| 1 | Research on protoplast preparation and culture in Gladiolus hybridus Hort显示文摘Protoplasts prepared from the aseptic leaves of Rose Supreme in Gladiolus hybridus Hort. were cultured. The yield of protoplasts with vigour was the highest under the following conditions: 1.5% cellulase Onzuka RS, 0.5% pectinase Y-23, 0.6 mol· L^(-1) mannitol, pH 5.8, digestion time 2 h, and temperature 27℃. After isolation and purification, protoplasts were cultured in solid and liquid combined medium, which included KM8P, 0.6 mol· L^(-1) mannitol, 1.0 mg· L^(-1) 2, 4-D, 0.2 mg· L^(-1) NAA and 0.2 mg· L^(-1) KT. | 3 | Study on Cell Suspension Culture of Floribunda Rose显示文摘Friable callus was induced when immature seeds of floribunda rose were inoculated on MS medium supplemented with 2,4-D 3.0 mg·L-1. When transfered onto subculture media, friable callus developed into embryogenic callus, which was used to establish cell suspension lines. Cell suspensions had to be subcultured at a interval of 4-5 days at the first several culture cycles. The best subculturing cycle for the stable cell suspensions was 8-10 days. | 4 | Study on Transformation of Oriental Hybrid Lily by Agrobacterium-mediated显示文摘An efficient procedure was described for the transformation of the monocotyledonous oriental hybrid lily,Lilium cv. Siberia. by Agrobacterium-mediated genetic transformation via leaves regeneration. The leaves of lea· ets which derived from bulbs were sliced into 1.0 cm long and were co-cultivated with A. tumefaciens strain LBA4404/pB2AE12,which harbored a vector carrying the neomycin phosphotransferase,DREB2A genes in the T-DNA region. The suitable genetic transformation condition was determined as follows:the bacterial concentration reached 0.5-0.6(OD600) ,15 min infection time,20 mg · L-1 acetosyingone,and 10.6 mmol · L-1 NH4NO3 medium was used for co-cultivation 3 days,delayed 7 days for selecting by 30 mg · L-1 kanamycin containing regeneration medium. Effi cient shoot regeneration was observed on MS medium supplemented with 1.0 mg · L-1 naphthaleneacetic acid,0.5 mg · L-1 benzyladenine and 0.1 mg · L-1 Kinetin after about 6 weeks culture. | 5 | Asponchimides A−E: new enantiomeric N-acetyldopamine trimers from Aspongopus chinensis显示文摘Five new racemic N-acetyldopamine (NADA) trimers, asponchimides A−E ( 1 − 5 ), were isolated from Aspongopus chinensis, a prominent traditional Chinese medicinal insect employed for alleviating pain, treating indigestion, and addressing kidney ailments. Compounds 1 − 5 were successfully resolved by chiral high-performance liquid chromatography (HPLC), yielding five pairs of enantiomers: (+)- and (−)-asponchimides A−E ( 1a / 1b − 5a / 5b ). Their structural identities were discerned by extensive spectroscopic analyses, including high-resolution mass spectrometry (HRMS), ultraviolet-visible (UV-Vis) spectroscopy, infrared (IR) spectroscopy, and nuclear magnetic resonance (NMR), and their absolute configurations were determined by electronic circular dichroism (ECD) calculations. Compounds 1 − 5 are pioneering instances of NADA trimers featuring a Δ7 double bond.
