| 1 | Abrogation of USP7 is an alternative strategy to downregulate PD-L1 and sensitize gastric cancer cells to T cells killing显示文摘Targeting immune checkpoints such as programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for treating melanoma,gastric cancer(GC)and bladder cancer with clinical benefit.Nevertheless,many patients failed to respond to anti-PD-1/PD-L1 treatment,so it is necessary to seek an alternative strategy for traditional PD-1/PD-L1 targeting immunotherapy.Here with the data from The Cancer Genome Atlas(TCGA)and our in-house tissue library,PD-L1 expression was found to be positively correlated with the expression of ubiquitin-specific processing protease 7(USP7)in GC.Furthermore,USP7 directly interacted with PD-L1 in order to stabilize it,Gastric cancer;Immunosuppression;Cancer biologywhile abrogation of USP7 attenuated PD-L1/PD-1 interaction and sensitized cancer cells to T cell killing in vitro and in vivo.Besides,USP7 inhibitor suppressed GC cells proliferation by stabilizing P53 in vitro and in vivo.Collectively,our findings indicate that in addition to inhibiting cancer cells proliferation,USP7 inhibitor can also downregulate PD-L1 expression to enhance anti-tumor immune response simultaneously.Hence,these data posit USP7 inhibitor as an anti-proliferation agent as well as a novel therapeutic agent in PD-L1/PD-1 blockade strategy that can promote the immune response of the tumor. | Zhiru Wang Wenting Kang Ouwen Li Fengyu Qi Junwei Wang Yinghua You Pengxing He Zhenhe Suo Yichao Zheng Hong-Min Liu | 2021 | Acta Pharmaceutica Sinica B2021,11,3: | 5 |
| 2 | Ripple-associated high-firing interneurons in the hippocampal CA1 region显示文摘By simultaneously recording the activity of individual neurons and field potentials in freely behaving mice, we found two types of interneurons firing at high frequency in the hippocampal CA1 region, which had high correlations with characteristic sharp wave-associated ripple oscillations (100―250 Hz) during slow-wave sleep. The firing of these two types of interneurons highly synchronized with ripple oscillations during slow-wave sleep, with strongly increased firing rates corresponding to individual ripple episodes. Interneuron type I had at most one spike in each sub-ripple cycle of ripple episodes and the peak firing rate was 310±33.17 Hz. Interneuron type II had one or two spikes in each sub-ripple cycle and the peak firing rate was 410±47.61 Hz. During active exploration, their firing was phase locked to theta oscillations with the highest probability at the trough of theta wave. Both two types of interneurons increased transiently their firing rates responding to the startling shake stimuli. | 3 | Numerical Analysis on Nanoparticles-laden Gas Film Thrust Bearing显示文摘Nanoparticles can be taken as additives and added into various fluids to improve their lubricating performances. At present, researches in this area are mainly concentrated on the improvement effects of nanoparticles on the lubricating performances of liquid such as oil and water. Nanoparticles will also affect gas lubrication, but few related studies have been reported. Nanoparticles-laden gas film (NLGF) is formed when adding nanoparticles into gas bearing. Then, the lubricating performances of gas bearing including pressure distribution and load-carrying capacity will change. The variations of pressure distribution and load-carrying capacity in nanoparticles-laden gas film thrust bearing are investigated by numerical method. Taking account of the compressibility of gas and the interactions between gas and nanoparticles, a computational fluid dynamics model based on Navier-Stokes equations is applied to simulate the NLGF flow. The effects of inlet nanoparticles volume fraction and orifice radius on film pressure distribution and load-carrying capacity of the NLGF are calculated. The numerical calculation results show that both of the film land pressure and the maximum film pressure both increase when the nanoparticles are added into gas bearing, and the film pressures increase with the rising of the inlet nanoparticles volume fraction. The nanoparticles have an enhancement effect on load-carrying capacity of the studied bearing, and the enhancement effect becomes greater as the film thickness decrease. Therefore, nanoparticles can effectively improve the lubricating performance of gas bearing. | 4 | GABAergic Interneurons are Required for Generation of Slow CA1 Oscillation in Rat Hippocampus显示文摘Neuronal oscillations are fundamental to hippocampal function. It has been shown that GABAergic interneurons make an important contribution to hippocampal oscillations, but the underlying mechanism is not well understood. Here, using whole-cell recording in the complete hippocampal formation isolated from rats at postnatal days 14–18, we showed that GABA_Areceptormediated activity enhanced the generation of slow CA1 oscillations. In vitro, slow oscillations(0.5–1.5 Hz) were generated in CA1 neurons, and they consisted primarily of excitatory rather than inhibitory membrane-potential changes. These oscillations were greatly reduced by blocking