1Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
2Air-sea heat flux exchange over the South China Sea under different weather conditions before and after southwest monsoon onset in 2000显示文摘With the data observed from the Second SCS Air-Sea Flux Experiment on the Xisha air-sea flux research tower, the radiation budget, latent, sensible heat fluxes and net oceanic heat budgetswere caculated before and after summer monsoon onset. It is discovered that, after summer monsoononset, there are considerable changes in air-sea fluxes, especially in latent heat fluxes and net oceanicheat budget. Furthermre, the analyzed results of five synoptic stages are compared. And the character-istics of the flux transfer during different stages around onset of South China Sea monsoon are discussed.The flux change shows that there is an oceanic heat accumulating process during the pre-onset and thebreak period, as same as oceanic heat losing process during the onset period. Moreover, latent fluxes,the water vapor moving to the continent, even the rainfall appearance in Chinese Mainland also can beinfluenced by southwester. Comparing Xisha fluxes with those obtained from the Indian Ocean and thewestern Pacific Ocean, their differences may be obeerved. 3m6A RNA methylation-mediated HNF3γreduction renders hepatocellular carcinoma dedifferentiation and sorafenib resistance显示文摘Hepatocyte nuclear factor 3γ(HNF3γ)is a hepatocyte nuclear factor,but its role and clinical significance in hepatocellular carcinoma(HCC)remain unclear.Herein,we report that HNF3γexpression is downregulated in patient HCC and inversely correlated with HCC malignancy and patient survival.Moreover,our data suggested that the HNF3γreduction in HCC could be mediated by METTL14-dependent m6A methylation of HNF3γmRNA.HNF3γexpression was increased during hepatic differentiation and decreased in dedifferentiated HCC cells.Interestingly,HNF3γdelivery promoted differentiation of not only HCC cells but also liver CSCs,which led to suppression of HCC growth.Mechanistic analysis suggested an HNF3γ-centered regulatory network that includes essential liver differentiation-associated transcription factors and functional molecules,which could synergistically facilitate HCC cell differentiation.More importantly,enforced HNF3γexpression sensitized HCC cells to sorafenib-induced growth inhibition and cell apoptosis through transactivation of OATP1B1 and OATP1B3 expression,which are major membrane transporters for sorafenib uptake.Clinical investigation showed that patient-derived HCC xenografts with high HNF3γexpression exhibited a sorafenib response and patients with high HCC HNF3γlevels benefited from sorafenib therapy.Together,these results suggest that HNF3γplays an essential role in HCC differentiation and may serve as a therapeutic target and predictor of sorafenib benefit in patients.Tengfei Zhou Shichao Li Daimin Xiang Junyu Liu Wen Sun Xiuliang Cui Beifang Ning Xiao Li Zhuo Cheng Weiqi Jiang Cheng Zhang Xijun Liang Liang Li Xin Cheng Liu Hui Hongyang Wang Jin Ding 2020Signal Transduction and Targeted Therapy2020,5,1:4
4Ternary organic solar cells based on polymer donor,polymer acceptor and PCBM components显示文摘In this work,ternary organic solar cells(OSCs)combining a fullerene derivative PC71BM with a nonfullerene acceptor N2200-F blended with a polymer donor PM6 were reported.Compared with the binary systems,the highest power conversion efficiency(PCE)of 8.11%was achieved in ternary solar cells with 30 wt%N2200-F content,mainly due to the improved short-circuit current density(Jsc)and fill factor(FF).Further studies showed that the improved Jsc could attribute to the complementary abso rption of the two acceptors and the enhanced FF was originated from the higher hole mobility and the fine-tuned morphology in the ternary system.These results demonstrate that the combination of fullere ne and nonfullerene acceptors in ternary organic solar cells is a promising approach to achieve high-performance OSCs.Feng Liu Cheng Li Junyu Li Chao Wang Chengyi Xiao Yonggang Wu Weiwei Li 2020Chinese Chemical Letters2020,31,3:3

