2Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well.Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang 2021Protein & Cell2021,12,4:12
3Correlation between muscular strength and basal nuclei ischemic/hemorrhagic stroke-induced corticospinal tract injury,as detected by diffusion tensor imaging and tractography显示文摘BACKGROUND:Conventional neuroimaging diagnosis does not assist with the monitoring or evaluation of basal nuclei ischemic and hemorrhagic stroke,or motor functional recovery.Magnetic resonance,diffusion tensor imaging,and diffusion tensor tractography have all been used to observe features of cerebral white matter fibrous structures.In addition,diffusion tensor tractography is the only non-invasive imaging method to display the corticospinal tract in vivo.OBJECTIVE:To evaluate the impairment degree of corticospinal tract induced by basal nuclei ischemic and hemorrhagic stroke through the use of magnetic resonance,diffusion tensor imaging,and diffusion tensor tractography,and to analyze the correlation to muscular strength.DESIGN,TIME AND SETTING:A retrospective case analysis was performed at the Department of Medical Imaging,Neurology and Neurosurgery,Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA between November 2002 and June 2008.PARTICIPANTS:A total of 15 patients with acute or subacute cerebral ischemic stroke and nine with hemorrhagic stroke in the basal nuclei were selected.METHODS:Magnetic resonance,diffusion tensor imaging,and diffusion tensor tractography results and data were analyzed.Fractional anisotropy and directionally encoded color maps were obtained.Three-dimensional tractography of bilateral corticospinal tract was created,and corticospinal tract integrity was graded.Fractional anisotropy of infarct region and corresponding contralateral normal regions were measured,and hematoma volume in hemorrhagic stroke patients was determined.Hand motor function ability was evaluated using Brunstorm criteria.MAIN OUTCOME MEASURES:Fractional anisotropy of infarct region and corresponding contralateral normal regions;hematoma volume in hemorrhagic stroke patients;correlation between muscular strength and corticospinal tract impairment degree in ischemic stroke and hemorrhagic stroke patients before and after treatment.RESULTS:In ischemic stroke patients,the fractional anisotropy value was significantly lower in the infarct area of white matter than in the normal hemisphere(P < 0.01).The impairment degree of corticospinal tract negatively correlated with muscular strength of the corresponding hand(r =-0.97 P < 0.01).The hematoma volume of hemorrhagic stroke patients significantly negatively correlated with Spearman test results for muscular strength of the corresponding hand(r =-0.88,P < 0.01).CONCLUSION:Corticospinal tract impairment severity negatively correlated with muscular strength and motor functional recovery,which suggested that diffusion tensor imaging and diffusion tensor tractography could be used to evaluate corticospinal tract motor function.Shangwen Xu Ziqian Chen Jinhua Chen Youqiang Ye Yuning Lin Ping Ni Hui Xiao Gennian Qian 2010Neural Regeneration Research2010,5,13:7
4Evaluating ischemic stroke with diffusion tensor imaging显示文摘Objective:In recent years,diffusion tensor imaging (DTI) has emerged as a promising tool to study ischemic stroke. The aim of this study is to evaluate the anisotropic changes of cerebral white matter tracks in patients with ischemic stroke using DTI,and investigate the correlation between corticospinal tract damage and muscle strength in such patients during acute ischemia.Method:9 patients with acute ischemic stroke and 9 healthy subjects were examined with T1 weighted magnetic resonance imaging (MRI),T2 weighted MRI and DTI. Fractional anisotropy (FA) was measured and the three-dimensional fibrous band images of bilateral corticospinal tracts were reconstructed. The muscle strength of the affected hand was assessed by Brunnstorm standard[1],which is an internationally recognized measure of movement in hemiplegic patients.Results:①In the control group,there was no significant difference in FA between the bilateral corticospinal tracts,but FA in different white matter structure of the same side was significantly different (t=3.12,P < 0.05). FA of posterior limb of internal capsule was higher than that of knee and anterior limb of internal capsule,FA of anterior limb of internal capsule was the lowest. while in the patient group,FA of the infarcted sites was significantly lower than the contralateral ones (t=5.570,P < 0.01). ②The ipsilateral corticospinal tract demonstrated continuous interruption and the loss of consistent anatomic structure. The involved severity of corticospinal tract had significant correlation with that of muscle strength of the ipsilateral hand (r=0.83,P < 0.01).Conclusion:This study shows that DTI can be used to investigate ischemic stroke and assess ischemic stroke-induced damage. 