| 4 | Influence of chronic intermittent hypoxia on growth associated protein 43 expression in the hippocampus of young rats显示文摘This study aimed to explore the pathological change to hippocampal neurons and the expression of growth associated protein 43 in 21-day-old young rats following chronic intermittent hypoxia.Hematoxylin-eosin staining results showed varying degrees of degeneration and necrosis in hippocampal neurons depending on the modeling time.Immunohistochemistry revealed that growth associated protein 43 expression in young rats following chronic intermittent hypoxia decreased,but that levels were still higher than those of normal rats at each time point,especially 4 weeks after modeling.During 1-5 weeks after modeling,a slow growth in rat weight was observed.Experimental findings indicate that chronic intermittent hypoxia may induce growth dysfunction and necrosis of hippocampal neurons,as well as increase the expression of growth associated protein 43 in young rats. | Yan Chen Chunling Zhao Chunlai Zhang Lirong Luo Guang Yu | 2012 | Neural Regeneration Research2012,7,16: | 4 |
| 5 | Cryptography on elliptic curves over p-adic number fields显示文摘In this paper we introduce a cryptosystem based on the quotient groups of the group of rational points of an elliptic curve defined over p-adic number field. Some additional parameters are taken in this system, which have an advantage in performing point multiplication while keeping the security of ECC over finite fields. We give a method to select generators of the cryptographic groups, and give a way to represent the elements of the quotient groups with finitely bounded storage by establishing a bijection between these elements and their approximate coordinates. | 7 | Structural insights into the CRISPR-Cas-associated ribonuclease activity of Staphylococcus epidermidis Csm3 and Csm6显示文摘The Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)-CRISPR-associated(Cas) system is an adaptive immune system in bacteria and archaea that resists exogenous invasion through nucleic acid-mediated cleavage. In the type III-A system, the Csm complex contains five effectors and a CRISPR RNA, which edits both single stranded RNA and double stranded DNA. It has recently been demonstrated that cyclic oligoadenylates(cOAs), which are synthesized by the Csm complex, act as second messengers that bind and activate Csm6. Here, we report the crystal structures of Staphylococcus epidermidis Csm3(SeCsm3) and an N-terminally truncated Csm6(SeCsm6 AN) at 2.26 and 2.0 , respectively. The structure of SeCsm3 highly resembled previously reported Csm3 structures from other species; however, it provided novel observations allowing further enzyme characterization. The homodimeric SeCsm6 AN folds into a compact structure. The dimerization of the HEPN domain leads to the formation of the ribonuclease active site, which is consistent with the reported Csm6 structures. We relocated the epicenters and focal depths of the three events by means of the reversal-time imaging technique using broadband digital seismic data from worldwide stations ranging from 30° to 90°, imaged the spatiotemporal variation of the energy radiation sources by means of the nonplane wave array technique using the broadband digital seismic data from a generalized array made up of 33 stations of the Capital Region Digital Seismograph Net-work (CRDSN), and obtained the rupture duration times, extents and rupture velocities. | 10 | Effect of Ginsenoside Rbl on Expressive Proteome of Rat Neurons显示文摘[Objective] This study aimed to explore the effect of ginsenoside Rbl on expressive proteome of rat neurons by technologies of proteomics, bioinformatics and MS peptide fingerprinting. [Method] Rat neurons were cultured conventionally and randomly separated into two groups. The experimental group was treated with 5 μg/ml Rbl for 20 min, while control group was added with the same amount of medium. After cell lysis, the whole-cell protein was extracted. Two-dimensional electrophoresis (2-DE) was used to separate the extracts. Differential expression of proteome between the two groups was analyzed by using ImageMaster 2D Platinum v5.0 software and the two protein spots expressed differently were selected for differential identification with MALDI-TOF-MS. [Result] Based on the matching and comparative analysis of the protein spots, 418 protein spots were detected in experimental group, including 226 protein spots with differently expressive levels; according to the mass spectrometry, the two ginsenoside Rbl-related and differentially-expressed protein spots were identified as cytochrome P-450 and phosducin-like protein, and both of them were phosphorylated proteins. | 12 | Deubiquitinase ubiquitin-specific protease 3 (USP3) inhibits HIV-1 replication via promoting APOBEC3G (A3G) expression in both enzyme activity-dependent and -independent manners显示文摘Background: Ubiquitination plays an essential role in many biological processes, including viral infection, and can be reversed by deubiquitinating enzymes (DUBs). Although some studies discovered that DUBs inhibit or enhance viral infection by various mechanisms, there is lack of information on the role of DUBs in virus regulation, which needs to be further investigated.Methods: Immunoblotting, real-time polymerase chain reaction,in vivo/in vitro deubiquitination, protein immunoprecipitation, immunofluorescence, and co-localization biological techniques were employed to examine the effect of ubiquitin-specific protease 3 (USP3) on APOBEC3G (A3G) stability and human immunodeficiency virus (HIV) replication. To analyse the relationship between USP3 and HIV disease progression, we recruited 20 HIV-infected patients to detect the levels of USP3 and A3G in peripheral blood and analysed their correlation with CD4^(+) T-cell counts. Correlation was estimated by Pearson correlation coefficients (for parametric data).Results: The results demonstrated that USP3 specifically inhibits HIV-1 replication in an A3G-dependent manner. Further investigation found that USP3 stabilized 90% to 95% of A3G expression by deubiquitinating Vif-mediated polyubiquitination and blocking its degradation in an enzyme-dependent manner. It also enhances the A3G messenger RNA (mRNA) level by binding to A3G mRNA and stabilizing it in an enzyme-independent manner. Moreover, USP3 expression was positively correlated with A3G expression (r= 0.5110) and CD4^(+) T-cell counts (r= 0.5083) in HIV-1-infected patients.Conclusions: USP3 restricts HIV-1 viral infections by increasing the expression of the antiviral factor A3G.
