| 1 | s-HBEGF/SIRT1 circuit-dictated crosstalk between vascular endothelial cells and keratinocytes mediates sorafenib-induced hand-foot skin reaction that can be reversed by nicotinamide显示文摘Hand-foot skin reaction(HFSR),among the most significant adverse effects of sorafenib,has been limiting the clinical benefits of this frontline drug in treating various malignant tumors.The mechanism underlying such toxicity remains poorly understood,hence the absence of effective intervention strategies.In the present study,we show that vascular endothelial cells are the primary cellular target of sorafenib-induced HFSR wherein soluble heparin-binding epidermal growth factor(s-HBEGF)mediates the crosstalk between vascular endothelial cells and keratinocytes.Mechanistically,s-HBEGF released from vascular endothelial cells activates the epidermal growth factor receptor(EGFR)on keratinocytes and promotes the phosphorylation of c-Jun N-terminal kinase 2(JNK2),which stabilizes sirtuin 1(SIRT1),an essential keratinization inducer,and ultimately gives rise to HFSR.The administration of s-HBEGF in vivo could sufficiently induce hyper-keratinization without sorafenib treatment.Furthermore,we report that HBEGF neutralization antibody,Sirtl knockdown,and a classic SIRTI inhibitor nicotinamide could all significantly reduce the sorafenib-induced HFSR in the mouse model.It is noteworthy that nicotinic acid,a prodrug of nicotinamide,could substantially reverse the sorafenib-induced HFSR in ten patients in a preliminary clinical study.Collectively,our findings reveal the mechanism of vascular endothelial cell-promoted keratinization in keratinocytes and provide a potentially promising therapeutic strategy for the treatment of sorafenib-induced HFSR. | Peihua Luo Hao Yan Xueqin Chen Ying Zhang Ziying Zhao Ji Cao Yi Zhu Jiangxia Du Zhifei Xu Xiaochen Zhang Su Zeng Bo Yang Shenglin Ma Qiaojun He | 2020 | Cell Research2020,30,9: | 5 |
| 2 | A study on the changes of dynamic feature code when fixing bugs: towards the benefits and costs of Python dynamic features显示文摘Dynamic features in programming languages support the modification of the execution status at runtime, which is often considered helpful in rapid development and prototyping. However, it was also reported that some dynamic feature code tends to be change-prone or error-prone. We present the first study that analyzes the changes of dynamic feature code and the roles of dynamic features in bug-fix activities for the Python language. We used an AST-based differencing tool to capture fine-grained source code changes from 17926 bug-fix commits in 17 Python projects. Using this data, we conducted an empirical study on the changes of dynamic feature code when fixing bugs in Python. First, we investigated the characteristics of dynamic feature code changes, by comparing the changes between dynamic feature code and non-dynamic feature code when fixing bugs, and comparing dynamic feature changes between bug-fix and non-bugfix activities. Second, we explored 226 bug-fix commits to investigate the motivation and behaviors of dynamic feature changes when fixing bugs. The study results reveal that(1) the changes of dynamic feature code are significantly related to bug-fix activities rather than non-bugfix activities;(2) compared with non-dynamic feature code, dynamic feature code is inserted or updated more frequently when fixing bugs;(3) developers often insert dynamic feature code as type checks or attribute checks to fix type errors and attribute errors;(4) the misuse of dynamic features introduces bugs in dynamic feature code, and the bugs are often fixed by adding a check or adding an exception handling. | 4 | Empirical analysis of network measures for predicting high severity software faults显示文摘Network measures are useful for predicting fault-prone modules. However, existing work has not distinguished faults according to their severity. In practice, high severity faults cause serious problems and require further attention. In this study, we explored the utility of network measures in high severity faultproneness prediction. We constructed software source code networks for four open-source projects by extracting the dependencies between modules. We then used univariate logistic regression to investigate the associations between each network measure and fault-proneness at a high severity level. We built multivariate prediction models to examine their explanatory ability for fault-proneness, as well as evaluated their predictive effectiveness compared to code metrics under forward-release and cross-project predictions. The results revealed the following:(1) most network measures are significantly related to high severity fault-proneness;(2) network measures generally have comparable explanatory abilities and predictive powers to those of code metrics; and(3) network measures are very unstable for cross-project predictions. | 5 | Engineering imaging probes and molecular machines for nanomedicine显示文摘Nanomedicine is an emerging field that integrates nanotechnology, biomolecular engineering, life sciences and medicine; it is expected to produce major breakthroughs in medical diagnostics and therapeutics. Due to the size-compatibility of nano-scale structures and devices with proteins and nucleic acids, the design, synthesis and application of nanoprobes, nanocarriers and