1Peripheral T-lymphocyte subpopulations in different clinical stages of chronic HBV infection correlate with HBV load显示文摘AIM: To characterize the peripheral T-cell subpopulation profiles and their correlation with hepatitis B virus (HBV) replication in different clinical stages of chronic HBV infection.METHODS: A total of 422 patients with chronic HBV infection were enrolled in this study. The patients were divided into three stages: immune-tolerant stage, immune active stage, and immune-inactive carrier stage. Composition of peripheral T-cell subpopulations was determined by flow cytometry. HBV markers were detected by enzyme-linked immunosorbent assay. SerumHBV DNA load was assessed by quantitative real-time polymerase chain reaction.RESULTS: CD8+ T-cells were significantly higher in patients at the immune-tolerant stage than in patients at the immune-active and -inactive carrier stages (36.87 ± 7.58 vs 34.37 ± 9.07, 36.87 ± 7.58 vs 28.09 ± 5.64, P < 0.001). The peripheral blood in patients at the immune-tolerant and immune active stages contained more CD8+ T-cells than CD4+ T-cells (36.87 ± 7.58 vs 30.23 ± 6.35, 34.37 ± 9.07 vs 30.92 ± 7.40, P < 0.01), whereas the peripheral blood in patients at the immune-inactive carrier stage and in normal controls contained less CD8+ T-cells than CD4+ T-cells (28.09 ± 5.64 vs 36.85 ± 6.06, 24.02 ± 4.35 vs 38.94 ± 3.39, P < 0.01). ANOVA linear trend test showed that CD8+ T-cells were signif icantly increased in patients with a high viral load (39.41 ± 7.36, 33.83 ± 7.50, 31.81 ± 5.95 and 26.89 ± 5.71, P < 0.001), while CD4+ T-cells were signif icantly increased in patients with a low HBV DNA load (37.45 ± 6.14, 33.33 ± 5.61, 31.58 ± 6.99 and 27.56 ± 5.49, P < 0.001). Multiple regression analysis displayed that log copies of HBV DNA still maintained its highly signif icant coefficients for T-cell subpopulations, and was the strongest predictors for variations in CD3+, CD4+ and CD8+ cells and CD4+/CD8+ ratio after adjustment for age at HBV-infection, maternal HBV-infection status, presence of hepatitis B e antigen and HBV mutation.CONCLUSION: Differences in peripheral T-cell subpopulation profi les can be found in different clinical stages of chronic HBV infection. 2Significance of dredging on sediment denitrification in Meiliang Bay,China:A year long simulation study显示文摘An experiment for studying the effects of sediment dredging on denitrification in sediments was carried out through a one-year incubation of undredged (control) and dredged cores in laboratory. Dredging the upper 30 cm of sediment can significantly affect physico-chemical characteristics of sediments. Less degradation of organic matter in the dredged sediments was found during the experiment. Denitrification rates in the sediments were estimated by the acetylene blockage technique, and ranged from 21.6 to 102.7 nmol N2/(g dry weight (dw)·hr) for the undredged sediment and from 6.9 to 26.9 nmol N2/(g dw·hr) for dredged sediments. The denitrification rates in the undredged sediments were markedly higher (p<0.05) than those in the dredged sediments throughout the incubation, with the exception of February 2006. The importance of various environmental factors on denitrification was assessed, which indicated that denitrification was regulated by temperature. Nitrate was probably the key factor limiting denitrification in both undredged and dredged sediments. Organic carbon played some role in determining the denitrification rates in the dredged sediments, but not in the undredged sediments. 3Chemical composition and in vitro fermentation characteristics of high sugar forage sorghum as an alternative to forage maize for silage making in Tarim Basin, China显示文摘The chemical composition, mineral profile and in vitro fermentation characteristics of maize(MZ), high sugar forage sorghum(HS) and forage sorghum(FS), and silages made from each forage type were measured. The MZ and MZ silage(MZS) had higher crude protein, starch and ether extract contents than both sorghum forages and sorghum silages. HS had higher ash and water-soluble carbohydrate concentrations than FS and MZ. MZ, MZS, HS and HS silage(HSS) had lower neutral detergent