Effect of Magnetic Field on Tribological Properties of Lubricating Oils with and without Tricresyl Phosphate显示文摘Tribological properties of neat 150 SN mineral base oil and the oils doped with different contents of tricresyl phosphate(TCP) under magnetic field or non-magnetic field were evaluated on a four-ball tribotester, on which an external magnetic field was applied. Furthermore, the morphology and the tribochemical characteristics of the worn surfaces were examined with a scanning electron microscope(SEM) and an X-ray photoelectron spectrograph(XPS). The tribological test results showed that the magnetic field improved anti-wear properties but impaired the friction-reducing properties of neat base oil and the TCP-doped oils. The worn surfaces tested in magnetic field were characterized by a slighter wear than those tested in normal condition, especially for the surfaces lubricated with the TCP-doped oils. Furthermore, the results of XPS analysis indicated that tribochemical films on the surfaces tested with TCP-doped oils were mainly composed of ferriccontaining compounds such as Fe_2O_3, Fe_3O_4 and FePO_4. Under the influence of a magnetic field, the atomic concentrations of oxygen and phosphorous in the frictional sub-surfaces were higher than those without magnetic impact. Thus it can be inferred that the improved anti-wear properties and impaired friction-reducing capabilities of lubricating oils with TCP under a magnetic field were related to the diffusion of phosphorus and oxygen into the substrate induced by magnetism.Jiang Zeqi Fang Jianhua Chen Boshui Zheng Zhe Li Hao Xu Lai 2016China Petroleum Processing & Petrochemical Technology2016,18,3:12
Protective effect of Jiaweibugan decoction against diabetic peripheral neuropathy显示文摘Oxygen free radical damage is regarded as a direct or indirect common pathway associated with diabetic neuropathy and is the main cause of complications in peripheral neuropathies. We speculate that Jiaweibugan decoction has a significant effect in treating diabetic peripheral neuropathy through an anti-oxidative stress pathway. In this study, a diabetic rat model was established by intraperitoneal injection of streptozotocin. Rats were treated with Jiaweibugan decoction via intragastric administration. The levels of malondialdehyde and glutathione, which are indirect indexes of oxidative stress, in serum were determined using a colorimetric method. The expression levels of nuclear factor kappa B p65 mRNA and p38 mitogen-activated protein kinase, which are oxidative stress associated factors, in the dorsal root ganglion of spinal S 4-6 segments were evaluated by reverse-transcriptase polymerase chain reaction and immunohistochemistry. Results showed that, Jiaweibugan decoction significantly ameliorated motor nerve conduction velocity in diabetic rats, effectively decreased malondialdehyde levels in serum and the expression of nuclear factor kappa B p65 mRNA and p38 mitogen-activated protein kinase mRNA in the dorsal root ganglion, and increased glutathione levels in serum. Therefore, our experimental findings indicate that Jiaweibugan decoction plays an anti-oxidative stress role in the diabetic peripheral neuropathy process, which has a protective effect on peripheral nerve injury.Yu Wang Zeqi Chen Renqun Ye Yulei He Yuhong Li Xinjian Qiu 2013Neural Regeneration Research2013,8,12:5
CoP nanoparticles embedded in P and N co-doped carbon as efficient bifunctional electrocatalyst for water splitting显示文摘Noble-metal-free hydrogen/oxygen evolution reaction(HER/OER) electrocatalysts, especially bifunctional electrocatalysts, are essential for overall water splitting, but their performance is impeded by many factors like poor electrical conductivity. Herein, we fabricated cobalt phosphide(Co P) nanoparticles embedded in P and N co-doped carbon(PNC) matrix(Co P@PNC) to fully realize the high activity of Co P by maximizing its conductivity. Simply a carbonization coupled phosphidation approach was utilized where Co ions and organic ligands of Co-MOF were transferred into Co P and P and N co-doped carbon. The synthesized material shows an ideal electrical conductivity, excellent HER(overpotential of-84 m V and-120 m V @10 m A cm^(-2) in acidic and alkaline medias, respectively) and OER(overpotential of 330 m V@10 m A cm^(-2) in alkaline media) performances. Further, Co P@PNC acts as a superior catalyst for both anode and cathode to catalyze overall water splitting and only requires an voltage of 1.52 V to deliver a current density of 10 m A cm^(-2), superior to the noble-metal catalysts system(Pt/C//IrO_2) and the reported noble-metal-free bifunctional electrocatalysts.Zeqi Zhou Nasir Mahmood Yongchao Zhang Lun Pan Li Wang Xiangwen Zhang Ji-Jun Zou 2017Journal of Energy Chemistry2017,26,6:4
