| 1 | CTLA4 Silencing with siRNA Promotes Deviation of Th1/Th2 in Chronic Hepatitis B Patients显示文摘To determine whether RNA interference (RNAi) could block cytotoxic T-lymphocyte antigen 4 (CTLA4) in human lymphocytes in vitro and promote IFN-γ and IL-2 secretions, three small interfering RNAs (siRNAs) were selected based on target specificity sequences of human CTLA4 and transfected into human lymphocytes of chronic HBV patients. As a result, the expression of human CTLA4 mRNA was efficiently suppressed by all the three siRNAs. Compared with negative control (siRNA-co), siRNA-1 inhibited the expression of CTLA4 most efficiently and was used in the further study. The expressions of IFN-γ and IL-2 were upregulated and the level of IL-4 was almost unchanged in lymphocytes transfected with siRNA-1 compared with the blank control. These results indicated that siRNA-1 led to IFN-γ and IL-2 secretions, which is a main response of Th1/Th2. In a conclusion, RNAi significantly suppressed the expression of human CTLA4 mRNA in human lymphocytes in vitro, and could induce Th1/Th2 response. | 5 | Parameters identification of chaotic systems based on artificial bee colony algorithm combined with cuckoo search strategy显示文摘Artificial bee colony(ABC) algorithm is motivated by the intelligent behavior of honey bees when seeking a high quality food source. It has a relatively simple structure but good global optimization ability. In order to balance its global search and local search abilities further, some improvements for the standard ABC algorithm are made in this study. Firstly, the local search mechanism of cuckoo search optimization(CS) is introduced into the onlooker bee phase to enhance its dedicated search; secondly, the scout bee phase is also modified by the chaotic search mechanism. The improved ABC algorithm is used to identify the parameters of chaotic systems, the identified results from the present algorithm are compared with those from other algorithms. Numerical simulations, including Lorenz system and a hyper chaotic system, illustrate the present algorithm is a powerful tool for parameter estimation with high accuracy and low deviations. | 8 | Cylindrical vector beam multiplexer/demultiplexer using off-axis polarization control显示文摘The emergence of cylindrical vector beam(CVB)multiplexing has opened new avenues for high-capacity optical communication.Although several configurations have been developed to couple/separate CVBs,the CVB multiplexer/demultiplexer remains elusive due to lack of effective off-axis polarization control technologies.Here we report a straightforward approach to realize off-axis polarization control for CVB multiplexing/demultiplexing based on a metal–dielectric–metal metasurface.We show that the left-and right-handed circularly polarized(LHCP/RHCP)components of CVBs are independently modulated via spin-to-orbit interactions by the properly designed metasurface,and then simultaneously multiplexed and demultiplexed due to the reversibility of light path and the conservation of vector mode.We also show that the proposed multiplexers/demultiplexers are broadband(from 1310 to 1625 nm)and compatible with wavelength-division-multiplexing.As a proof of concept,we successfully demonstrate a four-channel CVB multiplexing communication,combining wavelength-division-multiplexing and polarization-division-multiplexing with a transmission rate of 1.56 Tbit/s and a bit-error-rate of 10^(−6) at the receive power of−21.6 dBm.This study paves the way for CVB multiplexing/demultiplexing and may benefit high-capacity CVB communication. | Shuqing Chen Zhiqiang Xie Huapeng Ye Xinrou Wang Zhenghao Guo Yanliang He Ying Li Xiaocong Yuan Dianyuan Fan | 2021 | Light(Science & Applications)2021,10,12: | 6 |
| 9 | Myt1L Promotes Differentiation of Oligodendrocyte Precursor Cells and is Necessary for Remyelination After Lysolecithin-Induced Demyelination显示文摘The differentiation and maturation of oligodendrocyte precursor cells(OPCs) is essential for myelination and remyelination in the CNS. The failure of OPCs to achieve terminal differentiation in demyelinating lesions often results in unsuccessful remyelination in a variety of human demyelinating diseases. However, the molecular mechanisms controlling OPC differentiation under pathological conditions remain largely unknown. Myt1 L(myelin transcription factor 1-like), mainly expressed in neurons,has been associated with intellectual disability, schizophrenia, and depression. In the present study, we found that Myt1 L was expressed in oligodendrocyte lineage cells during myelination and remyelination. The expression level of Myt1 L in neuron/glia antigen 2-positive(NG^(2+))OPCs was significantly higher than that in mature CC1^+ oligodendrocytes. In primary cultured OPCs,overexpression of Myt1 L promoted, while knockdown inhibited OPC differentiation. Moreover, Myt1 L was potently involved in promoting remyelination after lysolecithin-induced demyelination in vivo. Ch IP assays showed that Myt1 L bound to the promoter of Olig1 and transcriptionally regulated Olig1 expression. | 11 | Shikimic Acid Promotes Oligodendrocyte Precursor Cell Differentiation and Accelerates Remyelination in Mice显示文摘The obstacle to successful remyelination in demyelinating diseases, such as multiple sclerosis, mainly lies in the inability of oligodendrocyte precursor cells(OPCs) to differentiate, since OPCs and oligodendrocytelineage cells that are unable to fully differentiate are found in the areas of demyelination. Thus, promoting the differentiation of OPCs is vital for the treatment of demyelinating diseases. Shikimic acid(SA) is mainly derived from star anise, and is reported to have antiinfluenza, anti-oxidation, and anti-tumor effects. In the present study, we found that SA significantly promoted the differentiation of cultured rat OPCs without affecting their proliferation and apoptosis. In mice, SA exerted therapeutic effects on experimental