fibre, acid detergent fibre and acid detergent lignin than FS and FS silage(FSS). FSS had higher dry matter(DM) and p H than MZS and HSS. HSS contained higher concentrations of P and K than FSS and MZS. MZS and HSS had higher in vitro dry matter and organic matter digestibility, CH4 production, total volatile fatty acids, acetate and propionate than FSS. p H was higher for FSS than for HSS, and ammonia was lower for HSS than for MZS and FSS. HSS had higher gas production than MZS and FSS after 2, 4, 6 and 8 h incubation. MZS had higher gas production than HSS and FSS after 26 and 28 h of incubation. The results indicate that HS may substitute for MZ to make good quality silage. 4Meta-analyses on prevalence of selected Parkinson’s nonmotor symptoms before and after diagnosis显示文摘Background:Nonmotor symptoms are common among patients with Parkinson’s disease(PD)and some may precede disease diagnosis.Methods:We conducted a meta-analysis on the prevalence of selected nonmotor symptoms before and after PD diagnosis,using random-effect models.We searched PubMed(1965 through October/November 2012)for the following symptoms:hyposmia,constipation,rapid eye movement sleep behavior disorder,excessive daytime sleepiness,depression,and anxiety.Eligible studies were publications in English with original data on one or more of these symptoms.Results:The search generated 2,373 non-duplicated publications and 332 met the inclusion criteria,mostly(n=320)on symptoms after PD diagnosis.For all symptoms,the prevalence was substantially higher in PD cases than in controls,each affecting over a third of the patients.Hyposmia was the most prevalent(75.5%in cases vs.19.1%in controls),followed by constipation(50%vs.17.7%),anxiety(39.9%vs.19.1%),rapid eye movement sleep behavior disorder(37.0%vs.7.0%),depression(36.6%vs.14.9%),and excessive daytime sleepiness(33.9%vs.10.5%).We observed substantial heterogeneities across studies and meta-regression analyses suggested that several factors might have contributed to this.However,the prevalence estimates were fairly robust in several sensitivity analyses.Only 20 studies had data on any symptoms prior to PD diagnosis,but still the analyses revealed higher prevalence in future PD cases than in controls.Conclusion:These symptoms are common among PD patients both before and after diagnosis.Further studies are needed to understand the natural history of nonmotor symptoms in PD and their etiological and clinical implications.Honglei Chen Edward J Zhao Wen Zhang Yi Lu Rui Liu Xuemei Huang Anna J Ciesielski-Jones Michele A Justice Deborah S Cousins Shyamal Peddada 2015Translational Neurodegeneration2015,4,1:18

6The Homo sapiens Cave hominin site of Mulan Mountain,Jiangzhou District,Chongzuo,Guangxi with emphasis on its age显示文摘One of the most hotly debated and frontal issues in paleoanthropology focuses on the origins of modern humans. Recently, an incomplete hominin mandible with a distinctly weaker mental protuberance than modern human and a great variety of coexisting fossil mammals were unearthed from the Homo sapiens Cave of Mulan Mountain, Chongzuo, Guangxi. The mammalian fauna from the Homo sapiens Cave characterized by the combination of Elephas kiangnanensis, first occurring Elephas maixmus, and Megatapirus augustus, and strikingly different from the Early Pleistocene Gigantopithecus fauna and the Middle Pleistocene Ailuropoda-Stogodon fauna of South China could be regarded as an early representive of the typical Asian elephant fauna. Faunal analysis, biostratigraphic correlation, and, most importantly, U-series dating all consistently support an estimate of ca. 110 ka for the age of the fossil Homo sapiens and coexisting mammalian fauna, that is, the early Late Pleistocene. The fauna is mainly made up of tropical-subtropical elements, but grassland elements have a much greater variety than forest elements, which probably indicates a drier climate at that time. 8Antibiotic Resistance Genes in the Human-Impacted Environment:A One Health Perspective显示文摘Antibiotic resistance and its environmental component are gaining more attention as part of combating the growing healthcare crisis. The One Health