2Protective effects of acupuncture and LGNHFD on expressions of vascular endothelial growth factor, basic fibroblast growth factor,and cluster of differentiation 34 in rats with cerebral ischemia-reperfusion injury显示文摘OBJECTIVE: To investigate the protective effects of electroacupuncture and LGNHFD on cerebral ischemia-reperfusion injury, and their effects on the expression of cluster of differentiation 34(CD 34), vascular endothelial growth factor(VEGF), and basic fibroblast growth factor(b FGF) in the peripheral tissue of the cerebral ischemic semi-dark zone in rats.The influence and mechanism of expression.METHODS: Healthy male Sprague-Dawley rats were randomly divided into the sham operation group, model group, Traditional Chinese Medicine (TCM) group, electroacupuncture group, and acupuncture group. The cerebral ischemia model was prepared by middle artery occlusion. The electroacupuncture and TCM groups received electroacupuncture at the 'Neiguan(PC6)', 'Baihui(GV20)', and 'Renzhong(GV26)' acupoints from 4 h after modeling, once daily for 14 d. After modeling,the acupuncture and TCM groups were administered LGNHFD once daily for 14 d. After treatment,the infarct volume and regional cerebral blood flow(r CBF) were measured in each group, the expression of CD34 in the surrounding ischemic penumbra was determined using immunohistochemistry,and the expression of angiogenesis-related factors in the ischemic hemisphere was detected by western blot.RESULTS: Compared with the sham operation group, r CBF was significantly decreased(P < 0.01)and cerebral infarction volume was significantly increased(P < 0.01) in the model group at four time points(5 min and 3, 7, and 14 d after modeling). At three postoperative time points(3, 7, and 14 d), the number of CD34 + cells in the model group were significantly increased(P < 0.01), and the relative expression levels of VEGF, b FGF, and CD34+ in the ischemic hemisphere were significantly increased(P < 0.01, P < 0.05). Compared with the model group, the r CBF of the acupuncture group was significantly increased at 3, 7, and 14 d after intervention(P < 0.01), while the r CBF of the electroacupuncture and TCM groups was significantly increased at 7 and 14 d after intervention(P < 0.01).After 3, 7, and 14 d of intervention, infarct volume was significantly reduced(P < 0.01), the number of CD34 + cells was significantly increased(P < 0.01,P < 0.05), and the relative expression levels of VEGF,b FGF, and CD34 in the ischemic hemisphere were significantly increased(P < 0.01, P < 0.05). Compared with the electroacupuncture and TCM groups, the r CBF of the acupuncture group was significantly increased(P < 0.01), the infarct volume was significantly reduced(P < 0.01), and the relative expression levels of VEGF, b FGF, and CD34 were significantly increased(P < 0.05, P < 0.01) after 3, 7,and 14 d of intervention.CONCLUSION: Acupuncture therapy is superior to simple drug and electroacupuncture therapy for reducing infarct volume, increasing r CBF and CD34 expression in cells, and promoting capillary regeneration in rats with cerebral ischemia. 3Pioglitazone ameliorates neuronal damage after traumatic brain injury via the PPARg/NF-kB/IL-6 signaling pathway显示文摘Traumatic brain injury(TBI)is the major cause of high mortality and disability rates worldwide.Pioglitazone is an activator of peroxisome proliferator-activated receptor-gamma(PPARγ)that can reduce inflammation following TBI.Clinically,neuroinflammation after TBI lacks effective treatment.Although there are many studies on PPARγin TBI animals,only few could be converted into clinical,since TBI mechanisms in humans and animals are not completely consistent.The present study,provided a potential theoretical basis and therapeutic target for neuroinflammation treatment after TBI.First,we detected interleukin-6(IL-6),nitric oxide(NO)and Caspase-3 in TBI clinical specimens,confirming a presence of a high expression of inflammatory factors.Western blot(WB),quantitative real-time PCR(qRTPCR)and immunohistochemistry(IHC)were used to detect PPARγ,IL-6,and p-NF-kB to identify the mechanisms of neuroinflammation.Then,in the rat TBI model,neurobehavioral and cerebral edema levels were investigated after intervention with pioglitazone(PPARγactivator)or T0070907(PPARγinhibitor),and PPARγ,IL-6 and p-NF-kB were detected again by qRT-PCR,WB and immunofluorescence(IF).The obtained results revealed that:1)increased expression of IL-6,NO and Caspase-3 in serum and cerebrospinal fluid in patients after TBI,and decreased PPARγin brain tissue;2)pioglitazone could improve neurobehavioral and reduce brain edema in rats after TBI;3)the protective effect of pioglitazone was achieved by activating PPARγand reducing NF-kB and IL-6.The neuroprotective effect of pioglitazone on TBI was mediated through the PPARγ/NF-kB/IL-6 pathway.Yongbing Deng Xue Jiang Xiaoyan Deng Hong Chen Jie Xu Zhaosi Zhang Geli Liu Zhu Yong Chengfu Yuan Xiaochuan Sun Changdong Wang 2020Genes & Diseases2020,7,2:6

