| 1 | Time resource management of OAR based on fuzzy logic priority for multiple target tracking显示文摘For coping with the multiple target tracking in the presence of complex time-varying environments and unknown target information, a time resource management scheme based on chance-constraint programming(CCP) employing fuzzy logic priority is proposed for opportunistic array radar(OAR). In this scheme,the total beam illuminating time is minimized by effective time resource allocation so that the desired tracking performance is achieved. Meanwhile, owing to the randomness of radar cross section(RCS), the CCP is used to balance tracking accuracy and time resource conditioned on the specified confidence level. The adaptive fuzzy logic prioritization, imitating the human decision-making process for ranking radar targets, can realize the full potential of radar. The Bayesian Crame ′r-Rao lower bound(BCRLB) provides us with a low bound of localization estimation root-mean-square error(RMSE), and equally important, it can be calculated predictively. Consequently, it is employed as an optimization criterion for the time resource allocation scheme. The stochastic simulation is integrated into the genetic algorithm(GA) to compose a hybrid intelligent optimization algorithm to solve the CCP optimization problem. | 4 | Genome-wide CRISPR activation screen identifies candidate receptors for SARS-CoV-2 entry显示文摘The outbreak of coronavirus disease 2019(COVID-19) caused by SARS-CoV-2 has created a global health crisis. SARS-CoV-2 infects varieties of tissues where the known receptor ACE2 is low or almost absent, suggesting the existence of alternative viral entry pathways. Here, we performed a genome-wide barcoded-CRISPRa screen to identify novel host factors that enable SARS-CoV-2 infection. Beyond known host proteins, i.e., ACE2, TMPRSS2, and NRP1, we identified multiple host components,among which LDLRAD3, TMEM30A, and CLEC4G were confirmed as functional receptors for SARS-CoV-2. All these membrane proteins bind directly to spike’s N-terminal domain(NTD). Their essential and physiological roles have been confirmed in either neuron or liver cells. In particular, LDLRAD3 and CLEC4G mediate SARS-CoV-2 entry and infection in an ACE2-independent fashion. | 9 | HSPA8 acts as an amyloidase to suppress necroptosis by inhibiting and reversing functional amyloid formation显示文摘Ultra-stable fibrous structure is a hallmark of amyloids.In contrast to canonical disease-related amyloids,emerging research indicates that a significant number of cellular amyloids,termed‘functional amyloids’,contribute to signal transduction as temporal signaling hubs in humans.However,it is unclear how these functional amyloids are effectively disassembled to terminate signal transduction.RHIM motif-containing amyloids,the largest functional amyloid family discovered thus far,play an important role in mediating necroptosis signal transduction in mammalian cells.Here,we identify heat shock protein family A member 8(HSPA8)as a new type of enzyme—which we name as‘amyloidase’—that directly disassembles RHIM-amyloids to inhibit necroptosis signaling in cells and mice.Different from its role in chaperone-mediated autophagy where it selects substrates containing a KFERQ-like motif,HSPA8 specifically recognizes RHIM-containing proteins through a hydrophobic hexapeptide motif N(X_(1))φ(X_(3)).The SBD domain of HSPA8 interacts with RHIM-containing proteins,preventing proximate RHIM monomers from stacking into functional fibrils;furthermore,with the NBD domain supplying energy via ATP hydrolysis,HSPA8 breaks down pre-formed RHIM-amyloids into non-functional monomers.Notably,HSPA8’s amyloidase activity in disassembling functional RHIM-amyloids does not require its co-chaperone system.Using this amyloidase activity,HSPA8 reverses the initiator RHIM-amyloids(formed by RIP1,ZBP1,and TRIF)to prevent necroptosis initiation,and reverses RIP3-amyloid to prevent necroptosis execution,thus eliminating multi-level RHIM-amyloids to effectively prevent spontaneous necroptosis activation.The discovery that HSPA8 acts as an amyloidase dismantling functional amyloids provides a fundamental understanding of the reversibility nature of functional amyloids,a property distinguishing them from disease-related amyloids that are unbreakable in vivo. | Erpeng Wu Wenyan He Chenlu Wu Zhangcheng Chen Shijie Zhou Xialian Wu Zhiheng Hu Kelong Jia Jiasong Pan Limin Wang Jie Qin Dan Liu Junxia Lu Huayi Wang Jixi Li Sheng Wang Liming Sun | 2023 | Cell Research2023,33,11: | 0 |
| 10 | 青蒿不同溶剂萃取粗提物对结肠癌HT-29、Lovo细胞NF-κB活性的影响(英文)显示文摘Objective: To study changes of NF-κB activity in colon carcinoma cell lines treated with different crude extracts of abrotani herba obtained through solvent extraction methods. Methods: Crude extracts of abrotani herba were extracted with ligarine, chloroform, acetoacetate and n-butanol in separating funnel. Exposure concentration of crude extracts were obtained through detecting viability of HT-29 cells by MTT. Then HT-29 cells and Lovo cells were treated with different crude extracts respectively. Changes of NF-κB activity in HT-29 cells and Lovo cells using different crude extracts were observed by EMSA. Results: Successfully extracted different crude extracts of abrotani herba and called them ligarine extract, chloroform extract, acetoacetate extract, n-butanol extract and remaining extract for short. NF-κB activity was significantly inhibited in HT-29 cells treated with chloroform extract, there were no significant differences in other groups compared with the control. The same change of NF-κB activity was observed in Lovo cells using different crude extracts of abrotani herba.
