1Laparoscopic total mesorectal excision of low rectal cancer with preservation of anal sphincter:A report of 82 cases显示文摘AIM: To assess the feasibility and efficacy of laparoscopic total mesorectal excision (LTME) of low rectal cancer with preservation of anal sphincter.METHODS: From June 2001 to June 2003, 82 patients with low rectal cancer underwent laparoscopic total mesorectal excision with preservation of anal sphincter. The lowest edge of tumors was below peritoneal reflection and 1.5-7 cm from the dentate line (1.5-5 cm in 48 cases, 5-7 cm in 34 cases).RESULTS: LTME with anal sphincter preservation was performed on 82 randomized patients with low rectal cancer, and 100 % sphincter preservation rate was achieved. There were 30 patients with laparoscopic low anterior resection (LLAR) at the level of the anastomosis below peritoneal reflection and 2 cm above from the dentate line; 27 patients with laparoscopic ultralow anterior resection (LULAR) at the level of anastomoses 2 cm below from the dentate line; and 25 patients with laparoscopic coloanal anastomoses (LC AA) at the level of the anastomoses at or below the dentate line. No defunctioning ileostomy was created in any case. The mean operating time was 120 minutes (ranged from 110-220 min), and the mean operative blood loss was 20 mL (ranged from 5-120 mL).Bowel function was restored and diet was resumed on day 1 or 2 after operation. The mean hospital stay was 8 days (ranged from 5-14). Postoperative analgesics were used in 45 patients. After surgery, 2 patients had urinary retention, one had anastomotic leakage, and another 2patients had local recurrence one year later. 2Roles of Fas signaling pathway in vitamin E succinateinduced apoptosis in human gastric cancer SGC-7901 cells显示文摘AIM: To investigate the roles of Fas signaling pathway in vitamin E succinate-induced apoptosis in human gastric cancer SGC-7901 cells.METHODS: Human gastric cancer SGC-7901 cells were treated with VES at 5, 10, 20 mg@L-1, succinic acid and vitamin E as vehicle control and condition media only as untreated (UT) control. Apoptotic morphology was observed by DAPI staining. Western blot analysis was applied to measure the expression of Fas, FADD and caspase-8 proteins. After the cells were transiently transfected with Fas and FADD antisense oligonucleotides, respectively, caspase-8 activity was determined by flurometric method.RESULTS: The morphologically apoptotic changes were observed after VES treatment by DAPI staining. 23.7 % and 89.6 % apoptosis occurred after 24 h and 48 h of 20 mg@L-1 VES treatment, respectively. The protein levels of Fas, FADD and caspase-8 were evidently increased in a dose-dependent manner after 24 h of VES treatment. The blockage of Fas by transfection with Fas antisense oligonucleotides obviously inhibited the expression of FADD protein. 5Meteorological control on CO_2 flux above broad-leaved Korean pine mixed forest in Changbai Mountains显示文摘The impacts of temperature, photosynthetic active radiation (PAR) and vapor pressure deficit (VPD) on CO2 flux above broad-leaved Korean pine mixed forest in the Changbai Mountains were studied based on eddy covariance and meteorological factors measurements.The results showed that, daytime CO2 flux was mainly controlled by PAR and they fit Michaelis-Menten equation. Meanwhile VPD also had an influence on the daytime flux. Drier air reduced the CO2 assimilation of the ecosystem, the drier the air, the more the reduction of the assimilation. And the forest was more sensitive to VPD in June than that in July and August. The respiration of the ecosystem was mainly controlled by soil temperature and they fit exponential equation. It was found that this relationship was also correlated with seasons; respiration from April to July was higher than that from August to November under the same temperature. Daily net carbon exchange of the ecosystem and the daily mean air temperature fit exponential equation. It was also found that seasonal trend of net carbon exchange was the result of comprehensive impacts of temperature and PAR and so on. These resulted in the biggest CO2 uptake in June and those in July and August were next. 6Establishment of a novel corneal endothelial cell line from domestic rabbit,Oryctolagus curiculus显示文摘To