| 3 | Circulating tumor cells:biology and clinical significance显示文摘Circulating tumor cells(CTCs)are tumor cells that have sloughed off the primary tumor and extravasate into and circulate in the blood.Understanding of the metastatic cascade of CTCs has tremendous potential for the identification of targets against cancer metastasis.Detecting these very rare CTCs among the massive blood cells is challenging.However,emerging technologies for CTCs detection have profoundly contributed to deepening investigation into the biology of CTCs and have facilitated their clinical application.Current technologies for the detection of CTCs are summarized herein,together with their advantages and disadvantages.The detection of CTCs is usually dependent on molecular markers,with the epithelial cell adhesion molecule being the most widely used,although molecular markers vary between different types of cancer.Properties associated with epithelial-tomesenchymal transition and stemness have been identified in CTCs,indicating their increased metastatic capacity.Only a small proportion of CTCs can survive and eventually initiate metastases,suggesting that an interaction and modulation between CTCs and the hostile blood microenvironment is essential for CTC metastasis.Single-cell sequencing of CTCs has been extensively investigated,and has enabled researchers to reveal the genome and transcriptome of CTCs.Herein,we also review the clinical applications of CTCs,especially for monitoring response to cancer treatment and in evaluating prognosis.Hence,CTCs have and will continue to contribute to providing significant insights into metastatic processes and will open new avenues for useful clinical applications. | Danfeng Lin Lesang Shen Meng Luo Kun Zhang Jinfan Li Qi Yang Fangfang Zhu Dan Zhou Shu Zheng Yiding Chen Jiaojiao Zhou | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 8 |
| 4 | 循环肿瘤细胞与子宫内膜癌临床病理特征关系分析显示文摘目的探讨循环肿瘤细胞的数目、分型与子宫内膜癌患者临床病理特征的关系。方法收集就诊的子宫内膜癌患者83例,与同期就诊的宫体良性肿瘤患者11例作为对照。采用CanPatrol^TM CTCs技术检测血液中的CTCs,并分析其数目、分型及其与临床病理特征之间的关系。结果(1)在宫体良性肿瘤患者中,仅混合型CTCs被检出(4/11);在子宫内膜癌患者中,CTCs总检出率高达95.2%(79/83),CTCs总数、上皮型CTCs、混合型CTCs、间质型CTCs相应的中位数分别为:6.00个、0.00个、3.00个、1.00个。与宫体良性肿瘤患者对比,子宫内膜癌患者CTCs总数、上皮型CTCs、混合型CTCs、间质型CTCs均较高,差异有统计学意义(P<0.05)。(2)分析发现,当CTCs总数>1、上皮型CTCs>0、混合型CTCs>1、间质型CTCs>0时,是诊断子宫内膜癌与宫体良性病变的临界值,其诊断的敏感度为97.37%,特异度为50.00%,准确度为83.30%,阳性预测值为89.16%。(3)进一步分析各分型CTC数在子宫内膜癌患者的临床病例特征中发现,在术前肿瘤标志物包括CA125水平及CA199水平、分期、分型、分级、以及是否脉管浸润、神经侵犯、深肌层、宫颈侵犯、附件转移、淋巴结转移、腹水情况,差异无明显统计学意义。结论采用CanPatrol^TM CTC技术检测子宫内膜癌患者CTCs检出率高,但其在良性患者中,仍有一定的假阳性率,通过上调CTCs阈值,可提高诊断子宫良恶性病变的敏感性,但其准确度、特异度仍较低。笔者未发现CanPatrol^TM CTCs技术检测各分型CTCs数目与子宫内膜癌临床病例特征有相关性。因此我们认为,CanPatrol TM CTCs检测技术在子宫内膜癌的应用仍有待进一步改进。 | 黄珊瑜 李小毛 叶辉霞 丁杰 方莉 | 2020 | 广东医学2020,41,3: | 7 |
| 5 | 基于上皮-间质转化标记的循环肿瘤细胞表型检测对原发性乳腺癌的预后评估意义显示文摘目的探讨基于上皮-间质转化(EMT)标记的循环肿瘤细胞(CTCs)表型检测对原发性乳腺癌的预后评估意义。方法回顾性分析16例具有化疗前后两次Canpatrol CTCs分型与计数结果的乳腺癌患者的临床病理及生存资料。结果肿瘤直径越大者(P=0.021)、淋巴结有转移者(P=0.017)、TNM分期越高者(P=0.020)的化疗前CTCs数量更多;P53阳性者(P=0.003)、表皮生长因子受体(EGFR)阳性者(P=0.027)的化疗前CTCs表型以混合型为主;复发/转移者的化疗后CTCs数量较多(P<0.001),且表型以混合型为主(P=0.041);化疗后CTCs表型以混合型为主的患者总体预后较差(DFS:P=0.021,OS:P=0.061)。化疗后CTCs总数减少者的预后较好,但差异无统计学意义(P>0.05)。结论基于EMT的CTCs表型检测对原发性乳腺癌预后具有预测价值,并能为治疗方案的选择、疗效的评估提供重要信息。 | 李军成 何升东 孙鹏 黄涛 李凌帆 薛刚 | 2021 | 成都医学院学报2021,16,2: | 6 |
