| 2 | Comparison of labeling methods and time course of traumatic brain injury-induced cell death in mice显示文摘BACKGROUND:Various molecular mechanisms of cell death following traumatic brain injury have been previously described.However,the time course of cell death remains unclear.TUNEL and Fluoro-Jade B labeling have been widely used to label apoptotic cells and neuronal degeneration.Propidium iodide (PI) functions as a biomarker of cell death in vivo.OBJECTIVE:To explore the role of PI labeling compared to TUNEL and Fluoro-Jade B staining for detecting neural cell death,and to observe time course of traumatic brain injury-induced cell death in mice.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Laboratory of Aging and Nervous Diseases,Soochow University from September 2007 to December 2008.MATERIALS:PI (B1221) was purchased from Sigma,USA.TUNEL kit was purchased from Roche Molecular Biochemicals,USA.Fluoro-Jade B was purchased from Chemicon,USA.METHODS:A total of 70 healthy,male,Kunming mice were randomly assigned to sham-surgery (n=5) and model (n=65) groups.Traumatic brain injury was established using the controlled cortical impact method.PI was intraperitoneally injected at 1 hour prior to animal sacrifice.MAIN OUTCOME MEASURES:TUNEL,Fluoro-Jade B,and PI-positive cells were quantified using a double-labeling method to determine the time course of traumatic brain injury-induced cell death.RESULTS:PI labeled cells in an earlier phase of cell death than TUNEL and Fluoro-Jade B labeling.PI-positive cells were observed immediately following injury,and the numbers rapidly increased in injured brain areas at 1 hour,peaked at 24-48 hours,and subsequently decreased at 3-21 days post-injury.TUNEL-labeled cells were significantly increased at 12 hours,while Fluoro-Jade B-labeled cells were increased at 6 hours after injury,with cells still visible at 6-48 hours post-injury.Moreover,a greater number of PI-positive cells were observed compared to TUNEL-and Fluoro-Jade B-labeled cells.CONCLUSION:PI labeling is more sensitive and reliable than TUNEL and Fluoro-Jade B staining for detecting cell death following traumatic brain injury.Moreover,PI labeling can function as a reliable marker to estimate the entire time course of cell death. | Chengliang Luo Xiping Chen Hong Ni Qianqian Li Rui Yang Yuxia Sun Guangyou Zhu Luyang Tao | 2010 | Neural Regeneration Research2010,5,9: | 6 |
| 3 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. | 4 | Associations of muscle mass,strength,and quality with all-cause mortality in China:a population-based cohort study显示文摘Background It remains unclear about the association of muscle mass,strength,and quality with death in the general Chinese population of diverse economical and geographical backgrounds.The present study aimed to comprehensively examine such associations across different regions in China.Methods Based on the China Kadoorie Biobank study,the present study included 23,290 participants who were aged 38 to 88 years and had no prevalent cardiovascular diseases or cancer.Muscle mass and grip strength were measured using calibrated instruments.Arm muscle quality was defined as the ratio of grip strength to arm muscle mass.Low muscle mass,grip strength,and arm muscle quality were defined as the sex-specific lowest quintiles of muscle mass index,grip strength,and arm muscle quality,respectively.Cox proportional hazards models yielded hazard ratios(HRs)and 95%confidence intervals(CIs)for risks of all-cause mortality in relation to muscle mass,strength,and quality.Results During a median follow-up of 3.98 years,739 participants died.The HR(95%CI)of all-cause mortality risk was 1.28(1.08–1.51)for low appendicular muscle mass index,1.38(1.16–1.62)for low total muscle mass index,1.68(1.41–2.00)for low grip strength,and 1.41(1.20–1.66)for low arm muscle quality in models adjusted for sociodemographic characteristics,lifestyle factors,and medical histories.Conclusion Low muscle mass,grip strength,and arm muscle quality are all associated with short-term increased risks of mortality,indicating the importance of maintaining normal muscle mass,strength,and quality for general Chinese adults. | Man Wu Yuxia Wei Jun Lv Yu Guo Pei Pei Jiachen Li Huaidong Du Ling Yang Yiping Chen Xiaohui Sun Hua Zhang Junshi Chen Zhengming Chen Canqing Yu Liming Li 无 | 2022 | Chinese Medical Journal2022,,11: | 2 |