nanomachines provide unprecedented opportunities for achieving a better control of biological processes, and drastic improvements in disease detection, therapy, and prevention. Recent advances in nanomedicine include the development of functional nanoparticle based molecular imaging probes, nano-structured materials as drug/gene carriers for in vivo delivery, and engineered molecular machines for treating single-gene disorders. This review focuses on the development of molecular imaging probes and engineered nucleases for nanomedicine, including quantum dot bioconjugates, quantum dot-fluorescent protein FRET probes, molecular beacons, magnetic and gold nanoparticle based imaging contrast agents, and the design and validation of zinc finger nucleases (ZFNs) and TAL effector nucleases (TALENs) for gene targeting. | 7 | LncRNA LINC00525 suppresses p21 expression viamRNA decay and triplex-mediated changes in chromatin structure in lung adenocarcinoma显示文摘Background:Emerging evidence suggests that long noncoding RNAs(lncRNAs)play crucial roles in various cancers.In the present study,we aim to investigate the function and molecular mechanism of an up-regulated and survivalassociated lncRNA,LINC00525,in lung adenocarcinoma(LUAD).Methods:The expression level of LINC00525 in tissueswas determined by quantitative reverse transcription polymerase chain reaction(RT-qPCR)and in situ hybridization(ISH).The functional role of LINC00525 in LUAD was investigated using gain-and loss-of-function approaches,both in vivo and in vitro.RNA pull-down,RNA immunoprecipitation(RIP),chromatin immunoprecipitation(ChIP),triplex-capture assay,dual-luciferase assay,gene expression microarray,and bioinformatics analysis were used to investigate the potential underlying mechanisms involved.Results:LINC00525 is highly expressed in LUAD cells and tissues.Survival analysis indicated that upregulation of LINC00525 was associated with poor prognosis in patients with LUAD patients.Knockdown of LINC00525 inhibited cell proliferation and cell cycle progression in vitro.In xenograft models,LINC00525 knockdown suppressed tumor growth and tumorigenesis of tumorbearing mice.Mechanistically,LINC00525 epigenetically suppressed p21 transcription by guiding Enhancer Of Zeste 2 Polycomb Repressive Complex 2 Subunit(EZH2)to the p21 promoter through an formation of RNA-DNA triplex with the p21 promoter,leading to increased trimethylation of lysine 27 on histone 3(H3K27me3)of the p21 promoter.In addition,LINC00525 repressed p21 expression post-transcriptionally by enhancing p21mRNA decay.LINC00525 promoted p21mRNAdecay by competitively binding toRNABindingMotif Single Stranded Interacting Protein 2(RBMS2).Conclusion:Our findings demonstrate that LINC00525 promotes the progression of LUAD by reducing the transcription and stability of p21 mRNA in concert with EZH2 and RBMS2,thus suggesting that LINC00525 may be a potential therapeutic target for clinical intervention in LUAD. | Panqi Fang Hao Chen Zhifei Ma Chencheng Han Wenda Yin Siwei Wang Hongyu Zhu Wenjia Xia Jie Wang Lin Xu Tongyan Liu Rong Yin | 2021 | Cancer Communications2021,41,7: | 2 |
| 8 | De novo strategy with engineering a multifunctional bacterial cellulose-based dressing for rapid healing of infected wounds显示文摘The treatment and healing of infected skin lesions is one of the major challenges in surgery.To solve this problem,collagen I(Col-I)and the antibacterial agent hydroxypropyltrimethyl ammonium chloride chitosan(HACC)were composited into the bacterial cellulose(BC)three-dimensional network structure by a novel membrane-liquid interface(MLI)culture,and a Col-I/HACC/BC(CHBC)multifunctional dressing was designed.The water absorption rate and water vapor transmission rate of the obtained CHBC dressing were 35.78±2.45 g/g and 3084±56 g m^(-2)⋅day^(-1),respectively.The water retention of the CHBC dressing was significantly improved compared with the BC caused by the introduced Col-I and HACC.In vitro results indicated that the combined advantages of HACC and Col-I confer on CHBC dressings not only have outstanding antibacterial properties against Staphylococcus aureus(S.aureus)compared with BC and CBC,but also exhibit better cytocompatibility than BC and HBC to promote the proliferation and spread of NIH3T3 cells and HUVECs.Most importantly,the results of in vivo animal tests demonstrated that the CHBC dressings fully promoted wound healing for 8 days and exhibited shorter healing times,especially in the case of wound infection.Excellent skin regeneration effects and higher expression levels of collagen during infection were also shown in the CHBC group.We believe that CHBC composites with favorable multifunctionality have potential applications as wound dressings to treat infected wounds. | Chen Zhou Zhifei Yang Xiaowei Xun Le Ma Zejing Chen Xiaoming Hu Xidong Wu Yizao Wan Haiyong Ao | 2022 | Bioactive Materials2022,7,7: | 1 |