framework, promulgated by many global health agencies, recognizes that antimicrobial resistance is a truly inter-domain problem in which human health, animal agriculture, and the environment are the core and interrelated components.This prospectus presents the status and issues relevant to the environmental component of antibiotic resistance, namely, the needs for advancing surveillance methodology: the environmental reservoirs and sources of resistance, namely, urban wastewater treatment plants, aquaculture production systems, soil receiving manure and biosolid, and the atmosphere which includes longer range dispersal.Recently, much work has been done describing antibiotic resistance genes in various environments;now quantitative, mechanistic,and hypothesis-driven studies are needed to identify practices that reduce real risks and maintain the effectiveness of our current antibiotics as long as possible. Advanced deployable detection methods for antibiotic resistance in diverse environmental samples are needed in order to provide the surveillance information to identify risks and define barriers that can reduce risks. Also needed are practices that reduce antibiotic use and thereby reduce selection for resistance, as well as practices that limit the dispersal of or destroy antibiotic-resistant bacteria or their resistance genes that are feasible for these varied environmental domains.James M. 9Mechanosignaling activation of TGFβmaintains intervertebral disc homeostasis显示文摘Intervertebral disc(IVD) degeneration is the leading cause of disability with no disease-modifying treatment.IVD degeneration is associated with instable mechanical loading in the spine, but little is known about how mechanical stress regulates nucleus notochordal(NC) cells to maintain IVD homeostasis. Here we report that mechanical stress can result in excessive integrin α_vβ_6-mediated activation of transforming growth factor beta(TGFβ), decreased NC cell vacuoles, and increased matrix proteoglycan production, and results in degenerative disc disease(DDD). Knockout of TGFβ type II receptor(TβRII) or integrin α_v in the NC cells inhibited functional activity of postnatal NC cells and also resulted in DDD under mechanical loading.Administration of RGD peptide, TGFβ, and α_vβ_6-neutralizing antibodies attenuated IVD degeneration. Thus,integrin-mediated activation of TGFβ plays a critical role in mechanical signaling transduction to regulate IVD cell function and homeostasis. 10Through-needle all-optical ultrasound imaging in vivo: a preclinical swine study显示文摘High-frequency ultrasound imaging can provide exquisite visualizations of tissue to guide minimally invasive procedures.Here,we demonstrate that an all-optical ultrasound transducer,through which light guided by optical fibers is used to generate and receive ultrasound,is suitable for real-time invasive medical imaging in vivo.Broad-bandwidth ultrasound generation was achieved through the photoacoustic excitation of a multiwalled carbon nanotube-polydimethylsiloxane composite coating on the distal end of a 300-μm multi-mode optical fiber by a pulsed laser.The interrogation of a high-finesse Fabry–Pérot cavity on a single-mode optical fiber by a wavelength-tunable continuous-wave laser was applied for ultrasound reception.This transducer was integrated within a custom inner transseptal needle(diameter 1.08 mm;length 78 cm)that included a metallic septum to acoustically isolate the two optical fibers.The use of this needle within the beating heart of a pig provided unprecedented realtime views(50 Hz scan rate)of cardiac tissue(depth:2.5 cm;axial resolution:64μm)and revealed the critical anatomical structures required to safely perform a transseptal crossing:the right and left atrial walls,the right atrial appendage,and the limbus fossae ovalis.This new paradigm will allow ultrasound imaging to be integrated into a broad range of minimally invasive devices in different clinical contexts.Malcolm C Finlay Charles A Mosse Richard J Colchester Sacha Noimark Edward Z Zhang Sebastien Ourselin Paul C Beard Richard J Schilling Ivan P Parkin Ioannis Papakonstantinou Adrien E Desjardins 2017Light(Science & Applications)2017,6,1:6
