3Effects of soil water and nitrogen availability on photosynthesis and water use efficiency of Robinia pseudoacacia seedlings显示文摘The efficient use of water and nitrogen (N) to promote growth and increase yield of fruit trees and crops is well studied.However,little is known about their effects on woody plants growing in arid and semiarid areas with limited water and N availability.To examine the effects of water and N supply on early growth and water use efficiency (WUE) of trees on dry soils,one-year-old seedlings of Robinia pseudoacacia were exposed to three soil water contents (non-limiting,medium drought,and severe drought) as well as to low and high N levels,for four months.Photosynthetic parameters,leaf instantaneous WUE (WUEi) and whole tree WUE (WUEb) were determined.Results showed that,independent of N levels,increasing soil water content enhanced the tree transpiration rate (Tr),stomatal conductance (Gs),intercellular CO2 concentration (Ci),maximum net assimilation rate (Amax),apparent quantum yield (AQY),the range of photosynthetically active radiation (PAR) due to both reduced light compensation point and enhanced light saturation point,and dark respiration rate (Rd),resulting in a higher net photosynthetic rate (Pn) and a significantly increased whole tree biomass.Consequently,WUEi and WUEb were reduced at low N,whereas WUE i was enhanced at high N levels.Irrespective of soil water availability,N supply enhanced Pn in association with an increase of Gs and Ci and a decrease of the stomatal limitation value (Ls),while Tr remained unchanged.Biomass and WUEi increased under non-limiting water conditions and medium drought,as well as WUEb under all water conditions;but under severe drought,WUEi and biomass were not affected by N application.In conclusion,increasing soil water availability improves photosynthetic capacity and biomass accumulation under low and high N levels,but its effects on WUE vary with soil N levels.N supply increased Pn and WUE,but under severe drought,N supply did not enhance WUEi and biomass.Xiping Liu Yangyang Fan Junxia Long Ruifeng Wei Roger Kjelgren Chunmei Gong Jun Zhao 2013Journal of Environmental Sciences2013,25,3:15
4P wave velocity structure of upper and middle crust beneath the Three Gorges reservoir dam and adjacent regions显示文摘The three-dimensional P wave velocity structures of upper and middle crust beneath the Three Gorges reservoir dam and adjacent regions were reconstructed by means of the seismic tomography, using the absolute P-arrivals from the events observed by the Three Gorges seismic network. Our preliminary study of the deep structure shows: (1) the lithology of the Huangling anticline, integral and highly so-lidified, exhibits a uniformity. Down to a depth of 20 km, the Huangling anticline still shows high- velocity anomalies. Note that the characteristics of the velocity images with the different depths are not the same; (2) the depth of the sedimentary rocks beneath the entire Zigui basin is not constant, which is less than 6 km; (3) the top boundary of the crystalline basement varies from 0 to 7 km, whereas the bottom one between 14 and 16 km; (4) the partially molten substances of the upper mantle are located in the Badong-Zigui and its adjacent regions as well as nearby Peishi area. The development and evo-lution of the current tectonics are associated with the cooling and solidifying process of the molten substances within the upper mantle; (5) the different velocity structures of the deep crust in both east and west side of the Jiuwanxi fault are to be investigated in order to understand the origin of gravita-tional gradient zone in the Three Gorge areas; and (6) the velocities within the upper crust would be affected by reservoir water permeability.LI Qiang ZHAO Xu CAI JinAn LIU RuiFeng LONG GuiHua AN YanRu 2009Science China Earth Sciences2009,52,4:11
