1Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research. 2Effect of climate change and railway embankment on the degradation of underlain permafrost显示文摘Because of the global warming and the increasing human activity, the air temperature and the precipitation along the Qinghai-Tibet Railway increased gradually in recent years, which endanger the permafrost table, the embankment of the railway. The statistics of the air temperature, the precipitation and the geothermal temperature in recent 50 years in this dissertation come from the four weather station along the railway, that are Wudaoliang Station, Fenghuo Mountains Station, Tuotuo River Station and Ando Station. This dissertation analyzes the change of climate along the railway and then develops a research on the effect of the changing geothermal temperature on permafrost table and its countermeasures. The experiment result shows that the air temperature of the permafrost region rise steadily in about 50 years, especially in this century, the tendency of rising temperature is more obvious. The precipitation fluctuates but it is also rising rapidly, for the largest precipitation reached 492.6 mm. For 30 years now, the Qinghai-Tibet Plateau has been in the megathermal period, which also affects the permafrost region along the railway. 3Low‐intensity pulsed ultrasound ameliorates angiotensin II-induced cardiac fibrosis by alleviating inflammation via a caveolin-1-dependent pathway显示文摘Objective:Cardiac hypertrophy and fibrosis are major pathological manifestations observed in left ventricular remodeling induced by angiotensin II(AngII).Low-intensity pulsed ultrasound(LIPUS)has been reported to ameliorate cardiac dysfunction and myocardial fibrosis in myocardial infarction(MI)through mechano-transduction and its downstream pathways.In this study,we aimed to investigate whether LIPUS could exert a protective effect by ameliorating AngII-induced cardiac hypertrophy and fibrosis and if so,to further elucidate the underlying molecular mechanisms.Methods:We used AngII to mimic animal and cell culture models of cardiac hypertrophy and fibrosis.LIPUS irradiation was applied in vivo for 20 min every 2 d from one week before mini-pump implantation to four weeks after mini-pump implantation,and in vitro for 20 min on each of two occasions 6 h apart.Cardiac hypertrophy and fibrosis levels were then evaluated by echocardiographic,histopathological,and molecular biological methods.Results:Our results showed that LIPUS could ameliorate left ventricular remodeling in vivo and cardiac fibrosis in vitro by reducing AngII-induced release of inflammatory cytokines,but the protective effects on cardiac hypertrophy were limited in vitro.Given that LIPUS increased the expression of caveolin-1 in response to mechanical stimulation,we inhibited caveolin-1 activity with pyrazolopyrimidine 2(pp2)in vivo and in vitro.LIPUS-induced downregulation of inflammation was reversed and the anti-fibrotic effects of LIPUS were absent.Conclusions:These results indicated that LIPUS could ameliorate AngII-induced cardiac fibrosis by alleviating inflammation via a caveolin-1-dependent pathway,providing new insights for the development of novel therapeutic apparatus in clinical practice.Kun ZHAO Jing ZHANG Tianhua XU Chuanxi YANG LiqingWENG TingtingWU XiaoguangWU Jiaming MIAO Xiasheng GUO Juan TU Dong ZHANG Bin ZHOU Wei SUN Xiangqing KONG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,10:2

5Modification of ZnS∶Mn Phosphors by Sb-Doped SnO2 Coating显示文摘The coating of ZnS∶Mn by Sb-doped SnO2 using an co-precipitation process was reported. ZnS∶Mn phosphor particles were prepared by solid reaction with ZnS and MnCl2·4H2O. Surface modification of the ZnS∶Mn powders was carried out by coating transparent conductive films of Sb-doped tin oxides which were formed by co-precipitation and heat treatment process. Tin tetrachloride and antimony trichloride were used as the precursor materials for the co-precipitation. The influences of coating molar ratio, Sb concentration in the coatings, annealing temperature and time on the resistivity of coated ZnS∶Mn phosphors were investigated. The optimum co-precipitation processing parameters and annealing conditions were determined. The phosphors were characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), photoluminescence(PL) observation and conductance measurements. 