4Methodology of drug screening and target identification for new necroptosis inhibitors显示文摘Apoptosis has been considered as the only form of regulated cell death for a long time. However, a novel form of programmed cell death called necroptosis was recently reported. The process of necroptosis is regulated and plays a critical role in the occurrence and development of multiple human diseases. Thus,the study on the molecular mechanism of necroptosis and its effective inhibitors has been an attractive field for researchers. Herein, we introduce the molecular mechanism of necroptosis and focus on the literature about necroptosis drug screening in recent years. 5Sphingosine-1-phosphate,a novel TREM2 ligand,promotes microglial phagocytosis to protect against ischemic brain injury显示文摘The mechanism of sphingosine-1-phosphate(S1P)-mediated phagocytosis remains unknown.Here,we found that S1P or FTY720(an analog of S1P)promoted microglial phagocytosis in stroke independent of S1PRs.First,we used computer simulation of molecular docking to predict that S1P might be a ligand for triggering receptor expressed on myeloid cells 2(TREM2).Next,microscale thermophoresis(MST),surface plasmon resonance(SPR)and liquid chromatography–tandem mass spectrometry(LC–MS/MS)were performed to reveal that S1P was a novel TREM2 ligand.Then,we confirmed the pro-phagocytosis of S1P targeting in Trem2-Dap12 transfected CHO cells and TREM2 knockdown microglia.Point mutation analysis showed that D104 was the critical binding residue.Trem2^(−/−)mice were used to demonstrate the role of S1P-induced phagocytosis targeting on TREM2 in protecting against ischemic brain injury.Finally,further studies revealed that apolipoprotein E(APOE)loaded with S1P was released by microglia and bound to apoptotic neurons via LDL receptor related protein 1B(LRP1B)and thereby induced microglia to phagocytose apoptotic neurons.Overall,the present work reveals for the first time that S1P acts as a novel endogenous ligand of TREM2 to effectively promote microglial phagocytosis.Our findings provide a new lead compound for developing immunomodulator targeting on TREM2.Tengfei Xue Juan Ji Yuqin Sun Xinxin Huang Zhenyu Cai Jin Yang Wei Guo Ruobing Guo Hong Cheng Xiulan Sun 2022Acta Pharmaceutica Sinica B2022,12,4:3
6Experimental and numerical analysis of the heat flux characteristic of the plume of a 120-N thruster显示文摘The convective heat transfer of the plume of a 120-N thruster is investigated experimentally and numerically. Numerical results agree well with experimental results in that there is a nonlinear decrease in convective heat transfer with an increasing cone angle. It is also found that convective heat transfer decreases with increasing distance from the thruster outlet. Furthermore, the convective heat transfer of the plume mainly concentrates within a 35° cone angle and the heat flux decreases to the same order as solar radiation at the Earth’s surface when the cone angle exceeds 60°. 7Smart ring resonator–based sensor for multicomponent chemical analysis via machine learning显示文摘We demonstrate a smart sensor for label-free multicomponent chemical analysis using a single label-free ring resonator to acquire the entire resonant spectrum of the mixture and a neural network model to predict the composition for multicomponent analysis. The smart sensor shows a high prediction accuracy with a low rootmean-squared error ranging only from 0.13 to 2.28 mg/m L. The predicted concentrations of each component in the testing dataset almost all fall within the 95% prediction bands. 8Mineralogical Characteristics of Exsolved Spinel in the Panzhihua V-Ti Magnetite Deposit, Sichuan: Implications for the Mineralization Process显示文摘Spinel exsolution is widespread in titanomagnetite from the Fe-Ti oxide gabbro of the Panzhihua intrusion, Emeishan Large Igneous Province, SW China. However, little research has been conducted into the implications of patterns in the mineralogical characteristics of the spinel for spatial variation in the controls on the exsolution mechanism and, hence, the formation process of the ore deposit. This study selected the Lanjiahuoshan Ore Block in the Panzhihua V-Ti magnetite deposit to explore this issue, systematically studying