Nonlinear energy harvesting based on a modified snap-through mechanism显示文摘A modified snap-through mechanism is used in an electromagnetic energy harvester to improve its effectiveness. It mainly comprises three springs that are configured so that the potential energy of the system has two stable equilibrium points. In particular, the small vibration behavior of the harvester around one of the equilibriums is of interest. A multi-scale method(MSM) is used to analyze the frequency response curve. Two snap-through mechanisms are considered. One has both horizontal and vertical springs. The other has only horizontal springs. The frequency response curves of these two classes are compared under the same excitation and electric loading conditions. The latter exhibits more bending of the frequency response curve than the former one. The results are also validated by some numerical work. The averaged power subject to the Gaussian white noise is calculated numerically, and the results demonstrate that bi-stable energy harvesting with only horizontal springs can outperform the mechanism with both horizontal and vertical springs for the same distance between two equilibriums.Zeqi LU Ke LI Hu DING Liqun CHEN 2019Applied Mathematics and Mechanics(English Edition)2019,40,1:3
Symptom characteristics and prevalence of qi deficiency syndrome in people of varied health status and ages: A multicenter cross-sectional study显示文摘Objective:Aging caused by a deficiency syndrome can be found in ancient and modern traditional Chinese medicine literature.Qi deficiency syndrome(QDS)is a vital factor in the aging process. This study aimed to establish a full-scale trial to evaluate the prevalence, symptom severity, frequency, and distribution of QDS in different age groups and varying health status to elucidate the role of qi deficiency in the aging process and deterioration of health. Methods:This cross-sectional study was conducted in four hospitals in China, and data from 1220 participants were included. The participants, aged between 20 and 79 years, completed questionnaires that recorded prevalence of QDS and severity or frequency of relevant symptoms, then were interviewed by investigators. We used frequency analysis and chi-squared tests to perform comparative analysis of prevalence in different age and health groups;we used a ranksum tests for quantitative analysis of symptoms severity and frequency scores;we performed a regression analysis of correlation between syndrome occurrence and potential factors using nonconditional binary logistic stepwise regression of numerical variables.P<0.05 was considered statistically significant. Results:Prevalence, symptom severity and frequency scores of QDS showed a rising trend when physical condition worsened, rather than when age increased.Health status, fatigue, shortness of breath or no desire to talk, spontaneous sweating, swollen tongue with teeth marks on side, and deficient and weak pulse, rather than increasing age were contributing factors to this syndrome.Distribution of QDS in certain health and age stages showed remarkable irregularities. Conclusions:Qi deficiency may be a contributing factor for sub-health(sub-optimal health)and chronic diseases rather than aging. It may play a crucial role in chronic disease pathogenesis of young and middle-aged people, and in sub-health pathogenesis of older adults.Recognition of the warning signs and symptoms of QDS may lead to early intervention and prevention of subhealth, and chronic diseases.Yin Zhang Li Zhang Xia Zhao Yue Liu Shiyu Du Jian Li Ting Liu Fusheng Liu Zeqi Su Yuyong Jiang Xia Ding 2015Journal of Traditional Chinese Medical Sciences2015,2,3:1