GABA_Areceptor-mediated activity with bicuculline and were enhanced by increasing such activity with midazolam, suggesting that interneurons are required for oscillation generation. Consistently, CA1 fast-spiking interneurons were found to generate action potentials usually preceding those in CA1 pyramidal cells. | 5 | Simulation of Evapotranspiration Based on BEPS-TerrainLab V2.0 from 1990 to 2018 in the Dajiuhu Basin显示文摘Accurate estimations of evapotranspiration(ET)are essential for understanding land-atmosphere coupling and atmosphere-underlying surface energy and water vapor exchanges.Based on input data processing,this paper simulates the temporal and spatial variation of ET in the Dajiuhu Basin from 1990 to 2018 using the BEPS-Terrain Lab V2.0 model.Compared with the ET measured by an eddy covariance(EC)tower,the model explained 80.1%of the ET variation.From 1990 to 2018,the average annual ET in the Dajiuhu Basin was 1262.7 mm/yr indicating a downward trend(–27.12 mm/yr).In 2005,a sudden change point was observed based on the Mann-Kendall(MK)test and 3-year moving t-test.Around 2005,the downward trend in ET slowed and the proportional trend of ET to precipitation changed from upward trend to downward trend.Regarding spatial distribution,the ET in the basin’s central part was smaller than that in the basin’s surrounding area,the ET of the southern slope was higher than that of the northern slope,and the decrease in the ET rate on the sunny side was lower than that on the shady side.ET decreased as the elevation increased,with the fastest decrease observed between 2184 and 2384 m.For different landcover types,the average ET exhibited the following order:deciduous forest>mixed forest>wetland>grass>agriculture land.Decreased solar radiation is the main reason for the decreased ET in the Dajiuhu Basin,followed by increased wind speed and relative humidity,which together contribute 83.9%to the ET trend.This paper provides a theoretical basis for the study of ET changes and the mechanism of ET and provides a decision-making reference for water resource management in the Dajiuhu Basin and even the South-to-North Water Transfer Project. | WU Zongfan ZHANG Lihua LIU Dandan ZHANG Kang ZHU Zhiru FU Yasheng MA Yongming | 2020 | Chinese Geographical Science2020,30,6: | 2 |
| 6 | Numerical Simulation of Carbon Concentration Profiles in Case Layer of Steel 20 RE-Carburized in Multipurpose Furnace with Drip-Feed Atmosphere显示文摘The carburizing process has been successfully carried out in multipurpose furnace employing the drip-feed atmosphere with the addition of rare earth (RE). The carbon transfer coefficients (BRE) and mean diffusion coefficient (DRE) were calculated respectively based on the kinetics of the specimens weigh gain per unit area and the carburized layer growth. The results show that the BRE and DRE are all increased on basis of the incorporation of rare earth. In addition, the mathematical models of the carbon concentration profile under different carburizing conditions and the carbon diffusion coefficient DSRE (at x=0 for carburizing time t>0) have been deduced using the profile simulation method. | 7 | Effects of adding sodium dichloroacetate to low-protein diets on nitrogen balance and amino acid metabolism in the portaldrained viscera and liver of pigs显示文摘Background:Identifying regulatory measures to promote glucose oxidative metabolism while simultaneously reducing amino acid oxidative metabolism is one of the foremost challenges in formulating low-protein(LP)diets designed to reduce the excretion of nitrogen-containing substances known to be potential pollutants.In this study,we investigated the effects of adding sodium dichloroacetate(DCA)to a LP diet on nitrogen balance and amino acid metabolism in the portal-drained viscera(PDV)and liver of pigs.To measure nitrogen balance,18 barrows(40±1.0 kg)were fed one of three diets(n=6 per group):18%crude protein(CP,control),13.5%CP(LP),and 13.5%CP+100 mg DCA/kg dry matter(LP-DCA).To measure amino acid metabolism in the PDV and liver,15 barrows(40±1.0 kg)were randomly assigned to one of the three diets(n=5 per group).Four essential amino acids(Lys,Met,Thr,and Trp)were added to the LP diets such that these had amino acid levels comparable to those of the control diet.Results:The LP-DCA diet reduced nitrogen excretion in pigs relative to that of pigs fed the control diet(P<0.05),without any negative effects on nitrogen retention(P>0.05).There were no differences between the control and LP-DCA groups with respect to amino acid supply to the liver and extra-hepatic tissues in pigs(P>0.05).The net release of ammonia into the portal vein and production rate of urea in the liver of pigs fed the LP-DCA diet was reduced relative to that of pigs fed the control and LP diets(P<0.05).Conclusion:The results indicated that addition of DCA to a LP diet can efficiently reduce nitrogen excretion in pigs and maximize the supply of amino acids to the liver and extra-hepatic tissues. | Weizhong Sun Yunxia Li Zhiru Tang Huiyuan Chen Ke Wan Rui An Liuting Wu Zhihong Sun | 2020 | Journal of Animal Science and Biotechnology2020,11,3: | 2 |