| 16 | TRIP Steel Retained Austenite Transformation under Cyclic V-bending Deformation显示文摘To investigate the transformation behavior of TRIP steel retained austenite under cyclic load, cyclic V-bending deformation of low carbon Si-Mn TRIP600 was studied by experiment and finite element in this paper. The results showed that, under cyclic V-bending deformation, retained austenite in TRIP steel transformed into martensite gradually with the increasing of bending times, and for the symmetrical characteristic, upper surface and lower surface presented the same transformation tendency. From the fi rst to the fourth V-bending deformation, retained austenite volume fraction decreased nearly linearly and then attained saturation step by step. Compressive stress state was helpful for martensite transformation than tension stress state with V-bending deformation, and strain magnitude was the determining factor for retaining austenite martensitic transformation. With the increasing of bending times effective stress increased and the relationship between maximum effective stress and bending times was nearly linear. Effective stress and effective strain distribution were non-uniform, the maximum effective stress and effective strain were present in the center of the samples. The relationships between retained austenite and V-bending times, and retained austenite with effective strain were set up as Eqs.(1)-(5). | 17 | Clay mineral assemblages of the oceanic red beds in the northern South China Sea and their responses to the Middle Miocene Climate Transition显示文摘The Middle Miocene Climate Transition(MMCT,~14 Ma)is the largest cooling event in the Cenozoic“Coolhouse”,which significantly impacts the global chemical weathering pattern.In this paper,the responses of the MMCT global cooling event in the deep South China Sea were studied by clay mineral assemblages analysis of the oceanic red beds(ORB)at International Ocean Discovery Program(IODP)Expedition 368 Site U1502.The results show that the clay mineral assemblages of the ORB at Site U1502 are mainly composed of smectite(56–88%),illite(7–29%),and kaolinite(6–20%),without chlorite.The contents of these clay minerals and illite crystallinity show a four-stage variation pattern during early-middle Miocene(22.8–10.8 Ma).Smectite decreased from average 81%during 22.8–16.2 Ma and 16.2–14.4 Ma to average 67%during 13.8–10.8 Ma,with a rapid decrease of~14%during 14.4–13.8 Ma.On the contrary,illite and kaolinite increased rapidly by~8%and~6%,respectively,during 14.4–13.8 Ma.Illite crystallinity increased from average 0.18°Δ2θduring 22.8–16.2 Ma to average 0.19°Δ2θduring 16.2–14.4 Ma,and then decreased rapidly by~0.02°Δ2θduring 14.4–13.8 Ma.The provenance analysis of clay minerals shows that illite and kaolinite mainly originated from South China landmass due to physical erosion,while smectite mainly came from the Luzon arc as the product of chemical weathering.Therefore,smectite/illite ratio and illite crystallinity are used as proxies of chemical weathering intensity in the early-middle Miocene.High values of the ratio and the crystallinity represent the enhanced chemical weathering,whereas low values indicate the weakened chemical weathering or the strengthened physical erosion process.The smectite/illite ratio and illite crystallinity both decreased rapidly during 14.4–13.8 Ma,indicating the chemical weathering in the surrounding area of the South China Sea weakened rapidly,which we believe is the result of the MMCT event forcing.In addition,their values increased slightly during 16.2–14.4 Ma,which is in response to the relatively enhanced chemical weathering during the Middle Miocene Climate Optimum(MMCO).The variation pattern of clay mineral assemblages of the early-middle Miocene ORB in the South China Sea and its rapid transformation during the MMCT reveal that the Cenozoic cooling played a specific role in controlling the chemical weathering of the Earth’s surface. | Lei HE Zhifei LIU Xuan LYU Pengfei MA | 2022 | Science China Earth Sciences2022,65,5: | 0 |

| 19 | Preparation of nZVI embedded modified mesoporous carbon for catalytic persulfate to degradation of reactive black 5显示文摘High-efficiency and cost-effective catalysts with available strategies for persulfate(PS)activation are critical for the complete mineralization of organic contaminants in the environmental remediation and protection fields.A nanoscale zero-valent iron-embedded modified mesoporous carbon(MCNZVI)with a core-shell structure is synthesized using the hydrothermal synthesis method and high-temperature pyrolysis.The results showed that nZVI could be impregnated within mesoporous carbon frameworks with a comparatively high graphitization degree,rich nitrogen doping content,and a large surface area and pore volume.This material was used as a persulfate activator for the oxidation removal of Reactive Black 5(RB5).The effects of the material dosage,PS concentration,pH,and some inorganic anions(i.e.,Cl^(−),SO_(4)^(2−))on RB5 degradation were then investigated.The highest degradation efficiency(97.3%)of RB5 was achieved via PS(20 mmol/L)activation by the MCNZVI(0.5 g/L).The pseudo-first-order kinetics(k=2.11×10^(−2)min^(−1))in the MCNZVI/PS(0.5 g/L,20 mmol/L)was greater than 100 times than that in the MCNZVI and PS.The reactive oxygen species(ROS),including^(1)O_(2),SO_(4)^(·−),HO^(.),and·O_(2)^(−),were generated by PS activation with the MCNZVI.Singlet oxygen was demonstrated to be the primary ROS responsible for the RB5 degradation.The MCNZVI could be reused and regenerated for recycling.This work provides new insights into PS activation to remove organic contamination. | Zhifei Ma Huali Cao Fengchun Lv Yu Yang Chen Chen Tianxue Yang Haixin Zheng Daishe Wu | 2021 | Frontiers of Environmental Science & Engineering2021,15,5: | 0 |
