4Integration mechanisms of transgenes and population fitness of GH transgenic fish显示文摘It has been more than 20 years since the first batch of transgenic fish was produced. Five stable germ-line transmitted growth hormone (GH) transgenic fish lines have been generated. This paper reviews the mechanisms of integration and gene targeting of the transgene, as well as the viability, reproduction and transgenic approaches for the reproductive containment of GH-transgenic fish. 6中国西北内陆干旱区大叶白麻野生居群遗传多样性研究(英文)显示文摘[Objective] Study on the genetic diversity in wild populations of Poacynum hendersonii.[Method] Random amplified polymorphic DNA(RAPD)technique was employed to analyze the genetic diversity in five wild populations of P.hendersonii sampled from Xinjiang,Gansu and Qinghai provinces.[Result] Totally 165 clear and repeatable bands were generated in RAPD reaction by using 20 primers screened from 80 primers,of which 110 were polymorphic,accounting for 66.67%.At species level,Nei's gene diversity index(H),Shannon's information index(I)and genetic differentiation coefficient(Gst)were 0.220 5,0.304 7 and 0.908 2,respectively.P.hendersonii germplasm resources share a high level of genetic diversity,and genetic differentiation mainly exists among the populations.Results from genetic distances and cluster analysis showed that relationships among P.hendersonii populations were to some extent related with their geographical and climatic characters.[Conclusion] This study suggests that the conservation of P.hendersonii should focus on the protection of many populations,particularly the Qinghai population.赵金凤 张卫明 彭雪梅 唐泽紫 顾龚平 陆长梅 2008Agricultural Science & Technology2008,9,4:21
7PBPK modeling and simulation in drug research and development显示文摘Physiologically based pharmacokinetic(PBPK) modeling and simulation can be used to predict the pharmacokinetic behavior of drugs in humans using preclinical data.It can also explore the effects of various physiologic parameters such as age,ethnicity,or disease status on human pharmacokinetics,as well as guide dose and dose regiment selection and aid drug–drug interaction risk assessment.PBPK modeling has developed rapidly in the last decade within both the field of academia and the pharmaceutical industry,and has become an integral tool in drug discovery and development.In this mini-review,the concept and methodology of PBPK modeling are briefly introduced.Several case studies were discussed on how PBPK modeling and simulation can be utilized through various stages of drug discovery and development.These case studies are from our own work and the literature for better understanding of the absorption,distribution,metabolism and excretion(ADME) of a drug candidate,and the applications to increase efficiency,reduce the need for animal studies,and perhaps to replace clinical trials.The regulatory acceptance and industrial practices around PBPK modeling and simulation is also discussed.Xiaomei Zhuang Chuang Lu 2016Acta Pharmaceutica Sinica B2016,6,5:21
8QTL analysis of the rice seedling cold tolerance in a double haploid population derived from anther culture of a hybrid between indica and japonica rice显示文摘A doubled haploid population, derived from anther culture of F hybrid between a typical indica cv. and a japonica cv. has been used to investigate the seedling cold tolerance (SCT) in growth cabinet. By dynamically analyzing every day’s survival percentages of the parents and DH lines under 7-d cold plus 9-d normal temperature condition, the guantitative trait loci (QTLs) for SCT have been mapped based on a molecular linkage map constructed from this population. The results show that two parents had significant differences in SCT and the segregation of SCT in DH lines was basically a continuous distribution with most serious injury on the 6th d of the cold treatment. A total of 4 QTLs for SCT have been identified on chromosomes 1, 2, 3 and 4 respectively. The additive effects of qSCT-1, qSCT-2 and qSCT-3 have been contributed by the japonica ev JX17, but that of qSCT-4 has been contributed by the indica cv ZYQ8. 