11Compliance among infants exposed to hepatitis B virus in a post-vaccination serological testing program in four provinces in China显示文摘Background:Mother to child transmission of hepatitis B virus(HBV)remains the most common form of HBV infection in China.Prevention of HBV vertical transmission involves timely administration of the complete hepatitis B vaccine(HepB)series and hepatitis B immunoglobulin.Post-vaccination serological testing(PVST)is utilized to determine an infant's outcome after HBV exposure and completion of HepB series.We aim to determine the frequency of compliance with a PVST testing cascade for HBV infected mothers and analyze factors associated with infant lost to follow up(LTFU).Methods:We conducted a retrospeaive cohort review of previously collected data in Fujian,Jiangxi,Zhejiang and Chongqing provinces in China from 1 June 2016-31 December 2017.The study population included all HBV-exposed infants and their mothers.SAS software was used for statistical analyses.Bivariate and multivariate regression analyses(presented in odds ratio[OR]with 95%confidence intervals[CI])were used to compare the proportional differences of factors associated with PVST not being completed.Results:Among enrolled 8474 target infants,40%of them transferred out of the study provinces without further information and 4988 were eligible for PVST.We found 20%(994)of infants were not compliant with the testing cascade:55%of LTFU occurred because parents refused venous blood sample collection or failure of sample collection in the field,16%transferred out after 6 months of age,and 10%of families chose to have independent,confidential PVST completed without reporting results.High PVST noncompliance rates were more likely to be from Fujian(aOR=17.0,95%CI:9.7-29.9),Zhejiang(aOR=5.7,95%Cl:3.2-10.1)and Jiangxi(aOR=1.9,95%CI:1.0-3.4),and from HBV e antigen positive mother(aOR=1.2,95%CI:1.1-1.4).Conclusions:This study found that the LTFU rate reached 20%in PVST program,which was a significant problem.We recommend implementing a national elearonic information system for tracking HBV at risk mother-infant pairs;encourage further research in developing a less invasive means of completing PVST,and take effective measures nationally to reduce HBV stigma.Without reducing the loss to follow up rate among infants eligible for PVST,elimination of vertical HBV transmission will be impossible.Hui Zheng Guo-Min Zhang Po-Lin Chan Fu-Zhen Wang Lance Everett Rodewald Ning Miao Xiao-Jin Sun Zun-Dong Yin Jeffrey Edwards Hua-Qing Wang 2019Infectious Diseases of Poverty2019,8,4:5

13Change patterns of oncomelanid snail burden in areas within the Yangtze River drainage after the three gorges dam operated显示文摘Background:An'integrated control'strategy has been implemented within seven provinces at highest risk for schistosomiasis along Yangtze River in Peoples'Republic of China(P.R.China)since 2004.Since O ncom elania hupensis is the only intermediate host of the blood fluke(Schistosom a japonicum),controlling the distribution of snails is considered an essential and effeaive way to reduce the risk of schistosomiasis infection.The study aimed to determine the snail area burden and annual trend among provinces with potential risk for schistosomiasis along the Yangtze River,above and below the Three Gorges Dam(TGD).Methods:This retrospective study utilized data previously collected from the National Parasitic Diseases Control Information Management System(NPDCIMS)on annual snail surveys from 2009 to 2017.Descriptive statistics were performed for analyzing the snail burden by provinces,counties,type of environmental location and year,and mapping was conducted to present the snails distribution.Results:From 2009 to 2017,the total snail infested area decreased by 4.22%,from 372253 hm^2 to 356553 hm^2 within the seven high risk provinces.The majority of snails were found in the marshland and lake regions,outside of control embankments.The total snail burden trend remained relatively stable in upstream regions above the TGD from 2010 to 2015,while the trend decreased within downstream regions during this period.In 2016 and 2017,the total snail burden trend