled in Children Hospital of Chongqing Medical University,Chongqing,China.The first six patients were treated with low-dose hUC-MSCs(1×10^(6) cells/kg)and the next seven patients were treated with high-dose hUC-MSCs(5×10^(6) cells/kg).This study is registered with ClinicalTrials.gov,number NCT03558334.No prespecified infusion-associated adverse events,immediate complication,respiratory or cardiovascular compromise were observed during infusion and 24 h after infusion.No significant changes in safety laboratory values were observed.One death event occurred in the low-dose group on study day 10,and one death event occurred in the high-dose group on study day 24,while,after review in detail,the two cases are not believed to be infusion-associated events.In conclusion,intravenous application of a single dose of hUC-MSCs was tolerated in thirteen patients with severe BPD. | Yunqiu Xia Tingyuan Lang Yuqin Niu Xian Wu Ou Zhou Jihong Dai Lei Bao Ke Yang Lin Zou Zhou Fu Gang Geng | 2023 | Genes & Diseases2023,10,2: | 1 |
| 4 | Climate Warming Mitigation from Nationally Determined Contributions显示文摘Individual countries are requested to submit nationally determined contributions(NDCs)to alleviate global warming in the Paris Agreement.However,the global climate effects and regional contributions are not explicitly considered in the countries’decision-making process.In this study,we evaluate the global temperature slowdown of the NDC scenario(ΔT=0.6°C)and attribute the global temperature slowdown to certain regions of the world with a compact earth system model.Considering reductions in CO_(2),CH_(4),N_(2)O,BC,and SO_(2),the R5OECD(the Organization for Economic Co-operation and Development in 1990)and R5ASIA(Asian countries)are the top two contributors to global warming mitigation,accounting for 39.3%and 36.8%,respectively.R5LAM(Latin America and the Caribbean)and R5MAF(the Middle East and Africa)followed behind,with contributions of 11.5%and 8.9%,respectively.The remaining 3.5%is attributed to R5REF(the Reforming Economies).Carbon Dioxide emission reduction is the decisive factor of regional contributions,but not the only one.Other greenhouse gases are also important,especially for R5MAF.The contribution of short-lived aerosols is small but significant,notably SO_(2)reduction in R5ASIA.We argue that additional species beyond CO_(2)need to be considered,including short-lived pollutants,when planning a route to mitigate climate change.It needs to be emphasized that there is still a gap to achieve the Paris Agreement 2-degree target with current NDC efforts,let alone the ambitious 1.5-degree target.All countries need to pursue stricter reduction policies for a more sustainable world. | Bo FU Jingyi LI Thomas GASSER Philippe CIAIS Shilong PIAO Shu TAO Guofeng SHEN Yuqin LAI Luchao HAN Bengang LI | 2022 | Advances in Atmospheric Sciences2022,39,8: | 1 |
| 5 | pPeOp inhibits HGC-27 cell proliferation,migration and invasion by upregulating miR-30b-5p and down-regulating the Rac1/Cdc42 pathway显示文摘Gastric cancer is the fifth most frequently occurring and the fourth most lethal malignant cancer worldwide.A bioactive protein(pPeOp)from Omphalia lapidescens exhibits significant inhibitory effects on gastric cancer cells.miRNA deep sequencing analysis shows that miR-30b-5p is significantly upregulated in HGC-27 cells treated with pPeOp.Verification results show that the expression level of miR-30b-5p is significantly increased in HGC-27 cells after pPeOp treatment.Additionally,miR-30b-5p is significantly downregulated in clinical gastric cancer tissues compared to that in adjacent normal tissues.Following pPeOp treatment and/or transfection with miR-30b-5p mimic,the proliferation,migration,and invasion of HGC-27 cells are significantly impaired.Immunofluorescence microscopy shows that pPeOp and/or miR-30b-5p destroy(s)microfilaments and microstructures and inhibit(s)the formation of pseudopodia.Bioinformatics analysis,dual-luciferase reporter assay,and western blot analysis confirm that miR-30b-5p downregulates