9毛果苔草种群地上生物量与株长或鞘高分形特征显示文摘By using the principles and methods of fractal geometry theory, the relationship between aboveground biomass and plant length or sheath height of Carex lasiocarpa population was studied. The results showed that there was a good static fractal relationship between them, and the resulted fractal dimension was an efficient description of the accumulation of aboveground biomass in each organ. The dynamic fractal relationship showed that during the whole growing season, the increase of aboveground biomass had a self-similarity, being a fractal growth process, and the pattern of its increase was the fractal dimension D. 10Chromosomal level assembly and population sequencing of the Chinese tree shrew genome显示文摘Chinese tree shrews (Tupaia belangeri chinensis) have become an increasingly important experimental animal in biomedical research due to their close relationship to primates. An accurately sequenced and assembled genome is essential for understanding the genetic features and biology of this animal. In this study, we used long-read single-molecule sequencing and high-throughput chromosome conformation capture (Hi-C) technology to obtain a high-qualitychromosome-scale scaffolding of the Chinese tree shrew genome. The new reference genome (KIZ version 2: TS_2.0) resolved problems in presently available tree shrew genomes and enabled accurate identification of large and complex repeat regions, gene structures, and species-specific genomic structural variants. In addition, by sequencing the genomes of six Chinese tree shrew individuals, we produced a comprehensive map of 12.8 M single nucleotide polymorphisms and confirmed that the major histocompatibility complex (MHC) loci and immunoglobulin gene family exhibited high nucleotide diversity in the tree shrew genome. We updated the tree shrew genome database (TreeshrewDB v2.0: http://gffzzb70c77447da74c53h5vnwpb0nbk966xq6.ffgz.tsg.suse.edu.cn) to include the genome annotation information and genetic variations. 11QTL analysis of rice low temperature germinability显示文摘A double haploid population, derived from anther culture of F, hybrid between a typical indica and a japonica (ZYQ8/JX17), has been used to investigate the low temperature germinability (LTG) at 15℃. The low temperature germinability of two parents was significantly different. In 6-11 d, the germination percentage of ZYQ8 was higher than that of JX17. In 12-16 d, the germination percentage of JX17 was higher than that of ZYQ8. The quantitative trait loci (QTLs) of every day for low temperature germinabilityhave been mapped based on a molecular linkage map constructed from this population. In 8-11 d, qLTG-9 was identified in C397B-RZ617B on chromosome 9, the additive effect was positive, showing that, the allele from JX17 could increase low temperature germinability. In 12-16 d, qLTG-4 was mapped between RG908 and CT563 on chromosome 4, the additive effect was negative, showing that the allele from ZYQ8 could increase low temperature germinability. 12Changes in China’s lakes: climate and human impacts显示文摘Lakes have played a critical role in providing water and ecosystem services for people and other organisms in China for millennia.However,accelerating climate change and economic boom have resulted in unprecedented changes in these valuable lakes.Using Landsat images covering the entity of the country,we explored the changes in China’s lakes and the associated driving forces over the last 30 years(i.e.mid-1980 s to 2015).We discovered that China’s lakes have changed with divergent regional trends:in the sparsely populated Tibetan Plateau,lakes are abundant and the lake area has increased dramatically from 38596 to46831 km^2(i.e.increased by 8235 km^2,or 21.3%),whereas,in the densely populated northern and eastern regions,lakes are relatively scarce and the lake area has decreased from 36659 to 33657 km^2(i.e.decreased by 3002 km^2,or 8.2%).In particular,severe lake decreases occurred in the Mongolia-Xinjiang Plateau and the Eastern Plain(-2151 km^2).Statistical analyses indicated that climate was the most important factor controlling lake changes in the Tibetan Plateau,the Yun-Gui Plateau and the Northeast Plain.However,the strength of climatic control on lake changes was low in the Eastern Plain and the Mongolia-Xinjiang Plateau,where human activities,e.g.impoldering,irrigation and mining,have caused serious impacts on lakes.Further lake changes will exacerbate regional imbalances between lake resources and population distribution,and thus may increase the risk of water-resource crises in China.Shengli Tao Jingyun Fang Suhui Ma Qiong Cai Xinyu Xiong Di Tian Xia Zhao Leqi Fang Heng Zhang Jiangling Zhu Shuqing Zhao 2020National Science Review2020,7,1:14