increased in both upstream and downstream provinces,however,upstream saw a larger increase.From 2009 to 2017,there were a total of 5990 hm^2 of newly developed snail areas in the seven study provinces and the majority were concentrated in regions below the TGD,accounting for 5610 hm^2(93.70%).Conclusions:There has been a decline in total snail counts from 2009 to 2017.Meanwhile,new snail breeding areas were formed mainly within provinces downstream the TGD due to spread of snails,indicated that the oncomelanid snail would be difficult to completely eliminate.We suggest that the national schistosomiasis integrated control strategy,including mollusciding and environmental modification,will need to be enhanced significantly going forward to achieve a greater reduaion in snail burden and ultimately to achieve elimination.Si-Min Dai Jeffrey Edwards Zhou Guan Shan Lv Shi-Zhu Li Li-Juan Zhang Jun Feng Ning Feng Xiao-Nong Zhou Jing Xu 2019Infectious Diseases of Poverty2019,8,3:4
14Accelerated evolution of an Lhx2 enhancer shapes mammalian social hierarchies显示文摘Social hierarchies emerged during evolution,and social rank influences behavior and health of individuals.However,the evolutionary mechanisms of social hierarchy are still unknown in amniotes.Here we developed a new method and performed a genome-wide screening for identifying regions with accelerated evolution in the ancestral lineage of placental mammals,where mammalian social hierarchies might have initially evolved.Then functional analyses were conducted for the most accelerated region designated as placental-accelerated sequence 1(PAS1,P=3.15 × 10^-18).Multiple pieces of evidence show that PAS1 is an enhancer of the transcription factor gene Lhx2 involved in brain development.PAS1s isolated from various amniotes showed different c/s-regulatory activity in vitro,and affected the expression of Lhx2 differently in the nervous system of mouse embryos.PAS1 knock-out mice lack social stratification.PAS1 knock-in mouse models demonstrate that PAS1s determine the social dominance and subordinate of adult mice,and that social ranks could even be turned over by mutated PAS1.All homozygous mutant mice had normal huddled sleeping behavior,motor coordination and strength.Therefore,PAS1-Lhx2 modulates social hierarchies and is essential for establishing social stratification in amniotes,and positive Darwinian selection on PAS1 plays pivotal roles in the occurrence of mammalian social hierarchies.Yuting Wang Guangyi Dai Zhili Gu Guopeng Liu Ke Tang Yi-Hsuan Pan Yujie Chen Xin Lin Nan Wu Haoshan Chen Su Feng Shou Qiu Hongduo Sun Qian Li Chuan Xu Yanan Mao Yong Edward Zhang Philipp Khaitovich Yan-Ling Wang Qunxiu Liu Jing-Dong Jackie Han Zhen Shao Gang Wei Chun Xu Naihe Jing Haipeng Li 2020Cell Research2020,30,5:4

17Identifying the Risk of SARS-CoV-2 Infection and Environmental Monitoring in Airborne Infectious Isolation Rooms(AIIRs)显示文摘Healthcare workers(HCWs)are at high risk of occupational exposure to the new pandemic human coronavirus,SARSCoV-2,and are a source of nosocomial transmission in airborne infectious isolation rooms(AIIRs).Here,we performed comprehensive environmental contamination surveillance to evaluate the risk of viral transmission in AIIRs with 115 rooms in three buildings at the Shanghai Public Health Clinical Center,Shanghai,during the treatment of 334 patients infected with SARS-CoV-2.The results showed that the risk of airborne transmission of SARS-CoV-2 in AIIRs was low(1.62%,25/1544)due to the directional airflow and strong environmental hygiene procedures.However,we detected viral RNA on the surface of foot-operated openers and bathroom sinks in AIIRs(viral load:55.00–3154.50 copies/mL).This might be a source of contamination to connecting corridors and object surfaces through the footwear and gloves used by HCWs.The risk of infection was eliminated by the use of disposable footwear covers and the application of more effective environmental and personal hygiene measures.With the help of effective infection control procedures,none of 290 HCWs was infected when working in the AIIRs at this hospital.This study has provided information pertinent for infection control in AIIRs during the treatment of COVID-19 patients.Zhi-Gang Song Yan-Mei Chen Fan Wu Lin Xu Bang-Fang Wang Lei Shi Xiao Chen Fa-Hui Dai Jia-Lei She Jian-Min Chen Edward CHolmes Tong-Yu Zhu Yong-Zhen Zhang 2020Virologica Sinica2020,35,6:3