5SIRT1 suppresses PMA and ionomycin-induced ICAM-1 expression in endothelial cells显示文摘Intercellular adhesion molecule-1 (ICAM-1) plays an important role in the recruitment of leukocytes to the endothelium, which causes inflammation and initiation of atherosclerosis. We have previously shown that endothelium-specific over-expression of class III deacetylase SIRT1 decreases atherosclerosis. We therefore addressed the hypothesis that SIRT1 suppresses ICAM-1 expression in the endothelial cells. Here, we found that expression of SIRT1 and ICAM-1 was significantly induced by PMA and ionomycin (PMA/Io) in human umbilical vein endothelial cells (HUVECs). Adenovirus-mediated over-expression of SIRT1 significantly inhibited PMA/Io-induced ICAM-1 expression in HUVECs. Knockdown of SIRT1 by RNA interference (RNAi) resulted in increased expression of ICAM-1 in HUVECs. Luciferase report assay showed that over-expression of SIRT1 suppressed ICAM-1 promoter activity both in basic and in PMA/Io-induced conditions. We further found that SIRT1 was involved in transcription complex binding on the ICAM-1 promoter by chromatin immunoprecipitation (ChIP) assays. Furthermore, SIRT1 RNAi increased NF-κB p65 binding ability to the ICAM-1 promoter by ChIP assays. 6KIF18B promotes tumor progression in osteosarcoma by activating β-catenin显示文摘Objective:Osteosarcoma is a common primary highly malignant bone tumor.Kinesin family member 18B(K1F18B)has been identified as a potential oncogene involved in the development and metastasis of several cancer types.While KIF18B overexpression in osteosarcoma tissue is clearly detected,its specific function in the disease process remains to be established.Methods:K IF18B expression was assessed in osteosarcoma tissues and cells.We additionally evaluated the effects of KIF18B on proliferation,migration,and invasion of osteosarcoma cells,both in vitro and in vivo.Results:Our results showed overexpression of KIF18B in osteosarcoma tissues and cells.Knockdown of K IF18B induced G1/S phase arrest and significantly inhibited proliferation,migration,and invasion of osteosarcoma cells,both in vitro and in vivo.K IF18B regulated P-catenin expression at the transcriptional level by controlling nuclear aggregation of ATF2 and at the post-transcriptional level by interacting with the adenomatous polyposis coli(APC)tumor suppressor gene in osteosarcoma cells.Conclusions:KIF18B plays a carcinogenic role in osteosarcoma by regulating expression ofβ-catenin transcriptionally via decreasing nuclear aggregation of ATF2 or post-transcriptionally through interactions with APC.Our collective findings support the potential utility of KIF18B as a novel prognostic biomarker for osteosarcoma.Tian Gao Ling Yu Zhiwei Fang Jiayong Liu Chujie Bai Shu Li Ruifeng Xue Lu Zhang Zhichao Tan Zhengfu Fan 2020Cancer Biology & Medicine2020,17,2:7

8Efficacy of vinorelbine combined with low-dose methotrexate for treatment of inoperable desmoid tumor and prognostic factor analysis显示文摘Objective: To assess the efficacy of conservative chemotherapy for inoperable desmoid tumor(DT) and analyze the prognostic factors.Methods:From November 2008 to April 2016,71 patients of inoperable DT were treated with vinorelbine and low-dose methotrexate in the Department of Bone and Soft Tissue Tumors,Peking University Cancer Hospital&Institute,and enrolled in this retrospective study.The chemotherapy duration is one year.The efficacy of chemotherapy and the prognosis were observed.Results:Of the 71 patients,55% were female.Age of onset varied from 1 to 47 years,and the median age was 14years.Only 11(15.5%)cases suffered primary tumor.The distribution of the site of tumors was:31(43.7%)in the trunk,36(50.7%)in the limbs,and 4(5.6%)in the peritoneal and pelvic cavity.The size of tumor(the maximum diameter)differed from 2 to 37 cm with a mean of 9.3 cm.The median follow-up duration was 28(range,6–87)months.Common side effects included:nausea and vomiting,liver injury,bone marrow suppression and oral ulcers.When the chemotherapy finished,1(1.4%)case achieved complete response,24(33.8%)achieved partial response,37(52.1%)achieved stable disease and 9(12.7%)had progressive disease.The overall response rate was 87.3%.The progression-free survival(PFS)of the participants were from 6 to 87 months,and the 2-,3-and 5-year PFS was 79.9%,68.4% and 36.3%,respectively.No significant difference was identified in PFS in subgroups of gender,age of onset,age of chemotherapy,tumor site and tumor size.Conclusions:For recurrent,inoperable and progressive DT,enough course of chemotherapy with vinorelbine combined with low-dose methotrexate was an optional choice for local control.Shu Li Zhengfu Fan Zhiwei Fang Jiayong Liu Chujie Bai Ruifeng Xue Lu Zhang Tian Gao 2017Chinese Journal of Cancer Research2017,29,5:6