Isotoosendanin exerts inhibition on triple-negative breast cancer through abrogating TGF-β-induced epithelial—mesenchymal transition via directly targeting TGFβR1显示文摘As the most aggressive breast cancer,triple-negative breast cancer(TNBC) is still incurable and very prone to metastasis. The transform growth factor β(TGF-β)-induced epithelial—mesenchymal transition(EMT) is crucially involved in the growth and metastasis of TNBC.This study reported that a natural compound isotoosendanin(ITSN) reduced TNBC metastasis by inhibiting TGF-β-induced EMT and the formation of invadopodia.ITSN can directly interact with TGF-β receptor type-1(TGFβR1) and abrogated the kinase activity of TGFβR1,thereby blocking the TGF-β-initiated downstream signaling pathway. Moreover, the ITSN-provided inhibition on metastasis obviously disappeared in TGFβR1-overexpressed TNBC cells in vitro as well as in mice bearing TNBC cells overexpressed TGFβR1.Furthermore,Lys232 and Asp351 residues in the kinase domain of TGFβR1 were found to be crucial for the interaction of ITSN with TGFβR1.Additionally,ITSN also improved the inhibitory efficacy of programmed cell death 1 ligand 1(PD-L1) antibody for TNBC in vivo via inhibiting the TGF-β-mediated EMT in the tumor microenvironment. Our findings not only highlight the key role of TGFβR1 in TNBC metastasis, but also provide a leading compound targeting TGFβR1 for the treatment of TNBC metastasis. Moreover, this study also points out a potential strategy for TNBC treatment by using the combined application of anti-PD-L1 with a TGFβR1 inhibitor.Jingnan Zhang Ze Zhang Zhenlin Huang Manlin Li Fan Yang Zeqi Wu Qian Guo Xiyu Mei Bin Lu Changhong Wang Zhengtao Wang Lili Ji 2023Acta Pharmaceutica Sinica B2023,13,7:1

Proteomic analysis of effects of Chinese herbs to calm the liver and suppress hyperactive yang in a rat migraine model显示文摘BACKGROUND: The expression of ubiquitin and energy-associated protein can provoke migraines. Studies have suggested that expression is closely linked to 'hyperactivity of liver-yang theory' in Traditional Chinese Medicine (TCM), as well as the function of periphery sympathetic nerve medulla. OBJECTIVE: To observe proteomic changes in a rat migraine model with regard to hyperactivity of liver-yang when treated with Chinese herbs to calm the liver and suppress hyperactive yang compound. DESIGN, TIME AND SETTING: A randomized controlled study. This study was performed at the laboratory of Institute of Integrated Traditional Chinese and Western Medicine, Institute of Human Reproduction and Stem Cell Engineering and Key Laboratory of Cancer Proteomics of Ministry of Health, Xiangya Hospital Affiliated to Central South University between September 2006 and July 2007. MATERIALS: Thirty, male, healthy, Sprague-Dawley rats, aged eight weeks, were included in the final analysis. Aconite, to calm the liver and suppress hyperactive yang compound, was provided by the Dispensary of Traditional Chinese medicine, Xiangya Hospital, Central South University. A physiological electronic stimulator, type SDQ-1, was provided by Bengbu Practical Institute of Technology. The left trigeminal ganglion was localized and stimulated for 10 minutes, and the rats were orally administered an aconite concoction to establish a rat migraine model with hyperactivity of liver-yang. METHODS: Rats were randomly divided into a normal control group, model group, and TCM treatment group, with 10 rats in each group. The TCM treatment group was orally treated to calm the liver and suppress the hyperactive yang compound once a day for 28 days. In contrast, the model group and normal group were orally administered the same amount of distilled water once a day for 28 days. MAIN OUTCOME MEASURES: The total proteins from adrenal glands of the three groups were separated by two-dimensional gel electrophoresis (2-DE), and 2-DE images were analyzed by PDQuest 7.0 software. Matrix-Assisted Laser Desorption/Ionization-Time of Flight Mass Spectrometry (MALDI-TOF-MS) was used to obtain peptide mass fingerprints of the differential proteins. Databases were searched to identify the proteins. RESULTS: A total of 