| 3 | A Highly Pathogenic Strain of Porcine Deltacoronavirus Caused Watery Diarrhea in Newborn Piglets显示文摘Porcine deltacoronavirus(PDCoV) is a newly identified virus that causes watery diarrhea in newborn piglets and results in significant economic losses to the pig industry. Since first reported in Hong Kong in 2012, PDCoV has been subsequently detected in USA, South Korea, Thailand, and China's Mainland. Here we isolated a strain of PDCoV, named CHN-GD-2016,from the intestinal content of a diseased newborn piglet with severe diarrhea in a pig farm in Guangdong, China. PDCoV CHN-GD-2016 could be identified by immunofluorescence with PDCoV specific rabbit antisera, and typical crown-shaped particles with spiky surface projections of this PDCoV were observed with electron microscopy. Genomic analysis showed that the PDCoV CHN-GD-2016 was closely related to other Chinese PDCoV strains, with the highest sequence similarity with the strain CHN/Tianjin/2016. Importantly, inoculation of newborn piglets with 1×10~5 TCID_(50) of CHN-GD-2016 by oral feeding successfully reproduced clear clinical symptoms, including vomiting, dehydration, and severe diarrhea in piglets. In addition, the virus RNA in rectal swabs from 1 to 7 days post inoculation was detected, macroscopic and microscopic lesions in small intestine were observed, and viral antigen was also detected in the small intestines with immunohistochemical staining. | 4 | Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. | 5 | Infrared radiation characteristics of a hypersonic vehicle under time-varying angles of attack显示文摘Hypersonic vehicles emit strong infrared(IR) radiation signatures that can be treated as a detecting source for object identification and routine diagnosis. This paper is aimed at examining the intrinsic radiation characteristics of a Boost-Glide Vehicle(BGV) under the condition of various Angles of Attack(AOAs). A two-temperature model considering the thermal and chemical nonequilibrium effects is coupled with Navier-Stokes equations solved by the finite volume technique.A gas–solid conjunction heat transfer model is also added into the fluid solver to simulate the surface temperature of the vehicle. The radiative transfer equation is solved with Line of Sight(LOS)algorithm. The computational results for a Hypersonic Technology Vehicle-2(HTV-2) type vehicle show that radiances of the vehicle are strongly dependent on the surface temperature. The presence of AOA results in the significant difference of the surface temperature. Infrared radiation characteristics are also changed in intensity and spectral band due to the AOA. Simulations are performed with two time-varying AOAs. | 6 | Comparative Genome Analysis of Scutellaria baicalensis and Scutellaria barbata Reveals the Evolution of Active Flavonoid Biosynthesis显示文摘Scutellaria baicalensis(S.baicalensis)and Scutellaria barbata(S.barbata)are common medicinal plants of the Lamiaceae family.Both produce specific flavonoid compounds,including baicalein,scutellarein,norwogonin,and wogonin,as well as their glycosides,which exhibit antioxidant and antitumor activities.Here,we report chromosome-level genome assemblies of S.baicalensis and S.barbata with quantitative chromosomal variation(2 n=18 and 2 n=26,respectively).The divergence of S.baicalensis and S.barbata occurred far earlier than previously reported,and a whole-genome duplication(WGD)event was identified.The insertion of long terminal repeat elements after speciation might be responsible for the observed chromosomal expansion and rearrangement.Comparative genome analysis of the congeneric species revealed the species-specific evolution of chrysin and apigenin biosynthetic genes,such as the S.baicalensis-specific tandem duplication of genes encoding phenylalanine ammonia lyase and chalcone synthase,and the S.barbata-specific duplication of genes encoding 4-Co A ligase.In addition,the paralogous duplication,colinearity,and expression diversity of CYP82 D subfamily members revealed the functional divergence of genes encoding flavone hydroxylase between S.baicalensis and S.barbata.Analyzing these Scutellaria genomes reveals the common and species-specific evolution of flavone biosynthetic genes.Thus,these findings would facilitate the development of molecular breeding and studies of biosynthesis and regulation of bioactive compounds. | Zhichao Xu Ranran Gao Xiangdong Pu Rong Xu Jiyong Wang Sihao Zheng Yan Zeng Jun Chen Chunnian He Jingyuan Song | 2020 | Genomics, Proteomics & Bioinformatics2020,18,3: | 10 |
| 7 | Numerical simulation of mould filling process for pressure plate and valve handle in LFC显示文摘In lost foam casting (LFC), the distribution of polymer beads during the bead filling process is not uniform, and the collision between polymer beads determines the distribution of two-phase flow of gas and solid. The interaction between the gas and solid phases reveals as coupling effect of the force that gas exerts on particles or vice versa, or that among particles. The gas-solid flow in filling process is nonlinearity, which makes the coupling effect an essential point to carry out a simulation properly. Therefore, information of each particle's motion is important for acquiring the law of filling process. In bead filling process, compressed air is pressed into mold cavity, and discharged from gas vent, creating a pressure