9Activation of STAT3 stimulates AHSP expression in K562 cells显示文摘Studies on the chaperone proteinα-hemoglobin stabilizing protein(AHSP)reveal that abundant AHSP in erythroid cells enhance the cells’tolerance to oxidative stress imposed by excessα-hemoglobin in pathological conditions.However,the potential intracellular modulation of AHSP expression itself in response to oxidative stress is still unknown.The present study examined the effect and molecular mechanism of STAT3,an oxidative regulator,on the expression of AHSP.AHSP expression increased in K562 cells upon cytokine IL-6-induced STAT3 activation and decreased in STAT3 knock-down K562 cells.Regulation of AHSP in oxidative circumstance was then examined inα-globin-overloaded K562 cells,and real-time PCR showed strengthened expression of both AHSP and STAT3.ChIP analysis showed binding of STAT3 to AHSP promoter and binding was significantly augmented with IL6 stimulation and uponα-globin overexpression.Dual luciferase reporter assays of the wildtype and mutated SB3 element,an IL-6RE site,in the AHSP promoter in K562 cells highlighted the direct regulatory effect of STAT3 on AHSP gene.Finally,direct binding of STAT3 to SB3 site of AHSP promoter was confirmed with EMSA assays.Our work reveals an adaptive AHSP regulation mediated by the redox-sensitive STAT3 signaling pathway,and provides clues to the therapeutic strategy for AHSP enhancement.CAO Cong ZHAO GuoWei YU Wei XIE XueMin WANG WenTian YANG RuiFeng LV Xiang LIU DePei 2014Science China(Life Sciences)2014,57,5:5
10A viral protein orchestrates rice ethylene signaling to coordinate viral infection and insect vector-mediated transmission显示文摘Arthropod-borne viruses cause serious threats to human health and global agriculture by rapidly spreading via insect vectors. Southern rice black-streaked dwarf virus (SRBSDV) is the most damaging rice-infecting virus that is frequently transmitted by planthoppers. However, the molecular mechanisms underlying its propagation in the host plants and epidemics in the field are largely unknown. Here, we showed that the SRBSDV-encoded P6 protein is a key effector that regulates rice ethylene signaling to coordinate viral infection and transmission. In early SRBSDV infection, P6 interacts with OsRTH2 in the cytoplasm to activate ethylene signaling and enhance SRBSDV proliferation;this also repels the insect vector to reduce infestation. In late infection, P6 enters the nucleus, where it interacts with OsEIL2, a key transcription factor of ethylene signaling. The P6-OsEIL2 interaction suppresses ethylene signaling by preventing the dimerization of OsEIL2, thereby facilitating viral transmission by attracting the insect vector. 11Probing the fractional topological charge of a vortex light beam by using dynamic angular double slits显示文摘Vortex beams with fractional topological charge(FTC) have many special characteristics and novel applications.However, one of the obstacles for their application is the difficulty of precisely determining the FTC of fractional vortex beams. We find that when a vortex beam with an FTC illuminates a dynamic angular double slit(ADS), the far-field interference patterns that include the information of the FTC of the beam at the angular bisector direction of the ADS vary periodically. 