3Experience of one single surgeon with the first 500 robot-assisted laparoscopic prostatectomy cases in China's Mainland显示文摘Objectives: To summarize the experience of the first 500 robot-assisted laparoscopicradical prostatectomy (RALP) cases by one surgeon and analyze the influencing factorsof functional and oncological outcomes.Methods: Between April 2012 and October 2017, 500 patients who underwent RALP wereincluded and divided sequentially into five equal groups. Patients’ preoperative, perioperativeand postoperative outcomes were analyzed and evaluated, and the Kruskal-Wallis test wasused to analyze and compare the effect of surgeon experience by case.Results: There is a statistically significant reduction in operative time, intraoperative estimatedblood loss and postoperative hospital stay time (all p<0.001) with the increased experience.The results show that experience was the most important influencing factor in bothoperative time and blood loss. Pelvic lymph node dissection (PLND) might increase the operativetime. The total positive surgical margin (PSM) rate was 21.8%. The PSM rate in pT3 tumorswas significantly higher than that in pT2 tumors (12.0% vs. 37.1%, p<0.001). The 5-yearbiochemical recurrence (BCR)-free rate was 70.8%. The results of Cox regression showed thatpreoperative prostate-specific antigen (PSA), postoperative Gleason score (GS), and pathologicT stage were independent risk factors for BCR.Conclusion: After approximately 200 cases, the surgeon reached a plateau for RALP, but theoutcomes could still improve after more cases. The surgeon’s experience was the most importantinfluencing factor for both operative time and blood loss. 5Microbial diversity in lake sediments detected by PCR-DGGE显示文摘In this study,PCR-denaturing gradient gel electrophoresis(DGGE)was applied to analyze the microbial communities in lake sediments from Lake Xuanwu,Lake Mochou in Nanjing and Lake Taihu in Wuxi.Sediment samples from seven locations in three lakes were collected and their genomic DNAs were extracted.The DNA yields of the sediments of Lake Xuanwu and Lake Mochou were high(10 mg/g),while that of sediments in Lake Taihu was relatively low.After DNA purification,the 16S rDNA genes(V3 to V5 region)were amplified and the amplified DNA fragments were separated by parallel DGGE.The DGGE profiles showed that there were five common bands in all the lake sediment samples indicating that there were similarities among the populations of microorganisms in all the lake sediments.The DGGE profiles of Lake Xuanwu and Lake Mochou were similar and about 20 types of micro-organisms were identified in the sediment samples of both lakes.These results suggest that the sediment samples of these two city lakes(Xuanwu,Mochou)have similar microbial communities.However,the DGGE profiles of sediment samples in Lake Taihu were significantly different from these two lakes.Furthermore,the DGGE profiles of sediment samples in different locations in Lake Taihu were also different,suggesting that the microbial communities in Lake Taihu are more diversified than those in Lake Xuanwu and Lake Mochou.The differences in microbial diversity may be caused by the different environmental conditions,such as redox potential,pH,and the concentrations of organic matters.Seven major bands of 16S rDNA genes fragments from the DGGE profiles of sediment samples were further reamplified and sequenced.The results of sequencing analysis indicate that five sequences shared 99%-100%homology with known sequences(Bacillus and Brevibacillus,uncultured bacteria),while the other two sequences shared 93%-96%homology with known sequences(Acinetobacter,and Bacillus).The study shows that the PCR-DGGE technique combined with sequence analysis is a feasible and efficient method for the determination of microbial communities in sediment samples.Xinqing ZHAO Liuyan YANG Can CHEN Lin XIAO Lijuan JIANG Zhe MA Haowei ZHU Zhenyang YU Daqiang YIN 2008Frontiers in Biology2008,3,3:0

7Reaching consensus for membership dynamic in secret sharing and its application to cross-chain显示文摘The communication efficiency optimization,censorship resilience,and generation of shared randomness are inseparable from the threshold cryptography in the existing Byzantine Fault Tolerant(BFT)consensus.The membership in consensus in a blockchain scenario supports dynamic changes,which effectively prevents the corruption of consensus participants.Especially in cross-chain protocols,the dynamic access to different blockchains will inevitably bring about the demand for member dynamic.Most existing threshold cryptography schemes rely on redefined key shares,leading to a static set of secret sharing participants.In this paper,we propose a general approach to coupling blockchain consensus and dynamic secret sharing.The committee performs consensus confirmation of both dynamic secret sharing and transaction proposals.Our scheme facilitates threshold cryptography membership dynamic,thus underlying support for membership dynamic of threshold cryptographybased BFT consensus schemes.We instantiate a dynamic HotStuff consensus to demonstrate the effectiveness of the scheme.After the correctness and security proof,our scheme achieves the secrecy and integrity of the threshold key shares while ensuring consensus liveness and safety.Experimental results prove that our scheme obtains dynamic membership with negligible overhead.Yan Zhu Bingyu Li Zhenyang Ding Yang Yang Qianhong Wu Haibin Zheng 2023High-Confidence Computing2023,3,3:0
