1Synergistic effects of magnesium ions and simvastatin on attenuation of high-fat diet-induced bone loss显示文摘Introduction:Magnesium(Mg)has a prophylactic potential against the onset of hyperlipidemia.Similar to statin,Mg is recommended as lipid-lowering medication for hypercholesterolemia and concomitantly exhibits an association with increased bone mass.The combination of statin with Mg ions(Mg^(2+))may be able to alleviate the high-fat diet(HFD)-induced bone loss and reduce the side-effects of statin.This study aimed to explore the feasibility of combined Mg^(2+)with simvastatin(SIM)for treating HFD-induced bone loss in mice and the involving mechanisms.Materials and methods:C57BL/6 male mice were fed with a HFD or a normal-fat diet(NFD).Mice were intraperitoneally injected SIM and/or orally received water with additional Mg^(2+)until sacrificed.Enzyme-linked immunosorbent assay was performed to measure cytokines and cholesterol in serum and liver lysates.Bone mineral density(BMD)and microarchitecture were assessed by micro-computed tomography(μCT)in different groups.The adipogenesis in palmitate pre-treated HepG2 cells was performed under various treatments.Results:μCT analysis showed that the trabecular bone mass was significantly lower in the HFD-fed group than that in NFD-fed group since week 8.The cortical thickness in HFD-fed group had a significant decrease at week 24,as compared with NFD-fed group.The combination of Mg^(2+)and SIM significantly attenuated the trabecular bone loss in HFD-fed mice via arresting the osteoclast formation and bone resorption.Besides,such combination also reduced the hepatocytic synthesis of cholesterol and inhibited matrix metallopeptidase 13(Mmp13)mRNA expression in pre-osteoclasts.Conclusions:The combination of Mg^(2+)and SIM shows a synergistic effect on attenuating the HFD-induced bone loss.Our current formulation may be a cost-effective alternative treatment to be indicated for obesity-related bone loss.Bingyang Dai Xu Li Jiankun Xu Yuwei Zhu Le Huang Wenxue Tong Hao Yao Dick Ho-kiu Chow Ling Qin 2021Bioactive Materials2021,6,8:4
2Single cell analysis reveals inhibition of angiogenesis attenuates the progression of heterotopic ossification in Mkx^(−/−)mice显示文摘Tendon heterotopic ossification(HO)is characterized by bone formation inside tendon tissue,which severely debilitates people in their daily life.Current therapies fail to promote functional tissue repair largely due to our limited understanding of HO pathogenesis.Here,we investigate the pathological mechanism and propose a potential treatment method for HO.Immunofluorescence assays showed that the Mohawk(MKX)expression level was decreased in human tendon HO tissue,coinciding with spontaneous HO and the upregulated expression of osteochondrogenic and angiogenic genes in the tendons of Mkx^(−/−)mice.Single-cell RNA sequencing analyses of wild-type and Mkx^(−/−)tendons identified three cell types and revealed the excessive activation of osteochondrogenic genes during the tenogenesis of Mkx^(−/−)tendon cells.Single-cell analysis revealed that the gene expression program of angiogenesis,which is strongly associated with bone formation,was activated in all cell types during HO.Moreover,inhibition of angiogenesis by the small-molecule inhibitor BIBF1120 attenuated bone formation and angiogenesis in the Achilles tendons of both Mkx mutant mice and a rat traumatic model of HO.These findings provide new insights into the cellular mechanisms of tendon HO and highlight the inhibition of angiogenesis with BIBF1120 as a potential treatment strategy for HO.Junxin Lin Yuwei Yang Wenyan Zhou Chao Dai Xiao Chen Yuanhao Xie Shan Han Huanhuan Liu Yejun Hu Chenqi Tang Varitsara Bunpetch Dandan Zhang Yishan Chen Xiaohui Zou Di Chen Wanlu Liu Hongwei Ouyang 2022Bone Research2022,10,4:1
3Estimating blue marlin(Makaira nigricans)sustainable yield in the Indian Ocean using a data-poor approach显示文摘Blue marlin(Makaira nigricans)is a common bycatch species in the global tuna longline fishery.In this study,we applied a common data-poor approach,i.e.,depletion-corrected average catch(DCAC)to assess stock status of the Indian Ocean blue marlin.Sustainable yield(Ysust),one reference point in this case,was estimated,and its uncertainty was integrated by using Monte Carlo simulation.The results revealed the estimate of Ysust by DCAC was lower than MSY of 11,926 t by BSP-SS and is also lower than the provisional reference point of 11,704 t by the management proposal.DCAC is reliable for blue marlin in driving precautionary management quantity based on the CPUE of Japan(1980–2015).This study also implies that DCAC could be applied to other billfish stocks and uncertainty be estimated for sustainable yield.However,data-poor methods could be adjusted with precautionary approaches.Yuwei Fan Zhe Geng Jiangfeng Zhu Xiaojie Dai Kindong Richard 2019Aquaculture and Fisheries2019,4,3:1
4Motion direction prediction through spike timing based on micro Capsnet networks显示文摘Neural activity extraction and neural decoding from neural signals are an important part of critical components of brain-computer interface systems.With the development of brain-computer interface technology,the demand for precise external control and nervous activities in macaque monkey during unilateral hand grasp has increased the complexity of control and neural decoding,which puts forward higher requirements for the accuracy and stability of feature extraction and neural decoding.In this study,a micro Capsnet network architecture that consists of a few network layers,a vector feature structure,and optimization network parameters,is proposed to decrease the computing time and complexity,decrease artificial debugging,and improve the decoding accuracy.Compared with KNN,SVM,XGBOOST,CNN,Simple RNN,and LSTM,the algorithm in this study improves the decoding accuracy by 98.03%,and achieves state-of-the-art accuracy and stronger robustness.Furthermore,the proposed algorithm can further enhance the control accuracy in the brain-computer interface.ZHANG HuaLiang LIU Ji WANG BaoZeng DAI Jun LIAN JinLing KE Ang ZHAO YuWei ZHOU Jin WANG ChangYong 2022Science China(Technological Sciences)2022,65,11:1