12Efficacy of inverso isomer of CendR peptide on tumor tissue penetration显示文摘The dense extracellular matrix and high interstitial fluid pressure of tumor tissues prevent the ability of anti-tumor agents to penetrate deep into the tumor parenchyma for treatment effects. C-end rule(CendR) peptides can enhance the permeability of tumor blood vessels and tumor tissues via binding to neuropilin-1(NRP-1), thus aiding in drug delivery. In this study, we selected one of the CendR peptides(sequence RGERPPR) as the parent L-peptide and substituted D-amino acids for the L-amino acids to synthesize its inverso peptide D(RGERPPR). We investigated the NRP-1 binding activity and tumorpenetrating ability of D(RGERPPR). We found that the binding affinity of D(RGERPPR) with NRP-1 and the cellular uptake was significantly higher than that of RGERPPR. Evans Blue tests revealed that D(RGERPPR) exhibited improved tumor-penetrating ability in C6, U87 and A549 tumor-bearing nude mice. Using nude mice bearing A549 xenograft tumors as a model, we found that the rate of tumor growth in the group co-administered with D(RGERPPR) and gemcitabine(Gem) was significantly lower than the gemcitabine-treated group with a tumor suppression rate(TSR%) of 55.4%. 13Effects of Solvent Molecules on the Interlayer Spacing of Graphene Oxide显示文摘Graphene oxide(GO) contains numerous functional groups that facilitate the intercalation of polar solvents. The properties and applications of GO are closely related to its interlayer spacing. We report on the changes in the interlayer spacing of GO after the adsorption of water molecules and the polar organic solvents C_2H_6O_2(EG), C_3H_7NO(DMF), C_5H_9NO(NMP). Experiments were conducted to investigate the variations in the functional groups and structure of GO after solvent adsorption, and they play a vital role in modeling and verifying the results of molecular dynamics simulation. The most stable GO structures are obtained through molecular dynamics simulation. The expansion of the interlayer spacing of GO after the adsorption of monolayer solvent molecules corresponds to the minimum three-dimensional size of the solvent molecules. The spatial arrangement of solvent molecules also contributes to the changes in interlayer spacing. Most adsorbed molecules are oriented parallel to the carbon plane of GO. However, as additional molecules are adsorbed into the interlaminations of GO, the adsorbed molecules are oriented perpendicular to the carbon plane of GO, and a large space forms between two GO interlayers. 14Engineered neutrophil apoptotic bodies ameliorate myocardial infarction by promoting macrophage efferocytosis and inflammation resolution显示文摘Inflammatory response plays a critical role in myocardial infarction(MI)repair.The neutrophil apoptosis and subsequent macrophage ingestion can result in inflammation resolution and initiate regeneration,while the therapeutic strategy that simulates and enhances this natural process has not been established.Here,we constructed engineered neutrophil apoptotic bodies(eNABs)to simulate natural neutrophil apoptosis,which regulated inflammation response and enhanced MI repair.The eNABs were fabricated by combining natural neutrophil apoptotic body membrane which has excellent inflammation-tropism and immunoregulatory properties,and mesoporous silica nanoparticles loaded with hexyl 5-aminolevulinate hydrochloride(HAL).The eNABs actively targeted to macrophages and the encapsulated HAL simultaneously initiated the biosynthesis pathway of heme to produce anti-inflammatory bilirubin after intracellular release,thereby further enhancing the anti-inflammation effects.In in vivo studies,the eNABs efficiently modulated inflammation responses in the infarcted region to ameliorate cardiac function.This study demonstrates an effective biomimetic construction strategy to regulate macrophage functions for MI repair.Lili Bao Geng Dou Ran Tian Yajie Lv Feng Ding Siying Liu Ruifeng Zhao Lu Zhao Jun Zhou Lin Weng Yan Dong Bei Li Shiyu Liu Xin Chen Yan Jin 2022Bioactive Materials2022,7,3:3
