1Gene expression of vascular endothelial growth factor A and hypoxic adaptation in Tibetan pig显示文摘Background: Vascular endothelial growth factor A(VEGFA) can induce endothelial cell proliferation, promote cell migration, and inhibit apoptosis. These processes play key roles in physiological blood vessel formation and pathological angiogenesis.Methods: In this study, we examined VEGFA gene expression in the heart, liver, and kidney of Tibetan pigs(TP), Yorkshire pigs that migrated to high altitudes(YH), and Yorkshire pigs that lived at low altitudes(YL).We used PCR and Sanger sequencing to screen for single nucleotide polymorphisms(SNPs) in 5'-flanking DNA and exons of the VEGFA gene. Quantitative real-time PCR and western blots were used to measure expression levels and PCR products were sequenced.Results: Results showed that the VEGFA m RNA and protein expression in heart, liver and kidney of TP was higher than that in YH and YL. In addition, the m RNA sequence of the pig VEGFA gene was conserved among pig breeds, and only five SNPs were found in the 5'-flanking region of the VEGFA gene, the allele frequency distributions of the 5 SNPs were not significantly different between the TP, Yorkshire(YL), and Diannan small-ear(DN) pig populations.Conclusion: In conclusion, the Tibetan pig showed high levels of VEGFA gene expression in several hypoxic tissues, which suggests that the VEGFA gene may play a major functional role in hypoxic adaptation.Bo Zhang Yangzong Qiangba Peng Shang Yunfeng Lu Yuzeng Yang Zhixiu Wang Hao Zhang 2016Journal of Animal Science and Biotechnology2016,7,4:4
2Different n-6/n-3 polyunsaturated fatty acid ratios affect postprandial metabolism in normal and hypertriglyceridemic rats显示文摘Postprandial metabolism plays major roles in many pathological conditions.The n-6/n-3 polyunsaturated fatty acid(PUFA)ratio is closely related to various physiological disorders.This study aimed to investigate the effects of high fat meals with different n-6/n-3 PUFA ratios on postprandial metabolism in normal control(NC)and hypertriglyceridemia(HTG)rats.The postprandial response of triglyceride(TG)in HTG groups was higher than that in NC groups after different n-6/n-3 PUFA ratio meals.The HTG groups showed higher postprandial total cholesterol(TC)responses than NC groups after 1:1 and 20:1 ratio meals.The 5:1 n-6/n-3 PUFA ratio elicited lower postprandial responses of tumor necrosis factorα(TNF-α)than 1:1 and 10:1 ratios in HTG groups.The postprandial malondialdehyde(MDA)response was lower after a 5:1 n-6/n-3 PUFA ratio meal than 1:1 and 20:1 ratio meals in HTG groups.The 1:1 ratio resulted in a lower postprandial reactive oxygen species(ROS)level than 5:1 and 10:1 n-6/n-3 PUFA ratios in NC groups.The results showed that a low n-6/n-3 PUFA ratio improved postprandial dysmetabolism induced by a high fat meal in NC and HTG rats.A high n-6/n-3 PUFA ratio increased the difference in postprandial metabolism between NC and HTG rats.Ligang Yang Chao Yang Zhixiu Song Min Wan Hui Xia Xian Yang Dengfeng Xu Da Pan Hechun Liu Shaokang Wang Guiju Sun 2023Food Science and Human Wellness2023,12,4:1
3The First Crested Duck Genome Reveals Clues to Genetic Compensation and Crest Cushion Formation显示文摘The Chinese crested(CC)duck is a unique indigenous waterfowl breed,which has a crest cushion that affects its survival rate.Therefore,the CC duck is an ideal model to investigate the genetic compensation response to maintain genetic stability.In the present study,we first generated a chromosome-level genome of CC ducks.Comparative genomics revealed that genes related to tissue repair,immune function,and tumors were under strong positive selection,indicating that these adaptive changes might enhance cancer resistance and immune response to maintain the genetic stability of CC ducks.We also assembled a Chinese spot-billed(Csp-b)duck genome,and detected the structural variations(SVs)in the genome assemblies of three ducks(i.e.,CC duck,Csp-b duck,and Peking duck).Functional analysis revealed that several SVs were related to the immune system of CC ducks,further strongly suggesting that genetic compensation in the anti-tumor and immune systems supports the survival of CC ducks.Moreover,we confirmed that the CC duck originated from the mallard ducks.Finally,we revealed the physiological and genetic basis of crest traits and identified a causative mutation in TAS2R40 that leads to crest formation.Overall,the findings of this study provide new insights into the role of genetic compensation in adaptive evolution.Guobin Chang Xiaoya Yuan Qixin Guo Hao Bai Xiaofang Cao Meng Liu Zhixiu Wang Bichun Li Shasha Wang Yong Jiang Zhiquan Wang Yang Zhang Qi Xu Qianqian Song Rui Pan Lingling Qiu Tiantian Gu Xinsheng Wu Yulin Bi Zhengfeng Cao Yu Zhang Yang Chen Hong Li Jianfeng Liu Wangcheng Dai Guohong Chen 2023Genomics, Proteomics & Bioinformatics2023,21,3:0