5Construction of PR domain eukaryotic expression vector and its inhibitory effect on esophageal cancer cells显示文摘Objective:PR domain is responsible for the tumor suppressing activity of RIZ1.The study aimed to construct human PR domain eukaryotic expression vectors,transfect human esophageal cancer cells(TE13),and evaluate the anticancer activity of PR domain on human esophageal cancer TE13 cells.Methods:First,mRNA was extracted from human esophageal cancer tissue by RT-PCR,then reversetranscribed to cDNA.After amplifying from the DNA template,PR domain was linked to T vector.Second,after extraction,PR domain was cut using enzyme and linked to pcDNA3.1(+).Then,the plasmid was transfered to Trans1-T1 phage resistant competent cells,following by extracting the ultrapure plasmid,and transfecting into TE13 cells.In the end,the protein expression of pcDNA3.1(+)/PR domain in TE13 was detected by Western blot,and the apoptosis of TE13 by technique of flow cytometry.Results:More than 5,000 bp purposed band of pcDNA3.1(+)/PR domain plasmid was found by agarose gel electrophoresis.After transfection,the PR domain(molecular weight of about 28 Da)was found only in 3,4 and 5 groups by Western blot.Flow cytometry assay showed apoptosis in experimental group was significantly more than that in the control group(P<0.05).Conclusions:The PR domain eukaryotic expression vector was constructed successfully.The protein of the PR domain could be expressed in esophageal cancer TE13 cells firmly after transfection,and a single PR domain could promote apoptosis of TE13 cells.Yuan Chen Peng Zhang Yuanguo Wang Shangwen Dong Yimei Liu 2013Chinese Journal of Cancer Research2013,25,5:6

7Highly concentrated graphene oxide ink for facile 3D printing of supercapacitors显示文摘3D printing of functional energy storage devices is receiving escalating attention over the years due to the customizable manufacturing flexibility and imparted high areal and gravimetric energy density of three-dimensional structured devices, which contribute to the creation of numerous new opportunities for futuristic electronics. Graphene-based inks are ideal elements for the realization of 3D printed energy storage devices if the attractive intrinsic physiochemical properties of graphene could be preserved. However, it is still a great challenge to prepare uniformly dispersed graphene-based materials with desired rheological properties for 3D printing. Here we report a facile strategy for 3D printing of supercapacitors from a highly concentrated graphene oxide (GO) ink. The GO is properly dispersed and the ink fulfills the stringent rheological specifications for 3D printing. The printed GO electrode is functionalized with enhanced structural stability for proper reduction to graphene. The printed supercapacitors deliver the potential to linearly scale up in areal capacitance without jeopardizing the gravimetric capacitance when increasing printed layers. 8Characters of MR diffusion tensor imaging in cerebral ischemic corticospinal tract injury显示文摘BACKGROUND: Diffusion tensor imaging (DTI) is one of the noninvasive methods to study the morphological structure of brain white matter fibrous bands in vivo, and it has been applied primarily in clinic. DTI is acknowledged as the more effective imaging method to diagnose ultra-acute and/or acute cerebral infarction. OBJECTIVE: To observe the anisotropic characters of cerebral white matter fibrous bands in patients with ischemic stroke by using DTI, and investigate the correlation between the damage of corticospinal tract and muscle strength in patients with ischemic stroke at acute period. DESIGN: A case-control observation. SETTING: Department of Medical Imaging, Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA. PARTICIPANTS: Nine inpatients with injury of motor function induced by acute ischemic stroke (patient group) at 6 hours to 2 weeks after the attack were selected from the Department of Neurology, Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA from September 2005 to March 