| 14 | Expression of c-erbB2 in Gestational Trophoblastic Disease and Its Clinical Significance显示文摘Summary: In order to explore a potential indicator of predicting the occurrence and development of gestational trophoblastic tumor, the expression of c-erbB2 oncogene in human normal placenta, hydatidiform mole and choriocarcinoma was investigated. The expression of c-erbB2 was detected immunohistochemically by monoclonal antibody against the gene on the formalin-fixed paraffin sections of 21 hydatidiform moles, 21 invasive moles, 20 choriocarcinomas and 30 normal placentas. Results showed that the expression level of c-erbB2 was significantly higher in gestational trophoblastic tumor than in hydatidiform mole and normal placenta of midterm and term pregnancy (P<0.05), while there was no significant difference between patients with gestational trophoblastic tumor of stage Ⅲ, Ⅳ and those of stage Ⅰ, Ⅱ. | 16 | Role of Elevated Thrombospondin-1 in Kainic Acid-Induced Status Epilepticus显示文摘Previous studies have suggested that thrombospondin-1(TSP-1) regulates the transforming growth factor beta 1(TGF-b1)/phosphorylated Smad2/3(p Smad2/3) pathway. Moreover, TSP-1 is closely associated with epilepsy. However, the role of the TSP-1-regulated TGFb1/p Smad2/3 pathway in seizures remains unclear. In this study, changes in this pathway were assessed following kainic acid(KA)-induced status epilepticus(SE) in rats.The results showed that increases in the TSP-1/TGF-b1/p Smad2/3 levels spatially and temporally matched the increases in glial fibrillary acidic protein(GFAP)/chondroitin sulfate(CS56) levels following KA administration.Inhibition of TSP-1 expression by small interfering RNA or inhibition of TGF-b1 activation with a Leu-Ser-Lys-Leu peptide significantly reduced the severity of KA-induced acute seizures. These anti-seizure effects were accompanied by decreased GFAP/CS56 expression and Smad2/3 phosphorylation. Moreover, inhibiting Smad2/3 phosphorylation with ponatinib or SIS3 also significantly reduced seizure severity, alongside reducing GFAP/CS56 immunoreactivity. | 20 | Fabrication of adsorbents with enhanced CuⅠ stability: Creating a superhydrophobic microenvironment through grafting octadecylamine显示文摘In atmospheric conditions, CuⅠis easily oxidized to CuⅡdue to the coexistence of moisture and oxygen.The poor oxidation inhibition of CuⅠrestricts the practical application of CuⅠ-containing materials.Herein we introduce an approach to construct a superhydrophobic microenvironment in CuⅠfunctionalized metal–organic frameworks by coordinatedly grafting organic amine compounds onto open metal sites(OMSs), which can hinder the accessibility of moisture to pores thereby enhancing the stability of CuⅠ. As a proof of concept, MIL-101(Cr) with abundant OMSs and octadecylamine(OA)with long hydrophobic alkyl groups are used as carrier and surface coating. As superhydrophobic porous materials, the resultant CuⅠM-OA exhibits improved CuⅠstability in addition to retaining high crystallinity and intact porosity while almost all CuⅠis oxidized in hydrophilic CuⅠM upon exposure in a humid atmosphere for 30 h. CuⅠM-OA owns excellent adsorption desulfurization performance(ADS) with regard to thiophene, benzothiophene, and 4,6-dimethyl dibenzothiophene. Even from hydrated fuel, the adsorption performance of CuⅠM-OA maintains well while the adsorption capacity of CuⅠM decreases to 7% after4 cycles.