develop a rabbit corneal endothelial (RCE) cell line, in vitro culture of RCE cells was initiated from Oryctolagus curiculus corneas and a novel RCE cell line was established in this study. To initiate the primary culture of RCE cells, corneas from rabbit eyes were sliced and attached into glutin-coated wells with endothelial cell surface down. After being cultured at a time-gradient interval from 48 to 6 h, the corneal slices were detached and reattached into new wells, respectively. Cells in the wells containing only a pure population of RCE cells were collected and cultured in 20% FBS-DMEM/F12 medium con- taining chondroitin sulfate, ocular extract, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), carboxymethyl-chitosan, N-acetylglucosamine hydrochloride, glucosamine hydrochloride, culture medium of rabbit corneal stromal cells and oxidation-degradation products of chondroitin sul- fate at 37℃, 5% CO2. The cultured RCE cells, in quadrangle and polygonal shapes, proliferated to con- fluence 3 weeks later. During the subsequent subculture, the shape of RCE cells changed gradually from polygonal to more fibroblastic. A novel RCE cell line, growing at a steady rate, with a population doubling time of 53.8 h, has been established and subcultured to passage 67. Chromosome analysis showed that the RCE cells exhibited chromosomal aneuploidy with the modal chromosome number of 44. The results of immuno-cytochemical staining with neuron specific enolase (NSE) confirmed that the RCE cells were in neuroectodermal origin. Combined with the results of vascular endothelial growth factor (VEGF) treatment and endothelial cell morphology recovery, it can be concluded that the cell line established here is an RCE cell line. 7The study of neighboring nucleotide composition and transition/transversion bias显示文摘Base substitution is one of the raw fuels that produce genetic variation and drive evolu-tion. Recent studies have shown that the genome components affect mutation patterns to some extent. In order to infer the correlation between the Transition/Transversion ratio (Ts/Tv) and the number of immediately adjacent A&T nucleotides, we investigated 3611007 Oryza sativa SNPs (including 45462 coding SNPs, and 242811 intronic SNPs) and 32019 Arabidopsis SNPs. The results show that Ts/Tv is negatively correlated with the number of immediately adjacent A&T in O. sativa and Arabidopsis. We further calculated AT2 (the number of SNPs whose immediately adjacent nucleotides are either A or T) and AT0 (the number of SNPs whose immediately adjacent nucleotides are either C or G) for all 6 types of SNPs. C/G SNP of O. sativa and Arabidopsis has the highest AT2/AT0, which denotes C/G SNP may be influenced by the adjacent A&T nucleotides mostly. 8Grain Size and Wettability of TiO_2/SiO_2 Photocatalytic Composite Thin Filing显示文摘The uniform transparent TiO2/SiO2 photocatalytic composite thin films are prepared by sol-gel method on the soda lime glass substrates, and characterized by UV-visible spectroscopy, X-ray diffraction (XRD), transmission electron microscopy (TEM), BET surface area, FTIR spectroscopy and X-ray photoelectron spectroscopy (XPS). It was found that the addition of SiO2 to TiO2 thin films can suppress the grain growth of TiO2 crystal, increase the hydroxyl content on the surface of TiO2 films, lower the contact angle for water on TiO2 films and enhance the hydrophilic property of TiO2 films. 9Aoyitake plagiogranite in western Tarim Block, NW China: Age, geochemistry, petrogenesis and its tectonic implications显示文摘SHRIMPP U-Pb zircon age and geochemical and Nd isotopic data are reported for the Aoyitake plagiogranite in western Tarim Block, NW China. The plagiogranite intruded the Middle Pro- terozoic and Lower Carboniferous with an exposure area of ca. 60 km2 and crystallized at 330.7±4.8 Ma. Rock types mainly include tonalite, trondhjemite and minor amounts of diorite and quartz-diorite. Feldspars in the rocks are dominated by oligoclase-andesine, and minor perthite observed locally. The granites are sodic with Na/K ratios (molar) between 4 and 87. Total REE (50-220 ppm) show a clear positive correlation with SiO2. There is no LRRE/HREE fractionation (LaN/YbN=0.5-1.5), me- dium negative Eu anomalies (δ Eu=0.3-0.6), high Y content