| 6 | 血清CA153和CA125表达对乳腺癌患者预后的判断价值显示文摘目的探讨乳腺癌患者血清糖蛋白抗原(carbohydrate antigen,CA)153和CA125表达对预后的判断价值。方法选择乳腺癌患者78例作为研究对象,记录患者入院时的临床资料,检测血清CA153、CA125表达水平。所有患者随访6个月,调查预后死亡情况并进行相关性分析。结果在78例患者中,随访6个月,生存60例,死亡18例,死亡率为23.1%。存活组与死亡组的性别、年龄、体重指数、WBC、PLT、ALT、TBA、Cr等对比差异无统计学意义(P>0.05),两组的临床分期、组织学分化、病理类型等对比差异有统计学意义(P<0.05)。死亡组的血清CA153、CA125含量显著高于存活组(P<0.05)。直线相关分析显示:乳腺癌患者预后与CA153、CA125、临床分期、组织学分化、病理类型有显著正相关性(P<0.05);血清CA153、CA125判断乳腺癌患者预后死亡的受试者工作特征曲线(receiver operating characteristic curve,ROC曲线)下总面积为0.890、0.873(95%CI值为1.263~1.917,1.093~1.556)。结论中晚期乳腺癌患者的预后死亡率比较高,血清CA153、CA125值与患者预后显著相关,且能对患者预后进行有效判断。 | 单洪涛 杨迪 滕璐心 | 2020 | 实用癌症杂志2020,35,8: | 6 |
| 7 | S100A8 promotes epithelial-mesenchymal transition and metastasis under TGF-β/USF2 axis in colorectal cancer显示文摘Background:The transforming growth factor-β(TGF-β)pathway plays a pivotal role in inducing epithelial-mesenchymal transition(EMT),which is a key step in cancer invasion and metastasis.However,the regulatory mechanism of TGF-βin inducing EMT in colorectal cancer(CRC)has not been fully elucidated.In previous studies,it was found that S100A8 may regulate EMT.This study aimed to clarify the role of S100A8 in TGF-β-induced EMT and explore the underlying mechanism in CRC.Methods:S100A8 and upstream transcription factor 2(USF2)expression was detected by immunohistochemistry in 412 CRC tissues.Kaplan-Meier survival analysis was performed.In vitro,Western blot,and migration and invasion assays were performed to investigate the effects of S100A8 and USF2 on TGF-β-induced EMT.Mouse metastasis models were used to determine in vivo metastasis ability.Luciferase reporter and chromatin immunoprecipitation assay were used to explore the role of USF2 on S100A8 transcription.Results:During TGF-β-induced EMT in CRC cells,S100A8 and the transcription factor USF2 were upregulated.S100A8 promoted cell migration and invasion and EMT.USF2 transcriptionally regulated S100A8 expression by directly binding to its promoter region.Furthermore,TGF-βenhanced the USF2/S100A8 signaling axis of CRC cells whereas extracellular S100A8 inhibited the USF2/S100A8 axis of CRC cells.S100A8 expression in tumor cells was associated with poor overall survival in CRC.USF2 expression was positively related to S100A8 expression in tumor cells but negatively related to S100A8-positive stromal cells.Conclusions:TGF-βwas found to promote EMT and metastasis through the USF2/S100A8 axis in CRC while extracellular S100A8 suppressed the USF2/S100A8 axis.USF2 was identified as an important switch on the intracellular and extracellular S100A8 feedback loop. | Si Li Jun Zhang Senmi Qian Xuesong Wu Liang Sun Tianyi Ling Yao Jin Wenxiao Li Lichao Sun Maode Lai Fangying Xu | 2021 | Cancer Communications2021,41,2: | 5 |