30 rats were included in the final analysis. Reproducible 2-DE patterns from rat adrenal gland of the three groups were obtained. Compared with the normal group, nine proteins were down-regulated and five proteins were up-regulated in the model group; however, these expressions returned to normal, or near normal levels, in the TCM treatment group. A total of eight differentially expressed proteins were identified: glycogen phosphorylase, ATP synthase D chain, isovaleryl-CoA dehydrogenase, ubiquitin, Annexin-3, Annexin-A1, Peroxirdoxin-II, and heat shock protein-27. CONCLUSION: Liver calming and suppression of the hyperactive yang compound may up-regulate expression of proteins related to energy metabolism and the ubiquitin system. Compounds that are used to treat migraines may contribute to protein functions in the peripheral sympathetic nervous system.Jianjun Hu Zeqi Chen Guangwei Zhong Wei Li Yaohui Yin 2008Neural Regeneration Research2008,3,5:1
How does family background affect children’s educational achievement?Evidence from Contemporary China显示文摘Education is a lasting process.Academic performance in primary education plays a crucial role in obtaining further educational opportunities. Thus,it is necessary to examine how family background affects children’s academic achievement at an early stage.Through analysis of data from the Chinese Family Panel Study in 2010(CFPS2010),this paper proposes two pathways through which family influences children’s academic performance.Firstly, parents compete for high-quality educational opportunities for their children and better educational opportunities lead to better academic performance.Secondly, parenting behavior and educational support for their children could cultivate children’s learning habits and affect academic performance. We also find urban students’academic performance are more heavily affected by their families’socioeconomic status compared with rural students. These findings bear important implications for how to reduce the class difference in students’academic performance and promote educational equity in contemporary China.Zhonglu Li Zeqi Qiu 2018The Journal of Chinese Sociology2018,5,1:1
Is the“seven-year itch”real?—a study on the changing divorce pattern in Chinese marriages显示文摘Drawing from the data collected in the 2010 China Family Panel Study(CFPS)baseline survey, this paper studies the changing divorce pattern in Chinese marriages as a whole and for four cohorts. We find an inverted U-shape pattern in the divorce risk for Chinese marriages with the peak coming earlier over time.Family life cycle theory, natural evolvement theory, and population heterogeneity theory provide three different perspectives on interpreting the“inverted-U”shape divorce pattern.Family life cycle theory and natural evolvement theory are not supported by the data;however, population heterogeneity theory fits the data quite well.Split-population model shows that divorce risk for divorced couples increase linearly with marriage duration. However, the curve must come down at the end since the majority of the population does not divorce.By comparing four cohorts divided by marriage year,we find that the proportion of divorced couples has increased over time and the slope of the risk function for divorced couples has increased rapidly.Qi Xu Zeqi Qiu Jianxin Li 2016The Journal of Chinese Sociology2016,3,1:0

Peroxisome proliferator-activated receptors gama ameliorates liver fibrosis in non-alcoholic fatty liver disease by inhibiting TGF-β/Smad signaling activation显示文摘Background:Nonalcoholic fatty liver disease(NAFLD)is a chronic condition characterized by a progressive decline in liver function, leading to disruptions in liver integrity and metabolic function, resulting in lipid deposition and excessive accumulation of extracellular matrix(ECM).The pathogenesis of NAFLD is complex and not yet fully understood, contributing to the absence of specific therapeutic strategies.Peroxisome proliferator-activated receptor gamma(PPARγ)is a ligand-activated transcription factor pivotal in regulating lipid and glucose metabolism. However, the impacts of PPARγon NAFLD remains insufficiently explored. Thus, this study aimed