and low Sr/Y ratio (~1.0). These granites exhibit relatively juvenile Nd T2DM model ages of 470 to 580 Ma and positive εNd(331 Ma) values of 6.23 to 7.65. The aforementioned characteristics are similar to those of ocean island or ocean ridge plagiogranites. However, the regional geology, especially its scale, precludes that the plagiogranite pluton was derived directly from fractionational crystallization of mantle-derived basaltic magma. We interpreted that the primary magma of the pluton might be tonalitic in composition generated by ca. 50% partial melting of the juvenile basaltic crust. The primary magma experienced intensive frac- tionational crystallization, and intruded into the middle to upper crusts to form the granite pluton. 10The high-resolution climate recorded in the δ^(18)O of Porites lutea from the Nansha Islands of China显示文摘A Porites lutea core from Yongshu Reef of Nansha Islands covering 50 years growth history was ana-ly/ed for oxygen isotopic composition with monthly and seasonally resolution. The calibration of the δ18O with the instrumental temperature indicated that the coral δ18O is a good indicator for sea surface temperature (SST) and air temperature (t). It can be used to reconstruct the SST and air temperature of the Yongshu Reef sea area. In addition, the coral δ18O provides signatures for the intensity of the East Asia monsoon and it is a record for the activities of El Nino events. With the calibrated SST and air temperature formulas, the most recent fifty years SST and air temperature were reconstructed based on the coral δ18O, thus back up the understanding of the climate of Nansha Islands to 1950. far beyond the limit of the instrumental recording since September 1988. 11Study on DNA Cytosine Methylation of Cotton (Gossypium hirsutum L.) 12A NUMERICAL STUDY ON THE PATH AND ORIGIN OF THE YELLOW SEA WARM CURRENT (YSWC)显示文摘A three dimensional baroclinic model (POM) is employed to study the path, orgin and strength of the Yellow Sea Warm Current (YSWC). The model produced results are compared with the observed temperature and salinity fields, the trajectories of drifters and moored current meter data. The results show: (1) It is indicated for the first time that the YSWC in winter orignates from the middle sector (28°N, 126°E), the truning directional sector (30°N, 127°E) of the Kuroshio in the East China Sea (ECS) and the separated sector of the Tsushima Warm Current (TsWC) at (31°N, 128°E), while in summer mainly originates from the Taiwan Warm Current (TaWC) and partly from the separated sector of the TsWC. This path is consistent with the main distribution feature of the quasi synoptic observed temperature and salinity. The model produced anticyclonic gyre (28°N, 125.5°E) branched from the middle sector of the ECS Kuroshio agrees with the observed results of the satellite drifters. (2) The model produced YSWC is located between 50~60m isobath on the West Side of the Yellow Sea Trough (YST), which is consistent with the location of the Warm and Saline tongue. The model produced cyclonic gyre centering at (36°N, 124.5°E) coincides with the observed results in Feb. 1997. The traditional view indicates that the Northern Jiangsu Coastal Current (NJCC) turns eastward at (32.5°N, 122°E). However, we think that the NJCC goes southward and only the coastal water invades near (32.5°N, 122°E). The model produced path is consistent with the warm tongue in the range of 34° 35°N, 122.5° 123.5°E in 30m layer. (3) The model produced YSWC is strong in winter and weak in summer, which coincides with the observations. The northward volume transport along 35°N section in winter and summer are 0.72sv and 0.38sv, respectively, with an annual flux of about 1.54·10 13 m 3. 