15Faceted Kurdjumov-Sachs interface-induced slip continuity in the eutectic high-entropy alloy,AlCoCrFeNi_(2.1)显示文摘Recently,the eutectic high-entropy alloy(EHEA),AlCoCrFeNi_(2.1),can reach a good balance of strength and ductility.The dual-phase alloy exhibits a eutectic lamellar microstructure with large numbers of interfaces.However,the role of the interfaces in plastic deformation have not been revealed deeply.In the present work,the orientation relationship(OR)of the interfaces has been clarified as the Kurdjumov-Sachs(KS)interfaces presenting〈111〉_(B2) 〈110〉_(FCC)and {110} _(B2){111}_(FCC) independent of their morphologies.There exist three kinds of interfaces in the EHEA,namely,The dominating interface and the secondary interface are both non-slip planes and atomistic-scale faceted,facilitating the nucleation and slip transmission of the dislocations.The formation mechanism of the preferred interfaces is revealed using the atomistic geometrical analysis according to the criteria of the low interfacial energy based on the coincidence-site lattice(CSL)theory.In particular,the ductility of the dual-phase alloy originates from the KS interface-induced slip continuity across interfaces,which provides a high slip-transfer geometric factor.Moreover,the strengthening effect can be attributed to the interface resistance for the dislocation transmission due to the mismatches of the moduli and lattice parameters at the interfaces.Ting Xiong Wenfan Yang Shijian Zheng Zhaorui Liu Yiping Lu Ruifeng Zhang Yangtao Zhou Xiaohong Shao Bo Zhang Jun Wang Fuxing Yin Peter K.Liaw Xiuliang Ma 2021Journal of Materials Science & Technology2021,,6:2
16Discovery of a series of dimethoxybenzene FGFR inhibitors with 5H-pyrrolo[2,3-b]pyrazine scaffold: structure–activity relationship, crystal structural characterization and in vivo study显示文摘Genomic alterations are commonly found in the signaling pathways of fibroblast growth factor receptors(FGFRs). Although there is no selective FGFR inhibitors in market, several promising inhibitors have been investigated in clinical trials, and showed encouraging efficacies in patients. By designing a hybrid between the FGFR-selectivity-enhancing motif dimethoxybenzene group and our previously identified novel scaffold, we discovered a new series of potent FGFR inhibitors, with the best one showing sub-nanomolar enzymatic activity. After several round of optimization and with the solved crystal structure, detailed structure–activity relationship was elaborated. 17Salivary microbiota may predict the presence of esophageal squamous cell carcinoma显示文摘The aim is to explore the predictive value of salivary bacteria for the presence of esophageal squamous cell carcinoma (ESCC). Saliva samples were obtained from 178 patients with ESCC and 101 healthy controls, and allocated to screening and verification cohorts, respectively. In the screening phase, after saliva DNA was extracted, 16S rRNA V4 regions of salivary bacteria were amplified by polymerase chain reaction (PCR) with high-throughput sequencing. Highly expressed target bacteria were screened by Operational Taxonomic Units clustering, species annotation and microbial diversity assessment. In the verification phase, the expression levels of target bacteria identified in the screening phase were verified by absolute quantitative PCR (Q-PCR). Receiver operating characteristic (ROC) curves were plotted to investigate the predictive value of target salivary bacteria. LEfSe analysis revealed higher proportions of Fusobacterium, Streptococcus and Porphyromonas, and Q-PCR assay showed significantly higher numbers of Streptococcus salivarius, Fusobacterium nucleatum and Porphyromonas gingivalis in patients with ESCC, when compared with healthy controls (all P < 0.05). The areas under the ROC curves for Streptococcus salivarius, Fusobacterium nucleatum, Porphyromonas gingivalis and the combination of the three bacteria for predicting patients with ESCC were 69%, 56.5%, 61.8% and 76.4%, respectively. The sensitivities corresponding to cutoff value were 69.3%, 22.7%, 35.2% and 86.4%, respectively, and the matched specificity were 78.4%, 96.1%, 90.2% and 58.8%, respectively. 