Rac1/Cdc42 expression and activation by targeting RAB22A.Available data indicate that miR-30b-5p plays an anti-gastric cancer role in mediating pPeOp.pPeOp upregulates miR-30b-5p expression,which in turn inhibits RAB22A expression,resulting in a reduction in the expression and activation of Rac1 and Cdc42 and their downstream targets,thus destroying the cytoskeletal structure and inhibiting the proliferation,migration,and invasion of cancer cells. | Wenjun Xu Zhenjie Fu Yuqin Xu Man Hei Cheung Yan Chen Meiai Lin Hang Wen Hang Lv Chun Liang Jianshu Lou Yitao Chen | 2022 | Acta Biochimica et Biophysica Sinica2022,54,12: | 0 |

| 8 | Therapeutic potential of an anti-HER2 single chain antibody-DM1 conjugates for the treatment of HER2-positive cancer显示文摘Antibody–drug conjugates(ADCs)take the advantage of monoclonal antibodies to selectively deliver highly potent cytotoxic drugs to tumor cells,which have become a powerful measure for cancer treatment in recent years.To develop a more effective therapy for human epidermal growth factor receptor 2(HER2)-positive cancer,we explored a novel ADCs composed of anti-HER2 scFv–HSA fusion antibodies conjugates with a potent cytotoxic drug DM1.The resulting ADCs,T-SA1–DM1 and T-SA2–DM1(drug-to-antibody ratio in the range of 3.2–3.5)displayed efficient inhibition in the growth of HER2-positive tumor cell lines and the half-maximal inhibitory concentration on SKBR-3 and SKOV3 cells were both at the nanomolar levels in vitro.In HER2-positive human ovarian cancer xenograft models,T-SA1–DM1 and T-SA2–DM1 also showed remarkable antitumor activity.Importantly,three out of six mice exhibited complete remission without regrowth in the high-dose group of T-SA1–DM1.On the basis of the analysis of luminescence imaging,anti-HER2 scFv–HSA fusion antibodies,especially T-SA1,showed strong and rapid tumor tissue penetrability and distribution compared with trastuzumab.Collectively,the novel type of ADCs is effective and selective targeting to HER2-positive cancer,and may be a promising antitumor drug candidate for further studies. | Hang Zhang Yuxi Wang Yangping Wu Xiaohua Jiang Yiran Tao Yuqin Yao Yujia Peng Xiangzheng Chen Yuyin Fu Lin Yu Ruixue Wang Qinhuai Lai Weirong Lai Wenting Li Yuhuan Kang Shuli Yi Ying Lu Lantu Gou Min Wu Jinliang Yang | 2017 | Signal Transduction and Targeted Therapy2017,2,1: | 0 |
| 9 | Tprn is essential for the integrity of stereociliary rootlet in cochlear hair cells in mice显示文摘Tprn encodes the taperin protein,which is concentrated in the tapered region of hair cell stereocilia in the inner ear.In humans,TPRN mutations cause autosomal recessive nonsyndromic deafness(DFNB79)by an unknown mechanism.To determine the role of Tprn in hearing,we generated Tprn-null mice by clustered regularly interspaced short palindromic repeat/Cas9 genome-editing technology from a CBA/CaJ background.We observed significant hearing loss and progressive degeneration of stereocilia in the outer hair cells of Tprn-null mice starting from postnatal day 30.Transmission electron microscopy images of stereociliary bundles in the mutant mice showed some stereociliary rootlets with curved shafts.The central cores of the stereociliary rootlets possessed hollow structures with surrounding loose peripheral dense rings.Radixin,a protein expressed at stereocilia tapering,was abnormally dispersed along the stereocilia shafts in Tprn-null mice.The expression levels of radixin andβ-actin significantly decreased.We propose that Tprn is critical to the retention of the integrity of the stereociliary rootlet.Loss of Tprn in Tprn-null mice caused the disruption of the stereociliary rootlet,which resulted in damage to stereociliary bundles and hearing impairments.The generated Tprn-null mice are ideal models of human hereditary deafness DFNB79. | Yuqin Men Xiujuan Li Hailong Tu Aizhen Zhang Xiaolong Fu Zhishuo Wang Yecheng Jin Congzhe Hou Tingting Zhang Sen Zhang Yichen Zhou Boqin Li Jianfeng Li Xiaoyang Sun Haibo Wang Jiangang Gao | 2019 | Frontiers of Medicine2019,13,6: | 0 |