difference between outer and inner space near the gas vent. This pressure difference directly changes the spatial distribution and motion trace of gas and solid phases. In this paper, Discrete Element Method (DEM) and Computational Fluid Dynamics (CFD) are employed to simulate the fluid dynamic character based on Newton's Third Law of Motion. The simulation results of some casting products such as pressure plate and valve handle are compared with the result obtained from practical experiment in order to test the feasibility of DEM. | 10 | Improving uniform scattering device for straw-smashing,back-throwing,no-tillage planter under complete straw mulching condition显示文摘With the objective of obtaining a completely straw-mulched field,when no-tillage mechanical sowing is implemented with straw smashing,delivering,and back-throwing approaches,it may be difficult to scatter the smashed straw uniformly during a succeeding wheat sowing step.This is because the previous rice straw is substantial in quantity and has a high humidity and toughness,which may easily result in non-uniform straw mulching and thus sparse and weak seedlings of wheat.Therefore a force-dispersing and uniform-scattering device was designed.With the number of scattering impellers,impeller angle,and impeller rotation speed as the main factors and the percentage of pass for the scattering width and non-uniformity of the straw mulching as the assessment indices,single-factor experiments and orthogonal regressive tests were performed,and a dual-index(percentage of pass for the scattering width and non-uniformity of the straw mulching)fitted regression equation was established.The test results suggested that the main factors(from primary to secondary)that influence the indices were the impeller rotation speed,number of scattering impellers,and impeller angle.The optimal parameter combination for the uniform scattering device was four rows of impellers with an angle of 15°,rotation speed of 1015 r/min,percentage of pass of 72.65%for the scattering width,and a non-uniformity of 13.8%in the straw mulching.This combination can be used to realize a uniform scattering of the smashed straw along the seedling rows on the after-sowing ground.According to the field investigation of the wheat growth,the wheat emergence rate was 90.7%.The research results can provide a reference for improving the uniform scattering device for a straw-smashing,back-throwing,no-tillage planter for obtaining a completely straw-mulched field,enhancing the quality of the machinery operation,and ensuring good and strong seedlings after sowing. | Fengwei Gu Xuemei Gao Feng Wu Zhichao Hu Youqing Chen Chong Zhang | 2018 | International Journal of Agricultural and Biological Engineering2018,11,6: | 5 |

| 12 | Nd-Sr Isotopic Geochemistry of the Late Archean-Paleoproterozoic Granitoids in the Lüliang-Wutai Terrain,North China Craton,and Implications for Petrogenesis显示文摘In this paper we report geochemical and Nd-Sr isotopic data for a late Archean gneissic granitic pluton (Hengling pluton), an early Paleoproterozoic complex (Xipan complex) and a late Paleoproterozoic granitic pluton (Yunzhongshan granites) from the Lüliang-Wutai terrain, North China, to trace the source of these late Archean-Paleoproterozoic granitoids and, particularly, to understand the nature and mechanism of continental growth at that time. The Hengling granitic gneisses (ca. 2.51 Ga) are characterized by high Na2O and LILEs, TTG-like REE patterns (highly depleted HREE and minor Eu anomalies) and moderately depleted Nd-Sr isotopic compositions (εNd(t) =1.2?2.7, ISr=0.7015?0.7019), and were considered as being products of arc magmatism that was developed upon the North China craton. The Xipan complex (ca. 2.2 Ga) contain gabbroic diorite and monzonite, mostly being Na2O-rich, highly fractionated REE patterns and isotopically enriched (εNd(t) =?1.5 to ?4.1, ISr=0.7038?0.706). The gabbroic diorites probably originated from melting of an enriched mantle source, but significantly contaminated by lower crustal material, and the monzonites probably represent a product of a mixture between the gabbroic dioritic magma and granitic melts of crustal origin. The Yunzhongshan post-collisional granitoids (ca. 1.8 Ga) are characterized by high-K affinity and highly-enriched and homogeneous Nd isotopic compositions (εNd(t)=?4.9 to ?5.7), although they split into two groups in terms of REE patterns: one group showing elevated HREE (and Sc, Y and Zr) with significant negative Eu anomalies and the other showing highly depleted HREE and, to a lesser extent, mid-REE with negligible Eu anomalies. These granites are genetically related to a process of extensional collapse of a thickened orogen. They formed through magma mixing between mantle- derived basaltic magmas and crust-derived granitic melts, followed by significant fractionation of ferromagnesian phases (like hornblende and Cpx) and feldspar and accessory zircons. | 15 | Rapid earthquake focal mechanism inversion using high-rate GPS velometers in sparse network显示文摘In this study, we demonstrate an approach for inverting earthquake source parameters based on high-rate global positioning system(GPS) velocity seismograms. The velocity records obtained from single-station GPS velocity solutions with broadcast ephemeris are used directly for earthquake source parameter inversion