1PC4 serves as a negative regulator of skin wound healing in mice显示文摘Background:Human positive cofactor 4(PC4)was initially characterized as a multifunctional transcriptional cofactor,but its role in skin wound healing is still unclear.The purpose of this study was to explore the role of PC4 in skin wound healing through PC4 knock-in mouse model.Methods:A PC4 knock-in mouse model(PC4^(+/+))with a dorsal full-thickness wound was used to investigate the biological functions of PC4 in skin wound healing.Quantitative PCR,Western blot analysis and immunohistochemistry were performed to evaluate the expression of PC4;Sirius red staining and immunofluorescence were performed to explore the change of collagen deposition and angiogenesis.Proliferation and apoptosis were detected using Ki67 staining and TUNEL assay.Primary dermal fibroblasts were isolated from mouse skin to perform cell scratch experiments,cck-8 assay and colony formation assay.Results:The PC4^(+/+)mice were fertile and did not display overt abnormalities but showed an obvious delay in cutaneous healing of dorsal skin.Histological staining showed insufficient re-epithelialization,decreased angiogenesis and collagen deposition,increased apoptosis and decreased cell proliferation in PC4^(+/+)skin.Our data also showed decreased migration rate and proliferation ability in cultured primary fibroblasts from PC4^(+/+)mice in vitro.Conclusions:This study suggests that PC4 might serve as a negative regulator of skin wound healing in mice.Fengying Liao Long Chen Peng Luo Zhongyong Jiang Zelin Chen Ziwen Wang Chi Zhang Yu Wang JintaoHe Qing Wang Yawei Wang Lang Liu Yu Huang Huilan Wang Qingzhi Jiang Min Luo YiboGan Yunsheng Liu Yang Wang Jie Wu Wentao Xie Zhuo Cheng Yali Dai Jialun Li Zujuan Liu Fan Yang Chunmeng Shi 2020Burns & Trauma2020,8,1:1

3Experimental and numerical investigation on the dynamic shear failure mechanism of sandstone using short beam compression specimen显示文摘In this study,a novel testing method is proposed to characterize the dynamic shear property and failure mechanism of rocks by introducing the short beam compression(SBC)specimen into the split Hopkinson pressure bar(SHPB)system.Firstly,the stress distribution of SBC specimen is comprehensively analyzed by finite element method(FEM),and the results show that the optimal notch separation ratio of SBC specimen is C/H?0.2 to achieve successful dynamic simple-shear tests.Then,dynamic shear tests are conducted on sandstone using the SBC-SHPB method.Via careful pulse shaping technique,the dynamic force balance is guaranteed for SBC specimens,and the testing results show that the dynamic shear strength of sandstone is significantly rate-dependent.Combining the results of dynamic compression and tension tests,the failure envelopes of sandstone under different loading rates are obtained in the principle stress plane.It is found that the failure envelope of sandstone constantly expands outwards with increasing loading rate.Moreover,the energy partition of SBC specimen is quantified by virtue of high-speed digital image correlation(DIC)technique.The results show that the kinetic energy portion is non-negligible,and the shear fracture energy increases with increasing loading rate.In addition,the microscopic shear cracking mechanism of SBC specimen is analyzed by the thin section observation:the intra-granular(TG)fracture of minerals dominates the dynamic shear failure of sandstone,and the portion of TG fracture increases with increasing loading rate.This study provides a convenient and reliable method to investigate the dynamic shear property and failure mechanism of rocks.Zelin Yan Feng Dai Yi Liu 2023Journal of Rock Mechanics and Geotechnical Engineering2023,15,8:0
4Study on the Form Factor Estimation Method of High-Speed Catamaran with Asymmetrical Hulls显示文摘The method used to estimate the form factor of low-speed vessel will cause a large error when estimating the form factor of high-speed catamaran because of the interference effects. A method based on computational fluid dynamics( CFD) method is proposed to estimate the form factor of high-speed catamaran with asymmetrical hulls. This paper focused on a 2000-toners catamaran with asymmetrical hulls to compare the difference between normal method and CFD method. The resistance of this catamaran is calculated by the CFD method,and it was compared to the model test data to verify the validity of this method. The form factors calculated by CFD method are very different from the results calculated by Prohaska method in high speed area.Thus,the method used to estimate the form factor of low-speed vessel is not applicative for high-speed catamaran.