6The genome of the warm-season turfgrass African bermudagrass (Cynodon transvaalensis)显示文摘Cynodon species can be used for multiple purposes and have high economic and ecological significance.However,the genetic basis of the favorable agronomic traits of Cynodon species is poorly understood,partially due to the limited availability of genomic resources.In this study,we report a chromosome-scale genome assembly of a diploid Cynodon species,C.transvaalensis,obtained by combining Illumina and Nanopore sequencing,BioNano,and Hi-C.The assembly contains 282 scaffolds(~423.42 Mb,N50=5.37 Mb),which cover~93.2%of the estimated genome of C.transvaalensis(~454.4Mb).Furthermore,90.48%of the scaffolds(~383.08 Mb)were anchored to nine pseudomolecules,of which the largest was 60.78Mb in length.Evolutionary analysis along with transcriptome comparison provided a preliminary genomic basis for the adaptation of this species to tropical and/or subtropical climates,typically with dry summers.The genomic resources generated in this study will not only facilitate evolutionary studies of the Chloridoideae subfamily,in particular,the Cynodonteae tribe,but also facilitate functional genomic research and genetic breeding in Cynodon species for new leading turfgrass cultivars in the future.Fengchao Cui Geli Taier Manli Li Xiaoxia Dai Nan Hang Xunzhong Zhang Xiangfeng Wang Kehua Wang 2021Horticulture Research2021,8,1:1

12ARHGEF38 as a novel biomarker to predict aggressive prostate cancer显示文摘Prostate cancer(PCa)metastasis is considered the leading cause of cancer death in males.Therapeutic strategies and diagnosis for stage-specific PCa have not been well understood.Rho guanine nucleotide exchange factor 38(ARHGEF38)is related to tumor cell polarization and is frequently expressed in PCa.Microarray data of PCa were downloaded from GEO and TCGA databases.A total of 243 DEGs were screened,of which,32 genes were upregulated.The results of enrichment analysis showed the participation of these DEGs in the tumor cell metastasis pathway.ARHGEF38 was significantly up-regulated in the four most prevalent cancers worldwide(p<0.05),and its expression was higher in the tumor samples with higher Gleason score(GS).IHC,qRT-PCR,and western-blot analyses showed the higher expression of ARHGEF38 in PCa than benign prostatic hyperplasia(BPH).In addition,IHC results demonstrated a higher expression of ARHGEF38 in high-grade PCa than the low-grade PCa.Kun Liu Aixiang Wang Longke Ran Wanfeng Zhang Song Jing Yujing Wang Xianqin Zhang Geli Liu Wang Sen Fangzhou Song 2020Genes & Diseases2020,7,2:0
13A large forage gap in forage availability in traditional pastoral regions in China显示文摘Forage supply has been stressed due to the rapid increase in China's livestock consumption.However,the long-term dynamics of the relationships between forage demand and multi-sourced supply are not understood.Here,we examine the annual forage demand,or practical carrying capacity(PCC),and supply,or theoretical carrying capacity(TCC)from 2000 to 2019 in China.We construct a forage supply-demand index(FSDI)to represent the forage supply pressure using MODIS-derived net primary productivity products and provincial statistical datasets,and we consider two scenarios.First,natural grasslands are the sole source of forage.Second,natural grassland forage supply is supplemented with straw crops.We find an increase in PCC in northwestern China's major pastoral and agropastoral provincial regions,including Inner Mongolia,Gansu,Ningxia and Qinghai,at rate of 0.24-3.59 million sheep units(SU)a year.As the primary source of forage,the theoretical carrying capacity of natural grasslands(TCCgrass)expanded at a rate of 3 million SU/yr nationally.Crop straws fed 126.58 million SU nationally in 2019,which accounted for 11.3%of the total practical carrying capacity and alleviated the forage supply pressure by reducing FSDI by 26.56%.During 2000–2019,the theoretical carrying capacity of straw crops(TCCcrop)increased rapidly from 76.5 million SU to 126.6 million SU,which accounted for 10%-15%of the total forage supply at the national scale.We also discovered large carrying capacity gaps(TCCgap)in the northwestern pastoral provincial regions of Inner Mongolia,Xinjiang,Gansu,and some agricultural provinces such as Shandong and Henan,when we considered forage supply from both natural grasslands and straw crops.Our findings showed a large forage gap in the traditional pastoral regions,and we also discussed green fodder as a potential solution for balancing the supply of and demand for forage,which may shed light on crop and forage planning.Tong Yang Jinwei Dong Lin Huang Yuzhe Li Huimin Yan Jun Zhai Jie Wang Zhenong Jin Geli Zhang 2023Fundamental Research2023,3,2:0