6Dynamics and biological relevance of epigenetic N^(6)-methyladenine DNA modification in eukaryotic cells显示文摘DNA methylation represents a major type of DNA modifications that play key roles in diverse biological processes.With the recent development of highly selective and sensitive bioanalytical techniques,N^(6)-methyladenine(6mA)has been characterized as an important internal DNA modification dynamically occurring in multiple eukaryotes including humans.Increasing evidence has indicated that 6mA may act as a novel epigenetic modification involved in regulation of development,stress response and diseases such as cancer and neurodegenerative disorders.We review herein the recent advances in the detection and functional studies of 6mA modification,with special emphasis on its biological consequences and human health relevance as well as its dynamic regulation by various types of methyltransferases,demethylases and 6mA-binding proteins.It can be envisaged that further chemical and biological studies of 6mA modification will lead to a better understanding about its potentially important roles in normal and pathological biological processes.Yuwei Sheng Meijuan Zhou Changjun You Xiaoxia Dai 2022Chinese Chemical Letters2022,33,5:0
7Next-generation sequencing-based analysis of the effect of N^(6)-methyldeoxyadenosine modification on DNA replication in human cells显示文摘N^(6)-methyldeoxyadenosine(6 mdA) modification is considered as a new epigenetic mark that may play important roles in various biological processes.However,it remains unclear about the effect of 6 mdA on DNA replication in human cells.Herein,we combined next-generation sequencing with shuttle vector technology to explore how 6 mdA affects the efficiency and accuracy of DNA replication in human cells.Our results showed that 6 mdA neither blocked DNA replication nor induced mutations in human cells.Moreover,we found that the depletion of translesion synthesis DNA polymerase(Pol) κ,Pol η,Pol ι or Pol ζ did not significantly change the biological consequences of 6 mdA during replication in human cells.The negligible impact of 6 mdA on DNA replication is consistent with its potential role in epigenetic gene expression.Juan Wang Yuwei Sheng Ying Yang Xiaoxia Dai Changjun You 2022Chinese Chemical Letters2022,33,4:0
8Development of window opening models for residential building in hot summer and cold winter climate zone of China显示文摘This study monitors the open/closed states of the windows in 16 residential apartments in representative cities of the hot summer and cold winter zone of China,along with outdoor air temperature,outdoor relative humidity and outdoor PM2.5 concentration in 2017,to investigate the window opening/closing behavior.Three window opening models are developed.All the models are validated by modeling a typical residential apartment to compare the predicted indoor CO_(2)concentration with monitored data.It is found that the window opening probability is negatively correlated to both outdoor PM2.5 concentration and outdoor relative humidity,and the probability of window opening reaches the peak value when the outdoor air temperature is 23.7℃.In terms of season,the shortest window opening duration occurs in winter due to the cold outdoor air temperature and high outdoor PM2.5 concentration.Finally,by comparing the correlation between the measured date and simulation results of indoor CO_(2)concentration,this study provides an appropriate window opening model for reference.Pengzhi Zhou Haidong Wang Fei Li Yuwei Dai Chen Huang 2022Energy and Built Environment2022,3,3:0

10Combining in-situ TEM observations and theoretical calculation for revealing the thermal stability of CeO_(2) nanoflowers显示文摘The thermal stability of Ce_(2) nanomaterials can directly impact both the uniformity of the supported catalysts and the catalytic behavior of Ce_(2) itself.However,knowledge about the thermal stability of Ce_(2) is still deficient.Here,we conduct in-situ transmission electron microscopy experiments and theoretical calculations to elucidate the thermal stability of Ce_(2) nanomaterials under different environments.A sinter(<700℃)and a structural decomposition(>700℃)are observed within Ce_(2) nanoflowers under O_(2).The sinter firstly occurs among the nanoflowers’monomers and then the sintered nanoparticles structurally decompose to tiny nanoparticles from the strain interface.Under a vacuum environment,the Ce_(2) nanoflowers firstly undergo a transition from cubic fluorite Ce_(2) to hexagonal Ce_(2)O_(3),accompanied by the oxygen release.The Ce_(2)O_(3) nanoparticles further atomically sublimate from the edges to the center under high temperatures.Theoretical calculation results reveal a considerably lower energy barrier for the structural decomposition under O_(2) and for the sublimation under vacuum.This work provides a perspective on the structural design and performance optimization of Ce_(2)-based catalysts.Mingyun Zhu Kuibo Yin Yifeng Wen Shugui Song Yuwei Xiong Yunqian Dai Litao Sun 2022Nano Research2022,15,2:0