2006, and they all accorded with the present diagnostic standard for cerebrovascular disease in China. There were 5 males and 4 females, aged 16-87 years. At the same time, nine healthy right-handed physical examinees matched by age and sex with the patients were taken as the control group, and they all had no nervous disease, mental diseases, cerebrovascular abnormalities and injury history, etc. All the subjects were informed with the detected items and agreed to participate in the study. METHODS: All the 9 patients with ischemic stroke at acute period and 9 healthy subjects were examined with MRI, T1 weighted imaging, T2 weighted imaging and DTI. And the data were processed offline with dTV.II software, the images of fractional anisotropy and directional encoded color (DEC) were obtained, and the three-dimensional fibrous band images of bilateral corticospinal tracts were reconstructed. In the control group, the values of fractional anisotropy of main white matter fibrous bands were measured in the region of interest (ROI) of the anterior limb, knee and posterior limb of internal capsule. In the patient group, the values of fractional anisotropy of white matter were measured in the infarcted sites and corresponding contralateral sites of the patients. The ROI was set in bilateral cerebral peduncles to reconstruct three-dimensionally the bilateral corticospinal tracts. The muscle strength of the affected hand was assessed with Brunnstorm standard in the stroke patients. MAIN OUTCOME MEASURES: The characters of DTI and images of the value of fractional anisotropy, and the manifestations of three-dimensional corticospinal tracts were observed in the two groups. RESULTS: All the data from the 9 patients and 9 healthy volunteers were involved in the analysis of results. ① In the control group, the white matter and gray matter could be distinguished clearly in the image of fractional anisotropic values, the fibers of different directions were shown by different colors in DEC picture, which clearly demonstrated the normal anatomic structure and direction of white matter fibers. In the patient group, the infarctions occurred in the gray matter or white matter could be distinguished in the images of fractional anisotropic values, DEC picture could clearly show the direct influence of the infarcted site on the white matter fibers. The fractional anisotropic values in different white matter structure of the same side were significantly different in the control group (t=3.12, P < 0.05), and the reconstructed images fractional anisotropic values and DEC picture could show most of the main white matter fibrous bands. The fractional anisotropic values of the infarcted sites were significantly lower than the contralateral ones in the patient group (t=5.570, P < 0.01). ② The reconstructed bilateral corticospinal tracts showed that the anatomic forms of the contralateral corticospinal tract of the patients were almost identical to those of normal people, it started from precentral gyrus, downward to the nternal capsule, and extended to pontine and medulla oblongata, each fibrous band was continuous, and the form had good consistency. Because of the involvement of infarction of different severity, the ipsilateral corticospinal tract manifested as continuous interruption and the loss of consistent anatomic structural form. The involved severity of corticospinal tract had significant correlation with that of muscle strength of the ipsilateral hand (r=1.30, P < 0.01). CONCLUSION: ① DTI can display the direction and distribution of cerebral white matter fibrous bands. ② DTI images of fractional anisotropic values and DEC can show the directions and anisotropic degree of white matter fibers in the infarcted sites of stroke patients. ③ The three-dimensional images of fibrous bands can show the conditions of pyramidal tracts more directly. ④ The damaged severity of corticospinal tracts is correlated with that of muscle strength.Ziqian Chen Ping Ni Hui Xiao Youqiang Ye Gennian Qian Shangwen Xu Xizhang Yang Jinhua Chen Biyun zhang 2006Neural Regeneration Research2006,1,2:3

