| 2 | A viral vector expressing hypoxia-inducible factor 1 alpha inhibits hippocampal neuronal apoptosis显示文摘Hypoxia-inducible factor 1(HIF-1) attenuates amyloid-beta protein neurotoxicity and decreases apoptosis induced by oxidative stress or hypoxia in cortical neurons. In this study, we constructed a recombinant adeno-associated virus(rAAV) vector expressing the human HIF-1α gene(rAAV-HIF-1α), and tested the assumption that rAAV-HIF-1α represses hippocampal neuronal apoptosis induced by amyloid-beta protein. Our results confirmed that rAAV-HIF-1α significantly reduces apoptosis induced by amyloid-beta protein in primary cultured hippocampal neurons. Direct intracerebral rAAV-HIF-1α administration also induced robust and prolonged HIF-1α production in rat hippocampus. Single rAAV-HIF-1α administration resulted in decreased apoptosis of hippocampal neurons in an Alzheimer's disease rat model established by intracerebroventricular injection of aggregated amyloid-beta protein(25–35). | 12 | Engineering vacancy and hydrophobicity of two-dimensional TaTe_(2)for efficient and stable electrocatalytic N_(2)reduction显示文摘Demand for ammonia continues to increase to sustain the growing global population.The direct electrochemical N2 reduction reaction(NRR)powered by renewable electricity offers a promising carbon-neutral and sustainable strategy for manufacturing NH3,yet achieving this remains a grand challenge.Here,we report a synergistic strategy to promote ambient NRR for ammonia production by tuning the Te vacancies(VTe)and surface hydrophobicity of two-dimensional TaTe_(2)nanosheets.Remarkable NH3 faradic efficiency of up to 32.2%is attained at a mild overpotential,which is largely maintained even after 100 h of consecutive electrolysis.Isotopic labeling validates that the N atoms of formed NH4+originate from N2.In situ X-ray diffraction indicates preservation of the crystalline structure of TaTe_(2)during NRR.Further density functional theory calculations reveal that the potential-determining step(PDS)is*NH_(2)+(H^(+)+e^(-))/NH3 on VTe-TaTe_(2)compared with that of*+N2+(H^(+)+e^(-))/*N-NH on TaTe_(2).We identify that the edge plane of TaTe_(2)and VTe serve as the main active sites for NRR.The free energy change at PDS on VTe-TaTe_(2)is comparable with the values at the top of the NRR volcano plots on various transition metal surfaces. | Zhenqing Zhao Jongseo Park Changhyeok Choi Song Hong Xiangchao Hui Hao Zhang Tsz Woon Benedict Lo Alex W.Robertson Zengxiang Lv Yousung Jung Zhenyu Sun | 2022 | The Innovation2022,3,1: | 1 |

| 18 | Clinical Application of Circulating Tumor Cells in Evaluating the Effect of Chemotherapy in Advanced Non-Small Cell Lung Cancer显示文摘Objective:To explore the value of circulating tumor cells in evaluating the effect of chemotherapy for advanced non-small cell lung cancer.Methods:Sixty-two patients with advanced non-small cell lung cancer who received chemotherapy in the Affiliated Hospital of Hebei University from January 2018 to December 2021 were selected as the research subjects.The positive rate of CTCs after two weeks of chemotherapy and four weeks of chemotherapy as well as the evaluation of imaging efficacy were observed and analyzed.Results:Based on the clinical data of the patients,the positive rate of CTCs in male patients was 77.78%,that in female patients was 80.77%,that in patients≥60 years old was 78.13%,that in patients<60 years old was 76.67%,that in squamous cell carcinoma was 79.31%,that in adenocarcinoma was 78.79%,and that in highly differentiated,moderately differentiated,and poorly differentiated+undifferentiated CTCs was 66.67%,84.21%,and 90.91%,respectively;there was no statistical difference in the general data.The positive rate of CTCs was 88.71%before chemotherapy and 66.13%after two weeks of chemotherapy,in which the difference was statistically significant;the positive rate of CTCs four weeks after chemotherapy was 59.68%,which was statistically significant compared with that before chemotherapy;however,there was no significant difference between two weeks after chemotherapy and four weeks after chemotherapy.After chemotherapy,35 patients had CR+PR,19 patients had SD,and 8 patients had PD.The proportions of CR+PR,SD,and PD in the imaging evaluation results were 56.45%,30.65%,and 12.90%,respectively.After kappa consistency test,it was found that the consistency was good.Conclusion:CTCs can be used as one of the indicators to evaluate the effect of chemotherapy for advanced non-small cell lung cancer.The results are consistent with those of imaging evaluation.The detection of CTCs can be widely used as one of the clinical indicators. | Xuguang Zhang Shaoyong Dong Zhenqing Sun | 2022 | Proceedings of Anticancer Research2022,6,5: | 0 |

| 20 | N-glycans released from glycoproteins using a commercial kit and comprehensively analyzed with a hypothetical database显示文摘The glycosylation of proteins is responsible for their structural and functional roles in many cellular activities.This work describes a strategy that combines an efficient release, labeling and liquid chromatography–mass spectral analysis with the use of a comprehensive database to analyze N-glycans. The analytical method described relies on a recently commercialized kit in which quick deglycosylation is followed by rapid labeling and cleanup of labeled glycans. This greatly improves the separation, mass spectrometry(MS) analysis and fluorescence detection of N-glycans. A hypothetical database, constructed using Glyc Resoft, provides all compositional possibilities of N-glycans based on the common sugar residues found in N-glycans. In the initial version this database contains > 8,700 N-glycans, and is compatible with MS instrument software and expandable. N-glycans from four different well-studied glycoproteins were analyzed by this strategy. The results provided much more accurate and comprehensive data than that had been previously reported. This strategy was then used to analyze the N-glycans present on the membrane glycoproteins of gastric carcinoma cells with different degrees of differentiation. Accurate and comprehensive N-glycan data from those cells was obtained efficiently and their differences were compared corresponding to their differentiation states.