to investigate the role of PPARγin NAFLD and its underlying molecular mechanisms. Methods:Chemical detection kits were utilized to quantify collagen content, alanine aminotransferase(ALT),and aspartate aminotransferase(AST)level variations.Quantitative real-time polymerase chain reaction(qRT-PCR)was employed to assess alterations in extracellular matrix-related genes and inflammatory response genes in liver tissue and HepG2 cells, while western blotting was conducted to analyze the levels of both PPARγand the TGF-β/Smad signaling pathway. Results:Our findings unveiled significantly reduced PPARγexpression in a rat model of NAFLD,leading to subsequent activation of the TGF-β/Smad signaling pathway.Furthermore,PPARγactivation effectively mitigated NAFLD progression by inhibiting inflammation and fibrosis-related gene expression and collagen production. On a cellular level,PPARγactivation was found to inhibit the expression of extracellular matrix-related genes such as matrix metalloproteinase 2(MMP2)and matrix metalloproteinase 9(MMP9),along with inflammatory response genes interleukin(IL)-1βand IL-6.Additionally,PPARγactivation led to a significant decrease in the levels of ALT and AST.At the molecular level,PPARγnotably down-regulated the TGF-β/Smad signaling pathway, which is known to promote liver fibrosis. Conclusion:These groundbreaking findings underscore PPARγactivation as a promising therapeutic approach to delay NAFLD progression by targeting the TGF-β/Smad signaling pathway in hepatic cells. This highlights the potential of PPARγas a promising therapeutic target for NAFLD management in clinical settings.Qingwei Zhang Wenjie Zhao Zeqi Sun Xinxin Dong Liwei Zhu Zhen Zhang Ximing Chen Yingying Hu Menghan Du Jiamin Li Yong Zhang 2024Frigid Zone Medicine2024,4,1:0
High-Risk LEO Satellite Network Path Detection Based on Spatial and Temporal Delay Anomaly Analysis显示文摘The gradual deployment of Low-Earth Orbit(LEO)mega constellations with inter-satellite links(ISLs)promises ubiquitous,low-latency,and high-throughput satellite network services. However, networked LEO satellites with ISLs are also at risk of routing attacks such as hijacking.Existing defenses against route hijacking in terrestrial networks can hardly work for the LEO satellite network due to its high spatiotemporal dynamics. To deal with it,we propose RPD,a high-risk routing path detection method for LEO mega-constellation networks.RPD detects abnormal high-risk LEO network paths by checking the consistency between the path delay and the geographical distance. This is efficiently achieved by combining in-band measurements and out-of-band statistical processing to detect the anomaly of the clustering feature in the reference delay matrix.RPD avoids the recalculation of the header cryptographic marks when the handover occurs, thus greatly reducing the cost and improving the performance of highrisk path detection.Experiments showed that the proposed RPD mechanism achieves an average detection accuracy of 91.64%under normal network conditions, and maintain about 89%even when congestion occurs in multiple areas of the network and measurement noise is considered. In addition,RPD does not require any cryptographic operation on the intermediate node, only minimal communication cost with excellent scalability and deployability.Tianyu Zhang Hewu Li Jun Liu Lu Lu Qian Wu Shaowen Zheng Zeqi Lai Yuanjie Li 2023China Communications2023,20,7:0
A novel Tb^(3+) and Eu^(3+) co-doped dual-emitting phosphate K_(3)SrBi(P_(2)O_(7))_(2) phosphor for application in FIR thermometers显示文摘Rare earth co-doped phosphor for fluorescence intensity ratio(FIR) thermometer has gained increasing attention in recent years. Herein, the novel Tb^(3+)and Eu^(3+)co-doped K_(3)SrBi(P_(2)O_(7))_(2)(KSBP) phosphate phosphors were reported. The crystal structure of the title phosphor was determined using Rietveld refinement and proved to have a three-dimensional structure. The time-resolved spectroscopy reveals that there is almost no energy transfer between Tb^(3+)and Eu^(3+). More importantly, Tb^(3+)and Eu^(3+)emissions show different thermal quenching behaviors, which claims the potential of this material for application in optical thermometer. The FIR of the typical KSBP:0.02Tb^(3+),0.04Eu^(3+)sample demonstrates a polynomial relationship as a function of temperature and the absolute and relative sensitivity are0.025 K^(-1) and 0.59%/K, respectively. In general, our study reports a novel and potential KSBP:Tb^(3+),Eu^(3+)phosphate phosphor that is promising for use in high-sensitive FIR thermometers.Baochen Wang Shifeng Sun Zeqi Li Zongwang Li Weiwei Lin Chao Zeng Yan-gai Liu Ruiyu Mi 2024Journal of Rare Earths2024,42,3:0
