1Coupled GIMP and CPDI material point method in modelling blast-induced three-dimensional rock fracture显示文摘Three-dimensional rock fracture induced by blasting is a highly complex problem and has received considerable attention in geotechnical engineering.The material point method is firstly applied to treat this challenging task.Some inherent weaknesses can be overcome by coupling the generalized interpolation material point(GIMP)and the convected particle domain interpolation technique(CPDI).For the media in the borehole,unchanged GIMP-type particles are used to guarantee a homogenous blast pressure.CPDITetrahedron type particles are employed to avoid the fake numerical fracture near the borehole for the rock material.A blasting experiment using three-dimensional single-borehole rock was simulated to examine the applicability of the coupled model under realistic loading and boundary conditions.A good agreement was achieved between the simulation and experimental results.Moreover,the mechanism of three-dimensional rock fracture was analyzed.It was concluded that rock particle size and material parameters play an important role in rock damage.The reflected tensile waves cause severe damage in the lower part of the model.Rayleigh waves occur on the top face of the rock model to induce a hoop failure band.Duanying Wan Meng Wang Zheming Zhu Fei Wang Lei Zhou Ruifeng Liu Weiting Gao Yun Shu Hu Xiao 2022International Journal of Mining Science and Technology2022,32,5:2
2Eco-friendly biodegradable polyurethane based coating for antibacterial and antifouling performance显示文摘Biofouling, which comprises the absorption of proteins and the adhesion of bacteria to the surface of living entities, is a severe concern for the maritime sector since it ultimately leads to hydrodynamic drag,resulting in a higher increase in fuel consumption. As a result, polymer resins are crucial in the marine sector for anti-biofouling coatings. In this work, the poly(caprolactone-ethylene glycol-caprolactone)-p olyurethane(PECL-PU) are prepared through ε-caprolactone(CL), poly(ethylene glycol)(PEG), 4,4'-methylene bis(cyclohexyl isocyanate) and 1,4 butanediol. Our study demonstrate that the PECL-PU copolymer degraded in artificial seawater(5.21%), enzymatic solution(12.63%), and seawater(13.75%)due to the presence of PEG segments in the laboratory-based test under static condition. Because the addition of PEG segments are increased the polymer's amorphous area and decreased the crystallization of the polycaprolactone(PCL) in the copolymer, as demonstrated by differential scanning calorimetry, X-ray diffraction, and water contact angle studies. Therefore, the hydrolysis rates of PECL-PU were higher than the caprolactone-co-polyurethane(CL-PU). The antifouling test showed that PECL-PU3 copolymer had about 90.29% protein resistance, 85.2% Escherichia coli(E. coli) reduction and 94.61% marine diatom Navicula incerta reduction comparison to the control. 3A ribosomal gene panel predicting a novel synthetic lethality in non-BRCAness tumors显示文摘Poly(ADP-ribose)polymerase(PARP)inhibitors are one of the most exciting classes of targeted therapy agents for cancers with homologous recombination(HR)deficiency.However,many patients without apparent HR defects also respond well to PARP inhibitors/cisplatin.The biomarker responsible for this mechanism remains unclear.Here,we identified a set of ribosomal genes that predict response to PARP inhibitors/cisplatin in HR-proficient patients.PARP inhibitor/cisplatin selectively eliminates cells with high expression of the eight genes in the identified panel via DNA damage(ATM)signaling-induced pro-apoptotic ribosomal stress,which along with ATM signaling-induced pro-survival HR repair constitutes a new model to balance the cell fate in response to DNA damage.Therefore,the combined examination of the gene panel along with HR status would allow for more precise predictions of clinical response to PARP inhibitor/cisplatin.The gene panel as an independent biomarker was validated by multiple published clinical datasets,as well as by an ovarian cancer organoids library we established.More importantly,its predictive value was further verified in a cohort of PARP inhibitor-treated ovarian cancer patients with both RNA-seq and WGS data.Furthermore,we identified several marketed drugs capable of upregulating the expression of the genes in the panel without causing HR deficiency in PARP inhibitor/cisplatin-resistant cell lines.These drugs enhance PARP inhibitor/cisplatin sensitivity in both intrinsically resistant organoids and cell lines with acquired resistance.Together,our study identifies a marker gene panel for HR-proficient patients and reveals a broader application of PARP inhibitor/cisplatin in cancer therapy.Chao Zhang Qiang Guo Lifeng Chen Zheming Wu Xiao-Jian Yan Chengyang Zou Qiuxue Zhang Jiahong Tan Tian Fang Qunxian Rao Yang Li Shizhen Shen Min Deng Liewei Wang Huanyao Gao Jia Yu Hu Li Cheng Zhang Somaira Nowsheen Jake Kloeber Fei Zhao Ping Yin Chunbo Teng Zhongqiu Lin Kun Song Shuzhong Yao Liangqing Yao Lingying Wu Yong Zhang Xiaodong Cheng Qinglei Gao Jian Yuan Zhenkun Lou Jin-San Zhang 2023Signal Transduction and Targeted Therapy2023,8,6:0
4Phytoestrogen-derived multifunctional ligands for targeted therapy of breast cancer显示文摘Nano-targeted delivery systems have been widely used for breast tumor drug delivery.Estrogen receptors are considered to be significant drug delivery target receptors due to their overexpression in a variety of tumor cells.However,targeted ligands have a significant impact on the safety and effectiveness of active delivery systems,limiting the clinical transformation of nanoparticles.Phytoestrogens have shown good biosafety characteristics and some affinity with the estrogen receptor.In the present study,molecular docking was used to select tanshinone IIA(Tan IIA)among phytoestrogens as a target ligand to be used in nanodelivery systems with somemodifications.Modified Tan IIA(Tan-NH2)showed a good biosafety profile and demonstrated tumor-targeting,anti-tumor and anti-tumor metastasis effects.Moreover,the ligand was utilized with the anti-tumor drug Dox-loaded mesoporous silica nanoparticles via chemical modification to generate a nanocomposite Tan-Dox-MSN.Tan-Dox-MSN had a uniform particle size,good dispersibility and high drug loading capacity.Validation experiments in vivo and in vitro showed that it also had a better targeting ability,anti-tumor effect and lower toxicity in normal organs.These results supported the idea that phytoestrogens with high affinity for the estrogen receptor could improve the therapeutic efficacy of nano-targeted delivery systems in breast tumors.Ying Zhang Hao Pan Changxiang Yu Rui Liu Bin Xing Bei Jia Jiachen He Xintao Jia Xiaojiao Feng Qingqing Zhang Wenli Dang Zheming Hu Xiuping Deng Pan Guo Zhidong Liu Weisan Pan 2023Asian Journal of Pharmaceutical Sciences2023,18,4:0
