| 1 | A method for the production and expedient screening of CRISPR/Cas9-mediated non-transgenic mutant plants显示文摘Developing CRISPR/Cas9-mediated non-transgenic mutants in asexually propagated perennial crop plants is challenging but highly desirable.Here,we report a highly useful method using an Agrobacterium-mediated transient CRISPR/Cas9 gene expression system to create non-transgenic mutant plants without the need for sexual segregation.We have also developed a rapid,cost-effective,and high-throughput mutant screening protocol based on Illumina sequencing followed by high-resolution melting(HRM)analysis.Using tetraploid tobacco as a model species and the phytoene desaturase(PDS)gene as a target,we successfully created and expediently identified mutant plants,which were verified as tetra-allelic mutants.We produced pds mutant shoots at a rate of 47.5%from tobacco leaf explants,without the use of antibiotic selection.Among these pds plants,17.2%were confirmed to be non-transgenic,for an overall non-transgenic mutation rate of 8.2%.Our method is reliable and effective in creating non-transgenic mutant plants without the need to segregate out transgenes through sexual reproduction.This method should be applicable to many economically important,heterozygous,perennial crop species that are more difficult to regenerate. | Longzheng Chen Wei Li Lorenzo Katin-Grazzini Jing Ding Xianbin Gu Yanjun Li Tingting Gu Ren Wang Xinchun Lin Ziniu Deng Richard J.McAvoy Frederick G.Gmitter Jr. Zhanao Deng Yunde Zhao Yi Li | 2018 | Horticulture Research2018,5,1: | 12 |

This is achieved by mounting a two-dimensional shock-shock interaction generator below the lifting surface. The shock-shock interaction theory in conjunction with a three dimensional correction and checked with computational fluid dynamics (CFD) is used to analyze the lift and drag forces as function of the geometrical parameters and inflow Mach number. | 4 | Generalized Kutta–Joukowski theorem for multi-vortex and multi-airfoil flow with vortex production——A general model显示文摘By using a special momentum approach and with the help of interchange between singularity velocity and induced flow velocity, we derive in a physical way explicit force formulas for twodimensional inviscid flow involving multiple bound and free vortices, multiple airfoils, and vortex production. These force formulas hold individually for each airfoil thus allowing for force decomposition, and the contributions to forces from singularities(such as bound and image vortices,sources, and doublets) and bodies out of an airfoil are related to their induced velocities at the locations of singularities inside this airfoil. The force contribution due to vortex production is related to the vortex production rate and the distance between each pair of vortices in production, thus frameindependent. The formulas are validated against a number of standard problems. | 5 | Citron C-05 inhibits both the penetration and colonization of Xanthomonas citri subsp.citri to achieve resistance to citrus canker disease显示文摘Citrus canker,caused by Xanthomonas citri subsp.citri(Xcc),is a serious bacterial disease that affects citrus production worldwide.Citron C-05(Citrus medica)is the only germplasm in the Citrus genus that has been identified to exhibit strong resistance to Xcc.However,it has not been determined when,where,and how Xcc is restricted in the tissues of Citron C-05 during the infection process.In the present study,we investigated the spatiotemporal growth dynamics of an eGFP-labeled virulent Xcc(eGFP-Xcc)strain in Citron C-05 along with five susceptible biotypes(i.e.,lemon,pummelo,sour orange,sweet orange,and ponkan mandarin)upon inoculation via the spraying or leaf infiltration of a bacterial suspension.The results from extensive confocal laser scanning microscopy analyses showed that while Xcc grew rapidly in plants of all five susceptible genotypes,Xcc was severely restricted in the epidermal and mesophyll cell layers of the leaves of Citron C-05 in the early stage of infection.Not surprisingly,resistance against Xcc in Citron C-05 was found to be associated with the production of reactive oxygen species and hypersensitive response-like cell death,as well as greater upregulation of several defense-related genes,including a pathogenesis-related gene(PR1)and a glutathione S-transferase gene(GST1),compared with sweet orange as a susceptible control.Taken together,our results not only provide further valuable details of the spatiotemporal dynamics of the host entry,propagation,and spread of Xcc in both resistant and susceptible citrus plants but also suggest that resistance to Xcc in Citron C-05 may be attributed to the activation of multiple defense mechanisms. | Hongyan Fu Mingming Zhao Jing Xu Limei Tan Jian Han Dazhi Li Meijun Wang Shunyuan Xiao Xianfeng Ma Ziniu Deng | 2020 | Horticulture Research2020,7,1: | 2 |