| 10 | The molecular tumor burden index as a response evaluation criterion in breast cancer显示文摘Circulating tumor DNA(ctDNA)is a potential biomarker of prognosis and therapeutic response.We conducted this study to explore the role of the molecular tumor burden index(mTBI)in ctDNA as a therapeutic response and prognostic biomarker in a larger cohort prospective phase III randomized multicenter study.We collected 291 plasma samples from 125 metastatic breast cancer patients from the CAMELLIA study(NCT01917279).Target-capture deep sequencing of 1021 genes was performed to detect somatic variants in ctDNA from the plasma samples.The pretreatment mTBI value was correlated with tumor burden(P=0.025).Patients with high-level pretreatment mTBI had shorter overall survival than patients with low-level pretreatment mTBI,and the median overall survival was 40.9 months and 68.4 months,respectively(P=0.011).Patients with mTBI decrease to less than 0.02%at the first tumor evaluation had longer progression-free survival and overall survival(P<0.001 and P=0.007,respectively).The mTBI has good sensitivity to identify complete response/partial response and progressive disease based on computed tomography scans(88.5%and 87.5%,respectively).The patients classified as molecular responders had longer progression-free survival and overall survival than the nonmolecular responders in the overall cohort(P<0.001 and P=0.036,respectively),as well as in the cohort in which computed tomography scans were defined as representing stable disease(P=0.027 and P=0.015,respectively).The mTBI in ctDNA detected in liquid biopsies is a potential biomarker of therapeutic response and prognosis in patients with metastatic breast cancer. | Zongbi Yi Fei Ma Guohua Rong Binliang Liu Yanfang Guan Jin Li Xiaoying Sun Wenna Wang Xiuwen Guan Hongnan Mo Jiani Wang Haili Qian Binghe Xu | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 1 |

| 13 | Epithelial-mesenchymal transition contributes to malignant phenotypes of circulating tumor cells derived from gastric cancer显示文摘Circulating tumor cells(CTCs)are crucial to tumor metastasis,and they usually undergo epithelial-mesenchymal transition(EMT)in order to disseminate from the primary tumor.However,very little is currently known about the relationship between EMT and malignant phenotypes of CTCs in the context of gastric cancer.Therefore,this study aimed to investigate the contribution of EMT to malignant phenotypes of CTCs derived from gastric cancer cells.We xenografted MKN28 gastric cancer cells pretreated with transforming growth factor-beta 1(TGFβ-1)into nude mice by intravenous injection.Next,we isolated CTCs from the blood of nude mice by gradient centrifugation and found that CTCs derived from MKN28 cells pretreated with TGFβ-1 had a significantly increased viability and invasion ability compared to MKN28 cells without TGFβ-1 treatment.Immunocytochemical staining showed lower expression of E-cadherin and higher expression of N-cadherin,vimentin,and β-catenin in CTCs derived from MKN28 cells pretreated with TGFβ-1.Furthermore,the expression of Wnt3a,β-catenin,cyclin D1,and c-Myc was significantly higher in CTCs derived from MKN28 cells pretreated with TGFβ-1.Taken together,these findings suggest that TGFβ promotes EMT and malignant phenotypes of gastric cancer cells.Furthermore,the malignant phenotypes of gastric cancer cells induced by TGFβ are maintained in CTCs derived from these cells.Targeting EMT in CTCs is a new approach to the treatment of gastric cancer relapse and metastasis. | TIANGEN WU TIANHAO BAO DAOMING LIANG LIN WANG | 2019 | BIOCELL2019,43,4: | 0 |