6HMGB1 from Astrocytes Promotes EAE by Influencing the Immune Cell Infiltration-Associated Functions of BMECs in Mice显示文摘High mobility group box 1(HMGB1)has been reported to play an important role in experimental autoimmune encephalomyelitis(EAE).Astrocytes are important components of neurovascular units and tightly appose the endothelial cells of microvessels by their perivascular endfeet and directly regulate the functions of the blood-brain barrier.Astrocytes express more HMGB1 during EAE while the exact roles of astrocytic HMGB1 in EAE have not been well elucidated.Here,using conditional-knockout mice,we found that astrocytic HMGB1 depletion decreased morbidity,delayed the onset time,and reduced the disease score and demyelination of EAE.Meanwhile,there were fewer immune cells,especially pathogenic T cells infiltration in the central nervous system of astrocytic HMGB1 conditional-knockout EAE mice,accompanied by up-regulated expression of the tight-junction protein Claudin5 and down-regulated expression of the cell adhesion molecules ICAM1 and VCAM1 in vivo.In vitro,HMGB1 released from astrocytes decreased Claudin5 while increased ICAM1 and VCAM1 expressed by brain microvascular endothelial cells(BMECs)through TLR4 or RAGE.Taken together,our results demonstrate that HMGB1 derived from astrocytes aggravates EAE by directly influencing the immune cell infiltration-associated functions of BMECs.Junyu Shi Yifan Xiao Na Zhang Mengya Jiao Xuhuan Tang Chan Dai Chenchen Wang Yong Xu Zheng Tan Feili Gong Fang Zheng 2022Neuroscience Bulletin2022,38,11:3

8Enlightenment from the COVID-19 Pandemic:The Roles of Environmental Factors in Future Public Health Emergency Response显示文摘The coronavirus disease 2019(COVID-19)pandemic is challenging the current public health emergency response systems(PHERSs)of many countries.Although environmental factors,such as those influencing the survival of viruses and their transmission between species including humans,play important roles in PHERSs,little attention has been given to these factors.This study describes and elucidates the roles of environmental factors in future PHERSs.To improve countries’capability to respond to public health emergencies associated with viral infections such as the COVID-19 pandemic,a number of environmental factors should be considered before,during,and after the responses to such emergencies.More specifically,to prevent pandemic outbreaks,we should strengthen environmental and wildlife protection,conduct detailed viral surveillance in animals and hotspots,and improve early-warning systems.During the pandemic,we must study the impacts of environmental factors on viral behaviors,develop control measures to minimize secondary environmental risks,and conduct timely assessments of viral risks and secondary environmental effects with a view to reducing the impacts of the pandemic on human health and on ecosystems.After the pandemic,we should further strengthen surveillance for viruses and the prevention of viral spread,maintain control measures for minimizing secondary environmental risks,develop our capability to scientifically predict pandemics and resurgences,and prepare for the next unexpected resurgence.Meanwhile,we should restore the normal life and production of the public based on the“One Health”concept,that views global human and environmental health as inextricably linked.Our recommendations are essential for improving nations’capability to respond to global public health emergencies.Xiaolei Wang Fengchang Wu Xiaoli Zhao Xiao Zhang Junyu Wang Lin Niu Weigang Liang Kenneth Mei Yee Leung John P.Giesy 2022Engineering2022,8,1:2

10Structural insights into secretory immunoglobulin A and its interaction with a pneumococcal adhesin显示文摘Secretory Immunoglobulin A(SIgA)is the most abundant antibody at the mucosal surface.It possesses two additional subunits besides IgA:the joining chain(J-chain)and secretory component(SC).SC is the ectodomain of the polymeric immunoglobulin receptor(pIgR),which functions to transport IgA to the mucosa.How the J-chain and pIgR/SC facilitate the assembly and secretion of SIgA remains incompletely understood.Furthermore,during the infection of Streptococcus pneumoniae,the pneumococcal adhesin SpsA hijacks pIgR/SC and SIgA to gain entry to human cells and evade host defense.How SpsA targets pIgR/SC and SIgA also remains elusive.Here we report a cryo-electron microscopy structure of the Fc region of IgA1(Fcα)in complex with the J-chain and SC(Fcα-J-SC),which reveals the organization principle of SIgA.We also present a structure of Fcα-J-SC complexed with SpsA,which uncovers the specific interactions between SpsA and human pIgR/SC.These results advance the molecular understanding of SIgA and shed light on S.pneumoniae pathogenesis.Yuxin Wang Guopeng Wang Yaxin Li Qinyu Zhu Hao Shen Ning Gao Junyu Xiao 2020Cell Research2020,30,7:1
