| 2 | QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. | 4 | ALDH2 contributes to melatonin-induced protection against APP/PS1 mutation-prompted cardiac anomalies through cGAS-STING-TBK1-mediated regulation of mitophagy显示文摘Ample clinical evidence suggests a high incidence of cardiovascular events in Alzheimer’s disease(AD),although neither precise etiology nor effective treatment is available.This study was designed to evaluate cardiac function in AD patients and APP/PS1 mutant mice,along with circulating levels of melatonin,mitochondrial aldehyde dehydrogenase(ALDH2)and autophagy.AD patients and APP/PS1 mice displayed cognitive and myocardial deficits,low levels of circulating melatonin,ALDH2 activity,and autophagy,ultrastructural,geometric(cardiac atrophy and interstitial fibrosis)and functional(reduced fractional shortening and cardiomyocyte contraction)anomalies,mitochondrial injury,cytosolic mtDNA buildup,apoptosis,and suppressed autophagy and mitophagy.APP/PS1 mutation downregulated cyclic GMP-AMP synthase(cGAS)and stimulator of interferon genes(STING)levels and TBK1 phosphorylation,while promoting Aβaccumulation.Treatment with melatonin overtly ameliorated unfavorable APP/PS1-induced changes in cardiac geometry and function,apoptosis,mitochondrial integrity,cytosolic mtDNA accumulation(using both immunocytochemistry and qPCR),mitophagy,and cGAS-STING-TBK1 signaling,although these benefits were absent in APP/PS1/ALDH2−/−mice.In vitro evidence indicated that melatonin attenuated APP/PS1-induced suppression of mitophagy and cardiomyocyte function,and the effect was negated by the nonselective melatonin receptor blocker luzindole,inhibitors or RNA interference of cGAS,STING,TBK1,and autophagy.Our data collectively established a correlation among cardiac dysfunction,low levels of melatonin,ALDH2 activity,and autophagy in AD patients,with compelling support in APP/PS1 mice,in which melatonin rescued myopathic changes by promoting cGAS-STING-TBK1 signaling and mitophagy via an ALDH2-dependent mechanism. | Shuyi Wang Lin Wang Xing Qin Subat Turdi Dongdong Sun Bruce Culver Russel JReiter Xiaoming Wang Hao Zhou Jun Ren | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 9 |
| 5 | Bacillus cereus AR156 primes induced systemic resistance by suppressing miR825/825 and activating defense-related genes in Arabidopsis显示文摘Small RNAs play an important role in plant immune responses. However, their regulatory function in induced systemic resistance(ISR) is nascent. Bacillus cereus AR156 is a plant growth-promoting rhizobacterium that induces ISR in Arabidopsis against bacterial infection. Here,by comparing small RNA profiles of Pseudomonas syringae pv. tomato(Pst) DC3000-infected Arabidopsis with and without AR156 pretreatment, we identified a group of Arabidopsis micro RNAs(mi RNAs) that are differentially regulated by AR156 pretreatment. mi R825 and mi R825 are two mi RNA generated from a single mi RNA gene.Northern blot analysis indicated that they were significantly downregulated in Pst DC3000-infected plants pretreated with AR156, in contrast to the plants without AR156 pretreatment. mi R825 targets two ubiquitin-protein ligases,while mi R825 targets toll-interleukin-like receptor(TIR)-nucleotide binding site(NBS) and leucine-rich repeat(LRR)type resistance(R) genes. The expression of these target genes negatively correlated with the expression of mi R825 and mi R825. Moreover, transgenic plants showing reduced expression of mi R825 and mi R825 displayed enhanced resistance to Pst DC3000 infection, whereas transgenic plants overexpressing mi R825 and mi R825 were more susceptible. | 6 | In uence of Particle Size on Laser Absorption and Scanning Track Formation Mechanisms of Pure Tungsten Powder During Selective Laser Melting显示文摘A three-dimensional laser absorption model based on ray tracing was established to describe the coupled interaction of a laser beam with particles in the powder layers of pure tungsten(W)material processed by selective laser melting(SLM).The influence of particle size on the powder-to-laser absorptivity and underlying absorption behavior was investigated.An intrinsic relationship between the absorption,distribution of absorbed irradiance within the powder layers,and surface morphology and geometric characteristics(e.g.,contact angle,width and height of tracks,and remelted depth)of the laser scanning tracks is presented here.Simulation conclusions indicate that the absorptivity of the powder layers considerably exceeds the single powder particle value or the dense solid material value.With an increase in particle size,the powder layer absorbs less laser energy.The maximum absorptivity of theWpowder layers reached 0.6030 at the particle size of 5 lm.The distribution of laser irradiance on the particle surface was sensitive to particle size,azimuthal angle,and the position of the powder particles on the substrate.The maximum irradiance in the powder layers decreased from 1.117×10^–3 to 0.85×10^–3W·μm^-2 and the contour of the irradiance distribution in the center of the irradiated area gradually contracted when the particle size increased from 5 to 45 lm.An experimental study on the surface morphologies and cross-sectional geometric characteristics of SLM-fabricated W material was performed,and the experimental results validated the mechanisms of the powder-to-laser-absorption behavior that were obtained in simulations.This work provides a scientific basis for the application of the ray-tracing model to predict the wetting and spreading ability of melted tracks during SLM additive manufacturing in order to yield a sound laser processability. | Jiayao Zhang Dongdong Gu Ying Yang Hongmei Zhang Hongyu Chen Donghuai Dai Kaijie Lin | 2019 | Engineering2019,5,4: | 6 |

| 11 | DEM simulation of particle flow and separation in a vibrating flip-flow screen显示文摘Vibrating flip-flow screens(VFFS)with stretchable polyurethane sieve mats have been widely used in screening fine-grained materials in recent years.In this work,the discrete element method(DEM)is used to study the screening process in VFFS to explain particle flow and separation behavior at the particle scale.Unlike traditional vibrating screens,for VFFS,the amplitude response of each point on the elastic sieve mat is different everywhere.This study measures the kinematics of the elastic sieve mat under different conditions such as different stretched lengths and material loads.To establish the elastic sieve mat model in a DEM simulation,the continuous elastic sieve mat is discretized into multiple units,and the displacement signal of each unit tested is analyzed by Fourier series.The Fourier series analysis results of each unit are used as the setting parameters for motion.In this way,the movement of the elastic sieve mat is approximately simulated,and a DEM model of VFFS is produced.Through the simulation,the flow and separation of different-sized particles in VFFS are studied,and the reasonability of the simulation is verified by a pilot-scale screening experiment.The present study demonstrates the potential of the DEM method for the analysis of screening processes in VFFS. | Chi Yu Dongdong Lin Ningning Xu Xinwen Wang Kunwei Pu Zhenghao Wang Guofeng Zhao Runhui Geng Sanpeng Gong | 2023 | Particuology2023,,2: | 2 |

They play pivotal roles in various biological processes, such as energy conservation, metabolism,and signal modulation. Due to the difficulty of detection caused by variety OPBs, investigation of their respective physiological effects in organisms has been restrained by the lack of efficient tools. Many small fluorescent probes have been employed for selective detection and monitoring of OPBs in vitro or in vivo due to the advantages of tailored properties, biodegradability and in situ high temporal and spatial resolution imaging.