10“Extreme utilization”development of deep shale gas in southern Sichuan Basin,SW China显示文摘To efficiently develop deep shale gas in southern Sichuan Basin,under the guidance of“extreme utilization”theory,a basic idea and solutions for deep shale gas development are put forward and applied in practice.In view of multiple influencing factors of shale gas development,low single-well production and marginal profit of wells in this region,the basic idea is to establish“transparent geological body”of the block in concern,evaluate the factors affecting shale gas development through integrated geological-engineering research and optimize the shale gas development of wells in their whole life cycle to balance the relationship between production objectives and development costs.The solutions are as follows:(1)calculate the gold target index and pinpoint the location of horizontal well drilling target,and shale reservoirs are depicted accurately by geophysical and other means to build underground transparent geological body;(2)optimize the drilling and completion process,improve the adaptability of key tools by cooling,reducing density and optimizing the performance of drilling fluid,the“man-made gas reservoir”is built by comprehensively considering the characteristics of in-situ stress and fractures after the development well is drilled;(3)through efficient management,establishment of learning curve and optimization of drainage and production regime,the development quality and efficiency of the well are improved across its whole life cycle,to fulfil“extreme utilization”development of shale gas.The practice shows that the estimated ultimate recovery of single wells in southern Sichuan Basin increase by 10%-20%than last year.MA Xinhua WANG Hongyan ZHAO Qun LIU Yong ZHOU Shangwen HU Zhiming XIAO Yufeng 2022Petroleum Exploration and Development2022,49,6:2

12Changes in brain function and anatomical structure following treatment of hyperbaric oxygen for visual pathway abnormalities in 16 cases Evaluation of functional magnetic resonance imaging combined with diffusion tensor imaging显示文摘BACKGROUND: There is a growing research focus on the combination of blood oxygenation level dependent functional magnetic resonance imaging (BOLD-fMRI) and diffusion tensor imaging (DTI) to evaluate visual cortical function and structural changes in the cerebrum,as well as morphological changes to the white matter fiber tracks,after visual pathway lesions. However,the combined application of BOLD-fMRI and DTI in treating of visual pathway abnormalities still requires further studies. OBJECTIVE: To observe and evaluate the effects of hyperbaric oxygen on visual pathway abnormalities,and to evaluate the characteristics of cerebral function and anatomic structural changes by using BOLD-fMRI combined with DTI technique. DESIGN: Case contrast observation. SETTING: Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA. PARTICIPANTS: Sixteen patients (9 males and 7 females,15-77 years old) with lateral or bilateral visual disorder induced by visual pathway lesions were selected from the Department of Neurology,Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA from January 2006 to May 2007. These patients comprised the lesion group. Measures of interventional therapy: hyperbaric oxygen of two normal atmospheres for three courses (10 d/course) and routine internal medicine treatment. In addition,12 healthy subjects of similar sex and age to the lesion group were regarded as the control group. The control group underwent routine ophthalmological and ocular fundus examinations; diagnostic results were normal. The experiment received confirmed consent from the local ethic committee,and all patients provided informed consent. METHODS: BOLD-fMRI and DTI manifestations in the lesion group were observed before and after hyperbaric oxygen intervention,and the results were compared with the control group. The subjects were positioned on their back,and BOLD-fMRI images were collected with the following GRE EPI sequence: TR = 2 000 ms,TE = 40 ms,layer thickness = 5 mm,and 20-layer successive scanning to cover the whole brain. While,DTI images were collected with SE EPI sequence of single excitation: TR=10 000 ms,TE = 112 ms,layer thickness = 4 mm,layer spacing = 0.5 mm,and a 30-layer successive scanning,matrix = 128×128. A diffusion gradient was applied at 13 directions,and one layer without diffusion weighted imaging was collected at b =1 000 s/mm2. Numbers of activated voxels in cortical-activated regions,and fractional anisotropy in bilateral cerebral optical radiation regions,were calculated. Displacement,continuity,and destruction of fibrous tracts were analyzed. MAIN OUTCOME MEASURES: Results of BOLD-fMRI and DTI examinations. RESULTS: All 16 patients and 12 controls were included in the final analysis. ① Numbers of activated voxels through the use