13Lower clearance of sodium tanshinone IIA sulfonate in coronary heart disease patients and the effect of total bilirubin: a population pharmacokinetics analysis显示文摘This study developed a population pharmacokinetic model for sodium tanshinone IIA sulfonate(STS) in healthy volunteers and coronary heart disease(CHD) patients in order to identify significant covariates for the pharmacokinetics of STS. Blood samples were obtained by intense sampling approach from 10 healthy volunteers and sparse sampling from 25 CHD patients, and a population pharmacokinetic analysis was performed by nonlinear mixed-effect modeling. The final model was evaluated by bootstrap and visual predictive check. A total of 230 plasma concentrations were included, 137 from healthy volunteers and 93 from CHD patients. It was a two-compartment model with first-order elimination. The typical value of the apparent clearance(CL) of STS in CHD patients with total bilirubin(TBIL) level of 10 μmol×L^(–1) was 48.7 L×h^(–1) with inter individual variability of 27.4%, whereas that in healthy volunteers with the same TBIL level was 63.1 L×h^(–1). Residual variability was described by a proportional error model and estimated at 5.2%. The CL of STS in CHD patients was lower than that in healthy volunteers and decreased when TBIL levels increased. The bootstrap and visual predictive check confirmed the stability and validity of the final model. 16Spatial heterogeneity of soil chemical properties between Haloxylon persicum and Haloxylon ammodendron populations显示文摘Spatial heterogeneity is a ubiquitous feature in natural ecosystems, especially in arid regions. Different species and their discontinuous distribution, accompanied by varied topographic characteristics, result in soil resources distributed differently in different locations, and present significant spatial heterogeneity in desert ecosystems. In this study, conventional and geostatistical methods were used to identify the heterogeneity of soil chemical properties in two desert populations, Haloxylon persicum Bunge ex Boss., which dominates on the slopes and tops of sand dunes and Haloxylon ammodendron (C. A. Mey.) Bunge, which inhabits interdunes in the Gurbantunggut Desert of Xinjiang, China. The results showed that soil pH, electrical conductivity (EC), soil organic carbon (SOC), available nitrogen (AN) and available phosphorus (AP) were significantly higher in H. ammodendron populations than that in H. persicum. The coefficient of variation (CV) indicated that (1) most parameters presented a moderate degree of variability (10% < CV < 100%) except pH in both plots, (2) the variability of soil pH, EC and AP in H. ammodendron populations was higher than that in H. persicum populations, and (3) SOC and AN in H. ammodendron populations were lower than that in H. persicum populations. Geostatistical analysis revealed a strong spatial dependence (C0/(C0+C) < 25%) within the distance of ranges for all tested parameters in both plots. The Kriging-interpolated figures showed that the soil spatial distribution was correlated with the vegetation distribution, individual size of plants, and the topographic features, especially with the plants nearest to sampling points and the topographic features. In each plot, soil EC, SOC, AN and AP presented similar distributions, and fertile islands and salt islands occurred in both plots but did not affect every individual plant, since the sampling distance was larger than the size of such fertile islands. The results of topographic effects on soil heterogeneity suggested significant differences between the interdunes and dune-tops. Different topographic characteristics (physical factors) between plots result in the differences in SOC, AN and AP, while the heterogeneity of soil pH and EC arise from plant species and their distribution (biotic factor). Such biotic and physical factors did not occur in isolation, but worked together on soil heterogeneity, and played important parts in improving the soil properties. 17Helicobacter pylori infection and inflammatory bowel disease in Asians:A meta-analysis显示文摘AIM: To investigate the relationship between Helicobacter pylori infection and inflammatory bowel disease(IBD) in an Asian population.METHODS: The Pub Med, EMBASE, and CochraneLibrary databases were searched for observational studies published up until June 2014, without language restrictions. Additional references were obtained from reviewed articles. RESULTS: Ten studies involving 1299 IBD patients and 1817 controls were included in the meta-analysis(24.9% of IBD patients had H. pylori infection vs 48.3% of the controls). The pooled risk ratio for H. pylori infection in IBD patients compared with controls was 0.48(95%CI: 0.43-0.54; P < 0.001). There was no significant heterogeneity in the included studies(I2= 21%). 