18Ultrasonic concentration measurement of citrus pectin aqueous solutions using PC and PLS regression显示文摘This work demonstrated the use of multivariate statistical techniques called principal component(PC)and partial least squares(PLS)to extract the acoustic features of citrus pectin water solution.The concentration of citrus pectin water solution was predicted by PC and PLS regression method using the spectra of ultrasound pulse echoes travelling through mixtures.The values of root mean square error of validation(RMSEV)were 0.0675 g/100 g and 0.0662 g/100 g for PC and PLS regression model,respectively.Since the response variable was taken into account,PLS regression model was more accurate than PC regression model.Also,a method for temperature compensation was proposed to correct the impact of temperature variation on analyzed data.The proposed methods for pectin concentration measurement are easily adaptable to similar applications using existing hardware.Meng Ruifeng Zhong Jianjun Zhang Lifen Ye Xingqian Liu Donghong 2012International Journal of Agricultural and Biological Engineering2012,5,2:2
19Coupled GIMP and CPDI material point method in modelling blast-induced three-dimensional rock fracture显示文摘Three-dimensional rock fracture induced by blasting is a highly complex problem and has received considerable attention in geotechnical engineering.The material point method is firstly applied to treat this challenging task.Some inherent weaknesses can be overcome by coupling the generalized interpolation material point(GIMP)and the convected particle domain interpolation technique(CPDI).For the media in the borehole,unchanged GIMP-type particles are used to guarantee a homogenous blast pressure.CPDITetrahedron type particles are employed to avoid the fake numerical fracture near the borehole for the rock material.A blasting experiment using three-dimensional single-borehole rock was simulated to examine the applicability of the coupled model under realistic loading and boundary conditions.A good agreement was achieved between the simulation and experimental results.Moreover,the mechanism of three-dimensional rock fracture was analyzed.It was concluded that rock particle size and material parameters play an important role in rock damage.The reflected tensile waves cause severe damage in the lower part of the model.Rayleigh waves occur on the top face of the rock model to induce a hoop failure band.Duanying Wan Meng Wang Zheming Zhu Fei Wang Lei Zhou Ruifeng Liu Weiting Gao Yun Shu Hu Xiao 2022International Journal of Mining Science and Technology2022,32,5:2
20调强适形放疗在乳腺癌保乳术后应用的系统评价(英文)显示文摘Objective:The aim of this study was to assess the long-term effect of breast conservation with breast intensitymodulated radiotherapy (IMRT) for early-stage breast cancers.Methods:Pub Med,EMBASE,Cochrane Library,Chinese Biomedical Literature Database were searched to identify relevant original published trails,and the references of eligible studies were manually screened.Randomized controlled trails reported in any language,comparing breast IMRT with conventional radiotherapy (RT) in patients with early-stage breast cancer were eligible for inclusion.Two investigators independently assessed the quality of included trials and extracted data.The RevMan 5.0 software was used for statistical analysis.Results:Four trials involving 1440 patients were included.The results showed that in the IMRT group,the occurrence of acute moist dermatitis,the edema of the mammary gland and the occurrence of hyperpigmentation were lower than those of RT group,the pooled risk ratio value and 95% confidence interval were 0.28 (0.17-0.48),0.03 (0.00-0.21) and 0.06 (0.02-0.15) respectively.The use of IMRT did not correlate with pain and quality of life,but the presence of moist desquamation did significantly correlate with pain and reduced quality of life.Conclusion:The currently available evidence showed that IMRT significantly reduces the occurrence of moist acute dermatitis anywhere in the breast and alleviates the suffering of patients and improves the quality of life of patients.Future large-scale,high-quality,double-blind trials are needed.Xiupeng Ye Ling Su Xiaohu Wang Kehu Yang inhui Tian Qiuning Zhang Qiang Wang Xiaojun Li Yuqing Kang Liyun Guo Ruifeng Liu Shifang Feng 2010The Chinese-German Journal of Clinical Oncology2010,9,12:2
