| 4 | Fluxes of CO_2,CH_4 and N_2O from alpine grassland in the Tibetan Plateau显示文摘Using stat ic chamber technique, fluxes of CO 2 , CH 4 and N 2 O were measured in the alpine grassland area from July 2000 to July 2001, d eterminations of mean fluxes showed that CO 2 and N 2 O were gene rally released from the soil, while the alpine grassland accounted for a weak CH 4 sink. Fluxes of CO 2 , CH 4 and N 2 O ranged widely. The highest CO 2 emission occurred in August, whereas a lmost 90% of the whole year emission occurred in the growing season. But the variations of CH 4 and N 2 O fluxes did not show any clear patterns over the one-year-experim ent. During a daily variation, the maximum CO 2 emission occurred at 16:00, and then decreased to the minimum emi ssion in the early morning. Daily pattern analyses indicated that the variation in CO 2 fluxes was positively related to air temperatures (R 2 =0.73) and soil temperatures at a depth of 5 cm (R 2 =0.86), whereas daily variations in CH 4 and N 2 O fluxes were poorly explained by soil temperatures and climatic va riables. The differential expression has been confirmed by RT-PCR. One novel gene of 1096 bp has been cloned based on this EST. Bioinformatics analysis found that the new gene sequence contains a whole reading frame encoding 256 amino acids. There is a stop codon TAA in front of the 5’ end start codon, and a tailing signal AATAAA and poly A tail at the 3’ end. There is no homologous known gene found after searching by blasting this sequence to non-redundancy nucleotide database. | 9 | Profile of Methane Concentrations in Soil and Atmosphere in Alpine Steppe Ecosystem on Tibetan Plateau显示文摘The methane concentration profile from -1.5m depth in soil to 32m height in air was measured in alpine steppe lo-cated in the permafrost area. Methane concentrations showed widely variations both in air and in soil during the study period. The mean concentrations in atmosphere were all higher than those in soil, and the highest methane concentration was found in air at the height of 16m with the lowest concentration occur-ring at the depth of 1.5m in soil. The variations of atmospheric methane concentrations did not show any clear pattern both temporally and spatially, although they exhibited a more steady-stable state than those in soil. During the seasonal variations, the methane concentrations at different depths in soil were sig-nificantly correlated (R2>0.6) with each other comparing to the weak correlations (R2<0.2) between the atmospheric concentra-tions at different heights. Mean methane concentrations in soil significantly decreased with depth. This was the compositive influence of the decreasing production rates and the increasing methane oxidation rates, which was caused by the descent soil moisture with depth. Although the methane concentrations at all depths varied widely during the growing season, they showed very distinct temporal variations in the non-growing season. It was indicated from the literatures that methane oxidation rates were positively correlated with soil temperature. The higher methane concentrations in soil during the winter were deter-mined by the lower methane oxidation rates with decreasing soil temperatures, whereas methane production rates had no reaction to the lower temperature. | 11 | CO_2 processes in an alpine grassland ecosystem on the Tibetan Plateau显示文摘In this paper, the CO2 concentrations profile from 1.5 m depth in soil to 32 m height in atmosphere were measured from July 2000 to July 2001 in an alpine grassland ecosystem located in the permafrost area on the Tibetan Plateau, which revealed that CO2 concentrations varied greatly during this study period. Mean concentrations during the whole experiment in the atmosphere were absolutely lower than the CO2 concentrations in soil, which resulted in CO2 emissions from the alpine steppe soil to the atmosphere. The highest CO2 concentration was found at a depth of 1.5 m in soil while the lowest CO2 concentration occurred in the atmosphere. Mean CO2 concentrations in soil generally increased with depth. This was the compositive influence of the increasing soil moistures and decreasing soil pH, which induced the increasing biological activities with depth. Temporally, the CO2 concentrations at different layers in air remained a more steady state because of the atmospheric turbulent milking. | 15 | The development of an