of BOLD-fMRI: prior to hyperbaric oxygen therapy,the number of activated voxels in the bilateral cortex of occipital lobe were significantly less in the lesion group than in the control group (t =3.23,P < 0.01). In addition,the number of activated voxels significantly increased after treatment compared to before treatment (t = 2.46,P < 0.05). ② Fractional anisotropy in optical radiation regions of bilateral cerebrum: fractional anisotropy in the lesion group was significantly less than the control group (t =2.89,P < 0.05). In addition,fractional anisotropy after treatment was significantly higher than before treatment (t = 2.48,P < 0.05). Moreover,fractional anisotropy of optical neuropathy was significantly higher in 6 patients in the lesion group than the occipital lobe optical central lesion (t = 2.35,P < 0.05). CONCLUSION: BOLD-fMRI combined with DTI can indicate the occurrence,development,and therapeutic course of action for optical pathway lesions. 13Microcellular Foaming of Plasticized Thin PC Sheet I.Effects of Processing Conditions显示文摘Novel microcellular foams using thin plasticized PC sheet were prepared by compression molding. The measurement results showed that Tg of plasticized PC was decreased and the molecular chain mobility was increased. Decrease in Tg and increase in chains mobility were contributed to the widen of foaming temperature window. Effects of processing conditions on cell size, cell density and relative density were also investigated. The experimental results show that the temperature, tributyl citrate and foaming agent content have more effects on the structures and morphology of the plasticized PC microcellular foam. 14Determination of 18 Kinds of Amino Acids in Fresh Tea Leaves by HPLC Coupled with Pre-column Derivatization显示文摘A rapid and accurate quantitative method of high performance liquid chromatography( HPLC) with fluorescence detector has been developed for the analysis of 18 kinds of amino acids in fresh tea leaves. The samples were minced and mixed,and extracted with ultra pure water at 90℃ for 20 min. The 6-aminoquinolyl N-hydroxy-succinimidyl carbamate( AQC) was used as pre-column derivatization reagent. Gradient HPLC separation was performed on a C_(18) column( Symmetry C_(18),3. 9 mm × 15 cm,4 μm). Good linearity between concentrations and peak areas was achieved in the concentration range of 5. 0-250 μmol/L for 18 kinds of amino acids. The method was validated by the analysis of five replicates. The 18 kinds of amino acid standards were spiked in fresh tea leaf samples and the average recovery rate was 86. 25%-109. 05% with relative standard deviations( n = 5) ranging from 6. 03% to 10. 56%. The limit of detection( LOD) for the analytes was0. 05-1. 27 μmol/L. The method was successfully applied to the analysis of the 18 kinds of amino acids in fresh tea leaves from east Dongting and west Dongting mountains in Suzhou. 16Phosphoproteomics Reveals the AMPK Substrate Network in Response to DNA Damage and Histone Acetylation显示文摘AMP-activated protein kinase(AMPK)is a conserved energy sensor that plays roles in diverse biological processes via phosphorylating various substrates.Emerging studies have demonstrated the regulatory roles of AMPK in DNA repair,but the underlying mechanisms remain to be fully understood.Herein,using mass spectrometry-based proteomic technologies,we systematically investigate the regulatory network of AMPK in DNA damage response(DDR).Our system-wide phosphoproteome study uncovers a variety of newly-identified potential substrates involved in diverse biological processes,whereas our system-wide histone modification analysis reveals a link between AMPK and histone acetylation.Together with these findings,we discover that AMPK promotes apoptosis by phosphorylating apoptosis-stimulating of p53 protein 2(ASPP2)in an irradiation(IR)-dependent manner and regulates histone acetylation by phosphorylating histone deacetylase 9(HDAC9)in an IR-independent manner.Besides,we reveal that disrupting the histone acetylation by the bromodomain BRD4 inhibitor JQ-1 enhances the sensitivity of AMPKdeficient cells to IR.Therefore,our study has provided a resource to investigate the interplay between phosphorylation and histone acetylation underlying the regulatory network of AMPK,which could be beneficial to understand the exact role of AMPK in DDR.Yuejing Jiang Xiaoji Cong Shangwen Jiang Ying Dong Lei Zhao Yi Zang Minjia Tan Jia Li 2022Genomics, Proteomics & Bioinformatics2022,20,4:1
