| 7 | Au nanoclusters suppress chronic lymphocytic leukaemia cells by inhibiting thioredoxin reductase 1 to induce intracellular oxidative stress and apoptosis显示文摘Chronic lymphocytic leukaemia(CLL) is a rare blood cancer that always relapses as refractory disease and eventually leads to death. To date, therapeutic options for CLL patients are scarce and there is an urgent need to develop novel chemotherapeutics that are both effective and safe. Gold-containing compounds induce a lethal oxidative and endoplasmic reticulum stress response in cultured and primary CLL cells via inhibition of thioredoxin reductase(TrxR). However, traditional gold-containing medicines have revealed side effects during clinical applications. Therefore, safer gold-containing drugs are needed to overcome this challenge. In this study, a novel peptide templated gold cluster Au_(25)Sv_9 was synthesized and its therapeutic effect on CLL cells was evaluated. This nanocluster could induce cell apoptosis in MEC-1 cells in a dose-dependent manner which correlated with the uptake amount of clusters in cells.As expected, increasing intracellular reactive oxidative species(ROS) in MEC-1 cells was exhibited with the increase of cluster dosage. Further analyses demonstrated the underlying mechanism that the nanoclusters suppress the activity of TrxR1, increase the level of intracellular ROS, destroy the mitochondrial membrane potential and finally trigger the mitochondrial apoptotic pathway in MEC-1 cells.Furthermore, the direct interaction between Au_(25)Sv_9 clusters and TrxR1 was confirmed for the first time by isothermal titration calorimetry. | 9 | A viral protein orchestrates rice ethylene signaling to coordinate viral infection and insect vector-mediated transmission显示文摘Arthropod-borne viruses cause serious threats to human health and global agriculture by rapidly spreading via insect vectors. Southern rice black-streaked dwarf virus (SRBSDV) is the most damaging rice-infecting virus that is frequently transmitted by planthoppers. However, the molecular mechanisms underlying its propagation in the host plants and epidemics in the field are largely unknown. Here, we showed that the SRBSDV-encoded P6 protein is a key effector that regulates rice ethylene signaling to coordinate viral infection and transmission. In early SRBSDV infection, P6 interacts with OsRTH2 in the cytoplasm to activate ethylene signaling and enhance SRBSDV proliferation;this also repels the insect vector to reduce infestation. In late infection, P6 enters the nucleus, where it interacts with OsEIL2, a key transcription factor of ethylene signaling. The P6-OsEIL2 interaction suppresses ethylene signaling by preventing the dimerization of OsEIL2, thereby facilitating viral transmission by attracting the insect vector. | 10 | A botanical medicine dragon’s blood exhibited clinical antithrombosis efficacy similar to low molecular weight heparin显示文摘Deep vein thrombosis(DVT)is a common complication following traumatic fracture with a 0.5%–1%annual incidence.Low molecular weight heparin(LMWH)is the most commonly used anticoagulation drug for DVT prevention,but treatment with LMWH is invasive.Our aim is to compare the antithrombotic effect of dragon’s blood,an oral botanical anticoagulant medicine approved by the Chinese FDA,with LMWH in patients undergoing hip fracture surgery and to explore the molecular mechanisms of anticoagulation treatment.Our study recruited patients and divided them into LMWH and dragon’s blood treatment group.Coagulation index tests,Doppler ultrasound and mRNA sequencing were performed before and after anticoagulation therapy.There was no significant difference in postoperative DVT incidence between the two groups(23.1%versus 15.4%,P=0.694).D-dimer(D-D)and fibrinogen degradation product(FDP)showed significant reductions in both groups after anticoagulation treatments.We identified SLC4A1,PROS1,PRKAR2B and seven other genes as being differentially expressed during anticoagulation therapy in both groups.Genes correlated with coagulation indexes were also identified.Dragon’s blood and LMWH showed similar effects on DVT and produced similar gene expression changes in patients undergoing hip fracture surgery,indicating that dragon’s blood is a more convenient antithrombosis medicine(oral)than LMWH(hypodermic injection). | Jiangbin Liang Song Mei Xiangyu Qiao Wei Pan Yan Zhao Shaohui Shi Yaling Zhai Haizhao Wen Guoping Wu Chengyu Jiang | 2021 | Science China(Life Sciences)2021,64,10: | 4 |
| 11 | Spatial and Temporal Variations in Earthquake Stress Drops between the 2008 Wenchuan and 2013 Lushan Earthquakes显示文摘As a case study of spatial and temporal variations in earthquake stress drops between the 2008 Ms 8.0 Wenchuan and 2013 Ms 7.0 Lushan earthquakes,we computed 1828 stress-drop values for earthquakes with magnitudes 1.7≤ML≤5.0 during an eight-year time span before and after major earthquakes.We divide the study area into three subregions(the southern segment of the Longmen Shan fault zone;the southwestern junction of the Longmen Shan and Sichuan Basin;and the southwestern margin of the Sichuan Basin)and calculate individual event stress drops in each.The results show that regions of alternating high and low stress drop are found on either side of the southwestern segment of the Longmen Shan fault zone.During the two-year period after the 2008 Ms 8.0 Wenchuan earthquake,the stress state of the southern Longmen Shan fault shows no significant change.A marked increase in stress level appears about 18 months before the 2013 Ms 7.0 Lushan earthquake near the Lushan hypocenter zone.Two months after the Ms 7.0 event,the stress drops suddenly attenuate,with significantly less seismic energy release per event.We find that changes in the patterns of high and low stress drop values are consistent with the process of stress accumulation or transfer from the pre-mainshock to postmainshock periods.The results indicate that major earthquakes are the dominant cause of temporal and spatial evolution in stress levels.Stress drop variations show obvious temporal and spatial patterns that may suggests subtle changes in the character of stress fields on faults and spatial variations related to local intense compression and tectonic effects. | WU Weiwei LONG Feng LIANG Mingjian WEI Yaling SUN Wei CHEN Xuefen ZHAO Jing | 2020 | Acta Geologica Sinica(English Edition)2020,94,5: | 4 |