Functional groups and seasonal diversity of crustacean zooplankton in adjacent waters of Haizhou Bay,South Yellow Sea显示文摘Zooplankton are important linkages in the food web and can respond nonlinearly to environmental changes.Marine organisms thrive from spring to summer. Thus,it is crucial to understand how ecological functions of zooplankton communities may shift under seasonal environmental changes during this period.Samples were collected from May to August(May,June-Ⅰ,June-Ⅱ,July-Ⅰ,July-Ⅱ,and August)in 2018 in Haizhou Bay,Jiangsu,East China for zooplankton and environmental variables.Crustaceans accounted for 75 out of 134 zooplankton taxa and 91.8%of total zooplankton abundance. The average abundance of crustacean varied between 2824.6±635.4 inds./m3 in July-Ⅱand 6502.7±1008.8 inds./m3 in June-Ⅱ.Multivariate analyses results showed that the dissimilarity of community increased gradually in the time series.Body length, feeding type, trophic group, and reproduction mode were used to investigate crustacean community functions.Trait-based functional groups contained species with similar ecological roles.Functional diversity fused the differences of species and trait. The proportion of large-sized species(2-5 mm)decreased with the increasing proportion of medium-sized species(1-2 mm).The proportion of current feeders increased with the drop in the proportion of mixed feeders.Parthenogenesis species increased with decreasing free spawners, and omnivores-carnivores increased with decreasing omnivoresherbivores.Generalized additive models suggested that temperature was the main driver of variations in crustacean zooplankton function.Seven identified functional groups varied with increasing temperature.Omnivorous-herbivorous copepods declined(90.0%-68.0%),whereas the parthenogenetic cladocerans increased(0-24.1%).The small egg-brooding ambush copepods fluctuated(6.5%-9.3%)with increasing water temperature. The other functional groups changed slightly.Functional diversity also varied according to temperature changes. The community structure and ecological function of crustacean zooplankton community showed gradual changes with increasing temperature from spring to summer.Zeqi ZHENG Shunan FU Yixuan LI Ruping GE Hongju CHEN Zhenjiang YE Yunyun ZHUANG Guangxing LIU 2023Journal of Oceanology and Limnology2023,41,3:0
Size and shape effects of MnFe_(2)O_(4)nanoparticles as catalysts for reductive degradation of dye pollutants显示文摘The magnetic nanoparticles that are easy to recycle have tremendous potential as a suitable catalyst for environmental toxic dye pollutant degradation.Rationally engineering shapes and tailoring the size of nanocatalysts are regarded as an effective manner for enhancing performances.Herein,we successfully synthesized three kinds of MnFe_(2)O_(4)NPs with distinctive sizes and shapes as catalysts for reductive degradation of methylene blue, rhodamine 6G,rhodamine B,and methylene orange. It was found that the catalytic activities were dependent on the size and shape of the MnFe_(2)O_(4)NPs and highly related to the surface-to-volume ratio and atom arrangements.Besides, all these nanocatalysts exhibit selectivity to different organic dyes, which is beneficial for their practical application in dye pollutant treatment.Furthermore, the MnFe_(2)O_(4)NPs could be readily recovered by a magnet and reused more than ten times without appreciable loss of activity. The size and shape effects of MnFe_(2)O_(4)nanoparticles demonstrated in this work not only accelerate further understanding the nature of nanocatalysts but also contribute to the precise design of nanoparticles catalyst for pollutant degradation.Guowen Hu Zeqi Zhang Xuan Zhang Tianrong Li 2021Frontiers of Environmental Science & Engineering2021,15,5:0
