1A viral protein orchestrates rice ethylene signaling to coordinate viral infection and insect vector-mediated transmission显示文摘Arthropod-borne viruses cause serious threats to human health and global agriculture by rapidly spreading via insect vectors. Southern rice black-streaked dwarf virus (SRBSDV) is the most damaging rice-infecting virus that is frequently transmitted by planthoppers. However, the molecular mechanisms underlying its propagation in the host plants and epidemics in the field are largely unknown. Here, we showed that the SRBSDV-encoded P6 protein is a key effector that regulates rice ethylene signaling to coordinate viral infection and transmission. In early SRBSDV infection, P6 interacts with OsRTH2 in the cytoplasm to activate ethylene signaling and enhance SRBSDV proliferation;this also repels the insect vector to reduce infestation. In late infection, P6 enters the nucleus, where it interacts with OsEIL2, a key transcription factor of ethylene signaling. The P6-OsEIL2 interaction suppresses ethylene signaling by preventing the dimerization of OsEIL2, thereby facilitating viral transmission by attracting the insect vector. 2miR-100-5p in human umbilical cord mesenchymal stem cell-derived exosomes mediates eosinophilic inflammation to alleviate atherosclerosis via the FZD5/Wnt/β-catenin pathway显示文摘Exosomes derived from human umbilical cord mesenchymal stem cells(hUCMSC-Ex)play important roles in immune and inflammation diseases.However,the role of hUCMSC-Ex in atherosclerosis has not been elucidated.In this study,the isolated exosomes were identified by transmission electron microscopy and nanoparticle tracking analysis.Exosome marker protein levels were increased in the hUCMSC-Ex compared with those in hUCMSC suspension,indicating that exosomes were successfully isolated from hUCMSCs.Furthermore,eosinophils were treated with oxidized low-density lipoprotein(ox-LDL)to construct inflammation model and then incubated with hUCMSC-Ex derived from hUCMSCs which were transfected with miR-100-5p mimic or miR-100-5p inhibitor.We found that hUCMSC-Ex increased miR-100-5p expression,inhibited cell migration,promoted cell apoptosis,and reduced inflammatory cytokine levels in ox-LDL-treated eosinophils,and miR-100-5p overexpression in hUCMSCs enhanced these effects,while miR-100-5p inhibition reversed these effects.Moreover,frizzled 5(FZD5)was a target gene of miR-100-5p.FZD5 overexpression reversed the inhibitory effects of hUCMSC-Ex-miR-100-5p on cell progression and inflammation in eosinophils.Additionally,hUCMSC-Ex-miR-100-5p decreased the expression of cyclin D1 andβ-catenin proteins.Wnt/β-catenin pathway activator BML-284 effectively reversed the effects of hUCMSC-Ex-miR-100-5p on cell progression and inflammation in eosinophils.ApoE−/−mice were fed with high-fat diet to construct an atherosclerosis mice model,and hUCMSC-Ex was injected into mice.hUCMSC-Ex reduced atherosclerotic plaque area and inflammation response in atherosclerosis mice.This study demonstrates that hUCMSC-Ex-miR-100-5p inhibits cell progression and inflammatory response in eosinophils via the FZD5/Wnt/β-catenin pathway,thereby alleviating atherosclerosis progression.Heng Gao Zhanbiao Yu Yuanyuan Li Xue Wang 2021Acta Biochimica et Biophysica Sinica2021,53,9:3
3Genotypic variation in spatiotemporal distribution of canopy light interception in relation to yield formation in cotton显示文摘Background: Within-canopy interception of photosynthetically active radiation(PAR) impacts yield and other agronomic traits in cotton(Gossypium hirsutum L.). Field experiments were conducted to investigate the influence of 6 cotton varieties(they belong to 3 different plant types) on yield, yield distribution, light interception(LI), LI distribution and the relationship between yield formation and LI in Anyang, Henan, in 2014 and 2015.Result: The results showed that cotton cultivars with long branches(loose-type) intercepted more LI than did cultivars with short branches(compact-type), due to increased LI in the middle and upper canopy. Although loose-type varieties had greater LI, they did not yield significantly higher than compact-type varieties, due to decreased harvest index. Therefore, improving the harvest index by adjusting the source-to-sink relationship may further increase cotton yield for loose-type cotton. In addition, there was a positive relationship between reproductive organ biomass accumulation and canopy-accumulated LI, indicating that enhancing LI is important for yield improvement for each cultivar. 