13Differential Gene Expression Between Hybrids and Their Parents During the Four Crucial Stages of Cotton Growth and Development显示文摘The study aims to clarify the differential gene expression between cotton hybrids and their parents in order to better understand the molecular basis of cotton heterosis. The research focused on cotton heterotic and lower heterotic hybrids and their parents during the four crucial stages, which were analyzed using a differential display technique. The results indicated that there were both quantitative and qualitative differences in gene expression amongst them. The quantitative differences include over- and under-expression of parental genes and the dominant expression of highly-expressed parental genes in hybrids. In contrast, the qualitative differences are the following: (i) Bands were observed in both parents but not in the F1 hybrid (BPnF1); (ii) bands occurred in either of the parents but not in the F1 hybrid (UPnF1); (iii) bands presented only in the F1 hybrid but not in either of the parents (UF1nP); and (iv) bands were detected in either of the parents and the F1 hybrid (UPF1). Overall, the major differences of gene expression occurred in the qualitative level and four related differential patterns were observed. Furthermore, the amount of differential patterns during the flowering stage was relatively higher than those of other stages. At this juncture, both the amount of hybrid-specific expression patterns at flowering stage and the silenced expression patterns at boll-forming stage in highly heterotic hybrids were found higher than those in the lower heterotic ones. It was concluded that significant differences of gene expression in leaves were present between cotton hybrid and its parents during the whole growing stages. 14Determination of interface width value in phase-field simulation of dendritic growth into undercooled melt显示文摘The influence of the interface width value on the simulation results and its dependence upon thermo-physical parameters in the phase-field simulation of dendritic growth into undercooled melt are investigated. After choosing the reasonable interface width value, the tip velocities of dendritic growth in Ni melt under different undercoolings are calculated and compared with the experimental data in order to benchmark our results. It is shown that the reasonable interface width value, which is determined by the undercooling, anisotropy, interface kinetic, and thermal diffusivity, has to be taken low enough, and the agreement of our results with experimental data verifies that the credible results can be achieved as long as the interface width value is adequately low. 16Eddy flux corrections for CO_2 exchange in broad-leaved Korean pine mixed forest of Changbai Mountains显示文摘Based on analysis of mechanisms causing energy no-closure and nocturnal low fluxes issues for CO2 exchange studies by eddy covariance method, corrections were done with the raw data sets obtained from Changbai Mountains forest flux site, to evaluate the impacts of sonic anemometer tilt, frequency response limitations and advection on estimation of CO2 exchange, respectively. The results show that the planar fit coordinate transforming method is superior to the streamline coordinate transforming method in tilt correction. The latter could cause a systematical underestimation of eddy fluxes relating with the angle of sensor and terrain tilt. The underestimation of CO2 and energy fluxes for frequency response limitations average 3.0% and 2.0% during daytime, respectively, which increase by 9.0% and 5.5% during nighttime, respectively. The corrections of frequency response limitations are closely related to atmospheric stability. The advection loss of CO2 fluxes is dominated by nocturnal vertical advection, which is at least 18% when the horizontal advection is neglected. 20The gravitational gradient tensor’s invariants and the related boundary conditions显示文摘Here we report new approaches of recovering the Earth gravitational field from GOCE (Gravity field and steady-state Ocean Circulation Explorer) gradiometric data with the help of the gradient tensor’s invariants. Our results only depend on GOCE satellite’s position and gradiometry, in other words, they are completely independent of the satellite attitude. First, starting from the invariants, linearization models are established, which can be referred as the general boundary conditions on the satellite’s orbit. Then, the spherical approximation expressions for the models are derived, and the corresponding solving methods for them are discussed. Furthermore, considering effects of J2-term, the spherical approximation models are improved so that the accuracies of the boundary conditions can be theoretically raised to O ( J 2/2 T), which is approximately equivalent to O(T2).