exsolution textures in the titanomagnetite through petrographic observation and the integrated use of in-situ microanalysis. The results show that the exsolved spinel gradually becomes finer-grained and less abundant from the center to edge and the bottom to top of the ore bodies. Compositionally, there is an inverse correlation between the size of exsolved spinel grains and their Mg# value. In addition, there is compositional zonation in the spinel interiors, with a gradual increase in the Mg content and decrease in Fe content from the core to the rim. The analysis suggests that fractional crystallization of ferrotitanium magma with a high oxygen fugacity in a shallow magma chamber caused compositional differences in the primary magnetite solid solution in different parts of the Panzhihua intrusion. Additionally, the thermal evolution of the magnetite solid solution differed in different parts of orebody, bringing about variations in spinel development. Together, these effects resulted in spatial variation in the abundance, grain size, and morphology of spinel in different parts of the orebody and intrusion that follows an identifiable distribution law. Furthermore, the compositional zonation of exsolved spinels reflects the rapid growth of exsolution features in a high-temperature environment. 17Bi/BiVO_(4)/NiFe-LDH heterostructures with enhanced photoelectrochemical performance for streptomycin detection显示文摘Streptomycin(STR)plays an essential role in bacterial infection treatments.Selectivity and sensitivity of photoelectrochemical(PEC)sensors are the two most important parameters,which can be measured using the photosensitivity of its active material.We prepared a novel PEC sensor to detect STR using Bi/BiVO4/LDH(layered double hydroxides)heterostructures as an active material,which is photoactive in the visible light wavelength range.The simultaneous presence of LDH and Bi/BiVO4 enhanced the material photocurrent response,which was linear to the STR concentrations in the 0.01–500 nmol/L range.The STR detection limit by this sensor was 0.0042 nmol/L.Our novel PEC-based sensing strategy includes using an ultra-sensitive and highly selective sensor for STR detection.Additionally,the two-pot synthesis of Bi/BiVO4/LDH developed in this work is environmentally friendly.Liqin Yang Jun Zhao Caijun Wang Zheng Wang Cuijuan Xing Hao Guo Yuman Wang Zhiju Zhao Zhangji Hu Zhenyu Cai 2021Journal of Environmental Sciences2021,33,11:1

19Metformin escape in prostate cancer by activating the PTGR1 transcriptional program through a novel super-enhancer显示文摘The therapeutic efficacy of metformin in prostate cancer(PCa)appears uncertain based on various clinical trials.Metformin treatment failure may be attributed to the high frequency of transcriptional dysregulation,which leads to drug resistance.However,the underlying mechanism is still unclear.In this study,we found evidences that metformin resistance in PCa cells may be linked to cell cycle reactivation.Super-enhancers(SEs),crucial regulatory elements,have been shown to be associated with drug resistance in various cancers.Our analysis of SEs in metformin-resistant(MetR)PCa cells revealed a correlation with Prostaglandin Reductase 1(PTGR1)expression,which was identified as significantly increased in a cluster of cells with metformin resistance through single-cell transcriptome sequencing.Our functional experiments showed that PTGR1 overexpression accelerated cell cycle progression by promoting progression from the G0/G1 to the S and G2/M phases,resulting in reduced sensitivity to metformin.Additionally,we identified key transcription factors that significantly increase PTGR1 expression,such as SRF and RUNX3,providing potential new targets to address metformin resistance in PCa.In conclusion,our study sheds new light on the cellular mechanism underlying metformin resistance and the regulation of the SE-TFs-PTGR1 axis,offering potential avenues to enhance metformin’s therapeutic efficacy in PCa.Jianheng Ye Shanghua Cai Yuanfa Feng Jinchuang Li Zhiduan Cai Yulin Deng Ren Liu Xuejin Zhu Jianming Lu Yangjia Zhuo Yingke Liang Jianjiang Xie Yanqiong Zhang Huichan He Zhaodong Han Zhenyu Jia Weide Zhong 2023Signal Transduction and Targeted Therapy2023,8,9:0