18In vivo neuronal and astrocytic activation in somatosensory cortex by acupuncture stimuli显示文摘Acupuncture is a medical treatment that has been widely pra cticed in China for over 3000 years,yet the neural mechanisms of acupuncture are not fully understood.We hypothesized that neurons and astrocytes act independently and synergistically under acupuncture stimulation.To investigate this,we used two-photon in vivo calcium reco rding to observe the effects of acupuncture stimulation at ST36(Zusanli)in mice.Acupuncture stimulation in peripheral acupoints potentiated calcium signals of pyramidal neurons and astrocytes in the somatosensory cortex and resulted in late-onset calcium transients in astrocytes.Chemogenetic inhibition of neurons augmented the astrocytic activity.These findings suggest that acupuncture activates neuronal and astrocytic activity in the somatosensory co rtex and provide evidence for the involvement of both neurons and astrocytes in acupuncture treatment.Xiao-Yue Chang Kai Chen Tong Cheng Pui To Lai Li Zhang Kwok-Fai So Edward S.Yang 2022Neural Regeneration Research2022,17,11:2
19The influence of physiological environment on the targeting effect of aptamer-guided gold nanoparticles显示文摘Aptamer guided nano medicine shows great promise in targeted cancer therapies.However the loss of targeting capacity duri ng in vivo or clinical trials has largely hindered its popularity and there are no systematic studies to elucidate the causes.Herein,we investigated such loss of targeting capacity by examining how the physiological milieu affected targeting effect.Aptamer functionalized gold nanoparticle(AuNP)was chosen as the model and exposed to human blood serum that is used to mimic physiological milieu.Dynamic light scattering(DLS),flow cytometry and label-free liquid chromatography tan dem mass spectrometry(LC-MS/MS)were employed to determi ne variations of NPs'surface chemistry and biological identities changes after serum exposure.Results showed that the targeting ability loss was caused by protein corona blocking,replacement and enzymatic cleavage of surface aptamer targeting ligands.Noteworthy,the aggregation issue is critical for the smaller NPs.Analysis of the protein corona profile indicated the accumulation of immune-related proteins on the surface of aptamer-conjugated NPs,which could induce immune response,resulting in rapid clearanee of NPs.Ding Ding Yinling Zhang Edward A.Sykes Long Chen Zhuo Chen Weihong Tan 2019Nano Research2019,12,1:2
20The Process and Benefits of Developing Prototype Climate Services——Examples in China显示文摘Changes in climate pose major challenges to society, and so decision-makers need actionable climate information to inform their planning and policies to make society more resilient to climatic changes. Climate services are being developed to provide such actionable climate information. The successful development and use of climate services benefits greatly from close engagement between developers, providers, and users of the services. The Climate Science for Service Partnership China(CSSP China) is a China–UK collaboration fostering closer engagement between climate scientists, providers of climate services, and users of climate services. We describe the process within CSSP China of co-developing climate services through trials with users to revise and improve a prototype. Examples are provided covering various scientific capabilities, user needs, and parts of China. The development process is yielding many benefits, such as increasing the engagement between providers and users, making users more aware of how climate information can be of use in their decision-making, giving the climate service providers a better understanding of the users’ requirements for climate information, and shaping future scientific research and development. In addition to the benefits, we also document some challenges that have emerged, along with ways of alleviating them.