| 6 | Relationships between intensity of the Kuroshio current in the East China Sea and the East Asian winter monsoon显示文摘Based on satellite altimeter and reanalysis data,this paper studies the relationships between the intensity of the Kuroshio current in the East China Sea(ECS) and the East Asian winter monsoon(EAWM).The mechanisms of their possible interaction are also discussed.Results indicate that adjacent transects show consistent variations,and on an interannual timescale,when the EAWM is anomalously strong(weak),the downstream Kuroshio in the ECS is suppressed(enhanced) in the following year from February to April.This phenomenon can be attributed to both the dynamic effect(i.e.,Ekman transport) and the thermal effect of the EAWM.When the EAWM strengthens(weakens),the midstream and downstream Kuroshio in the ECS are also suppressed(intensified) during the following year from October to December.The mechanisms vary for these effects.The EAWM exerts its influence on the Kuroshio's intensity in the following year through the tropospheric biennial oscillation(TBO),and oceanic forcing is dominant during this time.The air-sea interaction is modulated by the relative strength of the EAWM and the Kuroshio in the ECS.The non-equivalence of spatial scales between the monsoon and the Kuroshio determines that their interactions are aided by processes with a smaller spatial scale,i.e.,local wind stress and heating at the sea surface. | YIN Ming LI Xin XIAO Ziniu LI Chongyin | 2018 | Acta Oceanologica Sinica2018,37,7: | 2 |
| 7 | Early embryonic transcriptomes of Zeugodacus tau provide insight into sex determination and differentiation genes显示文摘Zeugodacus tau(Walker)is an invasive pest.The sterile insect technique is an environment-friendly method for pest control.Understanding the mechanism of sex determination will contribute to improving efficiency of this technique.In this study,we identified the transformer(tra)gene in Z.tau.One female-specific and two male-specific isoforms of tra were found in Z.tau,and the male-specific splicing pattern of tra was found to occur 5 h after egg laying.We performed transcriptome sequencing at 1 h(E1),5 h(E5),and 9 h(E9)after egg laying and obtained high-quality transcriptome libraries of early embryo development.We identified 13297 and 11713 differentially expressed genes(DEGs)from E5 versus E1 and E9 versus E1 comparisons,respectively.To explore the potential functions of the DEGs during embryonic development,Gene Ontology,Clusters of Orthologous Groups of proteins,and Kyoto Encyclopedia of Genes and Genomes analyses were performed.Twenty-six genes potentially involved in sex determination or differentiation,including Maleness-on-the-Y(MoY),were identified in Z.tau.To verify the transcriptome results,15 genes were selected for quantitative real-time PCR validation.The results were consistent with the transcriptome sequencing results.Moreover,U2 small nuclear riboprotein auxiliary factor(U2AF-50),female lethal d(fl(2)d),and virilizer(vir)were highly expressed at E5,indicating that they may be related to the sex-specific splicing of tra.Further functional analysis is needed to confirm this speculation.Our data provide an insight into the mechanism underlying sex determination and differentiation in tephritid species. | Peipei Liu Wenping Zheng Jiao Qiao Ziniu Li Zhurong Deng Yimin Yuan Hongyu Zhang | 2022 | Insect Science2022,29,3: | 1 |

| 9 | Subsonic impulsively starting flow at a high angle of attack with shock wave and vortex interaction显示文摘Impulsively starting flow, by a sudden attainment of a large angle of attack, has been well studied for incompressible and supersonic flows, but less studied for subsonic flow. Recently,a preliminary numerical study for subsonic starting flow at a high angle of attack displays an advance of stall around a Mach number of 0.5, when compared to other Mach numbers. To see what happens in this special case, we conduct here in this paper a further study for this case, to display and analyze the full flow structures. We find that for a Mach number around 0.5, a local supersonic flow region repeatedly splits and merges, and a pair of left-going and right-going unsteady shock waves are embedded inside the leading edge vortex once it is sufficiently grown up and detached from the leading edge. The flow evolution during the formation of shock waves is displayed in detail. The reason for the formation of these shock waves is explained here using the Laval nozzle flow theory. | 10 | Tetrapioid induction of Crassostrea hongkongensis and C.sikamea by inhibiting the polar body 1 release in diploid fertilized eggs显示文摘The production of an all-triploid population by mating tetrapioid males with diploid females is the best and most fundamental method for the large-scale production of triploid oysters.Obtaining a stable tetrapioid population is essential for guaranteed production in industrialized triploid cultivation.C.hongkongensis and C.sikamea are important oyster breeding species in southern China,and have great economic value.However,there are not any published data on inducing tetrapioid C.hongkongensis or C.sikamea.Therefore,we investigated tetrapioid induction in these two oyster species by inhibiting the PB1 release in diploid fertilized eggs using Cytochalasin B(CB)under 31℃,15%o salinity.The results confirmed that the optimal tetrapioid induction conditions for C.hongkongensis were a CB concentration of 0.50 