autoimmune encephalomyelitis(EAE), such as alleviating clinical EAE scores, inhibiting inflammation, and reducing demyelination in the CNS. SA also promoted the differentiation of OPCs as well as their remyelination after lysolecithin-induced demyelination.Furthermore, we showed that the promotion effect of SA on OPC differentiation was associated with the up-regulation of phosphorylated m TOR. | 13 | Cyclin-dependent Kinase 18 Promotes Oligodendrocyte Precursor Cell Differentiation through Activating the Extracellular Signal-Regulated Kinase Signaling Pathway显示文摘The correct differentiation of oligodendrocyte precursor cells(OPCs) is essential for the myelination and remyelination processes in the central nervous system.Determining the regulatory mechanism is fundamental to the treatment of demyelinating diseases. By analyzing the RNA sequencing data of different neural cells, we found that cyclin-dependent kinase 18(CDK18) is exclusively expressed in oligodendrocytes. In vivo studies showed that the expression level of CDK18 gradually increased along with myelin formation during development and in the remyelination phase in a lysophosphatidylcholine-induced demyelination model, and was distinctively highly expressed in oligodendrocytes. In vitro overexpression and interference experiments revealed that CDK18 directly promotes the differentiation of OPCs, without affecting their proliferation or apoptosis. Mechanistically, CDK18 activated the RAS/mitogen-activated protein kinase kinase1/extracellular signal-regulated kinase pathway, thus promoting OPC differentiation. Based on Pro/ENGINEER software used widely in CAD/CAM systems,according to the curve equation of spatial cam follower’s motion specification,the follower displacement curve is drawn quickly and accurately,then the 3D modeling of spatial cam is created using Pro/Feature module in accordance with this curve,afterwards,the quality of contour surface of spatial cam is inspected by the use of surface analysis,the result of which is fed back to the modeling design of spatial cam; after analyzing the machined theory of spatial cam,the NC machining code for it is programmed automatically according to this theory and the modeling completed in the Pro/Manufacture module. | 18 | Approaching 18%efficiency of ternary organic photovoltaics with wide bandgap polymer donor and well compatible Y6:Y6-1O as acceptor显示文摘A series of ternary organic photovoltaics(OPVs)are fabricated with one wide bandgap polymer D18-Cl as donor,and well compatible Y6 and Y6-1O as acceptor.The open-circuit-voltage(V_(OC))of ternary OPVs is monotonously increased along with the incorporation of Y6-1O,indicating that the alloy state should be formed between Y6 and Y6-1O due to their excellent compatibility.The energy loss can be minimized by incorporating Y6-1 O,leading to the V_(OC)improvement of ternary OPVs.By finely adjusting the Y6-1O content,a power conversion efficiency of 17.91%is achieved in the optimal ternary OPVs with 30 wt%Y6-1O in acceptors,resulting from synchronously improved short-circuit-current density(JSC)of 25.87 mA cm^(-2),fill factor(FF)of 76.92%and V_(OC)of 0.900 V in comparison with those of D18-Cl:Y6 binary OPVs.The JSC and FF improvement of ternary OPVs should be ascribed to comprehensively optimal photon harvesting,exciton dissociation and charge transport in ternary active layers.The more efficient charge separation and transport process in ternary active layers can be confirmed by the magnetophotocurrent and impedance spectroscopy experimental results,respectively.This work provides new insight into constructing highly efficient ternary OPVs with well compatible Y6 and its derivative as acceptor. | Xiaoling Ma Anping Zeng Jinhua Gao Zhenghao Hu Chunyu Xu Jae Hoon Son Sang Young Jeong Caixia Zhang Mengyang Li Kai Wang He Yan Zaifei Ma Yongsheng Wang Han Young Woo Fujun Zhang | 2021 | National Science Review2021,8,8: | 3 |
| 19 | DEM simulation of particle flow and separation in a vibrating flip-flow screen显示文摘Vibrating flip-flow screens(VFFS)with stretchable polyurethane sieve mats have been widely used in screening fine-grained materials in recent years.In this work,the discrete element method(DEM)is used to study the screening process in VFFS to explain particle flow and separation behavior at the particle scale.Unlike traditional vibrating screens,for VFFS,the amplitude response of each point on the elastic sieve mat is different everywhere.This study measures the kinematics of the elastic sieve mat under different conditions such as different stretched lengths and material loads.To establish the elastic sieve mat model in a DEM simulation,the continuous elastic sieve mat is discretized into multiple units,and the displacement signal of each unit tested is analyzed by Fourier series.The Fourier series analysis results of each unit are used as the setting parameters for motion.In this way,the movement of the elastic sieve mat is approximately simulated,and a DEM model of VFFS is produced.Through the simulation,the flow and separation of different-sized particles in VFFS are studied,and the reasonability of the simulation is verified by a pilot-scale screening experiment.The present study demonstrates the potential of the DEM method for the analysis of screening processes in VFFS. | Chi Yu Dongdong Lin Ningning Xu Xinwen Wang Kunwei Pu Zhenghao Wang Guofeng Zhao Runhui Geng Sanpeng Gong | 2023 | Particuology2023,,2: | 2 |
| 20 | Double-side fabrication process and millimeter waveresponse of intrinsic Josephson junctions显示文摘We adopted double-side fabrication process to prepare intrinsic Josephson junctions (IJJs) based on Bi2Sr2CaCu2O8+x (BSCCO) single crystals. Using crystal cleavage and double-side argon ion milling, we have successfully fabricated very uniform IJJs with the thickness of single crystal slice less than 200 nm. Using quasi-optical system, the response of the IJJs to millimeter wave radiation was studied.