19Association between PPARG genetic polymorphisms and ischemic stroke risk in a northern Chinese Han population: a case-control study显示文摘Two common polymorphisms of the peroxisome proliferator-activated receptor gamma(PPARG) gene, rs1801282 and rs3856806, may be important candidate gene loci affecting the susceptibility to ischemic stroke. This case-control study sought to identify the relationship between these two single-nucleotide polymorphisms and ischemic stroke risk in a northern Chinese Han population. A total of 910 ischemic stroke participants were recruited from the First Hospital of China Medical University, Shenyang, China as a case group, of whom 895 completed the study. The 883 healthy controls were recruited from the Health Check Center of the First Hospital of China Medical University, Shenyang, China. All participants or family members provided informed consent. The study protocol was approved by the Ethics Committee of the First Hospital of China Medical University, China on February 20, 2012(approval No. 2012-38-1). The protocol was registered with the Chinese Clinical Trial Registry(registration number: ChiCTR-COC-17013559). Plasma genomic DNA was extracted from all participants and analyzed for rs1801282 and rs3856806 single nucleotide polymorphisms using a SNaPshot Multiplex sequencing assay. Odds ratios(ORs) and 95% confidence intervals(CIs) were calculated using unconditional logistic regression to estimate the association between ischemic stroke and a particular genotype. Results demonstrated that the G allele frequency of the PPARG gene rs1801282 locus was significantly higher in the case group than in the control group(P < 0.001). Individuals carrying the G allele had a 1.844 fold increased risk of ischemic stroke(OR = 1.844, 95% CI: 1.286–2.645, P < 0.001). Individuals carrying the rs3856806 T allele had a 1.366 fold increased risk of ischemic stroke(OR = 1.366, 95% CI: 1.077–1.733, P = 0.010). The distribution frequencies of the PPARG gene haplotypes rs1801282-rs3856806 in the control and case groups were determined. The frequency of distribution in the G-T haplotype case group was significantly higher than that in the control group. The risk of ischemic stroke increased to 2.953 times in individuals carrying the G-T haplotype(OR = 2.953, 95% CI: 2.082–4.190, P < 0.001). The rs1801282 G allele and rs3856806 T allele had a multiplicative interaction(OR = 3.404, 95% CI: 1.631–7.102, P < 0.001) and additive interaction(RERI = 41.705, 95% CI: 14.586–68.824, AP = 0.860;95% CI: 0.779–0.940;S = 8.170, 95% CI: 3.772–17.697) on ischemic stroke risk, showing a synergistic effect. Of all ischemic stroke cases, 86% were attributed to the interaction of the G allele of rs1801282 and the T allele of rs3856806. The effect of the PPARG rs1801282 G allele on ischemic stroke risk was enhanced in the presence of the rs3856806 T allele(OR = 8.001 vs. 1.844). The effect of the rs3856806 T allele on ischemic stroke risk was also enhanced in the presence of the rs1801282 G allele(OR = 2.546 vs. 1.366). 20Genome-wide association analysis identified molecular markers associated with important tea flavor-related metabolites显示文摘The characteristic secondary metabolites in tea(theanine,caffeine,and catechins)are important factors contributing to unique tea flavors.However,there has been relatively little research on molecular markers related to these metabolites.Thus,we conducted a genome-wide association analysis of the levels of these tea flavor-related metabolites in three seasons.The theanine,caffeine,and catechin levels in Population 1 comprising 191 tea plant germplasms were examined,which revealed that their heritability exceeded 0.5 in the analyzed seasons,with the following rank order(highest to lowest heritabilities):(+)-catechin>(−)-gallocatechin gallate>caffeine=(−)-epicatechin>(−)-epigallocatechin-3-gallate>theanine>(−)-epigallocatechin>(−)-epicatechin-3-gallate>catechin gallate>(+)-gallocatechin.The SNPs detected by amplified-fragment SNP and methylation sequencing divided Population 1 into three groups and seven subgroups.An association analysis yielded 307 SNP markers related to theanine,caffeine,and catechins that were common to all three seasons.Some of the markers were pleiotropic.The functional annotation of 180 key genes at the SNP loci revealed that FLS,UGT,MYB,and WD40 domain-containing proteins,as well as ATP-binding cassette transporters,may be important for catechin synthesis.KEGG and GO analyses indicated that these genes are associated with metabolic pathways and secondary metabolite biosynthesis.Moreover,in Population 2(98 tea plant germplasm resources),30 candidate SNPs were verified,including 17 SNPs that were significantly or extremely significantly associated with specific metabolite levels.These results will provide a foundation for future research on important flavor-related metabolites and may help accelerate the breeding of new tea varieties.Kaixing Fang Zhiqiang Xia Hongjian Li Xiaohui Jiang Dandan Qin Qiushuang Wang Qing Wang Chendong Pan Bo Li Hualing Wu 2021Horticulture Research2021,8,1:10