4Biofilm-overproducing Bacillus subtilis B12ΔYwcc decreases Cd uptake in Chinese cabbage through increasing Cd-immobilizing related gene abundance and root surface colonization显示文摘Biofilm-producing bacteria can decrease Cd uptake in vegetables, but mechanisms underlying this effect are poorly characterized. In this study, two mutant strains B12ΔYwcc and B12ΔSlr R were constructed from a biofilm-producing Bacillus subtilis strain B12. Then, the impacts of strain B12 and its high biofilm-producing mutant strain B12ΔYwcc and low biofilmproducing mutant strain B12ΔSlr R on Cd availability and uptake in Chinese cabbage and the related mechanisms were investigated in the Cd-polluted soil. Strain B12 and its mutants B12ΔYwcc and B12ΔSlr R increased the dry biomasses of edible tissues by 54%–130% compared with the controls. Strain B12 and its mutant B12ΔYwcc reduced the soil available Cd content by 36%–50% and root and edible tissue Cd contents by 23%–50% compared with the controls. Furthermore, the mutant strain B12ΔYwcc reduced the edible tissue Cd content by40% and increased the polysaccharide content by 23%, invertase activity by 139%, and gene copies of the cum A by 4.5-fold, eps A by 7.1-fold, and cad A by 4.3-fold, which were involved in Cd adsorption in the rhizosphere soils, respectively, compared with strain B12. The polysaccharide content and cum A, eps A, and cad A gene copy numbers showed significantly reverse correlations with the available Cd content. Notably, the mutant strain B12ΔYwcc showed better ability to colonize the vegetable root surface than strain B12. 14Experimental study of hysteresis and catastrophe in a cavity-based scramjet combustor显示文摘Hystereses and catastrophes were experimentally investigated in a cavity-based scramjet combustor.The inflow Mach number was 3.0.Fuel Equivalence Ratio(ER)was continuously regulated with multi-steps to explore influences of historical regulation directions on combustion states.Two divided hysteresis loops with catastrophes were observed.By 1-D flow estimations,the first loop occurred with shock-free/separated scramjet mode transitions,while the second kept in the separated scramjet mode.This breaks through the traditional knowledge that hysteresis and catastrophe were certainly related to ramjet/scramjet mode transitions.The first hysteresis and catastrophes were attributed to flame stabilization mode transitions between the cavity shearlayer stabilized and the jet-wake stabilized,with flow separation establishment/vanishment upstream the cavities.The obvious variations of flame and shock/separation structures meant large wall-pressure changes in the expansive duct,and generated obvious thrust catastrophes.Besides,transition ER and catastrophe were larger in historical ER-increasing path because combustion efficiency became obviously larger as flow separation established.Difference of critical transition ERs meant the first hysteresis.The second hysteresis and catastrophes in the jet-wake stabilized mode were attributed to flame/shock interaction mode transitions between the flame/shock weak interaction mode and intensive interaction mode.Each transition caused slightly stronger/weaker flame interacting with slightly larger/smaller flow separation,which meant small wall-pressure changes in the expansive duct,and thus thrust catastrophe was unobvious.Hysteresis occurred as the critical transition ER was slightly higher in historical ER-increasing path because of slightly lower combustion efficiency under slightly smaller separation.Xu ZHANG Qifan ZHANG Zhenjie WU Lianjie YUE Zhanbiao GAO Weihang LUO Hao CHEN 2022Chinese Journal of Aeronautics2022,35,10:0