8A novel dual serotonin transporter and M-channel inhibitor D01 for antidepression and cognitive improvement显示文摘Cognitive dysfunction is a core symptom common in psychiatric disorders including depression that is primarily managed by antidepressants lacking efficacy in improving cognition.In this study,we report a novel dual serotonin transporter and voltage-gated potassium Kv7/KCNQ/M-channel inhibitor D01(a 2-methyl-3-aryloxy-3-heteroarylpropylamines derivative)that exhibits both anti-depression effects and improvements in cognition.D01 inhibits serotonin transporters(K_(i)=30.1±6.9 nmol/L)and M channels(IC_(50)=10.1±2.4μmol/L).D01 also reduces the immobility duration in the mouse FST and TST assays in a dose-dependent manner without a stimulatory effect on locomotion.Intragastric administrations of D01(20 and 40 mg/kg)can significantly shorten the immobility time in a mouse model of chronic restraint stress(CRS)-induced depression-like behavior.Additionally,D01 dose-dependently improves the cognitive deficit induced by CRS in Morris water maze test and increases the exploration time with novel objects in normal or scopolamine-induced cognitive deficits in mice,but not fluoxetine.Furthermore,D01 reverses the long-term potentiation(LTP)inhibition induced by scopolamine.Taken together,our findings demonstrate that D01,a dual-target serotonin reuptake and M channel inhibitor,is highly effective in the treatment-resistant depression and cognitive deficits,thus holding potential for development as therapy of depression with cognitive deficits.Yaqin Wei Xiangqing Xu Qiang Guo Song Zhao Yinli Qiu Dongli Wang Wenwen Yu Yani Liu KeWei Wang 2024Acta Pharmaceutica Sinica B2024,14,3:0
9CC-chemokine receptor 7 and its ligand CCL 19 promote mitral valve interstitial cell migration and repair显示文摘The effect of CC-chemokine receptor 7(CCR7) and CC-chemokine ligand 19(CCL19) on rheumatic mitral stenosis is unknown.This study aimed to explore the roles of CCR7 and CCL19 in rheumatic mitral stenosis by measuring the expression of CCR7 and CCL19 in human mitral valves from rheumatic mitral stenosis patients.Additionally,we examined their effects on human mitral valve interstitial cells(hMVICs) proliferation,apoptosis and wound repair.CCR7 and CCL19 expression was measured in the mitral valves from rheumatic mitral stenosis patients(n=10) and compared to normal mitral valves(n=5).CCR7 was measured in cultured hMVICs from rheumatic mitral stenosis patients and normal donors by RT-PCR and immunofluorescence.The cells were also treated with exogenous CCL19,and the effects on wound healing,proliferation and apoptosis were assayed.In the rheumatic mitral valves,valve interstitial cells expressed CCR7,while mononuclear cells and the endothelium expressed CCL19.Healthy mitral valves did not stain positive for CCR7 or CCL19.CCR7 was also detected in cultured rheumatic hMVICs or in normal hMVICs treated with CCL19.In a wound healing experiment,wound closure rates of both rheumatic and normal hMVICs were significantly accelerated by CCL19.These effects were abrogated by a CCR7 neutralizing antibody.The CCR7/CCL19 axis did not influence the proliferation or apoptosis of hMVICs,indicating that wound healing was due to increased migration rates rather than increased proliferation.In conclusion,CCR7 and CCL19 were expressed in rheumatic mitral valves.The CCR7/CCL19 axis may regulate remodeling of rheumatic valve injury through promoting migratory ability of hMVICs.Xiaozhi Wang Liang Wang Liping Miao Rong Zhao Yanhu Wu Xiangqing Kong 2015The Journal of Biomedical Research2015,29,6:0
10Exploration of a Sequential Gp140-Gp145 Immunization Regimen with Heterologous Envs to Induce a Protective Cross-Reactive HIV Neutralizing Antibody Response In Non-human Primates显示文摘Raising a heterologous tier 2 neutralizing antibody(nAb)response remains a daunting task for HIV vaccine development.In this study,we explored the utility of diverse HIV-1 envelope(Env)immunogens in a sequential immunization scheme as a solution to this task.This exploration stemmed from the rationale that gp145,a membrane-bound truncation form of HIV Env,may facilitate the focusing of induced antibody response on neutralizing epitopes when sequentially combined with the soluble gp140 form as immunogens in a prime-boost mode.We first showed that gp140 DNA prime-gp145 Tiantan vaccinia(TV)boost likely represents a general format for inducing potent nAb response in mice.However,when examined in rhesus macaque,this modality showed little effectiveness.To improve the efficacy,we extended the original modality by adding a strong protein boost,namely native-like SOSIP.664 trimer displayed on ferritin-based nanoparticle(NP),which was generated by a newly developed click approach.The resulting three-immunization regimen succeeded in eliciting tier-2 nAb response with substantial breadth when implemented in rhesus macaque over a short 8-week schedule.Importantly,the elicited nAb response was able to effectively contain viremia upon a heterologous SHIV challenge.Collectively,our studies highlighted that diversification of Env immunogens,in both types and formulations,under the framework of a sequential immunization scheme might open new opportunity toward HIV vaccine development.Xiangqing Ding Kangli Cao Jing Wang Yanmin Wan Qinyun Chen Yanqin Ren Yongtang Zheng Mingzhao Zhu Renrong Tian Wenjun Wang Chen Zhao Xiaoyan Zhang Jianqing Xu 2021Virologica Sinica2021,36,4:0
