1LSD1 inhibition suppresses the growth of clear cell renal cell carcinoma via upregulating P21signaling显示文摘Histone lysine-specific demethylase 1(LSD1) has been implicated in the disease progression of several types of solid tumors. This study provides the first evidence showing that LSD1 overexpression occurred in 62.6%(224/358) of clear cell renal cell carcinomas(ccRCC). LSD1 expression was associated with the progression of ccRCC, as indicated by TNM stage(P ? 0.006), especially tumor stage(P ? 0.017) and lymph node metastasis(P ? 0.030). High LSD1 expression proved to be an independent prognostic factor for poor overall survival(Po0.001) and recurrence-free survival(Po0.001) of ccRCC patients. We further show that LSD1 inhibition by siRNA knockdown or using the small molecule inhibitor SP2509 suppressed the growth of ccRCC in vitro and in vivo. Mechanistically, inhibition of LSD1 decreased the H3 K4 demethylation at the CDKN1 A gene promoter, which was associated with P21 upregulation and cell cycle arrest at G1/S in ccRCC cells. 2Expression of TRAIL and its receptors in primary hepatic carcinoma and apoptosis-inducing effect of HrsTRAIL on hepatoma cell line HepG2显示文摘Objective: To investigate the expression of tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) and its receptors in primary hepatic carcinoma(PHC) and the apoptosis-inducing effect on hepatoma cell line HepG2. Methods: TRAIL and its receptors were detected by semiquantitive RT-PCR in 30 PHC and para-carcinoma tissues and two hepatoma cell lines of HepG2 and SMMC-7721. HepG2 cells were treated with human recombinant soluble TRAIL protein (HrsTRAIL) and then the viability of HepG2 cells was measured by microculture tetrazolium dye(MTT) assay and apoptosis index was demonstrated by fluorescence-activated cell sorting(FACS). Results:TRAIL and its receptors were detectable in all PHC and para-carcinoma tissues and hepatoma cell line HepG2. TRAIL, death receptor 4(DR4), DR5, and decoy receptor 2(DcR2) but not DcR1 were detectable in hepatoma cell line SMMC-7721. The expression patterns of TRAIL receptors in HepG2 were quite similar to PHC specimens. The semiquantitive results showed that the expression level of TRAIL and DcR were lower but DR was higher in hepatoma tissues than in para-carcinoma tissues. In PHC tissues, the expressions of DR were higher than DcR, while there was no difference in para-carcinoma tissues. HrsTRAIL had potent antitumor activity in a time- and dose-dependent manner. After co-incubations of the HepG2 cells in the presence of HrsTRAIL at concentration 1 000 ng/ml for 24 hours, the viability of HepG2 cells decreased to 45% and the apoptosis index reached 51%. Conclusion:TRAIL and its receptors were expressed in both PHC tissues and para-carcinoma tissues but the expression levels were different. The lower expression of TRAIL in PHC tissues suggested that insufficient apoptosis occured in the development of PHC. High expression of DR in PHC tissues may be a self-defense mechanism and may afford a theory of HrsTRAIL therapy for PHC. 3The biogenesis and secretion of exosomes and multivesicular bodies(MVBs):Intercellular shuttles and implications in human diseases显示文摘Exosomes carry and transmit signaling molecules used for intercellular communication.The generation and secretion of exosomes is a multistep interlocking process that allows simultaneous control of multiple regulatory sites.Protein molecules,mainly RAB GTPases,cytoskeletal proteins and soluble N-ethylmaleimide-sensitive fusion attachment protein receptor(SNARE),are specifically regulated in response to pathological conditions such as altered cellular microenvironment,stimulation by pathogenic factors,or gene mutation.This interferes with the smooth functioning of endocytosis,translocation,degradation,docking and fusion processes,leading to changes in the secretion of exosomes.Large numbers of secreted exosomes are disseminated by the flow of body fluids and absorbed by the recipient cells.By transmitting characteristic functional proteins and genetic information produced under disease conditions,exosomes can change the physiological state of the recipient cells and their microenvironment.The microenvironment,in turn,affects the occurrence and development of disease.Therefore,this review will discuss the mechanism by which exosome secretion is regulated in cells following the formation of mature secretory multivesicular bodies(MVBs).The overall aim is to find ways to eliminate disease-derived exosomes at their source,thereby providing an important new basis for the clinical treatment of disease.Minxue Xu Jie Ji Dandan Jin Yue Wu Tong Wu Renjie Lin Shengze Zhu Feng Jiang Yifei Ji Baijun Bao Mei Li Weisong Xu Mingbing Xiao 2023Genes & Diseases2023,10,5:0