using the Cut and Paste method, without requiring conversion of the velocity records into displacement records. Taking the El Mayor-Cucapah earthquake as an example, GPS velocity records from 10 stations with reasonable azimuthal coverage provide earthquake source parameters very close to those from the Global centroid moment tensor(Global CMT) solution. In sparse network tests, robust source parameters with acceptable bias can be achieved with as few as three stations. When the number of stations is reduced to two, the bias in rake angle becomes appreciable, but the magnitude and strike estimations are still robust. The results of this study demonstrate that rapid and reliable estimation of earthquake source parameters can be obtained from GPS velocity data. | 16 | Bacteria-Mediated Synergistic Cancer Therapy:Small Microbiome Has a Big Hope显示文摘The use of bacteria to specifically migrate to cancerous tissue and elicit an antitumor immune response provides a promising platform against cancer with significantly high potency.With dozens of clinical trials underway,some researchers hold the following views:“humans are nearing the first commercial live bacteria therapeutic.”However,the facultative anaerobe Salmonella typhimurium VNP20009,which is particularly safe and shows anticancer effects in preclinical studies,had failed in a phase I clinical trial due to low tumor regression and undesired dose-dependent side effects.This is almost certain to disappoint people’s inflated expectations,but it is noted that recent stateof-the-art research has turned attention to bacteria-mediated synergistic cancer therapy(BMSCT).In this review,the foundation of bacteria-mediated bio-therapy is outlined.Then,we summarize the potential benefits and challenges of bacterial bio-therapy in combination with different traditional anticancer therapeutic modalities(chemotherapy,photothermal therapy,reactive oxygen and nitrogen species therapy,immunotherapy,or prodrug-activating therapy)in the past 5 years.Next,we discuss multiple administration routes of BMSCT,highlighting potentiated antitumor responses and avoidance of potential side effects.Finally,we envision the opportunities and challenges for BMSCT development,with the purpose of inspiring medicinal scientists to widely utilize the microbiome approach in patient populations. | Xinyu Lou Zhichao Chen Zhonggui He Mengchi Sun Jin Sun | 2021 | Nano-Micro Letters2021,13,2: | 3 |
| 17 | Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors. | Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang | 2022 | Animal Models and Experimental Medicine2022,5,1: | 3 |

| 19 | Distribution Features of the Nanhua-Sinian Rifts and their Significance to Hydrocarbon Accumulation in the Tarim Basin显示文摘On the basis of reprocessing 34 new two-dimensional spliced long sections(20,191 km) in the Tarim Basin, the deep structure features of the Tarim Basin were analyzed through interpreting 30,451 km of two-dimensional seismic data and compiling basic maps. Seismic interpretation and geological analysis conclude that the Nanhua-Sinian strata are a set of rift-depression depositional systems according to their tectonic and depositional features. The rift valley formed in the Nanhua Period, and the transformation became weaker during the late Sinian Period, which eventually turned into depression. From bottom to top, the deposited strata include mafic igneous, tillite, mudstone, and dolomite. Three major depocenters developed inside this basin during the rift stage and are distributed in the eastern Tarim Basin, the Awati area, and the southwestern Tarim Basin. Among them, the rift in the eastern Tarim Basin strikes in the near east-west direction on the plane and coincides with the aeromagnetic anomaly belt. This represents a strong magnetic zone formed by upwelling basic volcanic rock along high, steep normal faults of the Nanhua Period. Controlled by the tectonic background, two types of sedimentary systems were developed in the rift stage and depression stage, showing two types of sequence features in the Sinian depositional stage. The Nanhua System appears as a wedge-shaped formation, with its bottom in unconformable contact with the base. The rifting event has a strong influence on the current tectonic units in the Tarim Basin, and affects the distribution of source rock in the Yuertus Formation and reservoir beds in the Xiaoerbulake Formation in Lower Cambrian, as well as the gypseous cap rock in Middle Cambrian. The distribution features of the rifts have important and realistic significance for determining the direction of oil and gas exploration in the deep strata of the Tarim Basin. | 20 | NiPt hollow nanocatalyst: Green synthesis, size control and electrocatalysis显示文摘Nearly monodispersed hollow nanospheres of bimetallic NiPt have been synthesized by a one-pot wet chemical method at room temperature with a precursor Ni nanocompound as a sacrificial template. The size control is carried out via the sacrificial template, from about 35 nm to nearly100 nm in diameter. The shell thickness of the NiPt hollow sphere reaches down to as thin as 2–3 nm slightly larger than a single layer of alloyed NiPt nanocrystallites. The product with the citric acid as surfactant exhibits enhanced oxygen reduction activities compared to a commercial Pt/C catalyst and the hollow nanospheres coated with PVP.