iridium(Ⅲ) complex functionalized G-quadruplex probe for the stability of G-quadruplex and lifetime image in cytoplasm显示文摘G-quadruplex(G4) is widely known as a non-classical secondary structure of nucleic acid. With the indepth study of G4, it is an urgent need for a phosphorescent probe with a high G4 binding ability to evaluate the level of G4 in the cytoplasm. Thus, this study designed and synthesized Ir-PDP where an Ir(Ⅲ)complex was used as a phosphorescent emitter. Meanwhile, two installed PDPs(pyridostatin derivatives)were used to improve the combination ability with G4 and reduced the cytotoxicity of the Ir(Ⅲ) complex.Compared with other nucleic acid secondary structures, Ir-PDP produced a higher phosphorescence lifetime after interacting with G4. Ir-PDP was distributed in the cytoplasm of living cells, and two-photon phosphorescence lifetime imaging can detect the binding events of the probe in the cytoplasm. The addition of G4 binder PDS significantly regulated cytoplasmic phosphorescence lifetime. Human PVT1 gene is located on region 8q24.21, a well-known cancer risk region, and encodes at least 26 linear ncRNA isoforms and 26 circular RNA isoforms, as well as 6 microRNAs. Several PVT1 functioning models have been reported recently such as competing endogenous RNA (ceRNA) activity and regulating protein stability of oncogenes, especially MYC oncogene. The promoter of PVT1 gene is a boundary element of tumor-suppressor DNA. CircPVT1 derived from PVT1 gene is also a critical non-coding oncogenic RNA. Although substantial advancements have been made in understanding the roles of PVT1 in cancer recently, the detailed mechanisms underlying its functions remain unclear. Herein, we summarize the recent progressions on the mechanisms underlying PVT1 regulated gene expression at different levels. | 18 | Interaction between Heparin and Fibronectin: Using Quartz Crystal Microbalance with Dissipation, Immunochemistry and Isothermal Titration Calorimetry显示文摘The adsorption behavior of heparin and fibronectin was studied by quartz crystal microbalance with dissipation(QCM-D), and the interaction between heparin and fibronectin was evaluated using immunochemistry and isothermal titration calorimetry(ITC) measurement. The results showed that there was competitive adsorption between heparin and fibronectin, and the preadsorption of fibronectin could prevent subsequent heparin adsorption to some extent, and the adsorbed Hep/Fn complex on the surface was in a rigid form. The bioactivity of heparin and fibronectin could be affected by the bulk concentration of each, and both heparin and fibronectin in Hep/Fn complex formed under p H 4 condition displayed larger bioactivity than that formed under p H 7 condition. Moreover, the fibronectin showed more exposed cell-binding sites at the p H value lower than physiological condition. The results of ITC further suggested that the interaction between heparin and fibronectin under p H 4 was stronger than under p H 7, and the complex was also more stable. The study brings forth the detailed interaction between heparin and fibronectin, which will be helpful for better understanding the interaction mechanism of the two biomolecules. | 19 | The R2R3-MYB transcription factor GaPC controls petal coloration in cotton显示文摘Although a few cases of genetic epistasis in plants have been reported, the combined analysis of genetically phenotypic segregation and the related molecular mechanism remains rarely studied. Here, we have identified a gene(named GaPC) controlling petal coloration in Gossypium arboreum and following a heritable recessive epistatic genetic model. Petal coloration is controlled by a single dominant gene,GaPC. A loss-of-function mutation of GaPC leads to a recessive gene Gapc that masks the phenotype of other color genes and shows recessive epistatic interactions. Map-based cloning showed that GaPC encodes an R2R3-MYB transcription factor. A 4814-bp long terminal repeat retrotransposon insertion at the second exon led to GaPC loss of function and disabled petal coloration. GaPC controlled petal coloration by regulating the anthocyanin and flavone biosynthesis pathways. Expression of core genes in the phenylpropanoid and anthocyanin pathways was higher in colored than in white petals. Petal color was conferred by flavonoids and anthocyanins, with red and yellow petals rich in anthocyanin and flavonol glycosides, respectively.