11Genome-wide CRISPR activation screen identifies candidate receptors for SARS-CoV-2 entry显示文摘The outbreak of coronavirus disease 2019(COVID-19) caused by SARS-CoV-2 has created a global health crisis. SARS-CoV-2 infects varieties of tissues where the known receptor ACE2 is low or almost absent, suggesting the existence of alternative viral entry pathways. Here, we performed a genome-wide barcoded-CRISPRa screen to identify novel host factors that enable SARS-CoV-2 infection. Beyond known host proteins, i.e., ACE2, TMPRSS2, and NRP1, we identified multiple host components,among which LDLRAD3, TMEM30A, and CLEC4G were confirmed as functional receptors for SARS-CoV-2. All these membrane proteins bind directly to spike’s N-terminal domain(NTD). Their essential and physiological roles have been confirmed in either neuron or liver cells. In particular, LDLRAD3 and CLEC4G mediate SARS-CoV-2 entry and infection in an ACE2-independent fashion. 12Algal community structure prediction by machine learning显示文摘The algal community structure is vital for aquatic management.However,the complicated environmental and biological processes make modeling challenging.To cope with this difficulty,we investigated using random forests(RF)to predict phytoplankton community shifting based on multi-source environmental factors(including physicochemical,hydrological,and meteorological variables).The RF models robustly predicted the algal communities composed by 13 major classes(Bray-Curtis dissimilarity=9.2±7.0%,validation NRMSE mostly<10%),with accurate simulations to the total biomass(validation R^(2)>0.74)in Norway's largest lake,Lake Mjosa.The importance analysis showed that the hydro-meteorological variables(Standardized MSE and Node Purity mostly>0.5)were the most influential factors in regulating the phytoplankton.Furthermore,an in-depth ecological interpretation uncovered the interactive stress-response effect on the algal community learned by the RF models.The interpretation results disclosed that the environmental drivers(i.e.,temperature,lake inflow,and nutrients)can jointly pose strong influence on the algal community shifts.This study highlighted the power of machine learning in predicting complex algal community structures and provided insights into the model interpretability.Muyuan Liu Yuzhou Huang Jing Hu Junyu He Xi Xiao 2023Environmental Science and Ecotechnology2023,,2:0

19m6A RNA methylation-mediated HNF3γ reduction rendershepatocellular carcinoma dedifferentiation and sorafenibresistance显示文摘Hepatocyte nuclear factor 3γ(HNF3γ)is a hepatocyte nuclear factor,but its role and clinical significance in hepatocellular carcinoma(HCC)remain unclear.Herein,we report that HNF3γexpression is downregulated in patient HCC and inversely correlated with HCC malignancy and patient survival.Moreover,our data suggested that the HNF3γreduction in HCC could be mediated by METTL14-dependent m6A methylation of HNF3γmRNA.HNF3γexpression was increased during hepatic differentiation and decreased in dedifferentiated HCC cells.Interestingly,HNF3γdelivery promoted differentiation of not only HCC cells but also liver CSCs,which led to suppression of HCC growth.Mechanistic analysis suggested an HNF3γ-centered regulatory network that includes essential liver differentiation-associated transcription factors and functional molecules,which could synergistically facilitate HCC cell differentiation.More importantly,enforced HNF3γexpression sensitized HCC cells to sorafenib-induced growth inhibition and cell apoptosis through transactivation of OATP1B1 and OATP1B3 expression,which are major membrane transporters for sorafenib uptake.Clinical investigation showed that patient-derived HCC xenografts with high HNF3γexpression exhibited a sorafenib response and patients with high HCC HNF3γlevels benefited from sorafenib therapy.Together,these results suggest that HNF3γplays an essential role in HCC differentiation and may serve as a therapeutic target and predictor of sorafenib benefit in patients.Tengfei Zhou Shichao Li Daimin Xiang Junyu Liu Wen Sun Xiuliang Cui Beifang Ning Xiao Li Zhuo Cheng Weiqi Jiang Cheng Zhang Xijun Liang Liang Li Xin Cheng Liu Hui Hongyang Wang Jin Ding 2021Signal Transduction and Targeted Therapy2021,6,1:0