5Insight into the influence of ether and ester electrolytes on the sodium-ion transportation kinetics for hard carbon显示文摘The electrochemical performance of hard carbon(HC)materials is closely related to the electrolyte used in the sodium ion batteries(SIBs).Conventional electrolytes carbonate(EC)demonstrates low initial Columbic efficiency(ICE)and poor rate performance,which is one of the main bottlenecks that limits the practical application of HCs.Ether electrolyte(diglyme)was reported to improve the rate performance of HCs.Nevertheless,the underlying mechanism for the excellent rate capability is still lack of in-depth study.In this work,the differences of sodium-ion diffusion between ether and carbonate-base electrolytes in HCs are analyzed layer by layer.Firstly,when sodium-ions are diffused in electrolyte,the diffusion coefficient of sodium-ion in ether electrolyte is about 2.5 times higher than that in ester electrolytes by molecular dynamics(MD)simulation and experimental characterization.Furthermore,when the solvated sodium-ions are diffused into the solid electrolyte interphase(SEI)interface and the HCs material,the enhanced charge transfer kinetics(thin SEI layer(4.6 vs.12 nm)and low RSEI(1.5 vs.24Ω))at the SEI combined with low desolvation energy(0.248 eV)are responsible for high-rate performance and good cycling stability of HC in ether electrolyte.Therefore,high diffusion coefficient,low desolvation energy,and good interface are the intrinsic reasons for enhanced rate performance in ether electrolyte,which also has guiding significance for the design of other high-rate electrolytes.Xiuping Yin Zhaomin Wang Yang Liu Zhixiu Lu Hongli Long Tao Liu Jiujun Zhang Yufeng Zhao 2023Nano Research2023,16,8:0
6Digital gene expression profiling analysis of A549 cells cultured with PM10 in moxa smoke显示文摘Background:Moxibustion is a traditional Chinese medicine therapy to cure diseases by fumigating meridians or affected parts via burning of moxa floss.Moxa smoke(MS)is one of the key factors in moxibustion.In this study,we adopted digital gene expression profiling,a next-generation gene sequencing technology,to investigate the effect of MS,inhalable particulate matter(PM10),on human lung adenocarcinoma A549 cells.Methods:The effects of MS PM10 on A549 cells,over different treatment durations were investigated in different groups:the 4-h group(4-h MS group and 4-h control group)and the 20-h group(20-h MS group and 20-h control group).Samples collected from the four groups were stored at80C for subsequent digital gene expression analysis.The differentially expressed genes(DEGs),identified after PM10 treatment,were screened,and their expression patterns analyzed by cluster analysis,Gene Ontology term enrichment,and Kyoto Encyclopedia of Genes and Genomes pathway analysis.Results:Compared with two control groups,1109 DEGs were identified after 4 h of MS intervention and 3565 DEGs were found after 20 h of MS intervention,respectively.Compared with that after 4-h intervention,2149 DEGs were identified after 20-h intervention.Cluster analysis demonstrated that PM10 can significantly inhibit cell cycle process with the prolongation of intervention time.Significant pathway enrichment analysis showed that MS PM10 can inhibit A549 cell cycle process at all phases.When MS PM10 exposure time prolongs,the inhibitory effect on cell cycle process becomes more obvious.Conclusion:MS PM10 has many biological activities,and may cause differential expression of genes involved in various biological processes.Nevertheless,further research on MS is warranted for better understanding of the mechanistic details.Xin Hui Ping Liu Li Han Chang Huang Zhihua Yang Maoxiang Zhu Bicheng Yang Ruoxi Li Zhixiu Lin Baixiao Zhao 2020Journal of Traditional Chinese Medical Sciences2020,7,4:0