mg/L with induction starting at 9.0 min after fertilization,and stopping at 21.0 min after fertilization;the induction efficiency index reached 0.123 under these conditions.The optimal tetrapioid induction conditions for C.sikamea were a CB concentration of 0.50 mg/L,with induction starting at 7.5 min after fertilization and stopping at 18 min after fertilization;the induction efficiency index could be as high as 0.281 under these conditions.However,we confirmed that the tetrapioid rate decreased with larval growth,and no tetrapioids were detected in the juvenile period of either C.hongkongensis or C.sikamea.This may be attributed to the very low survival of the tetrapioid larvae induced by this method,especially as most tetrapioid larvae died during the first three days.In summary,it is simple to directly induce tetrapioid C.hongkongensis and C.sikamea larvae by inhibiting the PB1 release of diploid zygotes,but the low survival rate makes it challenging to obtain viable juvenile tetrapioids. | Yanping Qin Zohaib Noor Xingyou Li Yuehuan Zhang Ziniu Yu Haitao Ma Jun Li Yinyin Zhou Riguan Mo | 2021 | Marine Life Science & Technology2021,3,4: | 1 |

| 13 | Review of theoretical achievements for starting flow problem for all Mach numbers显示文摘The typical behavior of unsteady flow and force evolution in a number of applications,such as aero-elastics, gust-wing interaction, flapping flight and flight maneuvering, can be understood using the starting flow model. Starting flow model is obtained either by setting rapidly an angle of attack for a wing moving at constant speed, or by accelerating a wing to a constant speed while gaining an angle of attack. In the limiting case of impulsively starting flow, the wing is assumed to gain suddenly an angle of attack in an initially uniform flow. Theories have been developed for impulsively starting flow at small angle of attack long before and at large angle of attack only recently, especially for incompressible and supersonic flow. This paper intends to provide a state-of-art overview of the typical flow phenomena, force evolution characteristics and developed theories for impulsively starting flow at any angle of attack and for both lower speed flow(vortex dominated) and high speed flow(compressible wave dominated). | 14 | Interdecadal correlation of solar activity with Tibetan Plateau snow depth and winter atmospheric circulation in East Asia显示文摘Studies on the impact of solar activity on climate system are very important in understanding global climate change. Previous studies in this field were mostly focus on temperature, wind and geopotential height. In this paper, interdecadal correlations of solar activity with Winter Snow Depth Index(WSDI) over the Tibetan Plateau, Arctic Oscillation Index(AOI) and the East Asian Winter Monsoon Index(EAWMI) are detected respectively by using Solar Radio Flux(SRF), Total Solar Irradiance(TSI) and Solar Sunspot Number(SSN) data and statistical methods. Arctic Oscillation and East Asian winter monsoon are typical modes of the East Asian atmospheric circulation. Research results show that on interdecadal time scale over 11-year solar cycle, the sun modulated changes of winter snow depth over the Tibetan Plateau and East Asian atmospheric circulation. At the fourth lag year, the correlation coefficient of SRF and snow depth is 0.8013 at 0.05 significance level by Monte-Carlo test method. Our study also shows that winter snow depth over the Tibetan Plateau has significant lead and lag correlations with Arctic Oscillation and the East Asian winter monsoon on long time scale. With more snow in winter, the phase of Arctic Oscillation is positive, and East Asian winter monsoon is weak, while with less snow, the parameters are reversed. | 18 | Kutta-Joukowski force expression for viscous flow显示文摘The Kutta Joukowski(KJ) theorem, relating the lift of an airfoil to circulation, was widely accepted for predicting the lift of viscous high Reynolds number flow without separation. However, this theorem was only proved for inviscid flow and it is thus of academic importance to see whether there is a viscous equivalent of this theorem. For lower Reynolds number flow around objects of small size, it is difficult to measure the lift force directly and it is thus convenient to measure the velocity flow field solely and then, if possible, relate the lift to the circulation in a similar way as for the inviscid KJ theorem. The purpose of this paper is to discuss the relevant conditions under which a viscous equivalent of the KJ theorem exists that reduces to the inviscid KJ theorem for high Reynolds number viscous flow and remains correct for low Reynolds number steady flow. It has been shown that if the lift is expressed as a linear function of the circulation as in the classical KJ theorem, then the freestream velocity must be corrected by a component called mean deficit velocity resulting from the wake. This correction is small only when the Reynolds number is relatively large. Moreover, the circulation, defined along a loop containing the boundary layer and a part of the wake, is generally smaller than that based on inviscid flow assumption.