Mitigating growth-stress tradeoffs via elevated TOR signaling in rice显示文摘Rice production accounts for approximately half of the freshwater resources utilized in agriculture,result-ing in greenhouse gas emissions such as methane(CH4)from flooded paddy fields. To address this chal-lenge,environmentally friendly and cost-effective water-saving techniques have become widely adopted in rice cultivation. However, the implementation of water-saving treatments(WsTs)in paddy-field rice has been associated with a substantial yield loss of up to 50%as well as a reduction in nitrogen use efficiency(NUE).In this study,we discovered that the target of rapamycin(TOR)signaling pathway is compromised in rice under WsT.Polysome profiling-coupled transcriptome sequencing(polysome-seq)analysis unveiled a substantial reduction in global translation in response to WST associated with the downregulation of TOR activity.Molecular, biochemical, and genetic analyses revealed new insights into the impact of the positive TOR-S6K-RPS6 and negative TOR-MAF1 modules on translation repression under WST.Intriguingly, ammonium exhibited a greater ability to alleviate growth constraints under WsT by enhancing TOR signaling, which simultaneously promoted uptake and utilization of ammonium and nitrogen allocation. We further demonstrated that TOR modulates the ammonium transporter AMT1;1 as well as the amino acid permease APP1 and dipeptide transporter NPF7.3 at the translational level through the 5'untranslated region.Collectively, these findings reveal that enhancing TOR signaling could mitigate rice yield penalty due to WST by regulating the processes involved in protein synthesis and NUE.Our study will contribute to the breeding of new rice varieties with increased water and fertilizer utilization efficiency.Wei Li Jiaqi Liu Zeqi Li Ruiqiang Ye Wenzhen Chen Yuqing Huang Yue Yuan Yi Zhang Huayi Hu Peng Zheng Zhongming Fang Zeng Tao Shiyong Song Ronghui Pan Jian Zhang Jumim Tu Jen Sheen Hao Du 2024Molecular Plant2024,17,2:0
RNA barcode segments for SARS-CoV-2 identification from HCoVs and SARSr-CoV-2 lineages显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),the pathogen responsible for coronavirus disease 2019(COVID-19),continues to evolve, giving rise to more variants and global reinfections.Previous research has demonstrated that barcode segments can effectively and cost-efficiently identify specific species within closely related populations. In this study,we designed and tested RNA barcode segments based on genetic evolutionary relationships to facilitate the efficient and accurate identification of SARS-CoV-2 from extensive virus samples, including human coronaviruses(HCoVs)and SARSr-CoV-2 lineages.Nucleotide sequences sourced from NCBI and GISAID were meticulously selected and curated to construct training sets, encompassing 1733 complete genome sequences of HCoVs and SARSr-CoV-2 lineages.Through genetic-level species testing,we validated the accuracy and reliability of the barcode segments for identifying SARS-CoV-2.Subsequently,75 main and subordinate species-specific barcode segments for SARS-CoV-2,located in ORF1ab,S,E,ORF7a,and N coding sequences, were intercepted and screened based on single-nucleotide polymorphism sites and weighted scores.Post-testing,these segments exhibited high recall rates(nearly 100%),specificity(almost 30%at the nucleotide level),and precision(100%)performance on identification. They were eventually visualized using one and two-dimensional combined barcodes and deposited in an online database(http://gffzz5c123ad9140e4e5fh9kck5x9fn0oq69vn.ffgz.tsg.suse.edu.cn/).The successful integration of barcoding technology in SARS-CoV-2 identification provides valuable insights for future studies involving complete genome sequence polymorphism analysis. Moreover, this cost-effective and efficient identification approach also provides valuable reference for future research endeavors related to virus surveillance.Changqiao You Shuai Jiang Yunyun Ding Shunxing Ye Xiaoxiao Zou Hongming Zhang Zeqi Li Fenglin Chen Yongliang Li Xingyi Ge Xinhong Guo 2024Virologica Sinica2024,39,1:0
